폴리(4-비닐피리딘)의 질소 원자에 메틸기를 부착시켜 4차아민화시켜서 부분적으로 개질된 폴리(4-비닐피리딘)을 얻었고, 부분 개질된 폴리(4-비닐피리딘)의 구조를 수용액상에서 레이저 광산란 및 형광법 등을 이용하여 조사한 결과, 자기 응집현상에 의하여 핵-껍질 구조를 가지고 있는 것으로 나타났다. 즉 사슬의 소수성 부분은 상호작용에 의하여 중심부에 모여있고, 4차화된 아민의 양전하를 가진 친수성 부분은 바깥 껍질을 형성하고 있었다. 이 고분자 수용액에 음이온 계면활성제인, 도데실 황산 소듐을 첨가하면 이들 사이에 임계 응집 농도가 존재하며, 이 임계 응집 농도는 특히 NaCl 농도 0.1 M 이상에서는 급격히 감소하는 경향을 보여주었다. 또한 계면활성제의 첨가에 따라 생성된 고분자-계면활성제의 복합 응집체의 입자크기 변화 등을 동적 광산란으로 측정 분석하였다.
락토페린 결합단백질(Lactoferrin-binding proteins, LBP)은 젖소유방염 원인균인 Streptococcus uberis의 막단백질로서 그 특성에 관해서는 잘 규명되어 있지 않지만, 특히 최근에는 스트렙토코커스성 유방염의 독성인자로서 중요시되고 있다. 본 연구에서는 S. uberis 네 가지 균주를 대상으로 LBP를 보다 효율적으로 추출하기 위하여 mutanolysin 및 sodium dodecyl sulfate(SDS)를 이용한 두 가지 다른 추출 방법을 사용하였다. 추출된 세균단백질을 SDS-polyacrylamide gel electrophoreis(SDS-PAGE)로 전기영동을 하였고, 겔을 니트로셀룰로스 막으로 이동시켰다. Rabbit anti-bovine lactoferrin 항체와 HRP-conjugated donkey anti-rabbit IgG 항체를 사용하여 LBP를 검출하였다. 이러한 웨스턴 블롯팅 분석을 통해 SDS 추출법이 mutanolysin 추출법에 비해 보다 효율적으로 110 kDa 및 112 kDa의 LBP를 추출할 수 있음을 증명하였다.
Objectives : Ulcerative colitis (UC) is an inflammatory bowel disease, which is one of chronic gastrointestinal disorders. Orostachys japonicus (OJ) has been used as a traditional medicine for various diseases including gastric cancer, gastric ulcers and intoxication. However, the regulatory effect of OJ on intestinal inflammation has not been fully understood, yet. The aim of this study was to investigate the effect of OJ on dextran sulfate sodium (DSS)-induced colitis in mice. Methods : To ascertain the pharmacological effects of OJ, the colitis mice were induced by drinking water containing 5% DSS for 7 days. Mice were randomized into groups receiving OJ (100 mg/kg), sulfasalazine (150 mg/kg) as a positive control, or water as a negative control. We evaluated the effects of OJ on DSS-induced the clinical signs, measuring weight loss and colon length. In addition, the inhibitory effect of OJ on the tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) level was determined by enzyme-linked immunosorbent assay in colitis tissue. Results : The results indicated that mice treated with DSS showed remarkable clinical signs, including weight loss, and reduced colon length. However, treatment with OJ significantly improved the weight loss and DAI as clinical symptoms. Moreover, OJ reduced the TNF-${\alpha}$ levels in DSS-treated colon tissues. Conclusions : Collectively, the findings of this study provide us with novel insights into the pharmacological actions of OJ as a potential medicine for use in the treatment of ulcerative colitis.
양이온성 수용성 5, 10, 15, 20-tetrakis(N-methylpyridinium-4-yl) porphyrin (TMPyP)의 존재 하에서 음이온 계면 활성제인 sodium n-dodecyl sulfate 의 용액 특성을 전도도법, UV-vis 및 공명 빛 산란 (RLS) 분광학적 방법을 이용하여 포괄적으로 연구하였다. 그 결과에 따르면 TMPyP 농도의 증가로 SDS 용액의 임계 미셀 농도는 감소하였는데, SDS 미셀의 안정화는 미셀 표면에 존재하는 음전하의 중화 때문이다. SDS용액 내에는 자유 포르피린 단량체, 미셀에 속박된 포르피린 단량체 또는 응집체, 그리고 nonmicellar porphyrin/계면 활성제 응집체와 같은 세 종류의 TMPy종이 분명히 존재하였다. 우리의 연구 결과는 SDS가 TMPyP의 응집을 유도한다는 것을 나타내었다. 실제로 두 종류의 J-aggregations이 관찰되었는데, 하나는 미셀에 속박된 포르피린 단량체 또는 응집체였으며, 다른 하나는 nonmicellar porphyrin/계면 활성제 응집체이다. 그러나, cmc이하에서는 TMPyP가 SDS 음이온과 정전기 상호작용을 나타내었다.
Background: Hemangioma and vascular malformation are benign vascular lesions that often occur in cephalic and cervical region. Currently, surgical resection, laser therapy, angiographic embolization, use of steroids, and sclerotherapy are used as treatments. Case presentation: This study reports three cases of benign vascular lesions that are remarkably treated by sodium tetradecyl sulfate (STS) injection, of which occurred in oral cavity and around the mouth. Three percent of STS was diluted with 0.9 % of normal saline, and it was injected to the lesion site at least once. The result of treatment was evaluated based on clinical findings. Conclusion: Surgical treatment of hemangioma and vascular malformation occurred in oral cavity is not normally used because of esthetic issues and potential hemorrhage. On the other hand, sclerotherapy using STS is an effective therapy compare to surgical treatment. Despite the number of STS injection was different for each patient, all three patients had reached satisfactory level through the treatment with gradual diminution of lesions.
Objectives : This study aimed to investigate the protective effects of an herbal mixture of Atractylodes macrocephala and Taraxacum spp. (ATC) on ulcerative colitis. We have previously screened traditional medicinal herbs to discover the effective candidate by the animal model. A. macrocephala and T. spp were identified as one of the effective herbs in the screening process. Methods : Experimental colitis was induced in male Balb/c mice by administering drinking water containing dextran sulfate sodium, which mimics the clinical and histological features of ulcerative colitis in human. ATC at doses of 30, 100 or 300 mg/kg were orally administered to mice twice per day for 10 consecutive days. To evaluate the damage from experimentla ulcerative colitis, body weight, colon length, disease activity index, myeloperoxidase and histological changes were measured and analyzed. Results : The administration of dextran sulfate sodium with drinking water resulted in markedly reduced colon length, severe body weight loss, increased levels of myeloperoxidase activity and histological damages in mice. ATC treatment significantly ameliorated the colon shortening, histological damage, body weight loss and disease activity index score in a dose-dependent manner. ATC also attenuated the colonic myeloperoxidase activity which reflects the severity and extent of inflammatory damage of colon. Conclusions : ATC exerts protective effects against inflammatory colonic structural damage induced by epithelial barrier integrity impairment. ATC also inhibits weight loss and related symptoms of UC which can be considered as the functional recovery of colon.
Electrodeposition of chromium from low concentration chromjum (III) sulfate complexess in aqueous using sodium formate-glycine mixtures as a complexing agent was studied. In the bath formation, it is found that the optimum again temperature and time for equilibration of the electrolytes are nrcessary for 24 hrs at 45$^{\circ}C$and the optimum elecrical charge for low current electrolysis which might be produced Cr(II) ions in the electrolytes is necessary for minum 2Ah/$\ell$. The optmun concentration off standard electrolyte for main composition is 0.05M chromium(III) sulfate, 0.2M sodium formate-0.2M glycine, and I mMNaSCN as a catalysea, respectivwly. The standard electrolyte is shows good covering, good throwing power, and 6% of current efficiency (Cr(III)basis). The oppearance of electrodeposits shows att active bright chromium. The SEM morphology of the chromjum coating is observed as smooth surfaces and dispersed micro prcro pores. X-ray diffraction analysis shows a bcc structure which the perferred orintation of the chromium electrodeposits is strongin(200)plane.
Magnetite nanoparticles ($Fe_3O_4$ NPs) were synthesized by co-precipitating method under optimized condition. The $Fe_3O_4$ NPs coated with sodium dodecyl sulfate-thenoyltrifluoroacetone ($Fe_3O_4$ NPs-SDS-TTFA) were then exerted as the magnetic solid phase extraction (MSPE) adsorbent for the extraction process prior to introducing to a flame atomic adsorption spectrometry (FAAS). The synthesized $Fe_3O_4$ NPs-SDS-TTFA were applied for the extraction of Pb(II) ions from different water samples. The characterization studies of nanoparticles were performed via scanning electron microscopy-energy dispersive micro-analysis (SEM-EDAX), X-ray diffraction (XRD) and vibrating sample magnetometer (VSM) techniques. The substantial parameters affecting the extraction efficiency were surveyed and optimized. A dynamic linear range (DLR) of $10-400{\mu}g\;L^{-1}$ was obtained and the limit of detection (LOD, n=7) and relative standard deviation (RSD%, n= 6, $C=20{\mu}g\;L^{-1}$) were found to be $2.3{\mu}g\;L^{-1}$ and 1.9%, respectively. According to the results, the proposed method successfully applied for the extraction of Pb(II) ions from different environmental water samples and satisfactory results achieved.
Sodium dodecyl sulfate-capillary gel electrophoresis (SDS-CGE) and capillary isoelectric focusing (CIEF) were employed to characterize and compare ricin E purified from the small grain seeds of Ricinus communis with ricin D isoform. During the purification of ricin E using ion-exchange and size-exclusion chromatography, SDS-CGE was found to be useful for monitoring the efficiencies of purification steps. SDS-CGE showed that the molecular size of ricin E was not significantly different from that of ricin D, which was confirmed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. CIEF was useful for discriminating ricin E from ricin D based on their isoelectric points (pI). The pI values of ricin E and D were 8.6-8.8 and 7.0-7.4, respectively. This study demonstrates the usefulness of SDS-CGE and CIEF for the characterization of ricin toxins.
