Serological Identification of Mastitic Streptococci by Lancefield Grouping and Antibiotic Sensitivity Test (Lancefield Grouping에 의한 유방염유래(乳房炎由來) Streptococcus의 동정(同定)과 항생물질(抗生物質)에 대한 감수성조사(感受性調査))

  • A Total of 173 streptococci isolated from 536 dairy cattle in Gyeonggi Province identified by Lancefield serological reaction and other biological properties. Antibiotics sensitivity test was also examined and the results obtained were as follows: 1. A total of 173 isolates of 167 strains (96.5%) were reacted with group A,B,C,D,E and G antisera. 2. Of the 167 positive strains, 75 strains were identified to Streptooccus uberis, 56 to Streptococcus agalactiage and 26 to Streptococcus dysgactiae, respectively. Also, Sreptococcus pyogenes, group D and group G streptococci were rarely isolated. 3. In results of antibiotic sensitivity tests, the most isolates were sensitive to leucomycin and erythromycin but less sensitive to streptomycin. and kanamycin.

A Comparative Study of the Detectable Methods of Residual Antibiotics in Milk (우유중 잔류 항생물질 분서방법에 관한 비교연구)

  • Recently, as concern about the residual antibiotics in milk increase, the detection methods of residual antibiotics used extensevely at the present time were investigated and compared to their properties and the detection limits of variable antibiotics. At first, comparactive tests of the detectable sensitivity of 4 test organisms, B. cereus, B. subtilis, M.luteus, B.stearothermophilus C-953, were performed by disc assay. As a result, B.stearothermophilus was the most sensitive strain of all other strains and showe the detect limit of 5-50 ppb for penlicillins (PCs). And also, B.subitilis was showed the more effective detection limit, 200-400 ppb, for aminoglycosides (AGs) and M.luteus was showed predominant sensitivity , 50-500 ppb for macrolides(MLs) and B.cereus was the most sensitive strain for tetracyclines (TCs) and showed the detection limit of 100-400 ppb. Therefore, each test strains were showed a different sensitivity in the detection of the different antibiotic families. When the detection limit of disc assay and other methods were compared, TTCmethod was less sensitive than other methods showing 5-50 ppb detectable lebel for PCs. Also, for the detection of other antibiotic families TTC method was showed the worst sensitivity and Delvo and Charm Farm tests were similar to the detectable properties of AGs and MLs. Although disc assay was showed the similar detection limit for PCs with Delvo and Charm Farm, it was more widely effective for the detection of kanamycin, erythromycin, chlortetracycline, doxycycline, verginiamycin and so on than Delvo or Charm Farm. CharmII test was showed the best sensitivity for the most of antibiotics except neomycin and gentamycin. But it was necessary that different tests must be performed to each antibiotic family and so it was regarded that the effectiveness of that method was low.

  • Phosphomannomutase (ManB) is involved in the biosynthesis of GDP-mannose, which is vital for numerous processes such as synthesis of carbohydrates, production of alginates and ascorbic acid, and post-translational modification of proteins. Here, we discovered that a deletion mutant of manB (BG101) in Streptomyces coelicolor (S. coelicolor) showed higher sensitivity to bacteriostatic chloramphenicol (CM) than the wild-type strain (M145), along with decreased production of CM metabolites. Deletion of manB also decreased the mRNA expression level of drug efflux pumps (i.e., cmlR1 and cmlR2) in S. coelicolor, resulting in increased sensitivity to CM. This is the first report on changes in antibiotic sensitivity to CM by deletion of one glycolysis-related enzyme in S. coelicolor, and the results suggest different approaches for studying the antibiotic-resistant mechanism and its regulation.

Distribution and Antibiotic Susceptibility Patterns of Genus Mycobacterium at a Private Hospital, Korea

  • Mycobacterium isolates were retrospectively identified, antibiotics susceptibility test results and basic clinical data were analyzed for the 715, excepted 308 in 1,023 specimens, from a mycobacterial laboratory at a tertiary care hospital from September 2002 to December 2008. Their male to female ratio was 1.12 to 1 (379 male, 336 female). The median age of study population was 47 years (range from 10 to 93 years). Distribution of Mycobacterium species was 90.1% of total were isolates Mycobacterium tuberculosis, and 9.9% of the total non-tuberculosis Mycobacterium isolated, and Among nontuberculosis Mycobacterium isolates, 60.6% were Mycobacterium avium complex, 14.1% were isolates Mycobacterium abscessus, and 12.7% were isolates Mycobacterium intracellulare. Among 526 Mycobacterium tuberculosis isolates, 81.7% isolates were susceptible to first line antibiotics, 18.3% were resistant to one or more antibiotics. Non-tuberculosis Mycobacterium isolates, all were resistant to two or more antibiotics. Multi-antibiotic resistant tuberculosis rate was show 10.2% of total specimens. Isolated Mycobacterium species, 19.2% were multi-antibiotic resistant tuberculosis, and the rate of nontuberculosis Mycobacterium resistant to isoniazid and rifampin was very highly 84.5%. Thus among acid fast bacilli culture positive cases, Mycobacterium tuberculosis and non-tuberculosis Mycobacterium were must exactly identification and antibiotic sensitivity test. It was considered to help to select of the antibiotic in preventive medicine.

Comparison of inactivation and sensitivity of antibiotic resistance bacteria by ultrasound irradiation (초음파 조사에 의한 항생제 내성균 불활성화 및 감수성 변화)

  • The 20-kHz ultrasonic irradiation was applied to investigate bacterial inactivation and antibiotic susceptibility changes over time. Applied intensities of ultrasound power were varied at 27.7 W and 39.1 W by changing the amplitude 20 to 40 to three bacteria species (Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus). By 15-min irradiation, E. coli, a gram-negative bacterium, showed 1.2- to 1.6-log removals, while the gram-positive bacteria, Enterococcus faecalis and Staphylococcus aureus, showed below 0.5-log removal efficiencies. Antibiotic susceptibility of penicillin-family showed a dramatic increase at E. coli, but for other antibiotic families showed no significant changes in susceptibility. Gram-positive bacteria showed no significant differences in their antibiotic susceptibilities after ultrasound irradiation. Bacterial re-survival and antibiotic susceptibility changes were measured by incubating the ultrasound-irradiated samples. After 24-hour incubation, it was found that all of three bacteria were repropagated to the 2- to 3-log greater than the initial points, and antibiotic inhibition zones were reduced compared to ones of the initial points, meaning that antibiotic resistances were also recovered. Pearson correlations between bacterial inactivation and antibiotic susceptibility showed negative relation for gram-negative bacteria, E. coli., and no significant relations between bacterial re-survival and its inhibition zone. As a preliminary study, further researches are necessary to find practical and effective conditions to achieve bacteria inactivation.

  • The study for a effect of monitoring on bovine mastitis was conduced for a year from Dec, 1989 to Nov, 1990, Sampling the bulk milk of 350 herds in Inchon city and out of 10 herds among them were carried out herds guidance, CMT, SCC, isolation of pathogens and antibiotic sensitivity tests. The results obtained were summarized as follows 1. Annual mean SCC of 1213 herds was 558, 000 cell /ml 2. The number of SCC below 500, 000 cell /ml to quarters for herds guidance was at 1st 77. 0%, End 84.8% and 3rd 80.4%. The is shown that milk quality was steadly improved. 3. The most number of isolated pathogens of bovine mastitis was Staphylococcus SPP - 402(47.2%) Streptococcus SPP - 80(18.7% ) 4. The highest rate of antibiotic sensitivity test was Stapylococcus SPP - cephalothin(76.7%) Streptococcus SPP - ampicillin(77.5%) Gram negative bacilli - tetracyclin(76.0%) 5. The effect of monitoring on bovine mastitis was improved showing that at 1st 49.0% to 3rd 72.0%

  • Spaceflights results in the reduction of immune status of human beings and increase in the virulence of microorganisms, especially gram negative bacteria. The growth of Klebsiella pneumoniae is enhanced by catecholamines and during spaceflight, elevation in the levels of cortisols occurs. So it is necessary to know the changes in physiology, virulence, antibiotic resistance and gene expression of K. pneumoniae under microgravity conditions. The present study was undertaken to study effect of simulated microgravity on growth, morphology, antibiotic resistance and cross stress resistance of K. pneumoniae to various stresses. The susceptibility of simulated microgravity grown K. pneumoniae to ampicillin, penicillin, streptomycin, kanamycin, hygromycin and rifampicin were evaluated. The growth of bacteria was found to be fast compared with normal gravity grown bacteria and no significant changes in the antibiotic resistance were found. The bacteria cultured under microgravity conferred cross stress resistance to acid, temperature and osmotic stress higher than the normal gravity cultured bacteria but the vice versa was found in case of oxidative stress.

One-Stage Treatment of Chronic Calcaneal Osteomyelitis with Bone Morphogenetic Protein 2 and Local Antibiotic Delivery in a Cat

  • An age-unknown, 4.8 kg, male, wild, domestic short-hair cat was presented for left hindlimb lameness. A physical examination revealed a draining tract which was suspected of bite on left calcaneal bone. The left tarsal joint was markedly swollen and exudates were observed around the draining tract. Sequestrum at left calcaneus bone, and osteolysis were identified by radiography. The sequestrum and its surrounding exudative tissue were debrided during surgery and the tissue was submitted for bacterial culture and sensitivity test. The debridement caused a bone defect ($1.5cm{\times}0.5cm{\times}0.5cm$) on the medial left calcaneal bone. Plate and screw fixation was performed to the calcaneus bone as buttress plate. Recombinant human bone morphogenetic protein-2 (rhBMP-2) loaded hydroxyapatite was implanted in the bone defect. Furthermore, Amikacin-impregnated collagen sponges were also placed around bone plate to deliver local antibiotics. A systemic antibiotic treatment regimen based on bacterial culture and sensitivity test results was administered for 4 weeks. The wound properly healed without any signs of infection, and the bone healing was confirmed by radiography. The patient showed normal weight bearing ambulation at 18 weeks after surgery. The use of rhBMP-2 and local antibiotic delivery system is a good surgical option for the one-stage treatment of chronic osteomyelitis.

Genetic Toxicity Studies of YH1226, a Cephalosporin Antibiotic (세파계 항생제, YH1226의 유전독성 평가)

  • The results of chromosome aberration test in mammalian cells in culture (Chinese hamster lung fibroblast cells) showed no induction of structural and numerical aberrations by YH1226, a cephalosporin antibiotic regardless of metabolic activation, while positive control group (mitomycin C and benzo(a)pyrene) showed structural chromosome aberrations of 25% and 10%, respectively. The in vivo induction of micronuclei was measured in polychromatic erythrocytes in bone marrow of male ddY mouse given YH1226 at 500, 250, 125 mg/kg by i.p. once. After 24 hours, animals were sacrificed and evaluated for the incidence of micronucleated polychromatic erythrocytes in whole erythrocytes. Although a positive response for induction of micronuclei in animals treated with mitomycin C demonstrated the sensitivity of the test system for detection of a chemical clastogen, YH1226 did not induce microunclei in bone marrow of ddY male mice.

In Vitro Inhibitory Activities of Essential Oils from Rosmarinus officinalis L. Against Antibiotic-Susceptible and Resistant Strains of Some Pathogenic Bacteria (Rosmarinus officinalis 정유의 수종 항생제 감수성 및 내성 균주에 대한 억제효과)

  • The in visto inhibitory activities of essential oils of the Rosmarinus officinalis as well as its main constituents were evaluated against antibiotic-susceptible and -resistant strains of Staphylococcus aureus, streptococcus pneumoniae, Salmonella enteritidis and S. typhimurium. The essential oil fraction of R. officinalis and its main components, 1,8-cineole and camphor, exhibited significant inhibitory activities against most of the tested strains in this study, with MICs(minimum inhibitory concentrations) racing from 0.5mg/ml to 16mg/ml. The total oil fraction showed higher activity than its main components, 1,8-cineole and camphor against S. aureus strains. No remarkable differences were evident between MICs of the susceptible and resistant strains of S. aureus. Among the tested strains, S. pneumoniae CCARM 3523, the resistant strain to norfloxacin, oxacillin and erythromycin exhibited significantly lower sensitivity to the tested oils than antibiotic-susceptible strain. The oils revealed mostly higher inhibitory activity against S. typhimurium than against S. enteritidis.
