Prevalence of toxoplasma antibody in swine by microplate latex agglutination test in Kyeongnam district (Latex 응집반응에 의한 경남중부지역 돼지의 톡소플라즈마병 항체조사)

  • This study was conducted to detect the serum antibody of toxoplasma in swine from breeding-pig, rearing-pig farm and slaughtered pig in abattior by latex agglutination(LA) test. The perfomance of LA test was carried out with commercial Toxo-MT kit(Eiken Chemical Co.)by Tsubota and Ozawa's method. The cut-off titer of positive and negative reactions by Toxo-MT antigen used in this experiment was determined as the serum titer of 1 : 32. Positive rate of toxoplasma antibody from the total of 823 serum samples by LA test was 17.0%(140 cases). And positive rates of toxoplasma antibody against serum samples of 194 from breeding-pig farm, 273 from rearing-pig farm and 356 from abattior were 91 cases(46. 9%), 23 cases(8.4%) and 26 cases(7.3%), respectively. The distributions of serum antibody titers in 823 test sera by LA test were shown 51 cases(36.3%) in 1:32, 40(28.6%) in 1:64, 17(12.1%) in 1:128, 14(10.0%) in 1:256, 3(2.1%) in 1:512, 5(3.6%) in 1:1024 and 3(2.1%) in 1:2048. The ranges of positive rate from the sera in each group of breeding-pig farms were 20~61.9%.

  • This study was conducted in May of 1996 to December of 1996 in order to investigate the status of rubella antibodies in the women of childbearing age. The subjects were 543 fertile women (Ages 21-42 years). ELISA method was used for the detection of rubella antibodies and then questionaire survey was performed to know about the variables of past history of rubella. past rubella immunization. parity and cognition. The results were as follows: 1. The Positive rate of rubella Ig G antibody in total subjects was $65.0\%$. The positive rate of rubella Ig G antibody was $72.2\%$ in 21-25 age group. $71.4\%$ in 26-30 age group. $54.5\%$ in 31-35 age group, $52.6\%$ in 36-42 age group. As age increased, the positive rate of rubella Ig G antibody was decreased. There was statistically significant difference by age group(P=0.00l). In the subjects with a history of rubella immunization, the positive rate of rubella IgG antibody was $81.8\%$, and in those with past history of rubella was $83.3\%$ of positive rate. 2, Cognition rate about rubella immunization showed $50.8\%$ in total subjects. and there was no significant difference between parity and cognition rate of rubella immunization(P=0.85l). observed a low positive rate of rubella IgG antibody as compaired with other studies. Therefore, to prevent congenital rubella infection, rubella immunization was needed for unmarried women.

Epidemiological study for spotted fever group rickettsia of wild rats(Rattus norvegicus and R rattus) in Korea (국내 야생 집쥐류(Rattus norvegicus와 R rattus)의 spotted fever group rickettsia 감염에 관한 역학조사)

  • Seventy wild rats were caught from Seoul city, Kyonggi, Kangwon, Honam, and yongnam provinces. All of them were in same species, Rattus norvegicus, except two R rattus from Kyonggi province. Seventy sera from wild rats were studied by immunofluorescent antibody assay for evidence of infection by spotted fever group rickettsia. The antidody prevalance was 37.14%(26/70) for spotted fever group rickettsia. The sero-prevalence rates for spotted fever group rickettsia antibody was the hightest in Kyonggi province with 55.56%(10/18), yongnam province with 50.00%(10/20), Kangwon province with 25.00%(2/8), Seoul city with 18.75%(3/16), and Honam province with 12.50%(1/8). The sero-positive rates difference between sexes were higher in female with 46.15%(12/26) than in male with 31.81%(14/44) for spotted fever group rickettsia. Twenty six of 68 Rattus norvegicus with antibody for spotted fever group rickettsia were in subadult with 50.00%(6/12), young adult with 38.89%(7/18), middle-aged adult with 35.29%(6/17), and old adult with 33.33%(7/21). No antibody was detected from R rattus.

Infection Patterns of Porcine Epidemic Diarrhea Virus (PEDV) by Sera-epidemiological Analysis in Korean Pig Farms (혈청역학적 분석을 통한 한국의 돼지 유행성 설사병 바이러스 장염양상)

  • To investigate the infection patterns of porcine epidemic diarrhea virus (PEDV) in Korean pig farms, a total of 4,768 swine sera samples from 159 pig farms were taken twice, in June (n=82) and October (n=77) in 2007. In each farm selected for the survey, 10 samples from breeding pigs and 4 from each of the 5 age groups (30, 60, 90, 120, and 150 days) were taken, and all serum samples were tested for PEDV by the serum neutralization test. The overall seroprevalence was 62.6% (2,983/4,768), with the highest prevalence in breeding pigs (93.5%, 1,485/1,589). The prevalence showed an increasing trend with increasing age (30.8, 27.2, 44.7, 61.6, and 71.2% respectively in the 30, 60, 90, 120, and 150 days age groups) (p<0.0001 for $x^2$ trend test). The association between age and PEDV prevalence was similar in both surveys, indicating that the infection of PEDV seemed to be occurring repeatedly in the farms surveyed. This inference could also be explained by the fact that prevalence in sows was very high despite low vaccination coverage, as they are continuously exposed to PEDV in potentially infected farms for a longer period. Based on the neutralizing antibody levels in sows and growing pigs, the majority of farms (91.8%, n=146 farms) were endemically infected with PEDV, and most of pigs seemed to be intensively infected with PEDV at around early growth (41.8%) and weaning (31.5%). On the other hand, serum neutralizing antibodies were not detected in pigs older than 30 days of age in farms classified as having no PEDV infection (n=13 farms), indicating the level of maternal antibody against PEDV is decreased on a non-detectable level before the piglet is 60 days old in the field situation. The results indicated that most farms surveyed in 2007 were affected with endemic PEDV infection. Therefore, a national monitoring and control program for the endemic type PEDV infection needs further attention.

  • Toxoplasmosis, caused by Toxoplasma gondii, is a parasitic zoonosis with worldwide distribution. The present study investigated the prevalence of T. gondii in dogs in Zhanjiang city, southern China, using both serological and molecular detection. A total of 364 serum samples and 432 liver tissue samples were collected from the slaughter house between December 2012 and January 2013 and were examined for T. gondii IgG antibody by ELISA and T. gondii DNA by semi-nested PCR based on B1 gene, respectively. The overall seroprevalence of T. gondii IgG antibody was 51.9%, and T. gondii DNA was detected in 37 of 432 (8.6%) liver tissue samples. These positive DNA samples were analyzed by PCR-RFLP at 3'- and 5'-SAG2. Only 8 samples gave the PCR-RFLP data, and they were all classified as type I, which may suggest that the T. gondii isolates from dogs in Zhanjiang city may represent type I or type I variant. This study revealed the high prevalence of T. gondii infection in dogs in Zhanjiang city, southern China. Integrated measures should be taken to prevent and control toxoplasmosis in dogs in this area for public health concern.

Correlation between Disease Prevalence and Production Performance in Korean Swine Farms (양돈 생산성에 따른 주요 질병 분포 조사)

  • Currently, various diseases reside in Korean swine farms and affect production performance of the farms greatly. These damages from disease are further aggravated by the concurrent infection of other disease. In this study, y investigating the distribution of porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Salmonella spp. in farms, correlation between the damage and the prevalence of disease was analyzed. Ten selected Korean swine farms that uses PCV2 vaccine were tested for presence of antibody and antigen of PRRSV, PCV2, Salmonella spp. per ages of pigs, 4weeks, 7weeks, 11weeks and 17weeks, respectively. The results were analyzed by dividing the farms in to groups with MSY above 19, and that with MSY below 19. Then calculating the distribution of disease each ages of pigs. Farms with MSY below 19 showed high prevalence of disease by PRRSV, PCV2 and Salmonella spp.. In this group, the detection rate of PCV2 and Salmonella spp. was increased by the activation/viremia of PRRSV in the young ages of pigs. The results are proved that the correlation between disease prevalence and production performance in Korean swine farms were very significant. The prevalence of PRRSV is more important index which influence to the productivity in current prevalence of diseases.

Prevalence of Leptospiral Antibodies in Pigs (돼지의 Leptospira 속균에 대한 혈중항체조사 -제천시 소재 도축장 출하돈 중심-)

  • This experiment was to investigate the Leptospiral antibody in the pigs with the serological test by the microscopic-agglutination-test (MAT) from November 1991. to Januaury 1992. Antigen(living antigen) was used L. icterohemorrhagiae, L. pomona, L. canicola, L. hardjo, L. ballum, L. australis, L. autumnalis, L. grippotyphosa, L. tarassovi, L. pyrogenes, L. bataviae, L. hebdomadis for the serolgical test in the pigs. The result obtained are summarized as follows of the total 202 serum samples examined, 1. Among the serum samples of 202 heads, 19 heads of the pigs(9.4%) were positive. 2. Among the positive samples of 19 heads, The detected were L. icterohemorhagiae 10 heads(5.0%), L. pomona 3 heads(1.5%), L. canicola 6 heads(3.0%). 3. Antibody titers of positive sera were ranging from 1:100 to 1 : 400. Serotiters appeared to be very low, 4. The seroprevalence of Leptospira in Chechon was higher than that other districts (5. 4% -5.8%), but the lower than Chung-nam, Kyonggi(13.7-15.9%)

Prevalence of dog erythrocyte antigen (DEA) 1 among Jindo dogs (진돗개에서 개적혈구항원(dog erythrocyte antigen) 1 혈액형 조사)

  • This study was conducted to investigate the prevalence of dog erythrocyte antigen (DEA) 1 with DEA 1.1 and DEA 1.2 on 122 Jindo dogs (29 males, 93 females) from 2014 to 2015 using a monoclonal antibody card kit (blood typing card kit, Korea Animal Blood Bank Inc., South Korea). Among the tested dogs, 14.8% (18/122) were positive for the DEA 1.1 antigen and 85.2% (104/122) were positive for the DEA 1.2 antigen. The prevalence of positive types for the DEA 1.2 antigen was significantly higher than the DEA 1.1 antigen (P<0.01). The prevalence of positive types for the DEA 1.1 antigen was higher in white-haired Jindo dogs than yellow-haired dogs (P<0.05). However, there was no gender difference in the prevalence of the DEA 1.1 antigen (P=0.665). The incidence of sensitization after the first transfusion without blood group test was 12.6% and the incidence of acute hemolytic transfusion reaction after the second transfusion in the same immunized dogs was 1.6%. Therefore, the blood group test for the DEA 1 antigen should be performed for Jindo dogs to ensure safe and effective transfusion therapy and further studies remain to be conducted for other DEAs among Jindo dogs.

Study on Infection of Theileria Sergenti in Neonatal Calves (신생 송아지에 있어서 Theileria sergenti의 감염에 관한 연구)

  • The rate of 58 neonatal calves in infection of Theileria sergenti was investigated in random samples on the farms located in Kyunggi, Chonbuk districts of Korea. 1. The criteria used in veryfying infection with T. sergenti included the detection of parasites by giemsa stain and acridine orange stain in the blood smear slides. 2. Further evidence of current or previous exposure to T. sergenti was based on demonstration of T. sergenti specific antibody and antigen by the western immunoblot and the directed immunofluorescent antibody test in the peripherial blood of the calves. 3. The prevalence rates were 35%, 50% in Kyunggi, Chonbuk provinces respectively and the overall prevalence in all the farms was 43.2% by means of acridine orange stain. 4. The parasites that were observed in the peripherial blood of calves was showen surely by the western immunoblot to the characteristic 34KD antigen among the proteins of T. sergenti (Korean isolate). 5. And the antigen of the neonatal calves reacted at the very highest titer(1 : 2, 560) 6. These data highlight the significances of T. sergenti in the neonatal calf disease in Korea.

Direct Immunofluorescence in Clinically Diagnosed Oral Lichen Planus

  • Purpose: Oral lichen planus (OLP) is relatively common mucosal disease in clinical dentistry. OLP is intractable and regarded having malignant potential. Until now, there is some debate on how far OLP can be malignant, and which characteristics can be a risk factor for malignant transformation. Clinician need to know some differences between OLP and lesions similar to OLP to manage properly and suppose prognosis correctly. Therefore, the aim of this study was to divide clinical OLP into two groups and to compare the results of direct immunofluorescence (DIF) between two groups. Methods: This study was conducted on outpatients who visited at the department of Oral Medicine in Chonbuk National University Hospital from January 2007 to November 2015. Patients with DIF result were retrospectively reviewed. The selected patients were classified 'clinical typical of OLP' (CTO) or 'clinical compatible with OLP' (CCW) by modified World Health Organization diagnostic criteria of OLP and oral lichenoid lesion. Results: DIF were classified by deposition intensity or pattern of anti-human antibody and fibrinogen. The classification of fluorescence pattern in each specimen was graded as positive, possibly positive or negative. Conclusions: Both CTO and CCW had positive and possibly positive pattern. Prevalence of positive pattern was 68.8% in CTO and 52.6% in CCW and that of possibly positive pattern was 9.4% in CTO and 5.3% in CCW. Prevalence of negative was 21.8% in CTO and 42.1% in CCW.
