The Possibility of TBC1D21 as a Candidate Gene for Teat Numbers in Pigs

  • Based on a quantitative traits locus (QTL) study using a $F_2$ intercross between Landrace and Korean native pigs, a significant QTL affecting teat numbers in SSC7 was identified. The strong positional candidate gene, TBC1D21, was selected due to its biological function for epithelial mesenchymal cell development. Sequence analysis revealed six single nucleotide polymorphisms (SNPs) in the TBC1D21 gene. Among these, two SNP markers, one silent mutation (SNP01) for g.13,050A>G and one missense mutation (SNP04) for c.829A>T (S277C), were genotyped and they showed significant associations with teat number traits (p value = 6.38E-05 for SNP01 and p value = 1.06E-07 for SNP04 with total teat numbers). Further functional validation of these SNPs could give valuable information for understanding the teat number variation in pigs.

논에서 우분액비시용이 총체 벼의 생산성, 사료가치 및 수질오염에 미치는 영향 (Effect of Application of Cattle Slurry on Dry Matter Yield and Nutritive Value of Whole Crop Rice and Water Pollution in Rice Paddy Land)

  • 본 연구에서는 논에서의 조사료용 총체 벼 생산을 위해 우분액비의 시용효과를 규명하기 위하여 총체 벼 화학비료 시용구를 대조구로 하고 우분액비 시용구를 처리구로 하여 총체 벼 생육특성, 생산성 및 사료가치를 조사하였다. 그리고 처리구별 토양의 지력증진 효과를 분석함과 동시에 화학비료 및 우분액비의 시용에 따른 배출수와 용탈수의 $NO_3$-N, $NH_4$-N 및 $PO_4$-P 등을 분석하여 환경영향평가를 실시하고자 수행하였다. 본 연구는 2006년 5월부터 2009년 4월까지 3년 동안 전라북도 김제군 백산면 시험포장에서 완전임의배치 3반복으로 수행되었다. 화학비료 시용구의 총체 벼의 수량은 우분액비 시용구와 비슷한 수준을 보였다. 총체 벼의 조단백질 함량은 화학비료 시용구보다 우분액비 시용구에서 현저하게 증가하였으나 (p<0.05) NDF, ADF 및 TDN 함량은 처리구간에 큰 차이를 보이지 않았다. 우분액비시용에 따른 토양내 pH, $P_2O_5$, T-N, OM 농도는 시험 전에 비해 시험종료 후에 증가하는 경향을 나타냈다. 그리고 우분액비시용에 따른 토양내 Ca, Na, Mg 및 K 농도는 시험 전에 비해 시험종료 후에 현저하게 증가하는 경향을 나타냈다. 총체 벼 화학비료 시용구 및 우분액비 시용구에서 배출되는 성분중 $NO_3$-N 및 $NH_4$-N 농도는 2007년에 비하여 2008년에 현저하게 높은 경향을 보였으나 배출수중 $PO_3$-P 농도는 2년간 모두 비슷한 수준을 유지하였다. 그리고 처리구간에도 농도차이는 나타나지 않았다. 우분액비시용에 따른 용탈수중 $NO_3$-N, $NH_4N$$PO_4$-P의 농도는 큰 차이를 보여주지 않았다.

  • 본 연구에서는 논에서의 조사료 생산을 위한 작부형태별 우분액비의 시용효과를 규명하기 위하여 총체보리+수수$\times$수단그라스 교잡종 이모작구 그리고 헤어리베치를 녹비작물로 이용한후 수수$\times$수단그라스 교잡종 일모작구로 하여 논에서의 다양한 조사료 작부 형태를 배치하고, 수수$\times$수단그라스 교잡종의 생산성 및 사료가치를 조사하였다. 그리고 처리구별 토양의 지력증진 효과를 분석함과 동시에 우분액비의 시용과 경작형태에 따른 용탈수중 $NO_3$-N, $NH_4$-N 및 $PO_4$-P 등을 분석하여 환경오염에 미치는 영향을 조사하였다. 본 연구는 수수$\times$수단 그라스 교잡종의 생산성과 환경에 미치는 영향을 평가하고자 2006년 5월부터 2009년 4월 까지 3년 동안 전라북도 김제군 백산면 시험포장에서 완전임의배치 3반복으로 수행되었다. 수수$\times$수단그라스 교잡종은 총체보리와 이모작의 경우가 헤어리베치를 이용한 일모작에 비해 수량이 현저하게 증가하였다(P<0.05). 그리고 수수$\times$수단그라스 교잡종의 조단백질, NDF, ADF 및 TDN 함량은 처리구간에 큰 차이는 나타나지 않았다. 총체보리와 이모작구에서의 OM, T-N 및 $P_2O_5$ 함량은 녹비작물로 이용된 일모작구에 비해 높았다. 그리고 우분액비시용에 따른 토양 내 pH와 OM 농도는 시험전에 비해 시험종료 후에 현저하게 증가하는 경향을 나타냈다. 또한 이모작구와 일모작구에서 T-N 함량은 시험전에 비해 시험종료 후에 현저하게 증가하는 경향을 나타냈다(P<0.05). 경작형태별 우분액비 시용에 따른 토양내 양이온치환용량 ($Ca^{2+}$, K, Mg 및 Na) 농도는 시험 전에 비해 시험종료 후에 현저하게 증가하는 경향을 나타냈다(P<0.05). 또한 경작형태별 우분액비 시용에 따른 용탈수중 $NO_3$-N, $NH_4$-N 및 $PO_3$-P의 농도는 경작형태에 따른 확실한 차이를 보여주지 않았다.

AndroMed를 이용한 흑우 동결 정액으로 체외수정란 생산 효과 (Effect of Production In Vitro Embryo with Frozen-thawed Semen using AndroMed Extender in Korean Black Cow Semen)

  • The aim of present experiment was to examine commercial synthetic extender(AndroMed) for semen cryopreservation of Korean Black Bull. Semen was collected from a Korean Black Bull using an artificial vagina and transported to the laboratory. The semen was diluted 1:1 by AndroMed. The pellect was diluted to final sperm concentration of $5{\times}10^5/ml$ by doubling in every 10 minutes at $4^{\circ}C$ cold chamber. The semen was equilibrated for 1 hr at cold chamber and packed to 0.5 ml straw. The semen straws were located above 5 cm of liquid nitrogen for 5 minutes, above 5 cm for 10 minutes and above 10 cm for 10 min. And then the frozen straw was plunged to $LN_2$. The presented straws were examined the viability and motility after thawed at $37^{\circ}C$ water bath. Hanwoo semen was used as KPN (Korea Proven Bull Number) in this experiment. The survival rates was significantly higher in fresh semen than frozen semen ($80{\pm}14%\;and\;43{\pm}11%$). However, the motility rates was similar (80.7% and 66.4%). The survival and motility rates were higher in 5cm, 10 min treatment group than the other two groups in straw-located height and duration above $LN_2$ ($50{\pm}14%$ and 70.7% vs, 33.18% and $65{\pm}7%$ vs, 30.14% and 65.7%, respectively). The development rates to cleavage was higher in Black Cow than Hanwoo semen (62.2%, 64.4%), However, The development rates to blastocyst was higher in Hanwoo than Black cow semen (25.9%, 23.0%). In conclusion. The present results that acceptable fertilization and cryopreservation could be obtained by in vitro fertilization with frozen-thawed semen using a synthetic semen extender (AndroMed).

Effects of Ecklonia cava as fucoidan-rich algae on growth performance, nutrient digestibility, intestinal morphology and caecal microflora in weanling pigs

  • Objective: In the present study, role of increasing levels of Ecklonia cava (seaweed) supplementation in diets was investigated on growth performance, coefficient of total tract apparent digestibility (CTTAD) of nutrients, serum immunoglobulins, cecal microflora and intestinal morphology of weanling pigs. Methods: A total of 200 weaned pigs (Landrace${\times}$Yorkshire${\times}$Duroc; initial body weight $7.08{\pm}0.15kg$) were randomly allotted to 4 treatments on the basis of body weight. There were 5 replicate pens in each treatment including 10 pigs of each. Treatments were divided by dietary Ecklonia cava supplementation levels (0%, 0.05%, 0.1%, or 0.15%) in growing-finishing diets. There were 2 diet formulation phases throughout the experiment. The pigs were offered the diets ad libitum for the entire period of experiment in meal form. Results: The pigs fed with increasing dietary concentrations of Ecklonia cava had linear increase (p<0.05) in the overall average daily gain, however, there were no significant differences in gain to feed ratio, CTTAD of dry matter and crude protein at both phase I and phase II. Digestibility of gross energy was linearly improved (p<0.05) in phase II. At day 28, pigs fed Ecklonia cava had greater (linear, p<0.05) Lactobacillus spp., fewer Escherichia coli (E. coli) spp. (linear, p<0.05) and a tendency to have fewer cecal Clostridium spp. (p = 0.077). The total anaerobic bacteria were not affected with supplementation of Ecklonia cava in diets. Polynomial contrasts analysis revealed that villus height of the ileum exhibited a linear increase (p<0.05) in response with the increase in the level of dietary Ecklonia cava. However, villus height of duodenum and jejunum, crypt depth, villus height to crypt depth ratio of different segments of the intestine were not affected. Conclusion: The results suggest that Ecklonia cava had beneficial effects on the growth performance, cecal microflora, and intestinal morphology of weanling pigs.

The Effects of Rhodobacter capsulatus KCTC-2583 on Cholesterol Metabolism, Egg Production and Quality Parameters during the Late Laying Periods in Hens

  • An experiment was conducted to investigate the effects of dietary supplementation of Rhodobacter capsulatus KCTC-2583 on egg-yolk and serum cholesterol, egg production and quality parameters during the late laying periods in hens. A total of 160 Hy-Line Brown layers (54 wk-old) were randomly allotted to 4 treatment groups on the basis of laying performance. Each treatment had 4 replicates with 10 birds each (40 birds per treatment). Two hens were confined individually with cage size $35{\times}35{\times}40$ cm and each 10 birds (5 cages) shared a common feed trough between them forming one experimental unit. Dietary treatments were; basal diet supplemented with 0 (control), 0.05, 0.10 and 0.15% R. capsulatus KCTC-2583. Experimental diets were fed in meal form for 56 d. Dietary supplementation of increasing levels of R. capsulatus KCTC-2583 reduced (linear, p<0.05) egg-yolk cholesterol and triglycerides (d 28, 42 and 56) concentrations. Also, serum cholesterol and triglycerides (d 21, 42 and 56) concentrations were linearly reduced (p<0.05) with increasing dietary R. capsulatus KCTC-2583. Laying hens fed a diet supplemented with increasing levels of R. capsulatus KCTC-2583 had increased (linear; p<0.05) overall egg production, egg weight, egg mass and feed efficiency. However, dietary treatments had no effect (linear or quadratic; p>0.05) on feed intake of laying hens. At d 28 and 56, breaking strength and yolk colour of eggs were linearly improved (p<0.05) in laying hens fed dietary increasing levels of R. capsulatus KCTC-2583. Dietary treatment had no effects (linear or quadratic; p>0.05) on albumin height, shell thickness and shell weight at any period of experiment. These results indicate that dietary supplementation of R. capsulatus KCTC-2583 has the potential to improve the laying hen performance and lead to the development of low cholesterol eggs during late laying period in Hy-Line Brown hens.

Association of Microsatellite Marker in FABP4 Gene with Marbling Score and Live Weight in Hanwoo

  • The bovine fatty acid binding protein 4 (FABP4) plays an important role to uptake intracellular fatty acid. It has been previously reported as a positional candidate gene for marbling score in livestock. The re-sequencing of FABP4 gene detected a polymorphic AT repeated sequence in intron II of FABP4 gene. Allelic distribution for this microsatellite marker was examined in other cattle breeds. A total of 8 alleles were detected with diverse repeat units (14 to 21 AT repeat) in Hanwoo and 7 breeds. Of the 8 alleles, the predominant alleles were $[AT]_{16}$, $[AT]_{18}$ and $[AT]_{19}$ in the Hanwoo and 7 cattle breeds. The linear mixed model for genotypic effect (3237AT) on carcass traits showed a significant effect on marbling score (MAR P=0.025) and live weight (LWT; P=0.04) in the 583 Hanwoo cattle population. Live weight (LW) was highest in the homozygous $(AT)_{17}$ genotype ($557.5{\pm}6.94$) and lowest in the heterozygous $(AT)_{16/17}$ genotype ($521.7{\pm}7.70$). On the other hand, the homozygous $(AT)_{17}$ genotype ($3.0{\pm}0.15$) has the highest effect on marbling score and the lowest effect was in homozygous (AT)$_{18}$ genotype ($2.2{\pm}0.15$). The marbling score difference between both groups was 0.8 which is around two times higher than SNP genotype effect on marbling score in Limousin $\times$ Wagyu crosses.

Metabolizable energy requirement for maintenance estimated by regression analysis of body weight gain or metabolizable energy intake in growing pigs

  • Objective: Feed energy required for pigs is first prioritized to meet maintenance costs. Additional energy intake in excess of the energy requirement for maintenance is retained as protein and fat in the body, leading to weight gain. The objective of this study was to estimate the metabolizable energy requirements for maintenance ($ME_m$) by regressing body weight (BW) gain against metabolizable energy intake (MEI) in growing pigs. Methods: Thirty-six growing pigs ($26.3{\pm}1.7kg$) were allotted to 1 of 6 treatments with 6 replicates per treatment in a randomized complete block design. Treatments were 6 feeding levels which were calculated as 50%, 60%, 70%, 80%, 90%, or 100% of the estimated ad libitum MEI ($2,400kJ/kg\;BW^{0.60}\;d$). All pigs were individually housed in metabolism crates for 30 d and weighed every 5 d. Moreover, each pig from each treatment was placed in the open-circuit respiration chambers to measure heat production (HP) and energy retained as protein ($RE_p$) and fat ($RE_f$) every 5 d. Serum biochemical parameters of pigs were analyzed at the end of the experiment. Results: The average daily gain (ADG) and HP as well as the $RE_p$ and $RE_f$ linearly increased with increasing feed intake (p<0.010). ${\beta}$-hydroxybutyrate concentration of serum tended to increase with increasing feed intake (p = 0.080). The regression equations of MEI on ADG were MEI, $kJ/kg\;BW^{0.60}\;d=1.88{\times}ADG$, g/d+782 ($R^2=0.86$) and $ME_m$ was estimated at $782kJ/kg\;BW^{0.60}\;d$. Protein retention of growing pigs would be positive while REf would be negative at this feeding level via regression equations of $RE_p$ and $RE_f$ on MEI. Conclusion: The $ME_m$ was estimated at $782kJ/kg\;BW^{0.60}\;d$ in current experiment. Furthermore, growing pigs will deposit protein and oxidize fat if provided feed at the estimated maintenance level.

Effect of L- or DL-methionine Supplementation on Nitrogen Retention, Serum Amino Acid Concentrations and Blood Metabolites Profile in Starter Pigs

  • The objective of the current study was to evaluate the effect of supplementation of either L-methionine (L-Met) or DL-methionine (DL-Met) to diets of starter pigs on nitrogen (N) balance, metabolism, and serum amino acid profile. Eighteen crossbred ($Duroc{\times}Landrace{\times}Yorkshire$) barrows weighing $15.45{\pm}0.88kg$ were randomly allotted to 1 of 3 diets with 6 pigs per treatment. The diets included a basal diet (Met-deficient diet) containing 0.24% standardized ileal digestibility Met with all other essential nutrients meeting the pig's requirements. The other two diets were produced by supplementing the basal diet with 0.12% DL-Met or L-Met. The experiment lasted for 18 days, consisting of a 13-day adaptation period to the diets followed by a 5-day experimental period. Pigs were fed ad libitum and free access to water throughout the experiment. Results showed that the supplementation of either L-Met or DL-Met improved N retention, and serum methionine concentration, and decreased N excretion compared with basal diet (p<0.01). The N retention of pigs fed diets supplemented with the same inclusion levels of DL-Met or L-Met were not different (p>0.05). In conclusion, on equimolar basis DL-Met and L-Met are equally bioavailable as Met sources for starter pigs.

EFFECT OF SUPPLEMENTING RUMEN-PROTECTED LYSINE ON GROWTH PERFORMANCE AND PLASMA AMINO ACID CONCENTRATIONS IN SHEEP

  • This experiment was carried out to investigate the effect of rumen-protected lysine (RPLys) on growth rate, feed efficiency and plasma amino acid concentrations in sheep. RPLys was supplemented at the level of 0% ($T_1$), 0.2% ($T_2$) and 0.4% ($T_3$) of total DMI with 24 sheep in a 56 day feeding trial. The results are summarized as follows: 1. live weight gain of sheep in groups $T_1$, $T_2$ and $T_3$ was 219, 216 and 244 g/d, and was significantly (p < 0.05) higher for $T_3$ through the entire experiment. 2. Feed intake was not affected by RPLys supplementation. 3. The group fed $T_3$ had a significantly (p < 0.05) better feed efficiency than the groups fed $T_1$ and $T_3$. The response of $T_3$ was higher in growing period II of feeding low protein basal diet than in period I. 4. Plasma lysine concentrations tended to be higher with supplementing RPLys, but there were no differences between $T_2$ and $T_3$. 5. Supplementing RPLys in the diets increased plasma concentrations of arginine, asparagines, threonine, serine, valine and leucine compared with sheep receiving no RPLys. In contrast, plasma histidine was lower in sheep fed the supplementing RPLys than fed the diet $T_1$ with significant (p < 0.05) difference.
