Studies on the Varietal Resistance of Rice to the Smaller Brown Planthopper, Laodelphax striatellus Fallen(IV) (애멸구에 대한 벼의 품종저항성에 관한 연구(VI))

  • Experiment was conducted to select the rice varieties and lines resistant to the smaller brown planthopper, Laodelphax striatellus Fallen, and study the cause of varietal resistance to the insect. The nature of varietal resistance was evaluated from the viewpoints of feeding and ovipositional preferences and antibiosis. Among the IRRI sources the varieties H 105, Muthumanikam, Vellailangalayan, Karsamba Red ASD-7. Manavari Co 22, Mudgo, PTB-18, IR 8 and IR 20 were selected as resistant sources. Among the domestic sources. only Suweon 213-1, Suweon 214 and Suweon 215 originated from the IR667 lines were resistant, and other leading varieties ail susceptible. The cause of resistance of rice to the insect seemed to be highly related with the non-feeding preference, not non-ovipositional preference. Nymphal mortality and rate of adult emergence were significantly different from the resistant and susceptible varieties. The rate of adult emergence was significantly lower in the resistant than In the susceptible varieties. The primary cause of lower adult emergence seemed to be due to the fact that the infects were suffered higher nymphal mortality in the resistant than in the susceptible varieties.

The Nature of the Variety Tongil (Suweon 213-1) in Resistance to the Striped Rice Border, Chilo suppressalis W. (이화명충에 대한 수도통일품종의 저항성)

  • This study was conducted to evaluated the nature of the variety Tongil(Suweon 213-1) in resistance to the striped rice borer, Chilo suppressalis Walker, comparing with those of Rexoro(susceptible check) and TKM-6(resistant check) selected at IRRI. 1. The striped rice borer moths much more preferred the variety Tongil for oviposition than the varieties Rexoro and TKM-6. The variety Tongil and more egg masses and number of eggs than the varieties Rexoro and TKM-6, while TKM-6 having more egg masses and more number of eggs than Rexoro. This reaction was consistent throughout the test regardless of the number of tillers per hill. 2. In laboratory, preference of larvae for feeding was studied with 5cm of stem pieces of the varieties. The results showed, in contrast to the case of ovipositional preference, that the striped rice borer larvae least preferred the stems of Tongil among the test varieties, while larvae much more preferred the stems of Rexoro than those of TKM-6. 3. The larval weights at 20 days later infested on the 40 day-old plants were the lowest on Tongil among the test varieties. On the variety Rexoro the larvae had heavier body weights(43.0mg), higher pupation(64.9%) and higher adult emergence(83.3%) than those on Tongil(larval weights 30.3mg, pupation 60%, adult emergence 60.7%) and TKM-6(larval weights 35.7mg, pupation 56.3%, adult emergence 51.9%). The pupal weights, however, were not consistent among the test varieties and/or sexes in comparison with the larval weights, pupation and adult emergence above mentioned. 4. Field experiments indicated that the incidence of dead hearts at 70 days after transplanting was relatively higher on the variety Tongil(11.1%) than those on Rexoro(8.9%) and TKM-6(8.4%), and the incidence of white heads at harvest was, in contrast to the dead hearts, lower on Tongil(9.8%) than those on Rexoro(27.4%) and TKM-6(13.9%). At harvest lower larval survival observed on Tongil (49 larvae/40 hills) than those on the susceptible variety Rexoro(104 larvae/40 hills) and on the resistant variety TKM-6(70 larvae/40 hills). The average larval weights collected from three test varieties at harvest were 80.5mg from Tongil, 83.7mg from TKM-6 and 99.6mg from Rexoro. 5. Increased nitrogen fertilizer application to the variety Tongil, the striped rice borer damage was increased. Also, preference of larvae for feeding significantly increased with the increase of nitrogen fertilizer application. 6. Any specific association between the plant characters and striped rice borer resistance could not be found. The variety Tongil even having large number of tillers, short plant height, large stem, broad leaf, etc, had still high preference of moths for oviposition, low preference of larvae for feeding, low damage, and relatively high antibiosis. 7. Resistance of the variety Tongil to the striped rice borers seemed to be associated with the low feeding preference and the relatively high antibiosis, not associated directly with the ovipositional preference.

Studies on Rice Varietal Resistance to Rice Stem Maggot, Chlorops oryzae Matsumura (벼줄기굴파리에 대한 수도(水稻)의 품종저항성(品種抵抗性))

  • To establish a method for measuring varietal resistance against rice stem maggot in field and pot, 20 varieties were used in $1981{\sim}1983$. The degree of injured stems by the 1st generation larvae was significantly correlated with that of injured panicles by the 2nd ($r=0.864^{**}$). There was no ovipositional preference in rice stem maggot. The larval mortality after artificial infestation varied widly from 5.6 to 100% in varieties and that of the resistant varieties was high after boring. There was a negative correlation between number of injured leaves and larval mortality after artificial infestation ($'81:r=-0.833^{**},\;'82:r=-0.918^{**}$). It is considered that larval mortality mainly caused by antibiosis and larval mortality 20 days after infestation on rice seedlings is ideal for evaluating the varietal resistance to rice stem maggot. Resistant varieties are Nagdongbyeo, Jinjubyeo and Chucheongbyeo susceptible varieties, Keumgangbyeo, Milyang 23, Baigyangbyeo and Milyang 30.

생물방제균 Bfacillus subtilis YB-70의 외부 Urease 유전자 도입과 길항력 증강

  • To genetically breed powerful multifunctional antagonistic bacteria, the urease gene of alkalophilic Bacillus pasteurii was transferred into Bacillus subtilis YB-70 which had been selected as a powerful biocontrol agent against root-rotting fungus Fusarium solani. Urease gene was inserted into the HindIII site of pGB215-110 and designated pGU266. The plasmid pGU266 containing urease gene was introduced into the B. subtilis YB-70 by alkali cation transformation system and the urease gene was very stably expressed in the transformant of B. subtilis YB-70(pGU266). The optimal conditions for the transfomation were also evaluated. From the in vitro antibiosis tests against F. solani, the antifungal activity of B. subtilis YB-70 containing urease gene was much efficient than that of the non-transformed strain. Genetic improvement of B. subtilis YB-70 by transfer of urease gene for the efficient control seemed to be responsible for enhanced plant growth and biocontrol efficacy by combining its astibiotic action and ammonia producing ability.

  • A potential biocontrol bacterium, YB-70 was isolated from a rhizosphere in suppressive soil and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on Fusarium solani with the culture filterates from B. subtilis YB-70, we found that antifungal mechanism of B. subtilis YB-70 was mediated by antibiotic substances produced from the bacterium. These antifungal substances were appeared to be hear-resistant, micromolecular, and ethy alcohol soluble. Antifungal agents produced by B. subtilis YB-70 showed strong inhibified against root-rotting fungi F. solani in in vivo pot test. An antifungal substance. YBS-1s, was purified from the culture broth of B. subtilis YB-70 by isoelectronic precipitation, silica gel column chromatography and Sephadex LH-20 column chromatography analysis by Fab-MASS, $^{1}$H-NMR, $^{13}$C-NMR, DEPT, and amino acid analyzer revealed that the YBS-1A was a peptide antibiotics of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin group composed of 11 amino acids with larher molecular weight of about 1, 500 dalton, which was lager than that of iturin A.

Isolation, Identification and Biological Control Activity of SKU-78 Strain against Ralstonia solanacearum (풋마름병균, Ralstonia solanacearum의 길항세균 SKU-78 균주의 분리 동정 및 특성)

  • Six stains of plant growth promoting rhizobacteria were selected through germinating seed assay and root colonization assay. Among them, SKU-78 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 60% reduction of bacterial wilt disease compared with the control. It was suggested that SKU-78 strain activated the host defense systems in plants, based on lack of direct antibiosis against pathogen. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, SKU-78 stain was identified as Bacillus sp. SKU-78.

Potential of Epicoccum purpurascens Strain 5615 AUMC as a Biocontrol Agent of Pythium irregulare Root Rot in Three Leguminous Plants

  • Epicoccum purpurascens stain 5615 AUMC was investigated for its biocontrol activity against root rot disease caused by Pythium irregulare. E. purpurascens greenhouse pathogenicity tests using three leguminous plants indicated that the fungus was nonpathogenic under the test conditions. The germination rate of the three species of legume seeds treated with a E. purpurascens homogenate increased significantly compared with the seeds infested with P. irregulare. No root rot symptoms were observed on seeds treated with E. purpurascens, and seedlings appeared more vigorous when compared with the non-treated control. A significant increase in seedling growth parameters (seedling length and fresh and dry weights) was observed in seedlings treated with E. purpurascens compared to pathogen-treated seedlings. Pre-treating the seeds with the bioagent fungus was more efficient for protecting seeds against the root rot disease caused by P. irregulare than waiting for disease dispersal before intervention. To determine whether E. purpurascens produced known anti-fungal compounds, an acetone extract of the fungus was analyzed by gas chromatography mass spectrometry. The extract revealed a high percentage of the cinnamic acid derivative (trimethylsiloxy) cinnamic acid methyl ester. The E. purpurascens isolate grew more rapidly than the P. irregulare pathogen in a dual culture on potato dextrose agar nutrient medium, although the two fungi grew similarly when cultured separately. This result may indicate antagonism via antibiosis or competition.

  • Several selected rice varieties Tong-il (Suweon #213-1), Jinheung, IR747, Suweon #229 and so forth were evaluated for their resistance to the striped rice borer, Cltilo suppressalis Walker. The resistance appears to be non-preference and antibiosis in nature. Under laboratory and greenhouse conditions, resistance to striped rice borer was manifested by the low larval weight, percentage of pupation and number of eggs recorded. In the cage test, varieties Suweon #240 and Shenshuraku appeared to be non-preferred by the borer moths for oviposition. Under field condition, variety Tetep received the lowest percentage of infested tillers.

Hanji made of mulberry fiber, a bast fiber of mulberry is a representative environment-friendly natural fiber. In addition, it has various functions similar to those of yellow earth such as emission of far infrared rays, antibiosis, deodorization, fast dry ability of sweat, and simple dyeing ability. Dyeing properties and colorimetric characteristics for cellulose fabrics dyed with indigo by different reducing conditions (쪽 염료의 환원조건에 따른 섬유소 직물의 염색성과 색채특성)

  • This study was carried out in order to provide useful data for planning fabrics of summer eco-friendly fashion products. The fabrics used in this study were four cellulose fibers: cotton, cotton/mulberry blended, flax, and flax/lyocell blended. Dyeing with natural indigo was carried out under three different reducing conditions (i.e., general, eclectic, and eco-friendly) that have different reducing agent and pH levels, and hydrosulfite and glucose were used as a reducing agent. The dye uptake (K/S value) of fabrics dyed with natural indigo by a reducing condition was the highest at 660nm. Regardless of the fabrics, dye uptake was the highest under the general reducing condition and the lowest under the eco-friendly reducing condition. Under different reducing conditions, the dye uptake of natural indigo fabrics with the maximum absorption wavelength indicated a difference. The colorfastness of cellulose fabrics that were dyed with natural indigo had a rate of 4 to 5 except for rubbing fastness, which indicated good colorfastness. Additionally, natural indigo-dyed cotton and flax fabrics had good antibiosis. When the color characteristics of fabrics dyed with natural indigo were measured, all of the three reducing conditions created purple blue (PB) colors, and the color characteristics of dyed fabrics by reducing condition and fabric showed significant differences.
