The Effect of Living Conditions on Stress and Behavior of Horses

  • Providing an adequate environment for horses is important to minimize the level of stress for domesticated horses. The objectives of this study were 1) to evaluate the effect of living conditions on stress level of horses, 2) to observe the effect of one month confinement on self-maintenance behavior and stereotypic behavior of horses. The experiment was conducted at National Institute of Animal Science, Equine Field Station (Seonghwan-eup, Korea). Horses were staying in the paddock prior to the experiment. On day 1, five horses were randomly selected and housed in metal fence panels stall. Six horses remained in the same paddock. The ratio of neutrophil to lymphocyte (on day 15) and cortisol (on day 1 and 29) from stalled horses were significantly higher than horses in the paddock. Duration or frequency of self-maintenance behaviors such as feeding, drinking, resting, walking was not significantly different between day 1 and day 29. However, the frequency of urination significantly decreased (p<0.05) on day 29 compared with day 1. The frequency of stereotypic behaviors was not different between day 1 and 29. Our data indicate that horses may be more stabled when they are staying in the paddock rather than staying in the stall, but the stress level of horses in the stall during one month confinement was not effective for horses to adapt stereotypic behavior. In conclusion, providing an adequate environment and stress-less horse management techniques can minimize the stress level of horses.

제주흑우 집단에서 모색 관련 유전자와 microsatellite marker의 다형현상을 이용한 수정란이식 및 인공수정 유래 후대우 검증 (Verification of ET and AI Derived Offspring Using on the Genetic Polymorphisms of Microsatellite and Coat Color Related Genes in Jeju Black Cattle)

  • 농가에 보급된 제주흑우 수정란이식 및 인공수정 생산축의 확인을 위하여 분자유전학적 실험기법을 이용한 개체 추척을 수행하였다. 유전자 marker 체계는 ISAG 권장 MS marker 11종, 예비시험 후 선발된 SAES marker 11종, 흑모색 관련 MC1R과 ASIP 유전자들을 조합하여 분석하였다. 분석결과 부모 정보가 없는 상태에서의 부권 부정율이 국제권장기준보다 높은 수준을 보였으며, 형매간 동일개체출현률은 $5.3{\times}10^{-10}$으로 조사되었다. 친자검정 결과 후보축에 대한 후보 부, 모, 부모 모두가 확인되는 경우는 각각 77.0, 54.0, 40.5%였다. 부-모-자간 trio-mismatch가 전혀 없는 수정란이식 개체는 공급 수정란 대비 14.7%로 확인되었고, 전체 후보축군 중 32.4%는 후보 부와의 mismatch가 없는 인공수정에 의해 생산된 개체들로 판정하였다. ISAG marker들만을 분석한 결과에서는 7두가 동일한 3가지 유전자형 조합을 나타내었으나, ISAG/SAES marker들을 조합했을 때에는 2두에서만 동일 유전자형 조합을 나타내었다. MS와 모색유전자 분석자료를 모두 조합했을 때는 조사된 모든 개체들이 서로 구분되었다. 현재의 제주흑우집단이 소수 핵군에서 인공수정과 수정란이식 등 생명공학 기법으로 육성된 집단이기에 제주흑우집단의 유전적 다양성은 낮게 나타났다. 본 연구는 유전자 개체식별과 혈통관리 체계의 구축을 위해서는 적어도 20개 이상의 MS marker와 모색관련 유전자형 자료가 필수적으로 활용되어야함을 제안하고 있으며, 연구결과는 향후 제주흑우의 분자육종에 있어 유용한 자료가 될 것으로 시사하고 있다.

Does vitamin blends supplementation affect the animal performance, carcass traits, and nutrient digestibility of young Nellore finishing bulls?

  • Objective: This study was realized to evaluate the effects of supplementation with blends of water and fat-soluble vitamins on animal performance and carcass traits of young Nellore bulls. Methods: Forty-three Nellore bulls, with an initial weight of 261±27.3 kg and a mean age of 8±1.0 months, were used. Five animals were slaughtered at the beginning of the experiment (reference group), to determine the initial empty body weight of the bulls that remained in the experiment. The remaining 38 bulls were fed ad libitum and distributed in a completely randomized design in a 2×2 factorial scheme, with or without supplementation of water-soluble (B-blend+ or B-blend-) and fat-soluble (ADE+ or ADE-) vitamin blends. Diets were isonitrogenous (120 g of crude protein/kg dry matter [DM] of total mixed ration) and consisted of a roughage:concentrate rate of 30:70 based on total DM of diet. The experiment lasted 170 days, with 30 days of adaptation and 140 days for data collection. At the beginning and end of the experimental period, the bulls were weighed to determine the average daily gain. To estimate the apparent digestibility of nutrients and microbial efficiency, spot collections of feces and urine were performed for five consecutive days. Results: DM, ashes, organic matter, crude protein, ethereal extract, neutral detergent fiber corrected for residual ash and residual nitrogenous, and N intake and apparent digestibility were not influenced by vitamin supplementation, but total digestible nutrients intake and non-fibrous carbohydrates digestibility were influenced by B complex vitamin supplementation. Nitrogen balance, microbial efficiency, and performance data were not influenced (p>0.05) by vitamin supplementation. Conclusion: Vitamin supplementation (a blend of water-soluble and fat-soluble vitamins or their combinations) does not influence the animal performance and carcass traits of young Nellore bulls.

Effects of Prepartum Energy Intake and Calving Season on Blood Composition of Periparturient Cows

  • Sixteen periparturient Holstein cows calving during summer and autumn were fed to meet maintenance plus last 2 month of gestation level of TDN (MP) and 1.2 time of MP level (HMP). Dry matter intake of cows fed at HMP level during summer and autumn decreased by 1 and 2% of the offered feed in 1 week prepartum, respectively, and cows fed at MP level consumed all of the offered feed. Rectal temperatures at 08 : 30 h of cows fed at HMP level were higher than those of cows fed at MP level. Blood hematocrit and hemoglobin of cows increased during summer and were higher for cows fed at HMP level. Plasma total protein and glucose of cows during summer were higher than those during autumn, and the increased feed intake elevated plasma total protein and glucose. Plasma urea-N of cows fed at MP level was higher than that of cows fed at HMP level during autumn. The increased feed intake decreased plasma nonesterified fatty acid of cows during summer and autumn. These results suggest that blood components of periparturient cows are altered by calving season and feeding level.

Supplementation of nano-zinc in lower doses as an alternative to pharmacological doses of ZnO in weanling pigs

  • A set of studies was performed to determine the influence of dietary ZnO concentration and source during two phases (days 0 to 14 and days 15 to 28). Experiment 1: 168 weaned piglets were allocated to four treatment groups in six replicates. The treatments included a basal diet without ZnO supplementation (control), 2,500 mg ZnO/kg (In2500), 500 mg nano-ZnO/kg (N500), and 150 mg nano-ZnO/kg (N150). Experiment 2: 168 weaned piglets were divided into three treatment groups with eight replicates. The treatments included control, In2500, N300, and 150 mg nano-ZnO/kg (N150). An in vitro trial showed that the growth of Listeria monocytogenes, Escherichia coli, and Salmonella typhimurium was inhibited when exposed to 300 and 500 ppm of ZnO after 24 h of incubation. In experiment 1, the average daily gain (ADG) by the pigs was improved in the N500 and IN2500 treatment groups. Colonization of coliforms and Clostridium spp. significantly decreased in the pigs fed the N500 and IN2500 diets in phase 1. The total plasma antioxidant capacity was greater in the IN2500 and N500 treatment groups than in the control. Superoxide dismutase (SOD) activity was greater in pigs fed the IN2500 (phase 1) or the IN2500 and N500 (phase 2) diets than in the control and N150 treatment group. In experiment 2, pigs in the N300 treatment group showed a higher ADG and lower fecal score colonization of coliforms and Clostridium spp. compared with those in the N150 treatment group. In conclusion, nano-ZnO at a dose of 300 ppm showed the same growth as the pharmacological dose of Zn. This provides an option to the pharmacological dose.

Effect of Cytochalasin B Treatment on the Improvement of Survival Rate in Vitrified Pig Oocyte

  • To improve survival rates of vitrified pig oocytes, the treatment of cytoskeletal stabilizer on an appropriate time is one of the possible approaches. However, the exact treatment timing and effect of cytoskeletal stabilizer such as cytochalasin B (CB) is not well known during oocyte vitrification procedures. Thus, the present study was conducted to determine optimal treatment timing of CB during vitrification and warming procedures. In experiment 1, the survival rates of the postwarming pig oocytes were analyzed by fluorescein diacetate (FDA) assays with 4 classifications. In results, post-warming oocytes showed significantly (p<0.05) decreased number of alive oocytes (31.8% vs. 86.4%) compared to fresh control. In detail, the significant difference (p<0.05) was found only in strong fluorescence (18.2% vs. 70.5%) not in intermediate fluorescence groups (13.6% vs. 15.9%). In experiment 2, CB was treated before (CB-Vitri) and after (Vitri-CB) vitrification. In results, group of Vitri-CB showed significantly (p<0.05) higher (91.6%) survival rates compared to group of CB-Vitri (83.7%), significantly (p<0.05) and comparable with group of Vitri Control (88.7%) by morphological inspection. In FDA assay results, group of Vitri-CB showed significantly (p<0.05) higher (44.2%) survival rates compared to groups of CB-Vitri (36.7%) and Vitri Control (35.1%). In conclusion, the increased survival rates of post-warming pig oocyte treated with Vitri-CB method are firstly described here. The main finding of present study is that the CB treatment during recovery could be helpful to refresh the post-warming pig oocyte resulting its improved survival rates.

Effects of Supplementing Vitamin E and Nanoparticle-Sized Vitamin E on Growth Performance, Blood Profile, and Meat Quality in Broilers

  • This study was conducted to investigate the effect of vitamin E and nanoparticle-sized vitamin E (NVE) in a broiler diet on growth performance, blood profiles, and meat quality. A total of 45 one-day-old Arbor Acres broilers (initial body weight of 37.00 ± 0.41 g) were used in this experiment for 28 days. All broilers were randomly allocated into three dietary treatments in a randomized complete block design. The dietary treatments were as follows: a basal diet (CON), a basal diet supplementing with 380 mg/kg of vitamin E (T1), and a basal diet supplementing with 380 mg/kg of NVE (T2). Each treatment had five replicates with three birds per cage. On days 0-7, the T1 and T2 groups significantly increased (P<0.05) body weight gain compared to the CON group. Also, the T1 and T2 groups significantly increased (P<0.05) vitamin E content in blood and breast meat compared to the CON group. In shearing force, the T2 group showed a lower tendency (P=0.070) than the CON group. The T1 group showed a higher tendency (P=0.086) in the b* (yellowness) value than the T2 group. On day 7 after the end of the experiment, the T2 group significantly decreased (P<0.05) TBA values compared to the CON group. In conclusion, supplementation with vitamin E or NVE can improve broiler growth performance in the starter period, reduce TBA value through the antioxidant action of vitamin E, prevent lipid oxidation, and improve shelf life.

Effects of tannin supplementation on growth performance and methane emissions of Hanwoo beef cows

  • The objective of this study was to investigate the effects of dietary hydrolysable tannin on growth performance and methane emissions of Hanwoo beef cows. Fifteen cows participated in a seven-week experiment. The cows were stratified by initial methane emissions and assigned to one of two treatments: Control and tannin supplementation. Commercial hydrolysable tannin was top-dressed to a concentrate mix at 3 g/kg based on the dry matter. Enteric methane production was measured for 4 consecutive days at 1 week before and 1, 3 and 7 weeks after the initiation of the experiment using a laser methane detector. The feed intake was measured daily during the methane measurement periods and an additional two days prior to each measurement. The body weight of the cows was measured every 4 weeks. Hydrolysable tannin had no effect (p > 0.05) on body weight, average daily gain, dry matter intake (DMI) and feed conversion ratio. After one week, the methane emission of the tannin supplementation group was 3.66 ppm-m / kg DMI, which was about 3.4% lower (p = 0.078) than that of the control group; however, this tendency disappeared at 3 weeks after the start of the experiment (p > 0.05). The results of this study show that hydrolysable tannin supplementation can reduce enteric methane emissions for a limited period in Hanwoo beef cows. More research, however, is needed to determine the optimal level of hydrolysable tannin supplementation to reduce enteric methane emissions for a longer period without adversely affecting the animal performance of Hanwoo beef cattle.

Influence of β 1-4 Galacto-oligosaccharides Supplementation on Nitrogen Utilization, Rumen Fermentation, and Microbial Nitrogen Supply in Dairy Cows Fed Silage

  • In a balanced incomplete block design, two dry Holstein cows were used to investigate the effect of $\beta$ 1-4 galactooligosaccharides (GOS) supplementation on nitrogen (N) utilization, rumen fermentation and microbial N supply in the rumen. During the experiment, cows were fed four diets: orchardgrass (Dactylis glomerata L.) silage (OS), OS with GOS supplementation (OSG), OS mixed with alfalfa (Medicago sativa L.) silage (MS) and MS with GOS supplementation (MSG). GOS was supplemented at 2% of dry matter intake. Diets were fed at maintenance level of protein and energy. Results showed that N digestion was affected by silage and interaction of silage and GOS supplementation. Cows fed OSG had the highest N digested (p<0.05) followed by MS, OS and MSG. Supplementation of GOS to OS or MS diets tended to improve N utilization through reducing the N losses on dairy cows. There was no effect of GOS supplementation on rumen fermentation parameters (i.e. pH, $NH_3$-N and total VFA) at 1 h and 6 h after feeding. Compared to cows fed MS, cows fed OS silage had higher (p<0.05) allantoin excretion (80.8 vs. 67.1 mmol/d) and higher (p<0.05) total purine derivatives excretion (92.9 vs. 78.5 mmol/d). The microbial N supply in cows fed OSG was higher (p<0.05) than those fed OS, MS and MSG.

Preservation of Simmental bull sperm at 0℃ in Tris dilution: effect of dilution ratio and long-distance transport

  • Objective: This study aimed to assess the impact of the dilution ratio of Tris diluent, storage at 0℃, and long-distance transportation on the spermatozoa of Simmental cattle. It also validated the feasibility of the regional distribution of fresh semen. Methods: In experiment 1, semen was diluted at four dilution ratios (1:6, 1:9, 1:12, and 1:15) to determine the optimal dilution ratio of Tris diluent. In experiment 2, we assessed sperm viability, progressive motility (objectively assessed by computer-assisted sperm analyzer), and acrosome intactness in Tris dilutions kept at constant 0℃ for 1, 3, 6, 9, and 12 days. We compared them to Tianshan livestock dilutions (Commercial diluent). In experiment 3, semen was diluted using Tris diluent, and sperm quality was measured before and after long-distance transport. Artificial insemination of 177 Simmental heifers compared to 156 using Tianshan Livestock dilution. Results: The outcomes demonstrated that 1:9 was the ideal Tris diluent dilution ratio. The sperm viability, Progressive Motility, and acrosome integrity of both Tris and Tianshan dilutions preserved at 0℃ gradually decreased over time. sperm viability was above 50% for both dilutions on d 9, with a flat rate of decline. The decrease in acrosome integrity rate was faster for Tianshan livestock dilutions than for Tris dilutions when stored at 0℃ for 1 to 6 days. There was no significant difference (p>0.05) in sperm viability between semen preserved in Tris diluent after long-distance transportation and semen preserved in resting condition. The conception rates for Tris dilution and Tianshan livestock dilution were 49.15% and 46.15% respectively, with no significant difference (p>0.05). Conclusion: This shows that Tris diluent is a good long-term protectant. It has been observed that fresh semen can be successfully preserved for long-distance transport when stored under 0℃ conditions. Additionally, it is feasible to distribute semen regionally.
