Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.

Anti-cancer and Anti-inflammatory Properties of Korean Citrus Fruits (Citrus aurantium L.)

  • Purpose. Citrus aurantium L.(familyRutaceae), alsoknownasbitter orange, have been used as traditional herbal medicine in many Asian countries since ancient times. Hence, the purpose of the study was to briefly discuss the new findings about anti-inflammatory and anti-cancer activities of Citrus aurantium L in-vitro. Methods. The articles for this study were collected from pubmed and scopus electronic resources. Results. Citrus aurantium L contains an abundant Flavonoids, including hesperidin, naringin and nobiletin. These Flavonoids has reported to have various medicinal benefits that include antioxidant, antimicrobial, anti-inflammatory, anticancer, anti-diabetic activities, and also used to treat cardiovascular diseases. Conclusion. Based on the above evidence, we propose that Flavonoids from Korea Citrus aurantium L would be a therapeutic potential for cancer treatment and pharmacological benefit for inhibiting the inflammatory effect.

Sinensetin Inhibits Interleukin-6 in Human Mast Cell - 1 Via Signal Transducers and Activators of the Transcription 3 (STAT3) and Nuclear Factor Kappa B (NF-κB) Pathways

  • Sinensetin, a pentamethoxyflavone, is known to exert various pharmacological activities including anti-angiogenesis, anti-diabetic and anti-inflammatory activities. However, its effects on the human mast cell - 1 (HMC-1) mediated inflammatory mechanism remain unknown. To explore the mediator and cellular inflammatory response of sinensetin, we examined its influence on phorbol 12-myristate 13-acetate (PMA) plus A23187 induced inflammatory mediator production in a human mast cell line. In this study, interleukin (IL)-6 production was measured using the enzyme-linked immunosorbent assay and reverse transcription polymerase chain reaction. Sinensetin inhibited PMA plus A23187 induced IL-6 production in a dose-dependent manner as well as IL-4, IL-5 and IL-8 mRNA expression. Furthermore, sinensetin inhibited signal transducer and activator of transcription 3 (STAT3) phosphorylation, suggesting that sinensetin inhibits the production of inflammatory mediators by blocking STAT3 phosphorylation. Moreover, sinensetin was found to inhibit nuclear factor kappa B activation. These findings suggest that sinensetin may be involved in the regulation of mast cell-mediated inflammatory responses.

산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성 (Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

Effects of Silkworm Extract on Disaccharidase Activities of Small Intestine and Blood Glucose-Lowering in C57BL/6J Mice

  • This study examined the anti-diabetic effect of a silkworm extract in C57BL/6J mice, an ob/㏈ model, fed a high fat diet for 8 weeks. The body weight was not significantly different with the silkworm-extract supplement, nor did food intake and body weight gains also did not differ significantly among the high-fat diet groups. However, the water intake by the silkworm-extract supplemented groups increased significantly compared with that by the distilled-water supplement group, nonetheless, the FER did not differ significantly. For all groups, the blood glucose increased the most after 30 minutes and yet returned to a fasting level within 90 minutes. The fasting time and resulting glucose tolerance for the silkworm-extract supplemented groups were significantly decreased compared to that for the high fat diet with distilled water supplement group, while the level of blood glucose in silkworm-extract supplemented groups was significantly decreased compared with than in the diabetic control group. The HbA1c and insulin levels were no different among the groups. The sucrase and lactase activities in the proximal small intestine were significantly decreased in the silkworm-extract supplement groups compared to that in the diabetic control group. There was no significant difference in the glycogen contents in the liver and muscle among the groups. In conclusion, it was found that the silkworm-extract supplement repressed the disaccharidase activity in the small intestines mucosa of the C57BL/6J mice.

Hypoglycemic Activities of a Mangrove Plant Rhizophora apiculata Blume

  • The leaves of Rhizophora apiculata, a plant belonging to the family Rhizophoraceae were collected from the mangrove forest of Sunderbans, West Bengal, India. Alcoholic extract of the leaves of this plant was prepared and hypoglycemic/anti-hyperglycemic activity was studied in fed rats, glucose loaded rats and streptozotocin induced diabetic rats. The results of this study reveal that this plant extract has potential hypoglycemic action.

노간주나무 에탄올추출물의 지방 흡수 억제 및 지방분해 효능 (Anti-lipase and Lipolytic Activities of EtOH Extract from Juniperus rigida)

  • Obesity is an important risk factor that significantly increases mortality and disease rates in the cardiovascular disease, diabetes, and various diseases. So far, the most powerful way to inhibit fat absorption is pancreatic lipase inhibitors. In this study, we investigated the anti-obesity effect of the extract of Juniperus rigida. Juniperus rigida extract (JRE) had a inhibitory effect on pancreatic lipase activity ($IC_{50}$=8.63 ${\mu}g$/ml). In in vivo oil-emulsion loading test, this extract also inhibited the intestinal fat absorption. In addition, we measured inhibitory effects of JRE on activity of phosphodiesterase (PDE) and hormone sensitive lipase (HSL) among the important enzymes associated with lipolysis. JRE strongly inhibited PDE activity ($IC_{50}$=4.56 ${\mu}g$/ml), whereas inhibitory effect on HSL activity was very weak compared with orlistat. As a result, JRE inhibited the absorption of fat by inhibiting the activity of pancreatic lipase and induced lipolysis through inhibition of PDE activity. Therefore, we suggest that Juniperus rigida may be a potential therapeutic agent improving obesity.

홍국발효 대두의 항산화 및 항당뇨 활성 (Antioxidant and Anti-Diabetic Activities of Soybean Fermented with Monascus)

  • This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.

항당뇨 효능이 있는 천연물의 탐색 및 활성물질의 분석 (Screening of Natural Products for Anti-diabetic Activity and Analysis of Their Active Compounds)

  • 현대인들의 식습관 변화로 인해 대사성 질환의 발생률이 증가하고 있으며, 특히 당뇨병이 가장 큰 문제로 꼽힌다. 하지만 기존의 당뇨병 치료제는 부작용을 동반하고 있어서 예로부터 쓰이던 천연물을 통해 부작용이 적은 항당뇨 활성 물질을 찾고자 하였다. 따라서 본 실험에서는 항당뇨 및 항산화 활성이 있는 천연물을 탐색하기 위하여, DPPH free radical scavenging assay와 α-glucosidase 및 PTP1B inhibition assay를 이용하였다. 항당뇨 효능이 예상되는 12가지의 시료를 메탄올로 추출하고 항당뇨 활성 및 항산화 활성을 측정하였다. 그 중 항당뇨 활성이 우수한 느릅나무, 벌나무 가지, 연 씨앗, 홍화 씨앗 등의 4가지 시료를 대상으로 에틸아세테이트, 부탄올을 이용하여 활성물질을 용매추출하여 항당뇨 활성을 측정하였다. 그 결과, α-glucosidase와 PTP1B 저해활성이 우수한 홍화 씨앗의 에틸아세테이트 분획물(MG-11-E)을 선정하였다. MG-11-E를 대상으로 preparative thin layer chromatography를 수행한 결과, 분획물 #6에서 α-glucosidase 및 PTP1B에 대한 우수한 저해활성을 보였다. 50% 메탄올을 이동상으로 하여 TLC 분획물 #6을 high performance liquid chromatography하여 각각을 분획하고 항당뇨 활성을 측정한 결과, 단일 peak를 보인 RT 4분에서 항당뇨 활성이 확인되었다. 따라서 홍화 씨앗의 추출물의 항당뇨 효능을 나타내는 항당뇨 활성 성분은 PTP1B 보다 α-glucosidase에 대한 저해 활성이 더욱 강하게 나타나 PTP1B를 저해하는 천연물과 조합하여 사용함으로써 당뇨병에 대한 효과를 높일 수 있을 것으로 사료된다.

볶음 치커리의 생리활성 (Biological Activities of Roasted Chicory Root)

  • 국내산 치커리의 생리적 기능성을 알아보기 위해 실험한 결과는 다음과 같다. 복음치커리의 물추출물은 간세포의 독성을 유발시키지 않았으며, 5 mM의 galactosamine을 5시간 배양하였을 때 LDH의 활성이 가장 높게 나타났다. 5 mM의 galactosamine으로 독성을 유발시킨 흰쥐의 간세포에 볶음치커리 물추출물을 투여한 결과 간세포로 부터 유리되는 LDH의 활성을 유의성 있게 감소시켜 간세포 보호 활성을 나타내었다. 볶음치커리 물추출물 800 mg을 당뇨 유발 흰쥐에 10일간 투여한 결과 혈당치는 297 mg/dl로 유의적으로 감소하는 경향을 나타냈었고, 체중은 대조구에 비해 20% 높게 나타났다. Salmonella thyphimurium YG 1024의 복귀돌연변이 콜로니수를 조사한 결과 복음치커리 물추출물 ($10{\sim}5,000\;{\mu}/g$)의 처리구에서는 돌연변이를 유발시키지 않았으며, 2-aminofluorene을 처리한 양성 대조군에 물추출물을 plate당 $1,000{\sim}5,000\;{\mu}/g$ 농도로 처리한 결과 돌연변이 억제 활성은 없었다.
