Relationship of Phenolic Compounds and Free-radical Scavenging Activity in Black and Red Rice Extract

  • Total phenolic content, total anthocyanin content and antioxidant activity were analyzed from rice samples collected in Korea, Japan and China. The results showed that the total phenolic content and free-radical scavenging activity differed significantly in these rice lines. The correlation between content and activity was subsequently investigated. The results showed that in black rice, anthocyanin was the major phenolic component and that both phenolic content and anthocyanin content were closely correlated with free-radical scavenging activity. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) data showed that cyanidin-3-O-glucoside and peonidin-3-O-glucoside composed about 90% of the total anthocyanin content in black rice and in red rice. In the red rice extract, the total phenolic content produced a high correlation coefficient with antioxidant activity but correlated very poorly with the total anthocyanin content. The $OD_{458}$ and the $OD_{500}$ values which represent the proanthocyanidin content of the rice extract, produced high correlation coefficients with antioxidant activity and total phenolic content. These results suggest that the $OD_{458}$ and the $OD_{500}$ values can be used to evaluate the quality of red rice. In addition, based on the data obtained, a competitive accumulation model of anthocyanin and proanthocyanidin in black and red rice was proposed.

Effects of Sucrose level and Nitrogen Source on Fresh Weight and Anthocyanin Production in Cell Suspension Culture of 'Sheridan' Grape (Vitis spp.)

  • To establish an in vitro mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of nitrogen source and sucrose on fresh weight and anthocyanin production in cell suspension culture of 'Sheridan' grape level were studied. When the medium was devoid of $NO_3^-$, cell fresh weight was either remained stable (1% sucrose) or slightly decreased with culture time (2,3, and 4% sucrose). When $NH_4^-$ was lacking, 3% sucrose was most favorable for cell growth. When $NH_4^-$ was supplied as N source, the anthocyanin content of 2% sucrose containing medium was maintained 2 times higher than other levels till day 8 in culture, then that of 3 and 4% sucrose which peaked at day 12 thereafter. The anthocyanin content was low than $NO_3^-$-free media. Total anthocyanin content in $NH_4^-$-free medium was just about a half of that of $NH_4^+$ medium. Anthocyanin production of 2% sucrose in $NH_4^+$ medium was maintained about 3-fold till day 8, then decreased thereafter. In $NH_4^+$ medium, pH decreased gradually with final pH of 3.5 to 4.0, while pH in $NH_4^+$-free medium increased with final pH of 6.5 to 7.5.

Silage Fermentative Quality and Characteristics of Anthocyanin Stability in Anthocyanin-rich Corn (Zea mays L.)

  • The fermentative quality and quantitative change in anthocyanin of anthocyanin-rich corn (Zea mays L.) during storage and in vitro ruminal fermentation were studied. The anthocyanin-rich corn silages in bag silo, drum silo and round bale had good fermentative qualities, such as low pH (5% DM) and butyric acid-free, and its quality was maintained for more than 370 d. The amount of anthocyanin in the anthocyanin-rich corn decreased after ensiling by about 45% (from 3.34 to 1.88 mg/g DM), but stayed constant after day 60. The in vitro incubation of the anthocyanin-rich corn with ruminal fluid revealed little degradation of anthocyanin. These results indicate that the anthocyanin had no negative effect on silage fermentation, and the anthocyanin-rich corn silage is utilizable for practical use as a feedstuff. Our results also demonstrate alteration of the anthocyanin content during storage, and show that anthocyanin-rich corn is a suitable antioxidant source for ruminants because of the high stability of the anthocyanin in ruminal fluid.

적갓김치 Anthocyanins의 항산화 특성에 관한 연구 (Antioxidative Activities of Anthocyanins in Red Mustard Leaf Kimchi)

  • 재래종 적갓(Brassica juncea czerniak et coss)을 주재료로 담금하여 발효시킨 적갓김치(red mustard leaf kimchi, RML kimchi)의 crude anthocyanin(CA) 및 refined anthocyanin(RA) 획분들에 대한 항산화성을 규명하였다. 발효된 적갓김치는 산도 0.72(%), pH 4.6 그리고 crude anthocyanin 함량(건조물기준)은 49.4 mg/g이었다. 동결건조 적갓김치로부터 얻어진 조색소용액 (crude pigment solution)을 계통 추출분획하여 증류수 획분, methanol 획분, TFA-methanol (CA) 획분을 얻었으며, CA 획분을 계속 분획하여 정제된 5개의 정제된 RA 획분(Fr.1, Fr.2, Fr.3, Fr,4, Fr.5)을 얻었다. 이들의 항산화 활성을 linoleic acid의 autoxidation system에서 peroxide formation 억제능과 DPPH($\alpha$, $\alpha$'-diphenyl-$\beta$-picrylhydrazyl)의 free radical 소거능에 대하여 실험하였다. 그 결과 조색소용액 분획물 중 CA 획분이 항산화능이 높았고, CA 획분으로부터 얻어진 RA 획분 중 Fr.3이 가장 높은 항산화성을 보였다. 특히 Fr.3은 적갓김치의 대표적인 anthocyanin이었고 항산화 활성은 $\alpha$-tocopherel보다는 낮았으나 그것에 접근하는 유사한 특성을 보였다. 그리고 그 활성은 지질의 과산화 억제능과 수소공여성에서 다같이 비슷한 수준의 항산화성을 보였다.

Effect of Nitrogen Source on Cell Growth and Anthocyanin Production in Callus and Cell Suspension Culture of 'Sheridan' Grapes

  • To establish in vitro mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of nitrogen amount and the ratio of $NO_3^-$/$NH_4^+$ in the medium on cell growth and anthocyanin production were investigated. Total nitrogen amount and the ratio of $NO_3^-$/$NH_4^+$ in the medium strongly affected anthocyanin production and cell growth. When $NH_4^+$ was fixed, the cell growth was promoted by 50 mM total nitrogen (20 mM $NO_3^-$ : 30 mM $NH_4^+$ ) than other nitrogen combinations, and was strongly inhibited when $NO_3^-$ was lacking (0 mM $NO_3^-$ : 60 mM $NH_4^+$ ) while anthocyanin production was increased. When $NO_3^-$ was fixed, the cell growth was promoted by 70 mM total nitrogen (40 mM $NO_3^-$ : 30 mM $NH_4^+$) than other nitrogen combinations, and was strongly inhibited when $NO_3^-$ was lacking (0 mM $NO_3^-$ : 60 mM $NH_4^+$ ) while anthocyanin production was increased. Cell growth was gradually increased by all nitrogen combinations, but anthocyanin production reached its peak on day 4 in culture. Anthocyanin content increased with decreasing cell density. Sucrose was rapidly hydrolyzed to fructose and glucose within 4 days. Glucose and fructose concentrations in the medium increased and peaked at the 4th day. The anthocyanin content of $NH_4^+$-free 2% sucrose media was 2 times (200 $\mu\textrm{g}$/g) higher than that of 1% sucrose. When $NO_3^-$ was lacking, the highest anthocyanin production was observed at 4% sucrose after 12 days of culture, and increased along with the sucrose concentration.

Black Raspberry 침출액의 Anthocyanin 및 Polyphenol 함량과 항산화능 (Anthocyanins, Polyphenols and Antioxidant Activities of Black Raspberry Exudates)

  • Black raspberry 과실을 원형으로 사용하거나, 손으로 으깨거나, 녹즙기로 갈아서 20%, 40%, 60%, 80% ethanol에 담가(용액 900 mL 당 과실 125 g) 30, 60, 90일 동안 침출한 후 각 침출액에 대한 monomeric anthocyanin 함량, polyphenol 함량, DPPH와 ABTS free radical scavenging activity를 측정하였다. 과실의 형태와 상관없이 20% ethanol을 사용하여 침출한 경우에 monomeric anthocyanin 함량과 polyphenol 함량이 유의적으로 낮았다. 침출기간이 길어질수록 monomeric anthocyanin 함량과 polyphenol 함량이 감소하는 경향을 보였으며, 50% DPPH와 ABTS free radical scavenging activity는 20% ethanol에서 침출한 경우에 비교적 높아 항산화능이 낮았다. 이러한 연구 결과로 볼 때 black raspberry 침출 시 20% ethanol을 제외한 용매를 사용하고 과실은 으깨거나 마쇄할 필요 없이 원형 그대로 사용하는 것이 효율적이며, black raspberry의 가공 시 침출기간도 중요한 고려사항인 것으로 생각된다.

옥수수 색소 2호 품종의 일반성분, 유리당, 지방산 및 안토시아닌 색소의 함량 (Proximate, Free Sugar, Fatty acids Composition and Anthocyanins of Saekso 2 Corn Kernels)

  • 본 연구는 흑자색 알곡 품종으로 등록된 색소 2호 옥수수 알곡의 일반성분 및 색소 함량을 분석하여 기초자료로 제공하고자 수행되었다. 색소 2호 알곡의 일반성분 분석결과, 수분, 조회분, 조지방, 조단백질은 각각 8.84%, 1.44%, 5.46%, 10.31%이었다. 색소 2호 알곡의 유리당 성분으로 fructose, glucose, maltose 및 sucrose가 검출되었으며, 함량은 sucrose (1.00%) > glucose (0.63%) > maltose (0.52%) > fructose (0.44%) 순이었다. 색소 2호 알곡의 지방산 분석결과 총 11종의 지방산이 검출되었으며 포화 지방산과 불포화 지방산의 비율은 16.09 : 83.91로 linoleic acid가 50.86%로 가장 높게 나타났다. 색소 2호 알곡 추출물의 총 안토시아닌 함량은 0.24%였고, 알곡 추출물을 HPLC-MS/MS로 분석한 결과 색소 2호 알곡에 함유된 안토시아닌 색소는 C-3-G, Pg-3-G, Pn-3-G인 것으로 확인되었다.

Effects of Drying Methods on Anthocyanin Contents of Colored Barley (Hordeum vulgare L.) cv. Boanchalbori

  • This study investigated the effects of drying methods and drying time on the changes in anthocyanin content in colored barley. Colored barley cultivar Boanchalbori was harvested at a time when the anthocyanin content was the most and dried in afield. The harvested barley was then treated by two methods, sun drying and shade drying, for 4, 8, 24, and 32 h. The moisture content of the sun-dried barley decreased slightly faster than shade-dried samples, but the difference was not statistically significant. Chemical analysis indicated that the samples dried under shaded conditions had slightly higher crude fiber and lower nitrogen free extract, but the difference was not statistically significant. There was no difference in the total digestible nutrients between the two methods. In the case of sun-dried barley, the anthocyanin content decreased compared to the control and shade-dried samples after drying for 4 h (p < 0.05), was maintained at a constant level at 24 h, and then decreased at 32 h. In case of shade-dried barley, the anthocyanin content decreased gradually with the drying time, and a significant decrease was found at 24 h of drying (p<0.05) as compared to the control. The shade-dried method was more successful in reducing anthocyanin loss than the sun-dried method (p<0.05). There was a slight decrease in 1,1-Diphenyl-2-Picrylhydrazyl radical scavenging with drying time in the shade-dried method, and a significant decrease after 4 h with the sun-dried method. These results showed that covering with a two-layer awning was advantageous to dry colored barley in the field conditions.

흑미(현진주벼)유래 안토시아닌의 항산화능 탐색 (Isolation of Anthocyanin from Black Rice (Heugjinjubyeo) and Screening of its Antioxidant Activities)

  • 유색미는 회색 빛 외에 붉은색 흑은 보라색 계통의 색소를 겨 부분에 함유하고 있다. 특히, 흑미는 예로부터 건강식품으로 여겨져 왔다. 흑미(흑진주벼)는 최근에 anthocyanin포함하는 생리활성 물질원으로서 관심이 증대되고 있다. Anthocyanin은 수용성의 색소로서 flavonoid계통의 대표 색소로 알려져 왔다. 흑미에 함유되어있는 Anthocyanin으로는 cyanidin 3-O-glucoside와 peonidin 3-O-glucoside, malvidin 3-O-glucoside, pelagonidin 3-O-glucoside, delphinidin 3-O-glucoside등이 알려져 있다. 본 연구에서는, 흑미로부터 amberlite XAD7 chromatography방법을 사용해서 anthocyanin을 추출 후 HPLC와 UV-Vis spectrophotometer를 이용하여 정량분석 및 정성분석을 실시하였다. 그 결과 흑미의 anthocyanin 추출물 중 95%가 cyanidin 3-O-glucoside, 5%가 peonidin 3-O-glucoside임을 확인하였다. 그리고 in vitro상의 여러 가지 실험방법을 통하여 anthocyani의 항산화 효과를 측정하였다. 그 결과 100g/ml 농도의 anthocyanin추출물은 90%이상 linoleic acid emulsion의 과산화를 억제하였고, 55.20%의 DPPH 자유 라디칼 제거효과, 54.96%의 superoxide 자유 라디칼 제거효과, 72.67%의 과산화수소 제거효과를 보였다. 그리고 또한 높은 환원 능력을 확인할 수 있었다. 표준 항 산화 물질로는 BHA, BHT, ${\alpha}-tocopherol$을 사용하였다. 이 결과로 보아 anthocyanin추출물은 새로운 항산화 물질로서 이용될 수 있을 것이다.

흑진미 과피 용매분획물 투여시 당뇨 유발 흰쥐의 혈당 및 혈중 지질함량에 미치는 영향 (Effects of the Fractions of Oryza sativa cv. Heugjinmi on Plasma Glucose and Lipid Levels in Streptozotocin-Induced Diabetic Rats)

  • 본 연구는 streptozotocin으로 유발된 당뇨 흰쥐에게 흑진미 과피에서 추출한 각 용매 분획물을 14일간 경구투여한 후 혈당과 지질함량을 분석하였으며 다음과 같은 결론을 얻었다. 당뇨 대조군과 비교시 모든 용매 분획물 투여군에서 실험동물의 체중이 증가되었으나 유의적인 차이를 보이지는 않았다. 또한 acetone과 anthocyanin 분획물 투여시 실험 14일째에 혈당, 중성지방 및 유리지방산이 당뇨 대조군과 비교시 유의적으로 감소되었으며, 혈장 HDL-cholesterol 함량은 유의적으로 증가하였다. 이같은 결과는 흑진미 과피에 존재하는 특정 생리활성 물질이 실험 당뇨쥐의 혈당 및 혈중 지질함량 감소에 효과가 있음을 보여주고 있다.
