위암발증 원인에 대한 생활습관 및 영양섭취에 있어서 농촌지역 주민 조사 -강원도 화천군 주민의 Helicobacter pylori 감염률을 중심으로- (Survey on the Risk Factor Affecting on the Gastric Cancer Incident to the Healthy Adults in Rural Area -Infection of Helicobacter pylori in Whachon Area Kang-Won Do-)

  • To examine the relationship between the casual infection of gastric cancer and lifestyle and nutritional status we surveyed 169 persons 90 females and 79 males aged 40∼69 years from June 7 to June 14 1997 in Whachon area Kang-Won Do Korea. For this purpose we investigated Helicobacter pylori infec-tion and identified the effect of sex age, smoking alcohol consumption economic status occupation life-style and food intake on Helicobacter phlori infection. Helicobacter pylori status was evaluated using an en-zyme-linked immunosorbant assay(ELISA) for anti-Helicobacter pylori immunoglobulin G(IgG). Helicob-acter pylori infection was present in 66.3% of total subjects namely 67.7% of female and 64.4% of male and its prevalence increased with smoking low education level and low monthly income. Energy total protein calcium and vitamin C intakes in the negative Helicobacgter phlori infection group were higher than those in positive Helicobacter pylori infection group. However iron and niacin intakes were lower in the helicobacter pylori negative group than in the positive group.

Fermentation Characteristics and anti-Helicobacter pylori Activity of Aqueous Broccoli Fermented by Lactobacillus plantarum MG208

  • Helicobacter pylori infection causes gastrointestinal diseases such as chronic gastritis, peptic ulcers, and may lead to gastric cancer. Several studies have reported that lactobacilli present on broccoli show inhibitory activity against H. pylori. Here, we evaluated aqueous broccoli, fermented by Lactobacillus plantarum MG208, for its fermentation characteristics and anti-H. pylori activities including antibacterial activity, growth inhibition, anti-adhesion, and urease inhibition. The results indicated that the fermentation characteristics changed significantly depending on the amount of aqueous broccoli used for fermentation (p <0.05). There was no significant difference between the samples before fermentation (p >0.05). However, a significant concentration-dependent difference was noted in antibacterial activity and urease inhibition (p <0.05) following the addition of aqueous broccoli. Growth inhibition in the 10 mg/mL sample was significantly higher as compared to the negative control and similar to that with amoxicillin (positive control) (p <0.05). Anti-adhesion activity of aqueous broccoli was also significantly different (p <0.05) from the negative control. Therefore, aqueous broccoli fermented by L. plantarum MG208 could prove useful as a functional diet for protection of the gastric environment against H. pylori infection.

Lactobacillus acidophilus BK13 and Lactobacillus paracasei BK57 균주가 생산한 항균물질의 anti-Helicobacter pylori 활성 및 위장상피세포에 대한 세포독성 평가 (Evaluation of the anti-Helicobacter pylori and cytotoxic properties of the antimicrobial substances from Lactobacillus acidophilus BK13 and Lactobacillus paracasei BK57)

  • Lactobacillus acidophilus BK13과 Lactobacillus paracasei BK57 균주로부터 얻은 세포, 배양상등액 및 박테리오신 용액의 anti-Helicobacter pylori 활성과 위장상피세포에 대한 세포독성을 평가하였다. 실험균주를 MRS 배지 상에서 30시간 배양한 결과, L. acidophilus BK57 ($126.8{\pm}7.9mM$) 보다 L. paracasei BK57 ($155.9{\pm}7.9mM$)가 더 많은 양의 유산을 생산하였다. 또한, BK13 균주의 최대 박테리오신 활성(128 AU/ml)은 $37^{\circ}C$에서 30시간 배양 후 관찰되었으나, 이는 BK57의 활성(256 AU/ml) 보다는 낮았다. BK13 및 BK57 균주의 살아있는 세포를 H. pylori와 혼합 배양한 결과, 유산균의 초기균수에 의존하여 H. pylori의 저해효과가 나타났다. 게다가 BK13과 BK57로부터 얻은 배양상등액과 박테리오신은 H. pylori의 성장을 억제할 뿐만 아니라 위장상피세포에 대한 부착력과 urease 활성도 저해하였다. 한편, 이러한 균주들이 생산한 유산은 위암세포에 대한 세포독성 효과가 대조구보다 유의한 수준으로 높게 나타났다. 따라서 BK13과 BK57 균주의 항균물질은 위장질환의 원인균인 H. pylori를 저해시키는데 효과적이므로 이들 유산균은 H. pylori 감염으로부터 위장을 보호하는데 유용할 것으로 사료된다.

  • This study investigated the effect that some polyacetylenes and protopanaxatriol, which were isolated from heated ginseng (family Araliaceae), have on inhibiting Helicobacter pylori (HP) growth. Among the compounds tested, panaxytriol was quite effective in inhibiting HP growth with an MIC of 50 ${\mu}g/ml$. Cinsenoside Rhl and protopanaxatriol weakly inhibited $H^{+}/K^{+}$-ATPase from a rat stomach.

  • 6종 한약재의 70% 에탄올 추출물의 Helicobacter pylori의 위내 서식을 도와주는 것으로 알려진 urease의 활성억제능은 오매(Mume Fructus)의 에탄올(70%) 추출물에서 가장 높았으며, 소목 및 연교의 추출물도 높았다. 특히, 오매의 경우 한약재 추출물을 넣지 않은 대조군과 비교하여 90% 이상의 urease 활성억제 효과를 나타내었다.

Comparative Proteome Analysis of Celastrol-Treated Helicobacter pylori

  • Various preclinical and clinical trials have been conducted the efficacy of celastrol. In data presented in the current manuscript is the first trial to inhibit Helicobacter pylori with celastrol. In this study, the quantitative change of various H. pylori proteins including CagA and VacA by the anti-bacterial effect of celastrol was determined. The anti-H. pylori effects of celastrol was investigated by performing 2-dimensional electrophoresis and additional supporting experiments. After 2-dimensional electrophoresis analysis, spot intensities were analyzed and then each spot was identified using matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS) or peptide sequencing using Finnigan LCQ ion trap mass spectrometer (LC-MS/MS). The results show that celastrol has multiple effects on protein expression in H. pylori.

소함흉탕 에탄올 추출물 및 황련 알칼로이드의 헬리코박터 파이로리에 대한 항균활성 (In Vitro Anti-Helicobacter pylori Activity of Ethanol Extract of Sohamhyoongtang and Coptidis Rhizoma Total Alkaloids)

  • The aim of this study was to evaluate the anti-helicobacter activity of the ethanol extract of Sohamhyoongtang (Coptidis Rhizoma, Pinelliae Tuber and Trichosanthis Semen) and Coptidis Rhizoma total alkaloids, which is one of the components of Sohamhyoongtang. Crude ethanol extract of Sohamhyoongtang (ESHHT) and Coptidis Rhizoma total alkaloids (CRTA) were used for this experiment. Five different types of H. pylori (including H. pylori 26695) were used as test strain. To determine anti-helicobacter activity, minimum inhibitory concentration (MIC) was determined by agar dilution method. The effect of ESHHT and CRTA on the gene expression of H. pylori was investigated by quantitative realtime-PCR (qRT-PCR). MICs of ESHHT against five H. pylori strains were $250{\sim}500{\mu}g/ml$ and MICs of CRTA against five H. pylori strains were $50{\sim}200{\mu}g/ml$. Four representative virulence genes of H. pylori, cagA, ureA, ureB and ureI were tested as target genes for qRT-PCR. According to the qRT-PCR results, both ESHHT and CRTA markedly repressed the expression of cagA gene of H. pylori 26695 (6.91 and 20 folds respecively). These results showed that the ESHHT and CRTA demonstrated antihelicobacter properties, suggesting their potential use in gastritis or duodenal ulcer.

약용식물 추출물의 Helicobacter pylori에 대한 항균 활성과 항산화 활성에 미치는 효과 (The Effect of Medicinal Herb Extract on Antimicrobial Activity against Helicobacter pylori and Antioxidant Activity)

  • This study was designed to investigate the effect of medicinal herb extract on the antioxidant and antimicrobial activities against Helicobacter pylori, which is known as a ulcerogenic pathogen. The concentration of total phenolic compound of Scutellaria baicalensis(13.14%) was highest among water extracts and that of Ulmus parvifolia(15.12%) was highest among the ethanol extracts. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 91.00% and 65.03%, respectively, in DPPH assay. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 32.90% and 27.70%, respectively, in SOD assay. The antioxidant activity of the water extract of Ulmus parvifolia and of the ethanol extract of Glycyrrhiza uralensis Fischer was 2.15 and 2.17, respectively, in TBARS assay. In disc method, Scutellaria baicalensis showed the highest anti-microbial activity against H. pylori, followed by Glycyrrhiza uralensis Fischer among the water extracts and Glycyrrhiza uralensis Fischer showed the highest anti-microbial activity followed by Radix puerariae among ethanol extract.
