Study on the Conservation and Restoration of Excavated Costume (출토복식의 보존.보수에 관한 연구)

  • This study is about the excavated costume representing fundamental "Conservation and Restoration". It's not a report of a specific case, but it is a guideline that contains the costume and textile of museum collection. It is not research based on experiments; however, this paper is basically consists of various reported-documents. Followings are the conclusions of this study 1. These are the factors that we have to know to prevent the causes of fabric's degradation. -Light, -Humidity and temperature, -Microscopic organism, -Insect and rodent animal, -Air pollution, -Ph, -Handling 2. Basic principles of conservation and restoration follows are: -Select the Reversible method, -Represent the easily distinguishable repaired place, -Should be acted by an expert or people with experience, -Before the restoration, accurate and specified records should be completed, -Procedure, treatment method, and materials used should be recorded prior to restoration, -Should be cared minimally, -Be cautious when using the conservation materials, -When caring, make sure nothing is against the principle of aesthetic, historic, and form of preservation 3. The types of restoration are type of straight or curve, type of hole, type of without warp or weft, type of special part damage something like sleeve, collar, type of form that is severely damaged, and type of separated pieces. 4. The method of restoration is sewing, stitching, and the combination of sewing and stitching. 5. The restoration seams are welt seam, plain seam, flat felled seam, french seam etc. And there are kinds of used-sewing, such as, broad stitching, backstitch, half backstitch, basking, hemming, saddle stitching etc.

Radiographic, MRI and Histopathologic Assessment to Standardize Canine Spinal Cord Injury Model (척수손상 모델 견 표준화를 위한 방사선, 자기공명영상 및 조직 병리 평가)

  • Previous studies could not offer available guideline to decide size of balloon and grade of injury before induction of spinal cord injury (SCI) because grade of SCI was assessed after inserting a catheter and each experimental animal were different in body size and weight as well as in species. This study was performed to provide guideline for standardized SCI model. Eight healthy adult beagle dogs that had 8 mm of spinal canal height were assigned to four groups according to the diameter of balloon and compression time: 4 mm/3hrs, 4 mm/6hrs, 4 mm/12hrs and 6 mm/3hrs group. Radiography was performed to standardize between experimental animal and balloon before selecting balloon diameter to induce SCI. Behaviors outcomes, somatosensory evoked potentials (SEPs), magnetic resonance imaging (MRI) and histopathological examination were evaluated. Behaviors outcomes and SEPs were not available to assess grade of SCI and those only indicate SCI. The damaged area was revealed clear hyperintensity on STIR image and T2WI after induction of SCI. The hyperintense area on MRI was cranially and caudally expanded with increasing of the diameter of balloon or the compression time. Well corresponded to expanding of hyperintense area on MRI, the damaged region and the numbers of caspase-3 and PARP immunoreactive cells were increased on histopathological findings. Therefore, these results will be considered fundamental data to induce standardized SCI model in experimental animal that has various weight and size.

Development of LC-MS/MS Quantitation Method for Ethoxyquin in Fishery Products (수산물 중 에톡시퀸의 LC-MS/MS 정량분석법 개발)

  • Ethoxyquin (EQ, 1,2-dihydro-6-ethoxy-2,2,4-trimethyl-quinoline) is quinoline-based antioxidant used in the animal feed and food industry to protect the raw materials and final products against oxidation. In recent years the use of synthetic antioxidants in fishmeal ingredients carry-over to farmed fish fillets has received increasing attention in food safety. This study was conducted to develop an analytical method to determine EQ in aquatic products. The analytes were confirmed and quantified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the positive ion mode using multiple reaction monitoring (MRM). The sample was extracted with 1 N HCl (in case of flatfish extracted with 1 N HCl containing 10% acetonitrile). Then, solid phase extraction (SPE) was used for the cleanup. Standard calibration curves presented linearity with the correlation coefficient ($r^2$) > 0.99, analyzed at 0.005-0.2 mg/kg concentration. The developed method was validated according to the Codex Alimentarius Commission (CAC) guideline. The limits of quantitation for EQ were 0.01 mg/kg. Average recoveries ranged from 81.3% to 107%. The repeatability of measurements, expressed as the coefficient of variation (CV, %), was below 10%. The analytical method was characterized with high accuracy and acceptable sensitivity to meet CODEX guideline requirements and would be applicable to analyze the EQ residue in aquatic products.

Dietary Intakes, Plasma Levels and Urinary Excretions of Taurine in Adolescents and Adults Residing in Seoul Area (서울지역 청소년 및 성인의 타우린 섭취량, 혈중 농도 및 소변내 배설량에 관한 연구)

  • As diverse physiological functions of taurine have been reported, taurine-containing health drinks and products are marketed worldwide for the treatment of various conditions such as improvements of liver, heart and circulatory functions or as an aid to athletic performance. Although animal studies have shown that taurine is fairly safe when supplemented in the diet for an extended period, the effective dose range of taurine for dietary supplements is in controversy. Reports on dietary taurine intakes have been sparse, and would serve as a guideline for determining an appropriate taurine dosage. The present study was aimed to estimate dietary intake level of taurine using the taurine content database of commonly used food stuffs established recently in our laboratory, and also to evaluate plasma concentration and urinary excretion of taurine in adolescents and adults residing in Seoul area. Dietary taurine intakes of the subjects were 219$\pm$16.9mg/day for 16-19 years old(n=123), 177$\pm$18.1mg/day for adults older than 20 years old(n=123). Male subjects(n=115) consumed 216$\pm$21.1mg of taurine/day, while female subjects(n=131) consumed 181$\pm$14.3mg of taurine/day(p<0.05). The level of dietary taurine intake was positively correlated with the levels of dietary intakes of energy, carbohydrate, total lipids, cholesterol, vitamin A, vitamin B$_1$, niacin, vitamin C, calcium, phosphorous, sodium and potassium at p<0.01, and with dietary intakes of iron and animal lipids at p<0.05, respectively. Plasma taurine concentration of subjects were 135$\pm$5.9$\mu$mol/L, which is considered to be within a normal range for healthy subjects. The subjects excreted 1158$\pm$72.7nmol/ of tarine mg creatinine in their urine, which is approximated as 150-170mg of taurine/24hr urine based on the assumption that 18mg creatinine/kg/day is excreted in the urine of healthy adults, and this would be about 80% of the daily taurine intake observed in the same subjects. Dietary taurine intake level was positively correlated with plasma taurine concentration, as well as with urinary taurine excretion corrected by creatinine excretion at p<0.05. The present study was the first report of taurine intake, and plasma concentration and urinary excretion of taurine in a Korean population so far, and these results would serve as an index for the future study evaluating taurine status in a diverse population within and outside Korea.   • Korea is striving to establish a research ethics system, but there is still a disagreement as to the level of government involvement in research ethics. Therefore, this study analyzed the formation process of research ethics system in the United States and sought to derive implications for Korea's research ethics policy. The analysis of the present status of the research ethics policy and laws of the United States showed that i) the establishment of regulatory system is done only after the principles of the research ethics or the guideline are presented first in the academic fields; ii) the responsibility for research ethics is to be shared by the federal government and the research institute, and the primary responsibility for research is given to the research institute and the secondary responsibility to the government; iii) the government has imposed a minimum extent of strong regulation on unethical research; and iv) it is necessary to extend the responsibility of unethical research from the individual level to the structural level.

Five Year Experience of Preexposure and Postexposure Rabies Prophylaxis in Korean Children at the National Medical Center (소아청소년에서 공수병 예방접종 및 교상 후 조치 현황: 최근 5년간 국립중앙의료원에서의 경험)

  • Purpose : This study aims to investigate preexposure prophylaxis and postexposure prophylaxis of rabies that the National Medical Center (NMC) handled and to check whether appropriate measures were performed according to the recent domestic and overseas guidelines after animal bites. Methods : This study surveyed 41 people who were 18 years or under and received preexposure and postexposure prophylaxis of rabies at the NMC from November 2006 to December 2011. Their medical records were reviewed for their age, gender, the reason for preexposure prophylaxis, the body sites of animal bite, the kind of the animal that bit children, the region where the biting occurred and rabies vaccination and inoculation of immunoglobulin. Results : Eleven children took rabies vaccination for preexposure prophylaxis and 30 children received post exposure prophylaxis of rabies. Of patients who were bitten by unvaccinated animals including wild animals or by animals which were not certain to be vaccinated, 50% (13 of 26 children) received postexposure prophylaxis, while 75% (3 of 4 children) of patients who were bitten by vaccinated animals received postexposure prophylaxis of rabies. Ten of 30 bitten patients knew whether or not the biting animals had received rabies vaccination. Of them, four people (40%) were bitten by animals which had received rabies vaccination. Conclusion : To prevent the occurrence of rabies, people and health care providers need to correctly understand latest guideline for rabies preexposure and postexposure prophylaxis and the information for bitten patient, biting animal and area at bitten by animal should be accurately recorded.

Safety Assessment of Foods Produced Using Recombinant DNA Techniques

  • The introduction of genetically modified crops has raised concerns regarding safety issues over the insertion of foreign genes into plant genomes using recombinant DNA technology. Since 1991 in Japan, 29 foods and 6 food additives have been evaluated, based on the "Guideline for Safety Assessment", before these foods were marketed. The MHW, however, decided that safety assessment of such foods and food additives should be legally imposed. because soon such foods and food additives are expected to circulate globally and a new system for assessing safety of such foods and food additives at a pre-market stage is necessary, in order to avoid the distribution of any genetically modified foods that have had no safety assessment. The MHW published relevant announcements to amend existing regulations on 1 May 2000. "Standards for safety assessment of seed plant" is established based on a concept of substantial equivalence, and applicable to the products which are regarded as equivalent to the existing products used as foods and food additives. The characterization of the food products entails consideration of the molecular characterization. phenotypic and compositional characteristics, key nutrients and toxicants, and toxicity and allergenicity of the introduced proteins, and if there are indications of unintended effects of the modification, whether further safety testing (animal studies etc.) is needed should be considered. Safety and wholesomeness studies with whole foods should be care fully designed in order to avoid nutritional imbalances causing artifacts and uninterpretable results as was the case of Dr. Pusztaiis report. A case study of genetically modified soybeans (glyphosate-tolerant soybeans) on the immune system of rats and mice is shown. Chemical compositions were also compared with those of the non-GM soybeans. The studies failed to detect any differences in immuno-toxic activity.muno-toxic activity.

  • Amphibian population declines globally. Aquatic contamination by organic pollutants including endocrine disrupters has been suspected to the one of the reason for distinction of amphibia which has obligate aquatic life style during larval period. Amphibians have been widely accepted as animal model for the study of endocrine disruption in aquatic ecosystem at molecular as well as individual levels. There are increasing need for toxicological data in amphibians at multiple endpoints for management of contamination and development of safety guideline for important EDs in aquatic media. Alkylphenols have been widely used in agricultural, industrial, and housekeeping activities, contaminating the aquatic media and evoking endocrine disruption in aquatic animals. In this review, we summarized data concerning the endocrine disruption by alkylphenol organic pollutants on amphibians according to route, concentration, terms, and developmental stage of exposure together with mechanism of endocrine disruption.

A Study on Establishment of Technical Guideline of the Installation and Operation for the Efficient Bio-gasification Facility of Pig Manure and Food Waste(II): - Results of the Precision Monitoring - (가축분뇨 병합처리 바이오가스화를 위한 설계 및 운전 기술지침 마련 연구(II) - 정밀모니터링 결과 중심으로 -)

  • The purpose of this study is to provide a design and operation technical guideline for meeting the appropriate design criteria to bio-gasification facilities treating organic wastes. 9 anaerobic digestion facilities which is normally operated during the field survey and 14 livestock manure farms were selected for precision investigation. the physicochemical analysis was performed on the moisture and organic contents, nutrients composition (carbohydrate, fat, protein), volatile fatty acids (VFAs), and nitrogen, etc. Volatile solids (VS) of organic wastes brought into the bio-gasification facilities were 2.81 % (animal manure only) and 5.92 % (animal manure+food waste), respectively. Total solids (TS) reults of samples from livestock farms were 5.6 % in piglets and 11~13 % in other kinds of breeding pigs. The actual methane yield based on nutrients contents was estimated to $0.36Sm^3CH_4/kgVS$ which is equivalent to 72 % of theoretical methane yield value. The optimum mixing ratio depending on the effect of the combined bio-gasification was obtained through the continuous stirred-tank reactor (CSTR) which is operated at different mixing ratio of swine manure and food waste leachate. The range of swine manure and food waste leachate from 60:40 to 40:60 were adequate to the appropriate conditions of anaerobic digestion; less than 100 gTS/, more than alkalinity of 1 gCaCO3/L, C/N ratio 12.0~30.0, etc.

Assessment of Soil and Groundwater Contamination at Two Animal Carcass Disposal Sites (가축 사체 매몰지 주변 토양 및 지하수의 오염도 평가)

  • Outbreak of contagious diseases to livestock animals is becoming prevalent worldwide and consequently, tremendous numbers of the infected or culled stocks are buried on the ground as the most common disposal method. The buried animals can generate a wide range of detrimental components such as leachate, nutrient salts, and pathogenic bacteria, consequently contaminating the surround environment. This implies that regular investigations are required to monitor any possible detrimental environmental aspect occurred around burial sites. Therefore, the current study was conducted to investigate whether the soil and groundwater nearby the burial sites had been contaminated by the substances originated from the burial sites, which can be applied for the establishment of the ideal burial site construction design and post management scheme. For this, two different burial sites located in Cheonan and Pyeongtaek were selected. Cheonan and Pyeongtaek sites were constructed in 2004 and 2008, respectively and both contained dead poultry infected by avian influenza (AI). Soil and groundwater samples were collected around the sites followed by determination of the nutrient concentrations and bacteria (Salmonella, Camphylobacter, and Bacillus) existence in both soil and groundwater. Some of the soil samples showed higher EC, $NH_4$-N, $NO_3$-N concentration compared to those of the background (control) soils. Also the concentration of $NH_4$-N in some of the groundwater samples appeared to exceed the USEPA guideline value for drinking water (10 mg $L^{-1}$). These results indicated that the soil and groundwater were influenced by the burial site originated nutrients. In the soil, Bacillus was isolated in most soil samples while there were no detections of Salmonella and Camplylobacter. Due to the Bacillus existing mainly as a spore in the soils, it was considered that the frequent detection of Bacillus in the soil samples was attributed to the nutrients originated from the burial sites.
