Effects of Ginseng Saponins on the Induction of Differentiation in Mammary Epithelial Cells and Mammary tumor Cells (홍삼 사포닌에 의한 유선상피 및 유선암세포의 분화 유도 효과 연구)

  • Using Ginseng saponins (crude saponin and total saponin) and ginsenoside Rbl Rb2, Rc, Rd, Re, Rhl, and Rh2 in this study, we have examined the effects of the compounds on the induction of differentiation in normal rat mammary epithelial cells and 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary tumor cells in culture. When normal rat mammary organoids were cultured in 100-mm culture plates in the presence or absence of ginseng saponins, there were four different cell colonies after two weeks in culture: cobble stone, spindle, honey comb, and senescence type colonies. Ginseng saponins showed different effects on the development of each colonies. Scrape-loading dye transfer tech-nique was performed to measure the effects of total saponin, Rhl, and Rh2 on intercellular junctional communication. Intercellular communication was not observed at short cultilral time, e.g., four or seven days, but when it cultured it up to two weeks, cell to cell communication was observed in saponin-treated cells. Reconstituted basement membrane, Matrigel, supported the growth and development several different multicellular structures from normal mammary organoids (e.g., ductal, webbed, stellate, and squamous colonies) or DMBA-induced mammary tumor (e.g., alveolar unit, foamy alveolar unit, squamous metaplasia, lobule-ductal, stellate, and webbed colony). In ginseng saponin-treated groups, webbed colonies were more and squamous colonies were less than control group. Moreover, the ductal colonies, marker tructure of well-differentiate mammary epithelial cells, were developed more in saponin-treated group than in control group. In conclusion, ginseng saponins affected on the differentiation of normal rat mammary epithelial cells and DMBA-induced mammary tumor cells in culture.

ACTIVATION OF H-RAS ONCOGENE IN RAT SALIVARY GLAND TUMORS INDUCED BY DMBA AND IRRADIATION (DMBA 매식과 방사선 조사로 유도된 백서 타액선 종양에서 H-ras 암유전자의 활성화)

  • Cellular transforming genes have been identified in a number of different tumor cell lines and tumor types. A significant number of these oncogenes belong to the ras gene family. The ras gene family consists of three closely related genes:H-ras, K-ras and N-ras which code for a related 21 kDa protein. Mutations in codon 12, 13 and 61 of one of the three ras genes convert these genes into acute oncogenes. The presence of H-ras gene mutations has important prognostic implications in various tumors. Each genomic DNA was isolated from tumors induced by implantation with DMBA, or by treatment with DMBA -implantation/irradiation. When genome DNA was transfected into NIH 3T3 cells and investigated by two-step PCR-RFLP, the fOllowing results were concluded: 1. Transformation foci developed in two groups when the genome DNA of two experimental groups were transfected into NIH 3T3 cells. 2. Transformation efficiency was 0.01-0.02 foci/㎍DNA in the experimental group with the DMBA-implantation, 0.01-0.03 foci/㎍lgDNA in the experimental group with the DMBA-implantation/irradiation according to results of transfection assay. 3. When the point mutation of H-ras gene was investigated by a two-step PCR-RFLP, there was 13.9% (5/36) in the experimental group with the DMBA implantation, 15.4 % (6/39) in the experimental group with the DMBA -implantation/irradiation. 4. The point mutation in codon 12 and 61 of H-ras was 5.6%(2/36) and 8.3%(3/36) in the experimental group with the DMBA implantation. 5. The point mutation in codon 12 and 61 of H-ras gene was 7.7%(3/39) in the experimental group with the DMBA -implantation/irradiation.

Human Risk Assessment of a Contaminated Site Using Korean Risk-Based Corrective Action (K-RBCA) Software (한국형 소프트웨어를 이용한 유류.중금속 복합오염지역의 인체위해성평가 및 RBCA Tool Kit과의 비교분석)

  • By using a newly developed Korean risk-based corrective action (K-RBCA) software (K-RBCA) and the RBCA Tool Kit, risk assessment was performed on a site that was contaminated with aromatic hydrocarbons and heavy metals. Eight chemicals including benzene, ethylbenzene, xylenes, naphthalene, benz(a) anthracene, benzo(b) fluoranthene, benzo(a) pyrene, and arsenic that exceeded the US EPA Soil Screening Level were chosen as the target pollutants. A conceptual site model was constructed based on the site-specific effective exposure pathways. According to the RBCA Tool Kit the carcinogenic risk of arsenic was larger than $10^{-6}$, which is the generally acceptable carcinogenic risk level. The K-RBCA estimated the same level of carcinogenic risk for arsenic. With the RBCA Tool Kit, the carcinogenic risk of benzo(a) pyrene was estimated to be about $1.3{\times}10^{-6}$. However, with the K-RBCA benzo(a) pyrene did not exhibit any risk. The inconsistency between the softwares was attributed to the different fundamental settings (i.e., medium division) between the two softwares. While the K-RBCA divides medium into surface soil, subsurface soil, and groundwater, the RBCA Tool Kit divides medium into only soil and groundwater. These differences lead to the different exposure pathways used by the two softwares. The K-RBCA considers the exposure pathways in surface soil and subsurface soil separately to estimate risk, however, the RBCA Tool Kit considers the surface soil and subsurface soil as one and uses the integrated exposure pathways to estimate risk. Thus the resulting risk is higher when the RBCA Tool Kit is used than when the K-RBCA is used. The results from this study show that there is no significant difference in the risks estimated by the two softwares, thus, it is reasonable to use the K-RBCA we developed in risk assessment of soil and groundwater. In addition, the present study demonstrates that the assessor should be familiar with the characteristics of a contaminated site and the assumptions used by a risk assessment software when carrying out risk assessment.

Exposure to PAHs and VOCs in Residents near the Shinpyeong·Jangrim Industrial Complex (신평·장림 산단 인근 주민의 PAHs 및 VOCs 노출)

  • Objectives: This study aims to investigate the atmospheric concentration of polycyclic aromatic hydrocarbons (PAHs) and volatile organic compounds (VOCs) and the urinary concentration of biomarkers in residents near the Shinpyeong·Jangrim Industrial Complex to compare them with those of residents in a control area. Methods: Hazardous air pollutants (PAHs and VOCs) were measured in an exposure area (two sites) and a control area (one site). Urine samples were collected from residents near the industrial complex (184 persons) and residents in the control area (181 persons). Multiple linear regression analysis was used to identify which factors affected the concentration of PAHs and VOCs metabolites. Results: The average atmospheric concentration of PAHs in Shinpyeong-dong and Jangrim-dong was 0.45 and 0.59 ppb for pyrene, 0.15 and 0.16 ppb for benzo[a]pyrene, and 0.29 and 0.35 ppb for dibenz[a,h]anthracene. The average atmospheric concentration of VOCs was 1.10 and 0.99 ppb for benzene, 8.22 and 11.30 ppb for toluene, and 1.91 and 3.05 ppb for ethylbenzene, respectively. The concentrations of PAHs and VOCs in residents near the Shinpyeong·Jangrim Industrial Complex were higher than those of residents in the control area. Geometric means of urinary 2-hydroxyfluorene, 1-hydroxypyrene, methylhippuric acid, and mandelic acid concentrations were 0.45, 0.22, 391.51, and 201.36 ㎍/g creatinine, respectively. Those levels were all significantly higher than those in the control area (p<0.05). In addition, as a result of multiple regression analysis, even after adjusting for potential confounding factors such as gender and smoking, the concentration of metabolites in urine was high in residents near the Shinpyeong·Jangrim Industrial Complex. Conclusion: The results of this study show the possibility of human exposure to VOCs in residents near the Shinpyeong·Jangrim Industrial Complex. Therefore, continuous monitoring of the local community is required for the management of environmental pollutant emissions.

Contamination Investigation and Risk Characterization on the Polycyclic Aromatic Hydrocarbon of Agricultural Products Used for Food and Medicine (식·약공용 농·임산물의 다환방향족탄화수소 오염도 조사 및 위해도 결정)

  • Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.

Ecological Risk Assessment of Residual Petroleum Hydrocarbons using a Foodweb Bioaccumulation Model (먹이연쇄 생물축적 모형을 이용한 잔류유류오염물질의 생태위해성평가)

  • Residual petroleum hydrocarbons after an oil spill may accumulate in the marine benthic ecosystem due to their high hydrophobicity. A lot of monitoring data are required for the estimation of ecosystem exposure to residual petrochemicals in an ecological risk assessment in the affected region. To save time and cost, the environmental exposure to them in the affected ecosystem can also be assessed using a simple food-web bioaccumulation model. In this study, we evaluated residual concentrations of four selected polycyclic aromatic hydrocarbons (phenanthrene, anthracene, pyrene, and benzo[a]pyrene) in a hypothetic benthic ecosystem composed of six species under two exposure scenarios. Body-residue concentration ranged 5~250 mg/kg body depending on trophic positions in an extreme scenario in which the aqueous concentrations of PAHs were assumed to be one-tenth of their aqueous solubility. In addition, bioconcentration factors (BCFs) and bioaccumulation factors (BAFs) were evaluated for model species. The logarithm of bioconcentration factor (log BCF) linearly increased with increasing the logarithm of 1-octanol-water partition coefficient (log $K_{OW}$) until log $K_{OW}$ of 7.0, followed by a gradual decrease with further increase in log $K_{OW}$ without metabolic degradation. Biomagnification became significant when log $K_{OW}$ of a pollutant exceeded 5.0 in the model ecosystem, indicating that investigation of food-web structure should be critical to predict biomagnifications in the affected ecosystem because log $K_{OW}$ values of many petrochemicals are higher than 5.0. Although further research is required for better site-specific evaluation of exposure, the model simulation can be used to estimate the level of the ecosystem exposure to residual oil contaminants at the screening level.

Dietary Risk Assessment for Polycyclic Aromatic Hydrocarbons in Foods (식품중 Polycyclic Aromatic Hydrocarbons의 위해성평가)

  • This study was executed to determine the cumulative dietary risk of PAHs exposed by food ingestion. Food samples including barbecued beef, barbecued pork, grilled chicken, ham, bacon and vegetable oil which were collected from food markets. These samples were saponified, extracted and cleaned up to purify PAHs, and then the purified sample solutions were analyzed by HPLC-FL. Generally, the levels of total PAHs in barbecued beef (0.2 ppb), bacon (0.3 ppb), barbecued pork (0.7 ppb), ham (0.8 ppb), and vegetable oil (1.2 ppb) were low, whereas the level of total PAHs in grilled chicken (9.3 ppb) was significantly high. For the exposure assessment of PAHs due to food ingestion, PAHs levels converted into TEQ$_{BaP}$, the average body weight for 20-73 age group and consumed levels of food proposed from report on the National Health and Nutrition Survey were used. The estimated lifetime average daily intake of dietary PAHs was 4.32${\times}$10$^{-4}$ $\mu\textrm{g}$-TEQ$_{BaP}$kg/day as the mean value. The dietary risk adjusted to cancer potency of benzo(a)pyrene as 7.3 (mg/kg/day)$^{-1}$ was 3.44${\times}$10$^{-6}$ based on current data.ata.

Study on the Screening System of Organic Resources for Agricultural Utilization (유기성 자원의 농업적 활용을 위한 선별체계 연구)

  • This study was conducted to find suitable methods for screening organic resources useful for compost. Twenty-seven industrial and domestic sludges were collected from various cities and industrial areas. Contents of organic matters in the sludges were in the range of 79.3-98.0%, and the contents were much higher than the regulation level (60%) for raw materials of compost. Contents of total nitrogen were in the range of 0.8-2.6%. Contents of Fe and Al were very high. Content of HEM was highest in textile sludge ($257mg\;kg^{-1}$) and the contents in the others were in the range of $12.6-90.3mg\;kg^{-1}$. Content of PAHs was lowest in food sludge ($739.1{\mu}g\;kg^{-1}$ and pulp-mill sludge had the highest PAHs content ($3461.8{\mu}g\;kg^{-1}$). $Microtox^{(R)}$ $EC_{50}$ values were higher in the sludges which were classified as a possible material in composting after analysis and investigation. Lettuce root elongation and $EC_{50}$ values were relatively lower in pulp-mill sludge, sewage sludge 3 (Large city), food sludge and leather sludge. Therefore, mineral nutrients, heavy metals, organic compounds (HEM, PAHs, PCBs), and bioassay ($Microtox^{(R)}$ $EC_{50}$, Relative root elongation test) are recommended to be included in the screening system of raw material of compost in addition to the current regulation with organic matter and 8 heavy metals.
