Screening of Medicinal Plants on the Anti-inflammatory Effect and Active Component (전남지역 자생식물들의 소염활성검색 및 활성화합물)

  • The anti-inflammatory effect of the methanolic extracts of some medicinal plants was investigated on the hydrolysis of N-benzoylarginine-p-nitroanilide by trypsin in vitro, the writhing syndrome induced by acetic acid in mice, and the carrageenin-induced paw edema in rats. Among tested medicinal plants, the methanoilc extract of Angelica keiskei, Rosa rugosa and Cudrania tricuspidata showed significant inhibitory effect. And we also found that adenosine, isolated from A. keiskei was identified as one active components.

  • Objectives : We review a result of studies until the present and suggest the method of medical treatment for the clinical treatment of Scolopendrid Herbalacupuncture. Methods : We analysis the paper of the bibliographic studies, the experiment studies and the clinical studies from 2001 developed Scolopendrid Herbalacupuncture and grope for the course of studies. Results : 1. Scolopendrid Herbalacupuncture is proved the clinical safety by the aninmal and human tests. 2. The pharmacological action of Scolopendra subspinipes mutilans L. Koch is anti-convulsive action, analgesic action, lowering blood pressure, anti-inflammatory action, anti-tumor action and microbe inhibition 3. Scolopendrid Herbalacupuncture has been a fine effect to the entrapment neuropathy and inflammatory. 4. Scolopendrid Herbalacupuncture was thought effective on a acute phase and to the excessive symptoms. The Sub-chronic toxicity experiment observing the response after hypodermic medication over 90 days, The Genetic-mutagenic toxity experiment and the clinical effect studies are necessary.

Synthesis of Mefenamic Acid Derivatives and Antioxidative and Anticoagulant Activities

  • Mefenamic acid has been widely used as clinical drug for anti-inflammatory and analgesic. This drug was known to non-steroidal anti-inflammatory drugs (NSAIDs) such as aspirin, ibuprofen and indomethacin. Although the drugs which comprise this group are of diverse chemical structures, they all share the antipyretic, analgesic and anti-inflammatory actions which are characteristic of aspirin. Action of this drugs is caused by inhibitory effect of biosynthesis of prostaglandin that are synthesized from arachidonic acid via the endoperoxide biosynthesis pathway, the initial step of which is catalysed by cyclooxygenase. Mefenamic acid has more potent inhibitory action of prostaglandin biosynthesis than aspirin. Therefore, mefenamic acid is expected to have anticoagulant activity as aspirin-like drugs. This study was carried out to investigate the sinthesis of mefenamic acid derivatives from mefenamic acid and aromatic compound of antioxidant and its antioxidative and anticoagulant activities. Synthesis of mefenamic acid derivatives was conformed by conjugation as using esterification method. Biological activities was examined using effect of anticoagulant on bleeding time and effect of antioxidant by TBA method. As a result, SJ-202 showed strong antioxidative activity and anticoagulant activity among tested 4 compounds and exhibited similar activity to aspirin at anticoagulant activity.

Pharmacological Studies on Ether Fraction of Corni Fructus (산수유 에텔분획물의 약리작용에 관한 연구)

  • The ether fraction obtained from dry fruit of Cornus officinalis was investigated for the anti-inflammatory action, acute toxicity and central nervous system activities. From several pharmacological examinations, it was found that the ether fraction suppressed considerably carrageenin edema at the dose of 600mg/kg p.o. in rats, inhibited the granuloma formation in rats as given 100mg/kg p.o. for 7 days and decreased the swelling of both of complete adjuvant injected and noninjected (contralateral) paws of the rats at the dose of 100mg/kg p.o. given for 14 days. The $LD_{50}$ of the fraction are estimated to be more than 2,000mg/kg p. o. and 642mg/kg i. p. in mice. And the fraction did not show any sedative, stimulative, analgesic and anticonvulsant action but exhibited hypothermic action. These results might be concluded that the ether fraction of Corni fructus showed anti-inflammatory action in both of acute and chronic type models without any considerable central nervous depressant activity and exhibited very weak acute toxicity in mice.

  • Experimental studies were conducted to investigate for the effect of Mahaengkamsuk-tang on analgesic, antipyretic, antiinflammatory, secretion of respiratory tract, isolated ileum. Mahaengkamsuk-tang was composed of Ephedrae Herba, Armenicae Semen, Glycyrrhizae Radix, Gypsum Fibrosum. The following results has been obtained; analgesic and antipyretic actions were obtained. Anti-inflammatory effects in the paw edema induced by 1% dextran was significantly shown in rats. Spontaneous motilities of isolated ileum of mice were strongly suppressed, and contractions of isolated ileum of mice induced by acetylcholine chloride, barium chloride and histamine were remarkably inhibited. Expectorant effect was shown in rabbits. Continuous hypotensive action was seen.

  • Ulmi Cortex has been used for edema, articular rhematism and acne, etc in oriental medicine. Here, experimentally the MeOH extract showed significant anti-inflammatory action, inhibitory effect of leukocyte emigration in rats and the growth inhibition of Sta. aureus, Strep. faecalis, Psu. aeruginosa and Bacillus sp. The ether layer showed analgesic effect stronger than that of MeOH extract and BuOH layer showed antimicrobial activity stronger than that of the other layer.

Studies on the Pharmacological Actions of Cactus:Identification of Its anti-Inflammatory Effect

  • The ethanol extracts of Opuntia ficus-indica fructus (EEOF) and Opuntia ficus-indica stem (EEOS) were prepared and used to evaluate the pharmacological effects of cactus. Both the extracts inhibited the writhing syndrome induced by acetic acid, indicating that they contains analgesic effect. The oral administrations of EEOF and EEOS suppressed carrageenan-induced rat paw edema and also showed potent inhibition in the leukocyte migration of CMC-pouch model in rats. Moreover, the extracts suppressed the release of $\beta$-glucuronidase, a lysosomal enzyme in rat neutrophils. It was also noted that the extracts showed the protective effect on gastric mucosal layers. From the results it is suggested that the cactus extracts contain anti-inflammatory action having protective effect against gastric lesions.

  • This study was conducted about the effect of Shihogesikungang-tang on the central nervous system and cardiovascular system for the investigation of its clinical effect based on the Oriental medicinal references. The results of this study were summerized as follows; Analgesic activity as evaluated by the writhing syndrome in mice was significantly noted. A decrease effect of the spontaneous movement as estimated by wheel cage method, muscle relaxant effect as evaluated by the rotor rod method and the prolonged effect of sleeping time induced by thiopental-Na were significantly shown in mice. A antipyretic activity in febrile rats induced by the endotoxin was recognized. Anti-inflammatory effect in carrageen-induced paw edema in rats was significantly noted. Negative inotropic action on the isolated heart of frogs was noted. A vasodilative action in rabbits peripheral blood vessels and hypotension in anesthetized rabbits were remarkably recognized.

  • Hyunggyeyungyotang has been used for treatment of sinusitis and otitis media in oriental medicine since ancient times. It is reported that Hyunggyeyungyotang has good effects on inflammatory and allergic diseases of otorhinolaryngology in clinical medicine. Kamihyunggyeyungyotang was made by adding several herbs to Hyunggyeyungyotang which has such good effects. To investigate the effects of Hyunggyeyungyotang and Kamihyunggyeyungyotang on inflammatory, algesic and allergic diseases, the author examined the analgesic effect by acetic acid reaction, studied the anti-inflammatory effect through the experiments of the protein thermo-denaturation and circumscribed edema. Besides researched the anti-allergic effect through the vascular permeability response to Chemical Mediator and the delayed type hypersensitivity response to Picryl Chloride. The obtained results were as follows ; 1. In the analgesic effect of Hyunggyeyungyotang and Kamihyunggyeyungyotang extract by acetic acid method, both of the sample groups showed the analgesia, but didn't show useful effect. 2. In the anti-inflammatory effect on the protein thermo-denaturation, the sample groups revealed the inhibitory effect in proportion to concentration as compared with the control group. 3. In the inhibitory action on circumscribed edema induced by Caraggeenin, both of Hyunggyeyungyotang and Kamihyunggyeyungyotang administration showed the significant effect after 4 hours in comparison to the control group. 4. In the delayed type hypersensitivity response to Picryl Chloride, both of the sample groups revealed the significant effects. 5. Both of the sample groups decreased the vascular permeability induced by Histamine in comparison with the control group, but the significancy was admitted in only Hyunggyeyungyotang administration. According to above results, Hyunggyeyungyotang and Kamihyunggyeyungyotang are considered to be used for treament of the inflammatory diseases including sinusitis.

Analgesic and anti-inflammatory effects of galangin: a potential pathway to inhibit transient receptor potential vanilloid 1 receptor activation

  • Background: Galangin, commonly employed in traditional Chinese medicine for its diverse medicinal properties, exhibits potential in treating inflammatory pain. Nevertheless, its mechanism of action remains unclear. Methods: Mice were randomly divided into 4 groups for 7 days: a normal control group, a galangin-treated (25 and 50 mg/kg), and a positive control celecoxib (20 mg/kg). Analgesic and anti-inflammatory effects were evaluated using a hot plate test, acetic acid-induced writhing test, acetic acid-induced vascular permeability test, formalin-induced paw licking test, and carrageenan-induced paw swelling test. The interplay between galangin, transient receptor potential vanilloid 1 (TRPV1), NF-κB, COX-2, and TNF-α proteins was evaluated via molecular docking. COX-2, PGE2, IL-1β, IL-6, and TNF-α levels in serum were measured using ELISA after capsaicin administration (200 nmol/L). TRPV1 expression in the dorsal root ganglion was analyzed by Western blot. The quantities of substance P (SP) and calcitonin gene-related peptide (CGRP) were assessed using qPCR. Results: Galangin reduced hot plate-induced licking latency, acetic acid-induced contortions, carrageenan-triggered foot inflammation, and capillary permeability in mice. It exhibited favorable affinity towards TRPV1, NF-κB, COX-2, and TNF-α, resulting in decreased levels of COX-2, PGE2, IL-1β, IL-6, and TNF-α in serum following capsaicin stimulation. Galangin effectively suppressed the upregulation of TRPV1 protein and associated receptor neuropeptides CGRP and SP mRNA, while concurrently inhibiting the expression of NF-κB, TNF-α, COX-2, and PGE2 mRNA. Conclusions: Galangin exerts its anti-inflammatory pain effects by inhibiting TRPV1 activation and regulating COX-2, NF-κB/TNF-α expression, providing evidence for the use of galangin in the management of inflammatory pain.
