Therapeutic effect of a Kampo medicine, Choto-san, in patients with vascular dementia was demonstrated by a double-blind and placebo-controlled clinical trial. To clarify the therapeutic efficacy of Choto-san, anti-ischemic effect in mice, hypotensive effect in spontaneously hypertensive rats (SHR), anti-oxidative effects in vitro, and N-methyl-D-aspartate (NMDA) receptor-blocking activity using Xenopus oocytes were studied. (1) Pretreatment with Choto-san (0.75-6.O g/kg, P.O.) or a component herb Chotoko (Uncaria genus: 75 - 600 mg/kg, P.O.) prevented ischemia-induced impairment of spatial learning behaviour in mice. Indole alkaloids- and phenolic fractions extracted from Chotoko also improved significantly the learning deficit. (2) Subchronic administration of Choto-san (0.5 g/kg, p.o.) caused a significant hypotensive effects in SHR. (3) Choto-san, Chotoko, and the phenolic constituent, (-) epicatechin, significantly protected the NG108-15 cell injury induced by $H_20_2$ exposure in vitro and also inhibited lipid peroxidation in the brain homogenate. (4) Indole alkaloids, rhynchophylline and isorhynchophylline (1-100 uM), reversibly reduced NMDA-induced current in the receptor-expressed Xenopus oocytes. These results suggest that anti-vascular dementia effects of Choto-san are mainly due to the effect of Chotoko. From these results, it is possible to make a novel dietary supplement through several extraction steps from Chotoko.
Skin flap necrosis remains a major complication of reconstructive surgery. Euterpe oleracea Mart., popularly known as "acai berry" contains hydroxybenzoic acid, antioxidant polyphenolics and anthocyanins. These and other compounds within the acai berry confer anti-inflammatory and anti-oxidative effects. In this current study, we evaluated the protective effect of acai berry extracts on survival of random-pattern skin flaps in a murine model by histologic analysis. ICR mice were subjected to skin elevation surgery and orally administered acai berry extract (100 mg/kg) daily for 7 days. Tissues were stained with hematoxylin-eosin or Masson's trichrome to observe tissue integrity and collagen deposition. In addition, $TGF-{\beta}$ and VEGF was stained by immunofluorescence to determine anti-inflammatory cell infiltration and neovascularization, respectively. We found a decrease in inflammatory cell infiltration and increase in collagen deposition in the acai berry extract treated mice compared to control mice. Immunofluorescence staining reveal a higher number of $TGF-{\beta}$ positive cells and enhanced VEGF staining in the acai berry extract treated mice. The results from this study indicate that oral uptake of acai berry extract can promote healing and survival of surgical skin flaps in mice providing an augmentative therapeutic approach to enhancing skin flap survival.
백삼 및 백삼에 함유된 페놀성 성분 6종의 산화적 손상에 대한 신경세포보호효과와 LPS로 유도된 대식세포에서의 NO 생성억제효과를 검정한 결과 백삼 추출물은 두가지 모두에서 농도의존적인 효과를 나타내었다. 백삼에 함유된 페놀성 성분 중에는 cinnamic acid 및 maltol이 우수한 신경세포보호활성을 나타내었고 NO 생성억제효과는 ascorbic acid 및 cinnamic acid가 높았다. 또한 페놀성성분의 백삼내 함량을 정량한 결과 ascorbic acid, cinnamic acid, esculetin가 다른 성분들에 비하여 비교적 많이 함유되어 있었으며 이를 종합하여 볼 때 백삼의 신경보호효과 및 항염증효과에는 높은 함량 및 우수한 활성을 보이는 성분인 cinnamic aicd가 크게 기여하는 것으로 판단된다.
다양한 원인에 의하여 발생하는 미세먼지(particulate matter, PM)는 강력한 환경 오염 물질로 대두되고 있다. PM은 서로 다른 구성과 크기의 고체 입자와 액체 방울로 구성되며, 유해 화학 성분에는 원소 및 유기 탄소, 유기 화합물, 생물학적 화합물 및 금속이 포함된다. 급성 및 만성 PM 노출 시 생체 내 생리학적 시스템에 의하여 유입되고 축적되어 세포내 소포체 스트레스, 미토콘드리아 기능 장애, 산화 스트레스, 염증, 지질 축적 및 세포 주기 정지와 함께 세포 구조 변화를 촉진한다. 궁극적으로 이러한 세포 반응은 노화의 주요 특성을 발달시키는 원인으로 작용하며, 세포 노화와 관련된 자가포식 플럭스 및 리소좀 기능 장애를 향상시킨다. 선행 연구에서 PM과 사망률 증가 또는 수명 감소 사이의 긍정적인 연관성을 강조했지만, 노화에 대한 PM의 직접적인 증거는 여전히 제한적이다. 이 총설에서는 관찰 연구뿐만 아니라 노화 진행 및 연령 관련 질병 발병에 대한 PM의 시험관 내 및 생체 내 증거를 평가하였다. 이러한 평가는 소포체 스트레스, 미토콘드리아 기능 장애, 산화 스트레스, 염증, 지방 축적, 자가포식을 포함하여 PM 노출과 노화 사이의 연관성을 강화하는 연령 관련 세포 변화를 기반으로 한다. 이러한 자료는 PM에 따른 기본적인 세포 반응의 이해를 토대로 PM으로 인한 노화에 대한 새로운 치료법 개발에 기여할 수 있을 것이다.
Background: Oxidative stress plays an important role in neuro-degenerative disorders such as Alzheimer's disease. Oxidative stress is mediated by reactive oxygen species (ROS), which are implicated in the pathogenesis of numerous diseases, and account for the toxicity of a wide range of compounds. Methods and Results: In order to study the neuro-protective effect of the complex extracts of Aster scaber Thunberg (AS) and Chaenoleles sinensis Koehne (CSK) against hydrogen peroxide in PC12 cells, cell viability was evaluated by the MTT assay using tetrazole, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and the intracellular ROS levels were determined the by 2',7'-dichlorofluorescein diacetate (DCF-DA) assay. In order to examine the anti-amnesic effects of the complex extracts of AS and CSK, behavioral tests were performed on male ICR mice. The ameliorating effect of the complex extracts against Aβ1-42-induced learning and memory impairment was analyzed by y-maze and passive avoidance tests. The AS and CSK extracts showed neuro-protective activity both in vitro and in vivo, and the neuro-protective effect of their 60 : 40 (AS : CSK) mixture was better than that of the other mixtures. Moreover, the complex extracts synergistically inhibited acetylcholinesterase activity and rapid peroxidation. Conclusions: A mixture of the AS and CSK extracts could be used to develop functional foods and serve as raw materials for the development of therapeutics against Alzheimer's disease.
Objectives : Evodia officinalis DODE (EO), an herbal plant, has been widely used in traditional Korean medicine for the treatment of vascular diseases such as hypertension. The crude extract of EO contains phenolic compounds that are effective in protecting liver microsomes, hepatocytes, and erythrocytes against oxidative damage. But EO has been little found to have an anti-inflammatory activity. We investigated anti-inflammatory activity of EO in RAW 264.7 cells and human umbilical vein endothelial cells (HUVECs). Methods : Cytotoxic activity of EO on RAW 264.7 cells was investigated by using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines were measured by ELISA kit. The levels of intracellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) expression were measured by flow cytometer. Results : EO decreased LPS-induced NO production in RAW 264.7 cells. The inhibitory activity of EO on LPS-induced NO release is probably associated with suppressing TNF-${\alpha}$, IL-6 and MCP-1 formation. These results indicate that EO has potential as an anti-inflammatory agent. Moreover, EO decreased TNF-${\alpha}$-induced IL-8, IL-6 production, and ICAM-1 and VCAM-1 expression in HUVECs. Conclusions : EO inhibits TNF-${\alpha}$-induced inflammation via decreasing cytokines production and adhesion molecules expression. These results indicate that EO has potential as an anti-inflammation and anti-artherosclerosis agent.
본 연구는 TAA 복강투여로 유발된 간 손상 동물모델에서 인진호 열수 추출물의 간보호 효능을 평가하였으며 다음과 같은 결론을 얻었다. TAA로 인해 줄어드는 체중은 인진호 열수 추출물을 투여한 군에서 유의하게 증가하였으며, 간손상에 의해 증가한 혈중 암모니아 함량과 MPO 활성은 인진호 열수 추출물 투여군에서 유의하게 감소하였다. 간 조직의 western blotting 결과, 인진호 열수 추출물 투여가 산화적 스트레스 관련 인자들의 발현을 유의적으로 감소시키고, 항산화 관련 인자들의 발현을 유의하게 증가시켰으며, MMPs의 발현은 감소시키고 TIMP-1의 발현은 증가시킴을 확인할 수 있었다. 따라서 인진호 열수 추출물은 TAA로 유발된 간손상 동물모델에서 항산화 작용을 통해 산화적 스트레스를 억제하여 간보호 효과를 보이는 것으로 판단된다.
Elicitor를 처리한 삼백초의 물과 ethanol 추출물들의 기능성화장품활성을 살펴보았다. Elicitor 처리한 삼백초 추출물의 항산화 효과는 1.5 mg elicitor/leaf 1 g (group A)에서 3.0 mg elicitor/leaf 1 g (group B)으로 처리 농도가 높아질수록 항산화 효과는 높아지는 것으로 확인되었으며, elicitor 처리에 의해 물 추출물보다 70% ethanol 추출물의 항산화 효과가 더 우수한 효과를 나타내었다. Elicitor 처리한 삼백초 추출물의 미용식품활성은 elastase 억제 효과와 미백효과가 비처리군인 대조구와 유사한 효소 억제 효과를 나타내어 elicitor 처리가 삼백초 추출물의 미백 및 주름개선 효과의 증대에는 영향을 미치지 않는 것으로 확인되었으나, 염증 억제 효과가 비처리군인 대조구에 비해 우수한 염증 억제 효과를 나타내었으며, grpoup A와 group B 모두에서 대조구에 비해 억제율이 큰 폭으로 상승하는 효과를 나타내어 elicitor 처리가 유용물질의 산업화라는 전제를 충족시키기 위한 biomass 기법으로 매우 적당하다는 것을 시사해주었다. 따라서 삼백초 재배 시에 본 연구에서 사용한 elicitor를 처리하면 수확되는 삼백초의 단위 무게 당 생산량을 높일 수 있고, 생리활성도 증대시킬 수가 있어 산업화를 위한 원료생산에 효율적인 재배기법에 활용할 수 있을 것이라 판단되었다.
초미세 분쇄를 하였을 때 입자가 작아질수록 추출수율이 높아져 약 2.5배 높은 추출수율을 나타내었다. 일반 분쇄한 시료 추출물에서 69.8%의 전자공여능 억제효과가 관찰되었고, 미세분쇄와 초미세분쇄 추출물에서는 각각 70.7과 83.8%의 억제효과를 나타내었다. 일반분쇄 추출물과 미세분쇄 및 초미세분쇄 후 추출물 모두 97% 이상의 높은 ABTS 억제효과를 나타내어 분쇄 방법에 따른 항산화력의 차이는 거의 없었다. 일반 분쇄한 시료 추출물 보다 미세분쇄와 초미세분쇄 추출물에서 더 높은 PF값을 확인하였으며, 50% ethanol 초미세분쇄 추출물에서 1.8 PF로 가장 높은 항산화력을 나타내었다. 미세분쇄와 초미세분쇄 추출물에서는 일반분쇄 추출물에 비해 입자크기가 작아질수록 TBARS 억제율이 높아지며, 물 추출물보다 ethanol 추출물의 효과가 더 우수한 것으로 확인되었다. Xanthin oxidase 저해의 경우 초미세분쇄 후 효소억제 증대 효과를 확인할 수 있었다. Angiotensin converting enzyme 억제활성은 일반분쇄 추출의 경우 물 추출물에서는 억제활성이 나타나지 않았고, 50% ethanol 추출물에서 24%의 억제율이 확인되었다. 또한, ethanol 추출물의 억제효과가 물 추출물에 비해 상대적으로 우수하였다. 50% ethanol 초미세분쇄 추출물에서 Staphylococcus aureus, Escherichia coli에 대해서 아주 약한 항균효과를 나타내었을 뿐 나머지 추출물에서의 항균효과는 거의 관찰되지 않았다.

The extract of white rose petals has an antioxidant effect and can be used to treat allergic disease. The purpose of this study was to identify optimal conditions for extracting antioxidative compounds from white rose petals with 2,2-diphenyl-1-picrylhydrazyl scavenging activities. A response surface methodology based on a central composite design was used to investigate the effects of three independent variables: ethanol concentration ($X_1$), extraction temperature ($X_2$), and extraction time ($X_3$). The estimated optimal conditions for obtaining phenolic compounds with antioxidant activities were as follows: ethanol concentration of 42% ($X_1$), extraction time of 80 min ($X_3$), and extraction temperature of $75^{\circ}C$ ($X_2$). The estimated optimal conditions for obtaining flavonoid compounds with antioxidant effects were an ethanol concentration of 41% ($X_1$), extraction time of 119 min ($X_3$), and an extraction temperature of $75^{\circ}C$ ($X_2$). Under these conditions, predicted response values for the phenolic and flavonoid contents were 243.5 mg gallic acid equivalent/g dry mass and 19.93 mg catechin equivalent (CE)/g dry mass, respectively.
