표고 톱밥배지 재배시 LED 광파장이 자실체 생육에 미치는 영향 (Effects of cultural characteristics of Lentinula edodes according to LED wavelength with sawdust substrate cultivation)

  • 표고 톱밥재배시 LED광파장에 따른 생육특성을 분석한 결과, 파장이 긴 적색광과 황색광에서 대길이가 길어지고 갓색이 연해지며, 파장이 짧은 청색광과 녹색광에서는 대길이가 짧아지고 갓색이 진해지는 현상을 보였다. 자실체 항산화활성은 광파장에 따른 큰 차이를 보이지 않았으며, 2주기까지 배지당 자실체 수량은 녹색에서 159 g으로 형광등 121 g 대비 31% 증수효과가 있었다.

Effect of Smoking and Alcohol Consumption on Seminal Quality in Young Mice

  • The purpose of this study is to evaluate the effects of alcohol or cigarette smoking on seminal parameters in a large group of mice model. Nine groups (n=20/group) of mice were treated intensive noxious materials that abdominal injection of 21% (v/v) of ethanol, cigarette smoke (10, 20, 30 minutes/day), and combination of ethanol and 30 minutes of smoking. In addition, vitamin C and selenium were also treated to mice exposed to combination of alcohol and smoking to identify the recovering effect. Sperm viability and motility were significantly decreased in either alcohol consumption or smoking exposed group, and combination of both materials have additive detrimental effects on seminal parameters. Mice groups that exposed to alcohol and smoking showed statistically significant decrease in motility and increase of static spermatozoa. Moreover, combination of both treatments showed cumulative effect in increase of static spermatozoa. Treatments of either vitamin C or selenium dramatically recovered detrimental effects of alcohol and smoking on seminal quality, although combination of both antioxidant molecules did not show any additive effect. In conclusion, detrimental effects of alcohol and cigarette consumption on sperm quality and motility were identified in mice model, and these detrimental effects can be compensated to uptake of anti-oxidant molecules.

  • RBL-2H3 cell에서 HRE에 의하여 histamine이 분비되는 과정에서 ROS가 생성되는지 실험해 본 결과, ROS가 HRF를 처리한지 5분대에 최대치를 보이며 생성되었다가 소멸되는 것을 관찰할 수 있었다. 따라서 HRF가 세포내 second messenger로써의 ROS를 생성하였다고 확인할 수 있었다. 또한 ROS는 단백질 정제 과정에서의 endotoxin오염에 의해 영향을 받지만,본 실험에서 규명한 HRF에 의한 ROS 생성은 endotoxin에 의한 것이 아닌, 순수하게 HRF에 의한 signaling의 결과라는 것도 확인할 수 있었다.

Antioxidant Supplementation Enhances the Porcine Semen Preservation Capacity

  • Preservation of liquid semen is an important factor for breeding management in swine industry. Oxidative stress of spermatozoa during liquid preservation has a detrimental effect on sperm quality and decreases fertility. Objective of this study was to determine the effect of antioxidant, Quercetin, on capability of porcine liquid semen preservation. Freshly collected porcine semen from boars (n=3), having proven fertility was counted, diluted to $3{\times}10^7/mL$ and divided into 5 different semen extenders. Aliquots of diluted semen with different extenders were subjected to measure the pH, motility, viability and sperm DNA structure status on elapse time after preservation for 10 days. For the first 3 days, semen preserved in all 5 different extenders maintained their initial pH and either gradually decreased or increased thereafter, indicating lipid peroxidation has started. Sperm motility (r=0.52, p=0.01) and viability (r=0.55, p=0.03) had positive correlation with semen pH. Sperm motility was maintained well (p<0.05) in especially 2 extenders containing Tris and antioxidant compared to other extenders, suggesting both Tris and antioxidant worked as pH regulator and had beneficial effects on sperm characteristic during preservation. Sperm DNA structure status accessed by sperm chromatin structure assay on elapsed time after preservation, tended to be higher in semen preserved without antioxidant. Taken together, addition of antioxidant to extender prevents the sperm from oxidative stress during storage in mechanism by which antioxidant slows the lipid peroxidation, and thus reduced the reactive oxygen species in preserved porcine semen resulted in maintaining semen pH, sperm motility and viability for 7~10 days.

퀴노아를 첨가한 청국장의 발효기간에 따른 품질 변화 (Changes in Quality Characteristics of Cheonggukjang added with Quinoa during Fermentation Period)

  • In this study, the quality characteristics of cheonggukjang with addition of different quinoa were investigated. We evaluated the quality and sensory characteristics of cheonggukjang, including the pH, amino nitrogen, slime contents, color value, and total aerobic bacteria. Moreover, The anti-oxidant activities were measured as total polyphenol content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and superoxide dismutase (SOD) activity. The pH and amino nitrogen content were noted to significantly increase during the fermentation period in all the samples. The slime content of cheonggukjang added with quinoa increased with increased fermentation time, but cheonggukjang with addition of 20% of quinoa was decreased. The L value and b value decreased significantly with increased fermentation time, but the a value increased significantly. The microbial tests of cheonggukjang with addition of quinoa showed that the aerobic micro-organisms count was 7.45~9.10 Log cfu/g. Total polyphenol contents increased in all groups during the fermentation period, and activity increased with an increased percentage of added quinoa. The DPPH radical scavenging activity and SOD-like activity of cheonggukjang (with addition of quinoa) were also significantly higher than those of the control. The sensory quality of quinoa 10% cheonggukjang was stronger in flavor, and taste, and demonstrated a higher level of overall acceptability, when compared to the other groups.

구리에 노출된 돌돔(Oplegnathus fasciatus)의 항산화 효소 및 acetylcholinesterase 활성의 변화 (Chronic Effects of Copper on Antioxidant Enzymes and Acetylcholinesterase Activities in Rock bream Oplegnathus fasciatus)

  • A laboratory experiment was conducted to determine chronic effects of waterborne copper exposure on rock bream Oplegnathus fasciatus using a panel of enzymes. The activities of the following biochemical biomarkers were determined at different concentrations of $CuSO_4$ for 10 and 20 days: alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH) in plasma; antioxidant enzymes including glutathione (GSH), superoxide dismutase (SOD) and glutathione peroxidase (GPx) in liver and gills; and acethylcholinesterase (AChE) in brain and muscle. After exposure to two $CuSO_4$ concentrations (200 and $400{\mu}g/L$), the activities of plasma ALT in the fish showed a tendency to increase with AST and LDH, depending on $CuSO_4$ concentration. Additionally, GSH levels and SOD activities significantly increased, depending on $CuSO_4$ concentrations in liver and gills. This involved the inactivation of reactive molecules formed during oxidative stress, which could provide protection against oxidative damage induced by $CuSO_4$. However, GPx and AChE activities significantly decreased with $CuSO_4$ in liver and gills. In conclusion, these enzymes may represent convenient biomarkers for monitoring heavy metal pollution in coastal areas. Such chronic exposure studies are necessary for improving our understanding of complementary or deleterious effects of pollutants, and for developing metal toxicity biomarkers.

청간해주환(淸肝解酒丸)의 알코올 유도 뇌신경세포 손상에 대한 보호 효과 (Effect of Chungganhaeju-hwan in Ethanol-induced Neuronal Cell Damage)

  • Objectives : In this study, we evaluated the effect of Chungganhaeju-hwan(CGHJH) on hydrogen peroxide($H_2O_2$)-induced and ethanol(EtOH)-induced neuronal damage in vitro and in vivo, respectively. Methods:We carried out the anti-oxidant effects of CGHJH against hydrogen peroxide($H_2O_2$)-induced toxicity in HT22 and PC12 cells using thiazolyl blue tetrazolium bromide. Then, to investigate the protective effect on CGHJH against EtOH-induced memory impairment and hippocampal cell damage in male ICR mice, we performed novel object recognition test(NORT), and analysed the brain tissues after immunohistochemistry and western blotting. Results:CGHJH showed protective effect from $H_2O_2$-induced cell toxicity at doses of $1\sim100{\mu}g$/mL in both HT22 and PC12 cells. CGHJH had also recovery effect from EtOH-induced memory impairment in ICR mice from NORT and it protected hippocampal cells against EtOH toxicity in the result of cresyl violet and NeuN immunoreactivity. Conclusion : These results demonstrate that CGHJH has protective effect in neuronal cells against $H_2O_2$ and EtOH toxicities and this effect could be a main role of recovery effect on EtOH-induced memory loss.

고려인삼의 비사포닌 성분에 대한 화학적 및 생화학적 연구 (Chemical and Biochemical Studies on Non-saponin Constituents of Korean Ginseng)

  • There has been general tendency to explain the traditional ginseng efficacy through the pharmacological and biochemical activities of ginsenosides. However, when we analyze the pharmacological and biological data on ginseng reported yet, we can easily arrive at the conclsion that most of the data on pharmacological and biological activities must have been obtained using impure ginsenoside samples, which should contain some non-saponin constituents as impurities. Based on the above back-ground, the non-saponin constituents of ginseng were studied in our laboratory. Phenolic substances including Maltol, Vanillic Acid, Salicylic Acid, Ferrulic Acid and Caffeic acid and impure ginsenoside samples were found to show strong antioxidant and anti-fatigue activities, while pure ginsenosides were devoid of the activities. Maltol, one of antioxidant components In Korean red ginseng drew a special interest due to its very low pro-oxidant activity. The antioxidant activity of ginseng may be considered as scientific basis for the antiageing activity which was described in traditional medicinal material book as "long-term medication of ginseng will improve bio-efficiency and extend life-span" The lignin components, another non-saponin consitutents, isolated from ginseng extract In our laboratory may eplain the hepato-protective activity of ginseng which has been repeatedly rtaimed as one of the efficacies of ginsenosides. The P-carboline alkaloids isolated in our laboratory as one of the non-saponin constituents of ginseng may play some pharmacological activities which should also be investigated. Present paper will include chemistry and biochemical aspects of the non-saponin constituents of ginseng with special interests for the explanation of traditional ginseng efficacy on modern scientific basis.fic basis.

짚신나물 물 추출물의 항산화 활성 및 미백효과에 관한 연구 (Antioxidant and Whitening Effects of Agrimonia pilosa Ledeb Water Extract)

  • This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

Microwave처리에 의한 쌀 가공 부산물의 항산화능의 변화 (Effect of Microwave Treatment on the Antioxidant Activity of Rice Processed By-products)

  • Microwave 처리가 쌀가공 부산물로 발생하는 왕겨와 미 의 항산화능에 미치는 영향을 조사하였다. 100 W, 200 W, 440 W의 출력으로 microwave를 처리하였을 때, 출력과 처리시간에 비례하여 갈변을 나타내는 색도의 변화가 관찰되었다 그러나, 왕겨와 미강의 전자공여능은 조사된 범위 내에서 microwave처리에 대해 큰 변화를 보이지 않았다. 활성산 소종($H_2O$$_2$,O$_2$$^{-}$, .OH)을 발생시켜 어유의 과산화에 대한 왕겨와 미강의 과산화 억제력을 조사한 결과, 전반적으로 왕겨가 미강보다 microwaye 처리에 안정함을 보였다 특히, $H_2O$$_2$에 대한 왕겨의 과산화 억제력은 미강에 비하여 매우 높게 측정되었다. 이상의 결과로 왕겨가 미강보다 항산화 물질을 많이 함유함으로써 미강보다 높고 안정한 항산화능을 나타내 고, 또한 microwave 처리로 인하여 항산화 물질이 일부 파괴되지만 항산화력을 가지는 갈변 반응 생성물이 생성되어 왕겨의 경우에는 microwave 처리에 대해 영향을 거의 받지 않 음을 알 수 있었다. 따라서, microwave 처리 공정은 왕겨나 미강의 항산화능을 이용한 가공 방법에 이용될 수 있음을 의미한다.
