한국의 서울에 사는 길 고양이와 집 고양이에서 고양이 범백혈구감소증 바이러스의 유병률 (Prevalence of Feline Panleukopenia Virus in Stray and Household Cats in Seoul, Korea)

  • 우리는 한국의 서울에 있는 다른 지역에 사는 길 고양이와 집 고양이의 범백혈구감소증 바이러스의 유병률을 조사하였다. 혈액 샘플은 200마리 고양이 (길 고양이 100마리와 집 고양이)에서 수집하였으며 모든 샘플은 중합효소 반응검사를 실시하였다. 전체적인 고양이 범백혈구감소증 바이러스의 유병률은 2%이다. 검사를 통해 양성 고양이 중 3% (100 분의 3)은 길 고양이이며 1% (100분의 1)은 집 고양이였다. 고양이 백혈구감소증 바이러스의 발생은 성묘 (1살이상, 0.5%) 보다 자묘 (1살이내, 1.5%)에서 더 높았다. 건강한 고양이의 양성률은 1.8% (3/166)이며 아픈 고양이의 양성률은 2.2% (1/44)이다. 예방 접종한 고양이의 양성률은 1.3% (1/77)이었고 예방 접종하지 않은 고양이의 양성률은 2.3% (3/133)이다. 항체검사와 비교시 고양이 범백혈구 바이러스 항원의 검출은 길 고양이와 집 고양이의 현재 질병 감염 상태를 암시할 수 있다. 따라서 항원검사는 질병의 진단과 치료에 도움을 줄 수 있다. 결론적으로 우리의 연구는 현재까지 서울에서 고양이 범백혈구감소증 바이러스의 유병률을 평가한 적이 없었기 때문에 비교적 중요하다. 고양이 범백혈구감소증 바이러스의 유병률은 길 고양이와 어린 고양이에서 높았다. 따라서 고양이 범백혈구감소증 바이러스 발생을 예방하기 위해서는 적절한 예방접종과 감시가 중요하다.

진단용 one-step RT-PCR을 통한 돼지 인플루엔자 바이러스의 아형 및 pandemic 유형에 대 한 신속한 결정 (Rapid Determining for Subtypes and Pandemic Type of Swine Influenza Virus by Diagnostic One-step RT-PCR)

  • Swine influenza virus (SIV)는 돼지 개체군에서 가장 흔한 질병을 일으키는 바이러스 중 하나이며 그 subtype은 hemagglutinin (HA)와 neuraminidase (NA)에 의해 결정됩니다. 최근 SIV subtype 진단 방법이 개발되고 있으나 SIV의 리보뉴클레오타이드 서열의 많은 변이로 인해 PCR 보다는 항원-항체 반응을 이용하는 방법이 주로 사용되고 있다. 본 연구에서는 SIV 하위 유형의 신속한 결정을 위하여 2008년 이후 국내에서 발생한 SIV의 다중염기서열 정렬을 통하여 10개의 subtype 진단 프라이머 세트를 개발하고 이를 이용한 one-step RT-PCR 반응을 최적화하였다. 또한 감염력이 높고 독성이 있는 인플루엔자 H1N1 (pH1N1)의 아형에서 확인된 독특한 M 유전자서열을 검출함으로써 pandemic SIV를 조기에 결정하도록 특이적 프라이머를 설계하였다. 2008년부터 2014년까지 한국에서 발생한 9종의 SIV RNA를 활용하여 SIV의 아형 및 pandemic 가능성을 결정하기 위해 시험 분석한 결과 모든 진단 프라이머 세트는 SIV 아형을 정확하게 결정하였으며 pandemic SIV를 검출할 수 있는 것으로 확인되었다. 결과적으로 이들 프라이머 세트를 이용한 최적화된 one-step RT-PCR 분석이 SIV 아형의 신속한 진단에 유용하다는 것이 확인하였다. 이러한 결과는 SIV 하위 유형 및 pandemic SIV가 확산되기 전에 조기 발견을 위한 키트로 개발될 수 있음을 시사한다.

면역크로마토그래피를 이용한 E. coli O157: H7 신속검출 키트의 유효성 평가 (Evaluation of the Efficiency of E. coli O157: H7 Rapid Detection Kit using Immunochromatography)

  • 식품으로부터 다양한 병원 미생물을 신속 검출하기 위하여 다양한 검출 원리를 이요한 키트들이 개발 시판되고 있다. 검사키트는 신속, 정확하고 간단하게 사용할 수 있으므로 검사기관이나 실험실 뿐 아니라 식품회사에서 QC 또는 QA를 수행하기 위하여 사용이 증가되고 있는 추세이다. 이에 본 연구에서는 E. coli 0157:H7의 단클론항체를 이용하여 면역크로마토글래피법에 의해 개발된 E. coli 0157:H7 검출 키트(Donga Co, Korea, D-kit)에 대한 검출감도 및 특이성을 확인하고 식품 시료에 적용 가능성을 평가하였다. 면역크로마토그래피법에 의하여 개발 시판되고 있는 Reveal E. coli 0157:H7 kit (Neogen Co., USA. R-kit)와 VIP EHEC kit(Biocontro Inc., USA. V-kit)를 비교 키트로 사용하였다. E. coli 0157:H7 표준균주를 사용하여 실시한 검출감도 확인시험 결과 R-kit 및 D-kit는 104/m/의 농도에서 양성으로 확인되었고 $10^3$/ml에서도 약한 양성 반응을 보였으나, V-lit는 $10^5$/ml농도로 검출감도가 낮았다. 또한, 배양액을 가열하여 kit에 적용하는 것이 가열하지 않은 경우보다 검출감도를 높일 수 있었다. E. coli 0157:H7 분리 22주, verotoxin 생성 E. coli 7주 E. coli 분리주 40주 중 3주를 제외한 모든 균에서 음성의 결과를 보여 특이성을 확인하였다. 세 키트에 위양성 반응을 보인 것은 E. coli 0157:H19, E. coli 0148:H18 및 Salmonella gallinarium으로 이들 혈청형과 0157:H7 사이에는 유사한 혈청학적 특성이 존재하는 것으로 추정되었다. 이상의 실험결과로 D-kit는 E. coli 0157:H7을 검출하는데 감도 및 특이성 면에서 기존 키트인 R-kit 및 V-kit와 같이 이용한 것으로 확인되었다.

Reversibility and safety of KISS1 metastasis suppressor gene vaccine in immunocastration of ram lambs

  • Objective: The aim of this study was to investigate the reversibility and safety of KISS1 metastasis suppressor (KISS1) gene vaccine in immunocastration. Methods: Six eight-week old ram lambs were randomly divided into vaccinated and control groups. The vaccine (1 mg/ram lamb) was injected at weeks 0, 3, and 6 of the study. Blood samples were collected from the jugular vein before primary immunization and at weeks 2, 4, 6, 10, 14, 22, and 30 after primary immunization. All ram lambs were slaughtered at 38 weeks of age, and samples were collected. Results: The specific anti-KISS1 antibody titers in vaccinated animals were significantly higher and the serum testosterone level was significantly lower than those in the control groups from week 4 to 14 after primary immunization (p<0.05). No significant difference was observed at weeks 22 and 30 after the primary immunization. Similar results were also found for scrotal circumference, testicular weight, length, breadth, and spermatogenesis in seminiferous tubules in week 30 after primary immunization. KS (KISS1-hepatitis B surface antigen S) fusion fragment of KISS1 gene vaccine was not detected in host cell genomic DNA of 9 tissues of the vaccinated ram lambs by polymerase chain reaction. Conclusion: The effects of KISS1 gene vaccine in immunocastration were reversible and no integration events were recorded.

C형 간염의 진단을 위한 역전사중합효소연쇄반응과 효소면역측정법의 타당성 평가 (Assessment of Validity of RT-PCR and EIA for The Detection of Hepatitis C Virus Infection)

  • This study was conducted to estimate the validity of reverse transcriptase-polymerase chain reaction(RT-PCR) compared to enzyme immunoassay(EIA) for the detection of hepatitis C virus (HCV) infection. EIA for antibody to HCV(anti-HCV) and RT-PCR for HCV was executed on the subjects from Pusan and Kyungnam area with questionnaire survey to collect some relating factors of HCV infection. As the result from 617 cases, the prevalence of HCV infection was 1.5% by EIA and 3.7% by RT-PCR(p<0.05), and the age standardized rate was 1.7% and 3.4% by EIA and RT-PCR, respectively. The prevalence of hepatitis B surface antigen(HBsAg) was 6.8% by enzyme linked immunosorbent assay(ELISA) and the age standardized rate was 7.7%. It was the higher in male group comparing to female group(p<0.01). Both of the prevalence of HCV and HBsAg were higher in elevated asparate aminotransferase(AST) and alanine aminotransferase (ALT) group than in normal AST and ALT group(p<0.01). There was no specific risk factor of HCV infection. Though the degree of agreement of EIA and RT-PCR by gamma statistics was 97.2%, it showed a significant difference between the two methods(p<0.01). For the detection of HCV infection, positive predictive value of EIA was 66.7% and negative predictive value of EIA was 97.2%. This study suggests that negative result to anti-HCV by EIA didn't mean the free state of HCV infection, therefore it would be helpful that further monitoring for HCV infection by RT-PCR in the case of elevated AST and ALT and/or clinically suspected.

Immune Response and Plasma Alpha Tocopherol and Selenium Status of Male Buffalo (Bubalus bubalis) Calves Supplemented with Vitamin E and Selenium

  • An experiment was conducted using 20 male buffalo calves to study the effect of vitamin E and selenium supplementation on their immune response and plasma ${\alpha}$-tocopherol and selenium status. These buffalo calves (10-12 months old, average body weight $75.30{\pm}2.20 $ kg) were randomly allotted to four treatments on the basis of their body weights and were fed on wheat straw and concentrate mixture to meet their nutrient requirements of 500 g/d body weight gain. The buffalo calves were fed either a control diet (neither supplemented with Se nor VE) or diets supplemented with Se at 0.3 ppm (+Se), DL-alpha tocopheryl acetate at 300 IU (+VE), and both DL-alpha tocopheryl acetate at 300 IU and Se at 0.3 ppm (+Se+VE). These experimental diets were fed for 180 days. Blood samples were collected at day 0 and subsequently at 45 day intervals up to 180 days of experimental feeding to monitor plasma ${\alpha}$-tocopherol and Se concentrations. To assess humoral immune response, all calves were sensitized with formalin inactivated Pasteurella multocida antigen at 135 days of experimental feeding and blood was collected on 0, 7, 14, 21 and 28 days post vaccination (DPV) to measure antibody production using indirect ELISA. Cell mediated immune response of calves was assessed after 180 days of experimental feeding by in vivo delayed type hypersensitivity (DTH) reaction using phytohaemaglutinin-P (PHA-P) as a mitogen. Results revealed that feeding of VE and Se improved the plasma levels of these nutrients. Plasma levels of Se were affected by supplementation of both VE (p<0.001) and Se (p<0.001); however, no interaction ($Se{\times}VE$) was observed. Supplementation of Se improved the humoral immune response (p<0.008), whereas, VE showed a tendency towards improvement in cell mediated immune response (p<0.064). It was concluded that vitamin E and Se supplementation improved the status of these micronutrients and humoral immune response in buffalo calves.

SARS-CoV-2의 진단기술 (Diagnostic Techniques for SARS-CoV-2 Detection)

  • 코로나바이러스감염증-19(COVID-19)는 SARS-CoV-2에 의해 발병된다. 지금까지 인간에게 감염되는 7 가지 종류의 코로나 바이러스가 보고되었다. 그 중, HCoV-229E, HCoV-OC43, HCoV-NL63, 그리고 HCoV-HKU1 등 4종류의 코로나바이러스는 감기와 같은 단순 호흡기 질환을 유발한다고 보고되었다. 반면, SARS-CoV는 2002년에, MERS-CoV는 2012년에 각각 대유행을 일으킨 바 있다. 가장 최근에는 2019년 12월 중국 우한에서 처음 보고된 SARS-CoV-2가 전세계적인 대유행의 원인이 되고 있다. 이러한 SARS-CoV-2를 진단하고, 치료하고, 예방하기 위해서는 신속 정확한 진단키트, 치료제, 그리고 안전한 백신의 개발의 필수적으로 요구된다. 이러한 강력한 도구들을 개발하기 위해서는 SARS-CoV-2의 표현형, 유전자형, 그리고 생활주기 등의 연구가 선행되어야 한다. SARS-CoV-2의 진단기술은 현재 크게 두가지의 큰 분야인 분자진단과 면역혈청학적 진단으로 구분할 수 있다. 분자진단의 경우 SARS-CoV-2의 유전체를 대상으로 하며, 면역혈청학적 진단은 SARS-CoV-2의 항원 단백질 혹은 SARS-CoV-2에 대한 항체를 대상으로 한다. 본 총설에서는 SARS-CoV-2의 표현형, 유전체 구조, 그리고 유전자 발현에 대해서 정리하고, SARS-CoV-2에 대한 다양한 진단 기술 등에 대한 기초지식을 제공하고자 한다.

국내에서 유행한 Respiratory Syncytial 바이러스의 염기서열 및 계통분석 (Sequence and Phylogenetic Analysis of Respiratory Syncytial Virus Isolated from Korea)

  • Respiratory Syncytial virus (RSV) is an important cause of acute lower respiratory tract infections in human, with infants and young children being particularly susceptible. In the temperate zones, sharp annual outbreaks of RSV occur during the colder months, in both the northern and the southern hemisphere. RSV is unusual in that it can repeatedly reinfect individuals throughout life and infect babies in the presence of maternal antibody. RSV isolates can be divided into two subgroups, A and B, on the basis of their reactions with monoclonal antibodies, and the two subgroups are also distinct at the nucleotide sequence level. The specific diagnosis of RSV infection was best made by isolation of virus in tissue culture, identification of viral antigen, or by specific serologic procedures. Recently, rapid detection of RSV and analysis of RSV strain variation became possible by development of methods of reverse transcription and polymerase chain reaction amplification. In this study, to determine the genetic diversity of RSV found in Korea, 173 bp and 164 bp spanning selected regions of the RSV F and SH genes were enzymatically amplified and sequenced, respectively. Eight for F gene and three for SH gene were detected in 66 nasopharyngeal swap samples tested. Two major antigenic subgroups, A and B were confirmed from Korean samples (seven for subgroup A and one for subgroup B). At the nucleotide level of the F gene region, Korean subgroup A strains showed 95-99% homologies compared to the prototype A2 strain of subgroup A and 93-100% homologies among Korean subgroup A themselves. For the SH gene region, Korean subgroup A strain showed 97.5% homology compared to the prototype A2 strain of subgroup A, and Korean subgroup B strain showed 97% homology compared to the prototype 18537 strain of subgroup B. Most of base changes were transition and occured in codon position 3, which resulted in amino acid conservation. Using the maximum parsimony method, phylogenetic analysis indicated that Korean RSV strains formed a group with other RSV strains isolated from the United States, Canada, the Great Britain and Australia.

2003년 국내 원종계 및 종계의 추백리-가금티푸스 감염실태 (The prevalence of pullorum disease-fowl typhoid in grand parent stock and parent stock in Korea, 2003)

  • Serum samples of 30 chickens per flock from 6 grand parent stock (GPS) farms and 70 parent stock (PS) farms were collected for seroprevalent study of pullorum disease-fowl typhoid (PD-FT) infection by serum plate agglutination test (SPA). The incidence of PD-FT infection in GPS flocks and PS flocks were 0% and 15.7%, respectively. Especially PS flocks infected with PD-FT showed age dependent patterns that 22.2% of flocks between 20 to 30 weeks of age and 38.9% of flocks between 30 to 40 weeks of age were positive. The incidence of GPS flocks and PS flocks using Salmonella (S.) gallinarum 9R (SG9R) live vaccine were 33.3% and 58.6%, respectively. The sero-positive rate of 11 flocks were 6.7-83.3% by SPA and 2.9-55.6% by enzyme linked immunosorbent assay (ELISA), and ELISA showed more lower antibody levels than SPA. Furthermore, specific antibodies produced by SG9R vaccination were detectable by SPA using SG9R antigen without cross-reaction with the PD-FT infection.

의사 돼지콜레라 발생농장 역학조사 (Epidemiological surveys of an outbreak of false positive classical swine fever in Gyeongbuk province)

  • This study was conducted to survey the farm which suffered from disease similar to classical swine fever(CSF) in Gyeongbuk province. Clinical signs appeared first in a few number of growing pigs which showed specific signs of diarrhea, depression, sleepiness, and reluctance to get up or to eat. Younger piglets may have appeared chilled, shiver and huddle together. As the disease progresses the affected pig's skin went red and purple. In histopathological signs, there were many haemorrhages throughout the body and larger haemorrhages in some organs such as lymph nodes. And there is a precipitous fall in the number of circulating leukocytes in the blood. In spite of insisting of farmer which did not vaccinate to classical swine fever, significant antibody production was detected in these affected pigs at enzyme-linked immuonsorbent assay. According to the above results at first glance, these affected pig suspected with CSF in clinical signs and histopathological lesions only. Because the symptoms and post-mortem picture were very similar to CSF, these false positive results would have been dangerous to diagnostician. But by reverse transcriptase polymerase chain reaction(RT-PCR) and comparative nucleotide sequence analysis, the disease was correctly diagnosed with post-weaning multisystemic wasting syndrome(PMWS) and porcine reproductive and respiratory syndrome(PRRS) compoundly. And the antigen which were detected the lesion similar to CSF virus, was confirmed with LOM vaccine strain of CSF. In most national CSF eradication program and in countries which are free of the CSF virus, vaccination against CSF is not practiced and generally is not allowed. But now in Korea, routine vaccination is practiced because of outbreaking the CSF repeatedly. When CSF is diagnosed the whole herd and other in contact animal are slaughtered continuously.
