Anti-stress effect of Hervchoke juice in ICR mice and SD rats

  • Herbchoke solution produced by Choa pharmaceutical company contained pyroligneous liquid and extracts from artichoke leaves. Pyroligneous liquid is produced by process carbonizing Oak in 350∼400$^{\circ}C$. It contain water 96%, acetic acid 3% and organic compound 1%. There are 200 kinds of constituents including minerals, vitamin B-complex and organic acids in it. The organic acids of them were presumed as active materials. It is traditionally used for treatment of stress related disorder, hepatic disease, immune disorder, G-I disorder and inflammatory disease. Extracts from artichoke leaves are traditionally used for treatment of stress related disorder, that is, hepatic disease, dyspeptic disorder, hyperlipidemic disorder and diuretic disorder. The aim of this study was to investigate anti-stress effects of Hervchoke juice produced from Choa pharmaceutical company.

  • Zingiber officinale is a spice which is having a strong historical medicinal background and used by different systems of medicine for various ailments. It is also used to maintain the characteristic sharpness and tangy essence of food and beverages and used in carbonated drinks, as a conserve in sugar syrup. Ginger considered as an incredible drug for inflammation, gastrointestinal problems, pain and sexual problems since centuries. There are many studies carried out in recent years, which proves that the active constituents of ginger shows a variety of pharmacological effects like antiemetic, hepatoprotective, anti hyperlipidemic and antibacterial. The most momentous among all the nutraceutical aspects of ginger are its positive control on gastrointestinal tract including digestive stimulant action, anticancer effect and anti-inflammatory effect.

  • High-dose extracts from artichoke leaves are traditionally used for treatment of stress related disorder, that is, hepatic disease, dyspeptic disorder, hyperlipidemic disorder and diuretic disorder. The aim of this study was to investigate anti-stress effects of Artichoke extract(Archoke juice produced from Choa company). The experiments were performed with the use of young(6-8 weeks of age) male mice of ICR stain weighing between 20 and 25 g at the time of first treatment with Archoke juice. The were grouped normal, control, Ginseng, diazepam and Arcoke juice group. (omitted)

  • Objectives :Ginkgo-Chunghyul-dan (GCHD) is newly developed herbal medicine to prevent and treat stroke. In this study, we investigated whether the GCHD had antioxidant activity and anti-platelet aggregation effect in vitro and hypolipidemic activities in vivo. Methods :Anti-oxidant activity of GCHD was measured using the Blois method, anti-platelet effect of GCHD was assessed by the Born method, and hypolipidemic activities of GCHD were evaluated in corn oil- or Triton WR-1339-induced and cholesterol-fed rats. Results :GCHD showed anti-oxidant activity in the study inhibiting the formation of 1-diphenyl-2-picrylhydrazyl radicals and xanthine oxidase activity. GCHD had anti-platelet aggregation activity. GCHD significantly lowered total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) in high cholesterol diet and Triton WR-1339 induced model TG in corn oil-induced model. GCHD had no acute toxicity at a single dosage. Conclusion : These results suggest that GCHD has the potential to treat hyperlipidemia and stroke.

Beneficial effects of natural Jeju groundwaters on lipid metabolism in high-fat diet-induced hyperlipidemic rats

  • BACKGROUND: Groundwater is believed to possess many beneficial effects due to its natural source of various minerals. In this study, we examined the effects of natural Jeju groundwater S1 (Samdasoo$^{TM}$), S2 and S3 pumped up from different locations of Jeju Island, Korea, along with local tap water, on body weight gain, serum lipids and lipoproteins, and liver histopathology in high-fat diet-induced hyperlipidemic rats. MATERIALS/METHODS: Rats were randomly and equally divided into 6 groups. Different water samples were supplied to the hyperlipidemic rats as their daily drinking water and the widely-used anti-hyperlipidemic drug simvastatin was used as a positive control. Body weight, serum lipids and lipoproteins were measured weekly. Liver weight, liver index and liver histopathology were examined after the execution of the rats. RESULTS: After drinking Jeju groundwaters for two months, S2 but not S3 significantly reduced weight growth and serum triglycerides levels and increased high density lipoprotein-C (HDL-C) without affecting total cholesterol or LDL-C. S1 and particularly S2 significantly reduced the severity of liver hypertrophy and steatosis. All Groundwaters had much higher contents of vanadium (S3>S2>S1>>tap water) whereas S1 and S2 but not S3 markedly blocked autoxidation of ferrous ions. CONCLUSION: Jeju Groundwater S1 and particularly S2 exhibit protective effects against hyperlipidemia and fatty liver and hypothesize that the beneficial effect of Jeju Groundwaters may be contributed from blockade of autoxidation of ferrous ions rather than their high contents of vanadium.

Salicornia herbacea Prevents High Fat Diet-Induced Hyperglycemia and Hyperlipidemia in ICR Mice

  • Salicornia herbacea L. (Chenopodiaceae) has been used as a seasoned vegetable by living in coastal areas. S. herbacea (SH) has been demonstrated to stimulate cytokine production, nitric oxide release, and to show anti-oxidative effect. In a series of investigations to develop potential anti-diabetic and/or anti-hyperlipidemic agents from Korean indigenous plants, 50% ethanol extract of Salicornia herbacea was found to prevent the onset of the hyperglycemia and hyperlipidemia induced by high fat diet in ICR mice. At 6 week old, the ICR mice were randomly divided into five groups; two control and three treatment groups. The control mice were to receive either a regular diet (RD) or high-fat diet (HFD), and the treatment groups were fed a high fat diet with either 350 mg/kg, 700 mg/kg of SH (SH350 and SH700) or 250 mg/kg of met-formin (MT250) for a 10-week period. SH not only reduced body weight but also corrected associated hyperglycemia and hyperlipidemia in a dose dependent manner. SH exerted beneficial effects on the plasma glucose and lipid homeostasis possibly ascribed to its specific effects on lipogenesis related genes (SREBP1a, FAS, GAPT), and PEPCK, glucose 6-phosphatase gene expressions in liver. Ethanol extract of S. herbacea has potential as a preventive agent for type 2 diabetes (and possibly hyperlipidemia) and deserves future clinical trial.

감국약침이 과산화지질을 급여한 흰쥐의 지질대사, 항산화 및 면역계에 미치는 영향 (Effects of gamgook(Chrysanthemum indicum L.) herbal-acupuncture on lipid lowering effect, anti-oxidative capacity and anti-inflammatory effect in rat fed high oxidized fat)

  • Objectives: To investigate the effects of Chrysanthemum indicum L. pharmacopuncture on lipids, antioxidant capacity and anti-inflammation in rats fed high-fat diet. Methods: Hyperlipidemic rats induced by high-fat diet were divided into 5 groups: no treatment control (normal, n=8), high-fat diet only control (control, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV4 group (TI, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV17 group (TII, n=8), and high-fat diet and Chrysanthemum indicum L. pharmacopuncture at EX-HN3 group (TIII, n=8). They were given pharmacopuncture accordingly every other day for two weeks followed by analyses of lowering lipids effects, oxidative capacity and anti-inflammatory effects. Results: Compared with the control, pharmacopuncture groups showed significantly decreased plasma total cholesterol (TC), liver thiobarbituric acid reactive substance (TBARS), catalase, glutathione peroxidase, neutrophils, monocytes, plasma and liver IL-$1{\beta}$, and plasma and liver IL-6. In other parameters including plasma and liver triglyceride, liver TC, LDL-cholesterol, HDL-cholesterol, liver TBARS, supraoxide dismutase, total protein, albumin, blood cell analysis, plasma and liver TNF-$\alpha$, and IL-10, there was no significant difference between control and pharmacopuncture groups. No clear acupoint-specificity was observed. Conclusions: Chrysanthemum indicum L. pharmacopuncture may improve control of hyperlipidemia.

Marein Prevented LPS-Induced Osteoclastogenesis by Regulating the NF-κB Pathway In Vitro

  • Many bone diseases such as osteolysis, osteomyelitis, and septic arthritis are caused by gram-negative bacterial infection, and lipopolysaccharide (LPS), a bacterial product, plays an essential role in this process. Drugs that inhibit LPS-induced osteoclastogenesis are urgently needed to prevent bone destruction in infective bone diseases. Marein, a major bioactive compound of Coreopsis tinctoria, possesses anti-oxidative, anti-inflammatory, anti-hypertensive, anti-hyperlipidemic, and anti-diabetic effects. In this study, we measured the effect of marein on RAW264.7 cells by CCK-8 assay and used TRAP staining to determine osteoclastogenesis. The levels of osteoclast-related genes and NF-κB-related proteins were then analyzed by western blot, and the levels of pro-inflammatory cytokines were quantified by ELISA. Our results showed that marein inhibited LPS-induced osteoclast formation by osteoclast precursor RAW264.7 cells. The effect of marein was related to its inhibitory function on expressions of pro-inflammatory cytokines and osteoclast-related genes containing RANK, TRAF6, MMP-9, CK, and CAII. Additionally, marein leads to markedly inhibited NF-κB signaling pathway activation in LPS-induced RAW264.7 cells. Concurrently, when the NF-κB signaling pathway was inhibited, osteoclast formation and pro-inflammatory cytokine expression were decreased. Collectively, marein could inhibit LPS-induced osteoclast formation in RAW264.7 cells via regulating the NF-κB signaling pathway. Our data demonstrate that marein might be a potential drug for bacteria-induced bone destruction disease. Our findings provide new insights into LPS-induced bone disease.

고지방과 고콜레스테롤 식이 급여에 의해 고지혈증이 유도된 흰쥐에서 꽃송이버섯 자실체의 항고지혈증과 항비만 효과 (Anti-hyperlipidemic and anti-obesity effects of Sparassis latifolia fruiting bodies in high-fat and cholesterol-diet-induced hyperlipidemic rats)

  • 꽃송이버섯의 자실체가 고지방과 고콜레스테롤을 급여한 Sprague-Dawley계 암컷 흰쥐의 지질대사와 비만에 미치는 효과를 조사하기 위해 생후 6주령의 흰쥐에 정상 식이를 급여한 군(ND), 정상 식이에 20%의 돈지(Lard)와 1%의 콜레스테롤을 첨가하여 급여한 군(HFD), HFD 식이에 꽃송이버섯 자실체 분말을 5% 수준으로 첨가하여 급여한 군(HFD+SL), HFD 식이에 simvastatin을 0.3% 수준으로 첨가하여 급여한 군(HFD+SS) 등 4개의 실험군으로 나누어 6주간 실험을 진행하였다. 각각의 실험군 간 체중의 증가율은 HFD+SL군과 HFD+SS군이 HFD군에 비해 유의하게 감소하여 ND군와 유사한 수준을 나타내 HFD 식이에 꽃송이버섯을 첨가하여 급여한 HFD+SL군의 비만이 억제되었다. ND군의 식이섭취량은 HFD군에 비해 유의하게 높았고 HFD+SL군과 HFD+SS군에 비해서는 고도로 유의하게 높았다. 혈청 총콜레스테롤(TC), 저밀도지단백질 콜레스테롤(LDL-C) 및 중성지방(TG)의 농도는 HFD군에 비해 ND군, HFD+SL군 및 HFD+SS군이 유의하게 낮았다. 심혈관계 질환의 위험도를 나타내는 동맥경화지수(AI)와 LDL-C/HDL-C 비율도 HFD군에 비해 ND군, HFD+SL군 및 HFD+SS군이 유의하게 낮아 꽃송이버섯 자실체의 급여가 심혈관계 질환의 발생 위험도를 낮출 수 있은 것으로 나타났다. 알부민, 크레아티닌, 요산 및 총단백질의 농도는 모든 실험군 간 정상적인 수치를 나타냈으나 혈당 농도는 HFD군에 비해 ND군, HFD+SL군, HFD+SS군 모두 유의하게 낮아 꽃송이버섯이 혈당을 낮추는 효과가 있었다. 간 기능 손상의 척도를 나타내는 AST와 ALT 효소의 활성은 모든 실험군 간 유의성은 없었으나 ALP 효소의 농도는 HFD에 비해 ND군와 HFD+SL군 및 HFD+SS군이 유의하게 낮아 꽃송이버섯을 첨가한 식이의 급여가 흰쥐의 ALP 농도를 유의하게 낮추는 효과가 있었다. 실험 5주와 6주 사이에 배출된 분변의 총 지질과 콜레스테롤의 양을 측정한 결과 HFD+SL군이 배출한 총 지질과 콜레스테롤의 양이 HFD군과 HFD+SS군에 비해 유의하게 높게 나타나 HFD식이에 함유된 일부의 지질과 콜레스테롤이 꽃송이버섯에 의해 장내로 흡수되지 않고 분변으로 배출된 것으로 나타났다. 각각의 실험군의 간 조직을 적출하여 oil red O로 염색하고 현미경으로 관찰한 결과 HFD군에서는 심한 지방간이 관찰되었으나 HFD+SS군은 ND군과 같이 지방간이 전혀 관찰되지 않았고 HFD+SL군의 간 조직에서는 소량의 지방 방울이 관찰되어 꽃송이버섯의 자실체에는 비알코올성지방간을 억제하는 효과가 있는 것으로 나타났다. 따라서 꽃송이버섯 자실체는 고지방과 고콜레스테롤 식이 섭취로 인해 발생하는 체중증가, 혈청의 콜레스테롤과 중성지방농도 및 비알코올성 지방간의 개선에 효과가 있어 심혈관계 질환과 비만의 예방과 치료에 도움을 줄 수 있을 것으로 판단된다.

Anticoagulant activities of piperlonguminine in vitro and in vivo

  • Piperlonguminine (PL), an important component of Piper longum fruits, is known to exhibit anti-hyperlipidemic, antiplatelet and anti-melanogenic activities. Here, the anticoagulant activities of PL were examined by monitoring activated-partial-thromboplastin-time (aPTT), prothrombin-time (PT), and the activities of thrombin and activated factor X (FXa). The effects of PL on the expressions of plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (t-PA) were also tested in tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) activated HUVECs. The results showed that PL prolonged aPTT and PT significantly and inhibited the activities of thrombin and FXa. PL inhibited the generation of thrombin and FXa in HUVECs. In accordance with these anticoagulant activities, PL prolonged in vivo bleeding time and inhibited TNF-${\alpha}$ induced PAI-1 production. Furthermore, PAI-1/t-PA ratio was significantly decreased by PL. Collectively, our results suggest that PL possesses antithrombotic activities and that the current study could provide bases for the development of new anticoagulant agents.
