광곽향 메탄올 추출물의 항산화, 항염증 및 암세포 증식 억제 효과 (Anti-oxidant, Anti-inflammatory and Anti-cancer Effect of Methanol Extract of Pogostemon cablin)

  • 본 연구에서는, 암세포 증식 억제효능과 항산화, 항염증 효능을 동시에 가지는물질을 탐색하였다. 그 결과, 광곽향 메탄올 추출물이 A549, HepG2, MCF7, HT29 등 다양한 암세포에 대하여 세포성장억제효과를 보였고, A549에 대해 특이적으로 뛰어난 사멸효과를 보였다. 광곽향 메탄올 추출물에 의한 A549에서의 항암 효과는 p38 - Cdc25A - Cdk - Cyclin - Rb pathway를 통해 G1 arrest 유도로 연결되는 것으로 사료된다. 또한 DPPH를 통한 free radical의 소거능 확인 결과 항산화 효과를 가지고 있는 것을 확인하였고, 대식세포(RAW 264.7)의 iNOS 발현을 감소시켜 LPS에 의해 유도되는 NO의 생성을 유의적으로 억제함을 확인했다. 이러한 결과들로부터 광곽향 메탄올 추출물이 항산화, 항염증, 항암 후보물질 소재로 활용가능 할 뿐 아니라, 다양한 건강기능성 소재로 활용가능할 것이라 사료된다.

Anti-Allergic Effect of Oroxylin A from Oroxylum indicum Using in vivo and in vitro Experiments

  • Oroxylum indicum has long been used in Asian traditional medicine to prevent and treat respiratory diseases, diabetes, diarrhea and other conditions. Oroxylin A is a flavone that is present in Oroxylum indicum and in Scutellaria baicalensis. Because the root extracts of both plants have been shown to have anti-allergic effects, the authors investigated whether oroxylin A is likely to have beneficial effects on allergic asthma using female Balb/c mice and rat RBL-2H3 mast cells. Antigen-induced degranulation was measured in vitro by measuring b-hexosaminidase activity. A murine ovalbumin-induced allergic asthma model was used to test the in vivo efficacy of oroxylin A. Sensitization and challenge of ovalbumin induced allergic asthma responses, the accumulations of eosinophils and Th2 cytokine levels in bronchoalveolar lavage fluid and lung tissues. Oroxylin A administration decreased numbers of inflammatory cells, especially eosinophils, and reduced the expression and secretion of Th2 cytokines, including IL-4 and IL-13, in lung tissues and bronchoalveolar lavage fluid. Histologic studies showed oroxylin A reduced inflammatory signs and mucin production in lungs. These findings provide evidence that oroxylin A has potential use as an anti-allergic therapeutic.

Anti-fatigue effects of Elaeagnus multiflora fruit extracts in mice

  • The fruit, leaves, and roots of Elaeagnus multiflora Thunb. have been used in traditional Chinese medicine to treat cough, diarrhea, and itching. However, the anti-fatigue effects of E. multiflora fruit (EMF) extract have not been studied in detail. The aim of this study was to examine the effect of EMF on fatigue and exercise performance in BALB/c mice. EMF was orally administered to mice at four doses (10, 50, 100, and 200 mg/kg/day) for 2 weeks. The anti-fatigue activity was evaluated by determining the exhaustive swimming time. Blood lactate and glucose levels and serum lactate levels after a 10 min swimming time, as well as ammonia, creatine kinase (CK), blood urea nitrogen (BUN), lactate dehydrogenase (LDH), and glycogen contents after exhaustive swimming time were measured. The exhaustive swimming time of the EMF 200 group was significantly increased (p <0.01). The EMF groups showed significantly low levels of CK, BUN, LDH, and lactate compared with the control group (p <0.05). Increased liver glycogen was observed in the EMF 200 group (p <0.05). These results suggest that EMF can be utilized as an efficacious natural resource for its anti-fatigue effects.

LPS로 유도된 RAW264.7세포주에서 황금뿌리 물추출물의 항염증활성 (Anti-inflammatory activity of Scutellaria Baicalensis root extract in lipopolysaccharide-induced RAW 264.7 cells)

  • Scutellaria baicalensis water extract (SWE)는 지질 다당류 LPS로 유도된 RAW 264.7 세포에서 NO 및 전 염증성 사이토카인인 TNF-α의 생성을 세포 독성을 유발하지 않고 유의하게 억제하였다. 또한, SWE는 iNOS 및 COX-2의 단백질발현을 농도의존적으로 감소시켰으며, ERK, JNK, p38과 같은 MAPKs 계열의 인산화 발현 수준을 조사한 결과 JNK와 p38의 발현 수준을 감소시켰다. 이는 SWE가 p38 인산화를 억제함으로써 iNOS, COX, 그리고 TNF-α와 같은 전 염증성 사이토카인의 발현을 감소시키며 결론적으로 NO의 생성을 억제시킨다는 결과를 도출할 수 있었다. 본 연구는 항염증 효능 검증뿐 아니라 염증대사기전의 주요인자를 탐색함으로써 황금의 기능성 소재로써의 가능성을 시사한다.

전라북도 소아과 개원의의 급성 위장염 환자에 대한 인식 조사 (Pediatrician Perspectives on the Evaluation and Treatment of Acute Gastrointestinal Infections, Jeonbuk, South Korea, 2002)

  • 목 적 : 전라북도 소아과 개원의의 급성 위장염에 대한 이해와 환자의 진단과 치료에 대한 경험과 인식을 알아보고 최근 개발된 로타 바이러스 백신에 대한 인식을 알아보고자 하였다. 방 법 : 2002년 4월부터 6월까지 전라북도에서 소아과 의원을 개원하고 있는 소아과 전문의를 대상으로 미리 작성된 설문지를 이용하여 우편배달을 통해 조사하였다. 배포된 설문지 수는 총 82매였고 이 중 63명의 소아과 전문의가 응답하여 회수율은 76.8%였다. 급성 위장염 환자의 비율, 선호하는 치료제 및 방법, 항생제 사용빈도 및 종류, 항생제 사용을 고려하는 임상소견, 급성 위장염 환자에서 시행해 보고 싶은 검사, 로타 바이러스 백신에 대한 관심도와 사용 계획에 대해 조사하였다. 결 과 : 급성 위장염 환자는 연중 발생하였으며 겨울과 봄에 약간 많았다. 주로 사용하는 치료제로는 유산균 제제(84.1%), 경구용 수액제(79.4%), 효소제(58.8%)를 선호하였다. 대부분 의사들이 전체 위장염 환자의 20% 이하에서 항생제를 사용하였고, 주로 사용하는 항생제는 amoxicillin(61.3%)과 trimethoprim/sulfamethoxazole(39.7%)이였으며, 항생제 사용을 고려하는 임상증상은 혈변이나 점액변, 고열 등의 증상이었다. 급성위장염 환아에서 중요하다고 생각하거나 가능하면 해보고 싶은 검사는 대변 검사(로타 바이러스 항원, 적혈구, 백혈구), 복부 방사선 검사, 혈청 전해질 검사 등이었다. 대부분의 의사들이 로타 바이러스 백신에 대해 긍정적인 관심을 가지고 백신이 도입될 경우 사용하겠다고 하였으며, 백신사용을 결정할 때 중요하게 생각하는 점으로는 예방효과와 부작용이었다. 결 론 : 전라북도 소아과 개원의들의 급성 위장염 환자에 대한 인식은 대체로 긍정적인 것으로 판단되었으며, 앞으로 보다 적극적인 검사방법의 개발과 이용이 중요할 것으로 생각된다.

양돈 임상 연구에 의한 천연물 오배자 추출물의 항균.항바이러스 효과 규명 (Antimicrobial Effects of the Extract of Galla rhois on the Long-term Swine Clinical Trial)

  • 천연물 오배자 추출물은 in vitro 선행 연구를 통하여 병원성 세균과 바이러스에 대한 강력한 항균 항바이러스 효과를 확인하였다. 본 연구는 양돈 농장에서 130일간의 장기 임상시험을 통하여 효능평가를 통한 천연물 오배자 추출물의 항균 항바이러스 효과를 규명하고자 수행되었다. 200 마리의 돼지를 천연물 오배자 추출물 투여군과 비투여군으로 2 그룹으로 나누어 130일 동안 양돈농장에서 연구가 수행되었다. 천연물 오배자 추출물 투여군에는 항생제가 없는 사료에 천연물 오배자 추출물을 0.2% 배합하여 급여하였고 비투여군은 항생제가 포함된 일반 사료를 급여하였다. 연구기간 동안 사료섭취량, 체중, 임상증상을 분석하고 시험종료일에 부검하여 육안검사와 미생물학적 검사 및 병리조직학적 검사를 수행하였다. 연구결과 천연물 오배자 추출물은 양돈 산업에서 심각한 문제가 되고 있는 병원성 세균과 바이러스에 대한 강력한 항균 항바이러스 효과를 가지고 있는 것을 확인할 수 있었다.

면역전자현미경(免疫電子顯微鏡)(IEM) 기법(技法) 및 immunogold conjugate 면역전자현미경(免疫電子顯微鏡)(IGC-IEM) 기법(技法)을 이용(利用)한 돼지 분변내 PED 바이러스의 검출(檢出) (Detection of PED virus by the immunoelectron microscopy and immunogold conjugate immunoelectron microscopy)

  • Both immunoelectron microscopy(IEM) and immunogold conjugate immunoelectron microscopy (IGC-IEM) techniques were developed for the detection of porcine epidemic diarrhea virus(PEDV) from the feces. Fecal samples were incubated sequentially with anti-PEDV monoclonal antibody(MoAb) and immunogold conjugated goat anti-mouse IgG+IgM. Then negatively stained, mounted on the formvar carbon-coated copper EM grids and observed by the transmission electron microscope. By the direct electron microscopy(DEM), coronavirus particles were observed from 17 cases of total 33 fecal samples of grower pigs and sows. The virons of coronavirus were moderately pleomorphic but mostly spherical, with a diameter ranged from 90 to 190nm. PED virus particles were identified from 15 cases of 17 DEM positive samples by the IEM and IGC-IEM techniques. Aggregates of PED virus coated with specific antibody were seen in fecal samples incubated with homologous anti-PED virus MoAb but not in control samples incubated with anti-TGE virus MoAb. Following incubation with immunogold-conjugated secondary antibody, the gold granules were usually distributed around and among the virus particles and soluble and viral particle-associated antigen. So, IEM and IGC-IEM techniques were proved a rapid and sensitive methods for detection and identification of PED virus from fecal and intestinal contents.

Antioxidant and Anti-Obesity Effects of Juglans mandshurica in 3T3-L1 Cells and High-Fat Diet Obese Rats

  • Juglans mandshurica Maxim. walnut (JMW) is well-known for the treatment of dermatosis, cancer, gastritis, diarrhea, and leukorrhea in Korea. However, the molecular mechanism underlying its antiobesity activity remains unknown. In the current study, we aimed to determine whether JMW can influence adipogenesis in 3T3-L1 preadipocytes and high-fat diet rats and determine the antioxidant activity. The 20% ethanol extract of JMW (JMWE) had a total polyphenol content of 133.33 ± 2.60 mg GAE/g. Considering the antioxidant capacity, the ABTS and DPPH values of 200 ㎍/ml of JMWE were 95.69 ± 0.94 and 79.38 ± 1.55%, respectively. To assess the anti-obesity activity of JMWE, we analyzed the cell viability, fat accumulation, and adipogenesis-related factors, including CCAAT-enhancer-binding protein alpha (C/EBPα), sterol regulatory element-binding protein-1c (SREBP1c), peroxisome proliferator-activated receptor-gamma (PPARγ), fatty acid synthase (FAS), and acetyl-CoA carboxylase (ACC). We found that total lipid accumulation and triglyceride levels were reduced, and the fat accumulation rate decreased in a dose-dependent manner. Furthermore, JMWE suppressed adipogenesis-related factors C/EBPα, PPARγ, and SREBP1c, as well as FAS and ACC, both related to lipogenesis. Moreover, animal experiments revealed that JMWE could be employed to prevent and treat obesity-related diseases. Hence, JMWE could be developed as a healthy functional food and further explored as an anti-obesity drug.

Reversible Hepatic Toxic Effect of Crocin Dyes in Rats

  • Gardenia jasminodes has been medically used for anti-inflammation, sedation and anti-diarrhea; The extract of this plant has been traditionally used as food colorant and referred as crocin dyes. In the present study, the possible hepatic toxicity of this dye has been evaluated on the basis of its alteration on the marker enzymes, namely, glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase, alkaline phosphatase, lactate dehydrogenase and gamma-glutamyltransferase. Crocin dyes did not affect hepatic function when they were orally administered to rats in a daily dose of 50 mg/kg for 8 days, but could induce acute hepatic discoloration. A high dose of 100 mg/kg for 2 weeks could induce both hepatic damage and black pigmentation, but a lower dose of 10 mg/kg for 40 days did not The induced black pigmentation and the acute hepatic damage were completely reversible. In conclusion, the crocin dyes have a very low hepatic toxicity in rats, even in high experimental dosages which could hardly happen in human practice. It is therefore suggested that the crocin dyes are safe for coloring foods.

  • For the purpose of examing on the efficancy of the Boolwhangumjeonggisan and on the effect of the Boolwhangumjeonggisan, dropsy animals given water boil ding abstraction exgis power. What made an experiment, the motility of isolate ileume, anticathartic action, the action of gastric juice, tied pylous ulcer and inhibited vomitting. 1. Boolwhangumjeonggisan displayed great suppresion effect in regard to automatic movement of the motility of isolated of mice and displayed anti-acethylcholine action, antibarium chloride action. Thus, the origin of muscle relaxation for internal smooth muscle is admitted. 2. It displayed great rexation effect in regard to fraction of rat's stomach and displayed contentional effect in regard to a cetylcholine and barium chloride. 3. It decreased rat of barium sulfate transport through the small intestine of mice. 4. It recognized anti-cathartic action, rat suffered from leading diarrhea by caster oil. 5. Total activity, pepsin secretion decreased and increased the pH of stomach in Shay's method. 6. It recognized powerful prevention effect on tied pylous ulcer. 7. It inhibited vomitting by administration of CuSo4 in frog.
