2,4,6-Trinitrotoluene(TNT) was reduced into intermediate products by mixed culture incubated in anaerobic condition. To test the effects of extracellular material to electron transfer between sulfide and TNT, filtered medium of mixed culture was loaded in the test tubes with TNT and sulfide. The transformation rate was measured under four different conditions. The rate under microbial activity was the fastest among under different conditions. With sulfide or filtrate alone and TNT, the reactions were measured as the slowest reactions or no reactions occured, respectively. The reaction rate coefficient were calculated by linear regression and the first order kinetic was fitted best. Also, the plot of rate coefficients (K$_{f}$) showed linear relationships when at time zero TNT and sulfide concentration were 20 mg/1 and 6.0 mM, respectively. By extrapolation, reaction rate coefficient of 100% filtrate could be calculated as 0.0054/minute. However, reaction rate was affected by different concentration of sulfide, so it is a dependent of sulfide concentration. The results of this test showed TNT reduction rate can be limited more by microbial reaction than by mediation of filtrate or sulfide and filtrate alone.
To evaluate training effect, the step exercise was loaded to three mem for nine weeks. Step score, cardiopulmonary functions and blood constituents were measured before, during and after the test exercise (50 cm-step exercise and treadmill running), and were compared with the pre-tranining values. The results were as follows: 1) By the training, Harvard step score increased remarkably, expecially in the early stage of training. 2) The post-training values of maximal oxygen uptake increased very significantly and it seemed to be due to increases of stroke volume and tissue oxygen extraction. 3) After the training, the degree of increase in expired volume was small during the treadmill exercise. 4) By the training, increasing rate of respiratory quotient lessened during the exercise and it was considered to be caused by the decreases of carbohydrate consumption and anaerobic metabolism. 5) The blood cholesterol concentrations were harldy changed with this degree of training. 6) The blood lactate level decreased during the recovery periods and the values of the recovery 0 and 5 minutes decreased remarkably, in comparison with the pre-trained values. The above results suggest that the 9 week-training of the step exercise brings about the enhancement of circulatory functions and tissue oxygen utilization, and changes of food-stuffs used during the exercise.
A glucose-limited “pH-stat” continuous culture study of Lactobacillus bulgaricus NLS-4 in an anaerobic condition showed the marked effects of environmental pH on end products, fermentation blances and bioenergetic aspects of the organism. Lactic acid was the major end product of fermentation with minor products, such as acetic acid, formic acid and ethanol throughout the pH range tested. In acidic conditions below pH 6.5, a typi-cal pattern of homofermentation was revealed whereas in alkaline conditions, the metabolic pattern was changed from homofermentation to heterofermentation and led to acquire much energy. This metabolic change was likely due to the pH-dependent lactate dehydrogenase activity. Molar growth yields (Yglc=35.5-44.4) and YATP, $18.5\pm2.5$ in average which was 80% higher than the value ever postulated seemed to be accounted for less requirement of maintenance energy of the organism in the culture conditions.
Gel bead의 효율적인 이용을 목적으로 산소요구성이 전혀 다른 두 균주로 구성된 혼합고정화 배양계의 개발을 시도하였다. 호기성 균으로서 A. awamori, 혐기성균으로서 Z. mobilis를 사용하여 생전분으로 부터 에탄올 생산을 행한 결과는 다음과 같다. 두 균주의 최적 혼합비율은 A. awamori $1.25{\times}10^{9}\;spore/L-gel$, Z. mobilis 0.5g cell/L-gel이었다. 배양이 완료된 후의 gel bead 내의 균주 분포는 각각 혐기부와 호기부, 즉 gel bead의 표면부와 중심부로 생육장소가 구분되어 있었다. A-Z계의 배양에서는 에탄올의 수율이 낮았고, 균사가 gel bead로 부터 누출되었다. 배양 개시 후 36시간째에 배양기의 면전을 특수 제조한 check valve가 부착된 실리콘 plug로 교환하여 혐기적 배양을 행한 A-Z 36계에서는 gel bead로 부터 균사의 누출이 상당히 억제되었고, pH가 4.3을 유지하면서, 에탄올의 수율이 대조구 보다 약2배 높게 나타났다.
To elucidate the role and mechanism of microbes, we combined culture-dependent and culture-independent approaches to investigate differences in gut bacterial composition between sows and weaned pigs. Under anaerobic conditions, several nonselective and selective media were used for isolation from fecal samples. All isolated bacteria were identified and classified through 16S rRNA sequencing, and the microbiota composition of the fecal samples was analyzed by metagenomics using next generation sequencing (NGS) technology. A total of 278 and 149 colonies were acquired from the sow and weaned pig fecal samples, respectively. Culturomics analysis revealed that diverse bacterial genus and species belonged to Firmicutes, Actinobacteria, Proteobacteria, and Bacteroidetes were isolated from sow and weaned pigs. When comparing culture-dependent and culture-independent analyses, 191 bacterial species and 2 archaeal bacterial species were detected through culture-independent analysis, and a total of 23 bacteria were isolated through a culture-dependent approach, of which 65% were not detected by metagenomics. In conclusion, culturomics and metagenomics should be properly combined to fully understand the intestinal microbiota, and livestock-derived microbial resources should be informed by culturomic approaches to understand and utilize the mechanism of host-microbe interactions.
경기도 김포 지역과 인천 서구 지역공단 주변에 위치한 소하천의 저니(sediment)에서 난분해성 염소화합물인 PCE (tetrachloroethylene)의 혐기성 탈염소화 능력이 있는 군집을 선별하고, 탈염소화에 관여하는 미생물을 탐색하였다. 혐기성 탈염소화 능력을 조사하기 위해 전자공여체로 lactate를 사용하여 혐기성 회분식 실험을 실시하였으며, 탈염소화 능력을 가진 군집을 선별하였다. 선발된 미생물군집은 분자생물학적 기법인 16S rRNA gene의 Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) 기법과 탈염소화 미생물을 선택적으로 탐색할 수 있는 species-specific primer를 이용하여 분석하였다. 총 16개의 시료 중에서 접종 8주 만에 3개의 시료에서 ethene까지 탈염소화시켰으며, 4개의 시료에서 cis-1,2-dichloroethene (cis-DCE)까지 탈염소화시켰다. 또한, 16S rRNA gene을 이용한 PCR-DGGE와 탈염소화 species-specific primer를 이용하여 분석한 결과, PCE 탈염소화 시료 내에는 Dehalococcoides sp.와 Geobacter sp.가 주로 존재하였으며, Dehalobacter sp.도 일부 시료에서 검출되었다.
오염된 양식 해수의 수질정화능력이 뛰어난 광합성세균의 고순도 대량배양을 위한 실험을 실험실 단위 및 파이롯트 단위에서 수행했다. Gas pack 반응기, flask 반응기 및 N-source로서 질소 가스를 사용한 실험 결과, 광합성세균은 현미경상 약 90%이상의 순도를 나타내었고, CFU 측정 결과에서도 붉은색을 띄는 광합성세균 colony만을 관찰 할 수 있었다. 배지의 교차에 의한 배양실험에서 기초배지를 이용한 배양이 광합성세균의 성장에 가장 효과적임을 알 수 있었다. 이러한 배양조건하에서의 5L bioreactor 배양에서 비증식속도가 $0.18h^{-1}$로 나타났고, 실험실 단위의 결과를 이용한 pilot 단위에서의 배양결과, 실험실 단위의 비증식속도와 동일한 값을 얻을 수 있었다.
본 실험에서는 균주의 특성에 따른 유산균수 시험방법의 적합 여부를 알아보기 위해 액상요구르트 제품들을 유효기간 중 여러 실험조건으로 비교 검토하였다. 비교한 실험조건은 배지(BCP, Elliker agar), 배양상태(aerobic, anaerobic), 희석수(saline, phosphate buffer), 희석방법 (10배, 100배)이었으며 $37^{\circ}C$에서 72시간 배양하였다. L. acidophilus 균주를 사용한 액상요구르트의 경우, 배지와 희석방법에 따른 차이는 거의 없었고 희석수와 배양상태에서는 약간의 차이가 있었다. L. jugurti 균주와 L. acidophilus +L. casei 혼합균주의 생우 배지 배양상태, 희석수에서 차이가 있었고 희석방법에는 거의 차이가 없었다. L. casei의 경우 배지, 희석방법에서 약간의 차이를 나타했으며 배양상태는 유산균수에 영향이 없었다. L. bulgaricus의 경우는 배지, 배양상태, 희석방법에 따라 차이가 있었고 희석수는 차이가 없었다. 그러므로 요구르트의 유산균수 측정은 균주의 종류에 따라 가장 좋은 실험조건을 선택하여 실시하는 것이 효과적인 시험방법으로 사료된다.
본 연구는 구취생성의 주요 원인균에 대한 nanosilver의 항균효과를 조사하기 위해서 시행되었으며, 이와 더불어 이들 세균의 증식 조건도 함께 연구하였다. 포항공과대학교 나노기술센터로부터 $NANOVER^{TM}$, WA 1000 현탁액을 제공 받아 희석하여 사용하였으며, 공시균으로는 주요 구취생성균으로 알려진 Fusobacterium nucleatum (KCTC 2640), Prevotella melaninogenica (KCTC 3689), Klebsiella pneumonia (KCTC 1560) 3종의 세균류를 한국생명공학연구원 생명자원센터로부터 구입하여 사용하였다. 공시균의 배양에 혐기성세균 배양에 이용되는 tryptone- yeast extract-ammonium acetate(TYA) 배지와 chopped beef meat(CBM) 배지를 이용하였다. 1. 최적배양조건으로 감압배양, CBM 배지가 적당한 것으로 나타났다. 2. 간균인 K. pneumonia에 Nanover(5 및 10 ppm)를 처리하고 주사형 전자현미경으로 균체의 형태를 관찰한 결과는 세균의 생장점 부근에서 세포벽에 손상을 주어 세포벽을 절단하여 다량의 막상 편린을 생성하고 autolysin을 불활성화 시켜 filament형 균체를 형성하여 균의 증식은 일시적으로 억제시킬 수 있었으나 완전히 사멸시키지는 못하였으며, 구취 세균을 전혀 다른 형태로 변화시키는 것으로 나타났다. 그러나 항균제로서의 역할은 가진 것으로 확인되었다.
Actinomyces is a part of the normal oral flora, but under certain circumstances it may become pathogenic. Actinomycosis is a chronic granulomatous infective disease caused by microaerophilic Gram-positive bacteria of the genus actinomyces. It can involve almost any system, but principally affects the head and neck. Because the lesions in the submandibular region and the angle of the jaw give the face a swollen, indurated appearance, actinomycosis of mandible can be easily misdiagnosed in its acute or early state of infection. In these cases the disease usually presented as a swelling suggestive of an abscess or mimicking a neoplasm. The yield from standard cultures was poor and repeated sampling and anaerobic culture may be needed to obtain a positive culture. So actinomycosis should always be considered in a differential diagnosis of all infections of the cervicofacial area. Diagnosis of actinomycosis is made based on the histopathology, the clinical presentation and past dental history. We experienced a case of actinomycosis in the masseter muscle and present the case with review of literature.
