Comparison of antibiotic resistance profiles for Escherichia coli isolated from wild boar and domestic pig fecal samples

  • Increasing presence of wild boar around cities and suburban areas is a growing concern with respect to agronomy, environmental ecology, and public safety. In this study, antibiotic resistance profiles of Escherichia (E.) coli isolated from wild boar and domestic pig fecal samples were compared. Eighty E. coli samples were isolated from wild boars. Resistance of the bacteria to 14 common antimicrobial agents used in human and veterinary medicine was evaluated. Ninety-five E. coli isolates from domestic pig farms were used for comparison. Common and distinct antibiotic resistance patterns were observed when comparing wild boar and domestic pig isolates, indicating that wild boars may significantly influence environmental microbiology.

  • Objectives: This study aims to examine the concentration, diversity, and antibiotic characteristics of penicillin G resistant enterobacteria present in pharmaceutical effluent. Methods: Water sampling was performed from a pharmaceutical company in Gyeonggi-do Province, Korea in March 2015. Water samples were plated in triplicate on tryptic soy agar plates with 32 mg/L of penicillin G. Penicillin G resistant enterobacteria were selected from the effluent and were subjected to 16S rRNA analysis for penicillin G resistant species determination. Identified resistant strains were tested for resistance to various antibiotics. Results: Penicillin G resistant enterobacteria were present at 6.2% as to culturable heterotrophic bacteria. Identified penicillin G resistant enterobacteria exhibited resistance to more than 10 of the antibiotics studied. These resistant bacteria are gram negative and are closely related to pathogenic species. Conclusion: Multi-antibiotic resistant bacteria in the effluent suggest a need for disinfection and advanced oxidation processes for pharmaceutical effluents.

  • In both untreated and conventionally stabilized swine manures antibiotic resistant (AR) microorganisms, Staphylococcus-like and Salmonella-like microorganisms were detected. Also pathogens with MAR phynotype were detected. Presence of such microorganisms suggest high level of pathogen-related health risk to farmers who may be in direct contact with the manure and its conventionally stabilized product In contrast the autothermal thermophilic aerobic digestion (ATAD) treatment have efficiently reduced AR and pathogenicity from the swine manure. When soil was fertilized using swine manure and its stabilized products, despite no detection of MAR-exhibiting pathogen-like microorganisms in fertilized soil, potential pathogen-related health risk could not be ruled out from the fertilized soil since the organic fertilization led to increase in AR and pathogenicity in the soil microbial communities. As conclusion, this microbiological study demonstrated that an ATAD process is applicable in control of pathogen-related health risk in livestock manure.

  • Antibiotic and physico-chemical properties of an active compound from actinomycetes isolate G-37, of which the culture filtrate had an inhibitory effect against tobacco mosaic virus(W) infection, were examined. The active compound, which was purified by ethylacetate extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography, showed strong antibacterial activities especially against Gram-positive bacteria including Bacillus subtillis, Sarcina lutea and Staphylococcus aureus. From the IH-NMR, FAB/RfS, UV spectral data, and physicochemical properties, the active compound of G-37 appears to belong to a peptide antibiotic group. Among the known peptide antibiotics in the antibiotic group, No. 280, A-30912, and Taitomycin showed molecular weights and ultra violet spectrum similar to those of the active compound from G-37, but was not identical to the compound, which suggests that it may be a new peptide antibiotics.

  • Recently, a number of countries are now considering the reuse of effluents from wastewater treatment for various water applications. To improve the reuse of wastewater effluent, the development of appropriate micro-pollutant removal technology is necessary. Although several researche have been studied for removing micro-pollutants in water, little study has been conducted for the removal of emerging contaminant such as antibiotic resistant genes (ARGs) by disinfection processes. Therefore, the aim of this study is to compare the capacity of disinfection technologies such as chlorination, ozone, and electron beam, for removing antibiotic resistant bacteria (ARB) and ARGs. Based on this study, better ARG removal can be achieved by ozonation and electron beam. Relatively, high CT values of chlorination or ozonation are needed to remove ARB and ARG compared to conventional pathogens.

  • Total 70 isolates of Escherichia coli obtained from pigs were studied. Forty four isolates had $\textregistered$-hemolytic activity which was heat labile. Minimum inhibitory concentration test indicated that 40 isolates (57.1%), 15 isolates (21.4%), 23 isolates (32.9%), and 5 isolates (7.1%) were resistant to ampicillin, cephalothin, gentamicin, and norfloxacin, respectively. None of them were extended spectrum $\textregistered$-lactamases (ESBLs) producer when the double disk synergy test (DDST) was performed.

Isolation and Properties of Antitumor Antibiotic YS-1649 from Penicillium sp. strain 1649

  • An antitumor antibiotic named YS-1649 was isolated from the culture filtrate of a newly isolated fungus identified as Penicillium sp.. The fermentation yield reached about 40 mg per liter of the broth. YS-1649, a $\gamma$-Iactone - structured antibiotic, has the molecular fomular of $C_7H_6O_4$, Its structure was determined to be patulin by spectral analysis. It is active against some bacteria and showed cytotoxic effect on the proliferation of human breast cancer cell line, MCF-7, at concentrations of more than 0.048 $mu g/ml$. This compound also showed strong cytotoxic effect on the proliferation of human cancer cell lines, A549 and ACHN.

Biotransformation of Rosamicin Antibiotic into 10,11-Dihydrorosamicin with Enhanced In Vitro Antibacterial Activity Against MRSA

  • A biotransformation approach using microbes as biocatalysts can be an efficient tool for the targeted modification of existing antibiotic chemical scaffolds to create previously uncharacterized therapeutic agents. By employing a recombinant Streptomyces venezuelae strain as a microbial catalyst, a reduced macrolide, 10,11-dihydrorosamicin, was created from rosamicin macrolide. Its chemical structure was spectroscopically elucidated, and the new rosamicin analog showed 2-4-fold higher antibacterial activity against two strains of methicillin-resistant Staphylococcus aureus compared with its parent rosamicin. This kind of biocatalytic approach is able to expand existing antibiotic entities and can also provide more diverse therapeutic resources.

  • China is the great powers of use and production of antibiotics.The current process of sewage treatment plants can not effectively remove antibiotics in water. Chinese scholars have detected different kinds of antibiotics in major waters of the country, which have potential harm to human body. Among all kinds of antibiotic treatment technologies, adsorption removal technology has the advantages of simple operation, low cost and high removal efficiency. It is a widely concerned antibiotic removal technology. However, at present, few materials have been put into practical application, and more materials with low cost and high efficiency need to be found. Different adsorptive materials have different adsorptivity to different antibiotics. For different antibiotics, different adsorptive materials can be integrated in the future, and the theory can be extended to application.

In Vitro Effects of Essential Oils from Ostericum koreanum against Antibiotic-Resistant Salmonella spp

  • The essential oil fraction of Ostericum koreanum was analyzed by GC-MS. Inhibiting activities of this oil and its main components were tested by the broth dilution assay and disk diffusion test against one antibiotic-susceptible and two resistant strains of Salmonella enteritidis and S. typhimurium, respectively. The GC-MS analysis revealed thirty-four compounds; the main components were $\alpha$-pinene (41.12%), $\rho$-cresol (17.99%) and 4-methylacetophenone (7.90%). The essential oil of O. koreanum and its main components were significantly effective against the tested antibiotic-susceptible strains as well as against the resistant strains of the two Salmonella species, with MICs (minimum inhibitory concentrations) ranging from 2 mg/mL to 16 mg/mL. The anti-Salmonella effects of the oils were dose-dependent on $M\"{u}ller-Hinton$ agar plates in this experiment. Additionally, checkerboard titer test results demonstrated significant combined effects of streptomycin and O. koreanum oil or cresol, one of the main components of this oil, against the two streptomycin resistant strains of S. typhimurium, with FICIs ranging from 0.12 to 0.37.
