피부의 노화와 주름 억제, 미백효과를 가지는 전통 한약재의 탐색 (Research of Traditional Herbal Medicines for Anti-aging, Inhibition Effect of Wrinkle and Whitening Effect in the Skin)

  • The aims of this study are to search herbal medicines that have anti-aging, anti-wrinkle, anti-oxidant and whitening effects. This is preliminary study as a finding for excellent and specific natural agents which inhibit the skin aging and whitening formation. In this study, the articles and the documents, which have been published in domestically or internationally, have been searched and analyzed. Articles were gathered by taking advantage of Society of Cosmetic Scientists of Korea's documents, Pub med database, Korean Medical Database and Korean Studies Information Service System. Among the many species of herbal medicines, we selected the herbal medicines showing superoxide scavenging activities, and 1.1-diphenyl-2-picrylhydrazyl(DPPH) scavenging effects. The herbal medicines were showed anti-wrinkle effects on Metrixmetallo proteinase (MMP-1) inhibition activity and elastase inhibition activity. Also we found herbal medicines that have highly effect of tyrosinase inhibition, L- DOPA inhibition and melanin synthesis inhibition.

자생식물 11종의 항산화 및 항주름에 관한 향장효능 검증 (A Verification of Cosmetic Effect about Anti-oxidant and Anti-wrinkle of 11 Native Plants)

  • 본 연구에서는 화장품의 천연소재로 사용 가능함을 알아보기 위해 추출부위가 각각 다른 총 11종의 자생식물의 항산화, 항주름에 관한 생리활성 효능을 스크리닝 하였다. DPPH scavenging assay 측정결과 100 μg/ml 농도에서 70% 넘는 활성을 나타낸 시료는 순비기나무, 송이고랭이, 방울고랭이, 큰고랭이, 자귀풀이었다. Hydrogen peroxide scavenging assay 측정결과 1,000 μg/ml 농도에서 물꼬챙이골을 제외한 10종이 90%넘는 활성을 나타내었다. Xanthine oxidase scavenging assay 측정결과 1,000 μg/ml 농도에서 순비기나무(Leaf; 86.3%)가 가장 높은 활성을 나타냈다. 4가지 항산화 실험에서 모두 효능이 우수하게 나온 시료는 순비기나무, 송이고랭이, 방울고랭이, 큰고랭이, 자귀풀이며 그중 collagenase 저해활성 측정결과 방울고랭이가 가장 높은 저해 활성을 나타내어 항산화 및 항주름에 효능이 있는 시료로 나타났다. 이러한 결과를 바탕으로 활성이 높은 자생식물은 화장품에 사용되는 천연소재로 이용이 가능한 좋은 수종으로 판단된다.

오배자의 항 주름 효과 및 미백 효과에 미치는 실험적 연구 (The Anti-wrinkle Effects and Whitening Effects of Galla Rhois)

  • Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search Galla Rhois (GR) inhibitory activity against aging process of skin in vitro. GR was screened for their inhibitory activity against elastase. GR (final concentration 1 mg/ml) appeared over 40% of inhibition of elastase activity. So we investigated anti wrinkle effects of GR look through MMP-1 inhibition activity. Also Extracts of GR showed higher anti-tyrosinase activity than arbutin (final concentration 1 mg/ml). These results suggest that GR could be potential sources of anti-aging and whitening effects for the skin. Objective : Galla Rhois extracts, Elastase (pancreatic from porcine pancreas), N-Succinyl-(Ala)3-p-nitroanilide, Ursolic acid, mushroom tyrosinase, L-tyrosine, arbutin, Amicon Ultra, Hand Hold UV lamp, MMP-1 primary antibody, MMP-1 secondary antibody Result : 1. GR appeared over 40% of elastase inhibitory activity. 2. GR had MMP-1 inhibition activity. 3. GR showed higher anti-tyrosinase activity than arbutin. Conclusion : According to above results, it is supposed that Galla Rhois could be strong potential sources of anti-aging and whitening effects for the skin.

오배산가미(五倍散加味)가 점막(粘膜) 및 피부질환(皮膚疾患)에 미치는 영향 (The effects of Ohbaesangami (OBSGM) on the mucosa and skin diseases)

  • Objectives : To investigate the effects of Ohbaesangami (OBSGM) on mucosa and skin diseases, anti-microbial and anti-inflammatory tests were performed using several in vitro test models. Results : In anti-microbial test, OBSGM showed the slight inhibitory effect against Propionibacterium acnes (P. acnes) and Staphylococcus aureus (S. aureus). In anti-oxidant test, OBSGM showed the potent radical scavenging activity. In anti-inflammatory test, OBSGM weakly inhibited the lipopolysaccharide (LPS)-induced nitric oxide(NO) release from the RAW 264.7 macrophage cells. OBSGM also inhibited the LPS-induced $interleukin-1{\beta}(IL-1{\beta})$ and cyclooxygenase-2 (COX-2) expressions. The inhibitory effects of OBSGM on macrophage activation was via the inhibition of $NF-{\kappa}B$, evidenced by transient transfection assay. Furthermore, OBSGM markedly inhibited the activation of Jun-N-terminal kinase (JNK) and p38 MAP kinase in RAW 264.7 cells. In skin wrinkle formation assay, OBSGM strongly inhibited collagnease and elastase, whose activities are tightly related with the wrinkle formation. In addition, OBSGM inhibited the activities of MMP-1, MMP-2 on the mRNA levels in RAW 264.7 cells. However, OBSGM did not show an inhibitory potential on tyrosinase activity and melanin synthesis, indicating that it could not be applicable for skin whitening. Conclusion : These results suggest that the anti-inflammatory effect of OBSGM may be due to its inhibitory potentials on the macrophage activation. And, the anti-wrinkle effects of OBSGM may be due to its inhibitory potential on the collagnease and elastase activities. Therefore, OBSGM could be applicable for the treatment of mucosa and skin diseases.

  • The purpose of this study was to investigate the effects of Cuscuta chinensis Lamark ethanol extract (CL) on wrinkle improvement. Cuscuta chinensis Lamark is known to contain dried saccharide, alkaloids, flavonoids, lignans and rein glycoside as major components of dried mature seeds of Cuscuta japonica Choisy. In this study, we evaluated the anti-wrinkle effects of CL and investigated bio-markers (e.g ; MMP-1, MMP-3, MMP-9, TIMP-1, type I procollagen) associated with skin wrinkle improvement. We tested the anti-wrinkle effect of CL using human fibroblast called CCD-986Sk cell. We observed an increase in MMPs, TIMP-1, and type 1 pro-collagen CL in CCD-986Sk cells irradiated with UVB at an intensity of $2mJ/cm^2$ for 60 seconds. As a result, CL decreased UVB-induced MMPs levels and mRNA expressions in CCD-986Sk cell. The levels and mRNA expressions of type I procollagen and TIMP-1 were increased by CL. These results suggest that CL has activities on improvement of skin wrinkle, which is induced by UVB radiation. Taken together, this study proposed the possibility of developing herbal medicine and functional herbal cosmetic materials with wrinkle-improving effects of Cuscuta chinensis Lamark.

바나나 잎 추출물의 주름개선 효과 (Antiaging Effects of Musa sapientum L. (Banana) Leaf Extract)

  • To examine the possibility of using Musa sapientum L. (Banana) leaf extract as a cosmetic raw material, banana leaves grown in Jeju Island were extracted with 70% ethanol. Polysaccharides present in banana leaf extract were discarded by precipitation with cold ethanol. Polysaccharide-discarded banana leaf extract promoted procollagen and COL1A1 gene expression, but inhibited matrix metalloproteinase (MMP)-1 and MMP-2 gene expression in human skin fibroblasts when examined by real-time reverse-transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The active compound in banana leaves was identified by fractionation with various solvents. The chloroform fraction showed the highest anti-wrinkle efficacy and the active compound of chloroform fraction was identified as corosolic acid by NMR, FT-IR, EA, and HPLC-MS. In addition, banana leaf extract showed anti-oxidative efficacy with an IC50 value of 67.91 ppm, as determined by DPPH free radical scavenging assay. Finally, the anti-wrinkle efficacy of banana leaf extract-containing cream was confirmed by clinical tests. Based on these results, banana leaves could have an application as a cosmetic raw material with anti-wrinkle efficacy.

호장근추출물이 피부질환 및 피부미용에 미치는 영향 (The Effects of Polygonum Cuspidatum on the Skin Functions)

  • Objectives : This study was carried out to investigate the effects of Polygonum cuspidatum extract on several skin functions including inflammation and wrinkle formation. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe $TNF-{\alpha}$, IL-8 by ELISA Results : Polygonum Cuspidatum extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Polygonum Cuspidatum extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Polygonum Cuspidatum extract could be applicable for improvement of several skin functions.

홍차 흑효모 발효물의 항산화, 항주름 및 미백효과 (Aureobasidium pullulans Ferment of Black Tea for Anti-oxidation, Whitening and Anti-wrinkle Effects)

  • 본 연구에서는 홍차 흑효모 발효물의 기능성화장품 소재로서의 가능성을 규명하고자 항산화, 미백, 항주름 효능을 확인하였다. 홍차 흑효모 발효물의 결과는 홍차 추출물, 흑효모 발효물과 비교하여 나타내었다. 항산화능 평가 결과 홍차 흑효모 발효물은 흑효모 발효물보다 뛰어난 DPPH 라디칼 소거능을 나타냈으며 홍차 추출물과는 유사한 결과를 나타냈다. 과산화수소로 유도된 세포 내 활성산소종 실험에서도 홍차 흑효모 발효물은 유의적 제거 활성을 보였다. 또한 홍차 흑효모 발효물은 tyrosinase 저해 활성을 보이며 Melan-a, B16F10 세포를 이용한 멜라닌 형성이 억제됨을 확인하였다. 항주름 효능 확인을 위해 콜라겐 분해 효소(MMP-1)를 측정한 결과 홍차 흑효모 발효물이 홍차 추출물, 흑효모 발효물보다 높은 항주름 활성을 보였다. 본 연구 결과를 통해 홍차 흑효모 발효물은 항산화능 효능을 보이며 미백, 항주름 효능을 보아 기능성 화장품 소재로 개발될 수 있음을 확인하였다.

해송자 오일의 피부 항노화 및 주름 개선 효과 (Skin Anti-aging and Anti-wrinkle Effects of Pinus koreaiensis Seed Oil)

  • Objectives : The aim of this study was to investigated the anti-aging and anti-wrinkles effects of the pinus koreaiensis seed(PKS) oil. Methods : The anti-oxidant effect was performed by beta-carotene bleaching assay and the intracellular proteome was analyzed expression of each 15 proteins by 2-D electrophoresis. And fatty acid was analysed by gas chromatography. Anti-wrinkle effect was analyzing human skin by the PRIMOS system. Results : Fatty acid analysis of PKS oil has shown oleic acid was 49.7% and linoleic acid was 34.1%. And the antioxidant effect was about 125% compared with alpha-tocoperol(0.1%) by beta carotene bleaching assay. In 2D PAGE analysis, fifteen protein changes in five mechanisms which was collagen synthesis pathway, MMPs, ECM-cell interaction, cytokine, antioxidant enzymes were analyzed. In case of anti-wrinkle effect was proved in vivo by analyzing human skin by the PRIMOS system. The analysis results of eye wrinkles for 4 weeks showed an improvement effect of over 6%. Conclusions : In this study, the amount of protein change in the five mechanism through the cell experiment and the skin anti wrinkle efficacy by the human in vivo test were investigated. As a result pinus koreaiensis seed oil by supercritical extraction could be used as a anti-aging and anti-wrinkle substance for the skin.

오미자 유산균 발효물의 주름개선 효과 (Anti-Wrinkle Effect of Schizandra chinensis Baillon Fermented with Lactobacillus plantarum)

  • 새로운 주름개선제 성분을 찾기 위해서 본 연구에서는 사람 피부 섬유아세포의 세포독성, 콜라겐 생합성, matrix metalloproteinase-I (MMP-1) 및 elastase 저해활성에 대한 Lactobacillus plantarum으로 발효된 오미자 발효액의 주름개선 효과를 평가하였다. 먼저, 오미자 추출물은 L. rhamnosus으로 $37^{\circ}C$에서 1일 동안 발효하였다. 발효물의 세포독성은 cytopathic effect reduction 방법에 의해 평가하였다. 콜라겐 생합성에 대한 발효물의 영향은 procollagen type-IC peptide EIA kit에 의해 평가하였으며, matrix metalloproteinase-I(MMP-1)에 대한 발효물의 영향은 Matrix Metalloproteinase-1 Biotrack activity Assay Kit에 의해 평가하였다. Elastase inhibition assay는 기질로써 N-Suc-$(Ala)_3$-nitroanilide을 사용하여 기질 반응에 의해 평가하였다. 결과로써 오미자 발효물은 사람 피부 섬유아 세포에 대해 $100{\mu}g/mL$의 농도에서 세포독성을 나타내지 않았다. 또한 오미자 발효물은 콜라겐 생합성을 촉진시켰으며, MMP-1의 저해 효과를 나타내었다(p < 0.05). Elastase inhibition assay에서 오미자 발효물의 $IC_{50}$$36.4{\mu}g/mL$이었다. 그러므로 본 연구에서 오미자 발효물은 주름개선 효과를 보유하고 있으며, 이것은 피부의 주름개선을 위해 사용가능하리라 사료된다.
