Anticancer and Antimicrobial Activities of 13(E)-labd-13-ene-8α,15-diol from Brachyglottis monroi

  • In a previous study, we reported that 13(E)-labd-13-ene-$8{\alpha}$,15-diol (13E) possesses antiviral and anticancer activities. In this study, the anticancer and antimicrobial activities of 13(E) were evaluated against 4 cancer cell lines and 6 bacteria. 13(E) showed inhibitory effect on a variety of cancer cell lines. The $IC_{50}$ values was 8.3-21.3 ${\mu}g/mL$. 13(E) was the most effective growth inhibitor of murine leukaemia cell lines P388, producing approximately 8.3 ${\mu}g/mL$ of $IC_{50}$ in the cytopathic effect (CPE) method. 13(E) also inhibited the growth of the gram-positive bacteria (Staphylococcus aureus, Bacillus cereus and Listeria monocytogenes) and gram-negative bacteria (Vibrio parahaemolyticus, Escherichia coli and Salmonella enteritidis) with a range of minimum inhibitory concentration (MIC) values from 0.092 to 0.598 mg/mL and gram-negative bacteria were more sensitive to the compound (MIC, 0.092 mg/mL).

Anticancer Activity of Monoterpenes and the Changes of Enzymes Activities Responsible for the Conversion of Reactive Oxygen Species (Monoterpenes의 항암작용과 활성산소 전환 효소의 활성 변화)

  • The present study was undertaken to investigate the anticancer activity of monoterepenes in the animal and the cancer cell line tests. Both of the noncyclic and cyclic monoterpenes showed significant life prolonging effects on ICR mouse with abdominal cancer induced by Sarcoma 180 cells up to 67.4% and 63.5% in case of linalool and geraniol, respectively. Linalool and geraniol also exhibited very excellent cytotoxicity against L1210 leukemic cells with $IC_{50}$/ value of 0.32 $\mu\textrm{g}$/mι in 5 days culture condition. In the presence of linalool and geraniol, the generation of $O_2$$^{[-10]}$ ion were found to be increased proportionally to the cytotoxicity arisen from these monoterpenes. Furthermore, the antioxidant enzymes activities such as superoxide dismutase (SOD) and glutathione peroxidase (GPx) responsible for the conversion of $O_2$$^{[-10]}$ ion to $H_2O$$_2$ and then to $H_2O$ augmented remarkably by linalool and geraniol. All data put together it can be postulated that monoterpenes may kill abdominal cancer cells of ICR mouse probably by activating anticancer system of the body, whereas the death of L1210 cells may be due to the detrimental attacks of reactive oxygen species (ROS) including $O_2$$^{[-10]}$ in spite of antioxidant enzymes activities to overcome the ROS attacks.

Changes in Biological Activities of Extracts from Herbal Chokong Pills by Manufacturing Stages (한방초콩환의 제조단계에 따른 추출물의 생리활성 변화)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from pickled soybean products by manufacturing stage. Methods : Yakkong (YK) was pickled for 15 days to prepare Chokong (CK) and Chokong pills (CKP) were mixed Chokong (CK) powder with vinegar. Herbal Chokong pills (HCKP) were made by addition five kinds of medicinal herbs to Chokong. Pickled soybean products by each manufacturing process were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest electron donating abilities (EDA) of the water and ethanol extracts were from HCKP among pickled soybean products and those were 81.2% and 91.5% at 1,000 $\mu$g/mL, respectively. The ethanol extracts of pickled soybean products had higher activities of EDA and superoxide dismutase (SOD) than water extracts. The highest anticancer activity was the water extracts of HCKP against both of MDA and A549 cells. Conclusions : These results suggest that herbal Chokong pills (HCKP) can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidative characteristics.

Progress on Understanding the Anticancer Mechanisms of Medicinal Mushroom: Inonotus Obliquus

  • Cancer is a leading cause of death worldwide. Recently, the demand for more effective and safer therapeutic agents for the chemoprevention of human cancer has increased. As a white rot fungus, Inonotus obliquus is valued as an edible and medicinal resource. Chemical investigations have shown that I. obliquus produces a diverse range of secondary metabolites, including phenolic compounds, melanins, and lanostane-type triterpenoids. Among these are active components for antioxidant, antitumoral, and antiviral activities and for improving human immunity against infection of pathogenic microbes. Importantly, their anticancer activities have become a hot recently, but with relatively little knowledge of their modes of action. Some compounds extracted from I. obliquus arrest cancer cells in the G0/G1 phase and then induce cell apoptosis or differentiation, whereas some examples directly participate in the cell apoptosis pathway. In other cases, polysaccharides from I. obliquus can indirectly be involved in anticancer processes mainly via stimulating the immune system. Furthermore, the antioxidative ability of I. obliquus extracts can prevent generation of cancer cells. In this review, we highlight recent findings regarding mechanisms underlying the anticancer influence of I. obliquus, to provide a comprehensive landscape view of the actions of this mushroom in preventing cancer.

Antioxidant, Anticancer and Immune Activation of Anthocyanin Fraction from Rubus coreanus Miquel fruits (Bokbunja) (복분자 안토시아닌 분획의 항산화, 항암 및 면역증진 효과)

  • This study was performed to determined the antioxidant activities, anticancer and immuno-activities of anthocyanin fraction from Rubus coreanus Miquel fruits (Bokbunja). Anthocyanin fraction extracted from Bokbunja revealed the presence of three anthocyanin components, which were tentatively identified as cyanidin 3-O-sambubioside, cyanidin 3-O-xylosylrutinoside and cyanidin 3-O-rutinoside using RP-HPLC/DAD/MS. The anthocyanin fraction from Bokbunja always showed reducing power and high scavenging activities against DPPH, hydroxy radical (OH) and superoxide anion radical ($O_2{^-}$) similar to general synthetic antioxidant and polyphenol compounds from plant origin. Anthocyanin fraction from Bokbunja showed high inhibition on proliferation of LNCaP and A549 cells and did not inhibit the proliferation of other cancer cells. Immuno-activities of Anthocyanin fraction from Bokbunja were investigated, it showed high promotion of human B and T cells growth about 50% and secretion of IL-6 and TNF-${\alpha}$ by treatment after 6 days. Over all, the result of the study suggest that anthocyanin fraction from Bokbunja displays antioxidant activity comparable to that general synthetic antioxidant, also, anthocyanin fraction from Bokbunja are expected to be good candidate for development into source of anticaner and immuno-activator agent in food industry.

Biological Activities of Water and Ethanolic Extracts from Allium victorialis L. Mature Leaves

  • Allium victorialis L. (A. victorialis) is a very popular vegetable in Korea. The most commonly used parts of this vegetable are the bulbs and young leaves. To determine if the mature leaves have any beneficial properties, we investigated antioxidant, anti-${\alpha}$-glucosidase, anti-inflammatory, and anticancer activities of water and ethanol extracts from A. victorialis. Antioxidant activity was evaluated by measuring total phenolic content, DPPH and superoxide radicals scavenging activities. The water extract from A. victorialis ($W{\cdot}A$. victorialis) exhibited higher antioxidant ability than the ethanol extract ($E{\cdot}A$. victorialis). Moreover, the water extract showed strong inhibitory effect on ${\alpha}$-glucosidase. On the other hand, the ethanol extract had greater anti-inflammatory activity on murine macrophage cells (RAW 264.7) and greater anticancer activities against human colon cancer cells (HT-29). These results suggest that mature leaves from E·A. victorialis may have health-enhancing effects.

Orthosiphon pallidus, a Potential Treatment for Patients with Breast Cancer

  • Objective: Orthosiphon pallidus (O. pallidus), which belongs to the Lamiaceae family, is a popular garden plant that is widely used for the treatment of various diseases, such as urinary lithiasis, fever, hepatitis, cancer and jaundice. The objective of the present work was to investigate the antioxidant free-radical scavenging and the anticancer activities of O. pallidus against human breast-cancer cell lines. Methods: The antioxidant activity of Orthosiphon pallidus aqueous extract (OPAE) was investigated using different models, such as the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the 2, 2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) models, as were the $Fe^+$ chelation, the hydroxyl radical and superoxide radical scavenging, and total reducing power activities. The anticancer activities of the extract were determined by using the 3-(4, 5- dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) and the sulforhodamine (SRB) assays on the MCF-7 and the MDA-MB-231 cancer cell lines. Results: The aqueous Orthosiphon pallidus extract showed potent activity in in-vitro models. It significantly inhibited the scavenging of hydroxyl and superoxide radicals, but induced a remarkable $Fe^+$ chelation activity. For both cell lines, the percent cytotoxicity was found to increase steadily with increasing OPAE concentration up to $240{\mu}g/mL$. Conclusion: These results suggest that Orthosiphon pallidus has excellent antioxidant, antimicrobial, and anticancer activities against human breast-cancer cell lines.

Thc Euect of Ginseng Extracts on the Enzyme Activity in the Liver of Swiss Mice (인삼성분이 생쥐 간의 수종 효소활성에 미치는 영향)

  • This study was prepared to observe some enzyme activities in the liver of mice treated with extracts of Ginseng anticancer compound, separated from the petroleum ether extracts by silicic acid chromatography, has the cytotoxic activity against cancer cells. Swiss mice, 72 heads were used (or this experiment and they were divdied into control, test group I and test group If, that test group I was injected crude extract and test group II was injected anticancer compound, while the control group was injected 0.9% NaCl solution. The injections were carried out 1,2,4 and 8 times once a day for 1-8 day, respectively. The liver was removed carefully from the mice at 24 hours after drugs injected, and homogenized at 4$^{\circ}C$ for enzyme study. The activities of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase(GPT) were determined by Reitmen and Frankel method and lactic acid dehydrogenase activity was determined by Wroblewski methods in vitro. The results obtained are summarized as follows; 1. The GOT activity was increased 26%, crude extract and 16%, anticancer compound than those of control at 1st injected groups and decreased gradually according to increase of injection time, at 8th injected groups, the GOT activity was decresed by 16%, crude extract and 12%, anticancer compound. 2. The GPT activity was not changed significantly at 1st and 2nd injected groups, but, at 4th injected groups, the GPT activity was decreased 20%, crude extract and 14%, anticancer compound. While the GPT activity was recovered to normal value at 8th injected groups. 3. At 1st injected groups, the LDH activity was increased 17%, anticancer compound, while those of crude extract was shown normal value. At 2nd injected groups, the LDH activity increased 35yo:, crude extract while those of anticancer compound was showed normal value. And the LOH activity was recovered gradually at 4th and 8th injected groups.

수종(數種) 한약재(韓藥材)의 항암활성(抗癌活性) 연구(硏究)

  • An extensive anticancer drug screening from natural resources has been carried out primarily using murine tumor model for past fourty years. Recently a new screening program from NCI, so called disease-oriented screening system. has been estabished to detect anticancer drugs that show selective growth inhibition toward variety of tissue specific human solid tumors originated from leukemia, lung, colon, CNS, melanoma, ovarian, renal, prostate amd breast. To develope the anticancer drugs from oriental medicinal herbs, we investigated the cytotoxic effects against human tumor cell panels with 23 kinds of MeOH extract of medicinal herbs. Evodiae Fructus, Meliae Toosendan Fructus, Saussureae Radix and Pharbitidis Semen showed strong activities against several tumor cell lines.

Physiological Activities of Opuntia humifusa Petal (천년초 꽃잎의 생리활성)

  • This study was conducted to investigate antimicrobial, antioxidant and anticancer activities of Opuntia humifusa (OH) petal extracts. The methanol and hexane extracts of OH petals showed their highest antimicrobial activity against Clostridium perfringens. The OH petal butanol fraction had the best antioxidative peroxynitrite scavenging activity among OH petal extracts. The DPPH scavenging activity of OH petals was lower than the peroxynitrite scavenging effect. The hexane and methanol fractions at a concentration of 200 ${\mu}g$/mL inhibited proliferation >80% in four kinds of human cervical cancer cells(B16F10, HepG2, HT-29 and MCF-7). In particular, the anticancer effect against B16F10 human skin cancer cells at the same concentration was higher than that in the other cancer cells.
