Wastewater containing kraft lignin (KL) discharged from pulp and paper industries could cause serious environmental contamination. Appropriate effluent treatment is required to reduce the pollution. Investigations on anaerobic bacteria capable of degrading KL are beneficial to both lignin removal and biofuel regeneration from the effluent. In this paper, an anaerobic strain capable of degrading KL was isolated from the sludge of a pulp and paper mill and identified as Dysgonomonas sp. WJDL-Y1 by 16S rRNA analysis. Optimum conditions for KL degradation by strain WJDL-Y1 were obtained at initial pH of 6.8, C:N ratio of 6 and temperature of 33℃, based on statistical analyses by response surface methodology. For a 1.2 g/l KL solution, a COD removal rate of 20.7% concomitant with biomass increase of 17.6% was achieved after 4 days of incubation under the optimum conditions. After the treatment by strain WJDL-Y1, KL was modified and degraded.
There are many advantages when using IIF and DNA probe methods over anaerobic culture method in that they are time-and effort-saving, more precise and more sensitive. Furthermore, in IIF and DNA probe methods, the detection is possible only with small amount of bacteria, the quantitative analysis is possible, and the cell viability is not necessary. The purpose of this study is to observe the incidence of P.endodontalis by carrying out anaerobic culture, IIF and colony lift using DNA probe method respectively, and to compare these 3 methods in terms of effectiveness and sensitivity in order to identify the most effective detection method. 30 teeth with at least one clinical symptoms, with single canal, and with pulp necrosis were sampled. For sampling bacteria, access cavity was prepared after disinfecting tooth and its surroundings. Then the paper point was inserted up to the periapical area, leave there for a while, and finally it was placed into PRAS Ringer's sol. and PBS sol. In anaerobic culture method, P.endodontalis was identified by biochemical tests after subculturing black and brown colonies which were produced after 7 days of incubation on BAP and Brucella BAP in anaerobic chamber. To identify P.endodontalis in IIF method, species-specific polyclonal rabbit-antisera of P.endodontalis(ATCC 35406) was reacted with sampled PBS sol. dispensed onto glass slide, and then P.endodontalis was examined by phase contrast microscopy after incubating with Goat anti-rabbit lgG conjugated to Fluorescein isothiocyanate. For colony lift using DNA probe method, membranes were laid over colonies on the surface of BAP and were hybridized with cloned DNA probe of P.endodontalis. The existence of P.endodontalis was then identified by the methods of chemiluminescent detection and color metric detection. Black colony was found in 11 teeth out of 30 teeth and P.endodontalis was detected in 6 teeth (20 %) by anaerobic culture method, 16 teeth (53 %) by IIF method, and 7 teeth (23 %) by DNA probe method. IIF method is significantly better in detecting P.endodontalis than DNA probe method and anaerobic culture method. There was no significant differences between DNA probe method and anaerobic culture method. There was significant correlation between the formation of black colony and the existence of P.endodontalis. The probability of detecting P.endodontalis when black colony being present is 2.89 times higher than when not being present. There was significant relationship between the foul odor of clinical symptoms and P.endodontalis. The sensitivity of existing P.endodontalis when foul odor being present was 93.75 %, while the specificity of not existing P.endodontalis when foul odor not being present was 28.57 %. These results suggested that the probes of P.endodontalis will be used to decide the method and prognosis in endodontic treatments.
Anoxic sediments collected from Shiwha-ho area were used to find the relationship between the heavy-metal, organic content and anaerobic respiration bacteria by most probable number (MPN) method. Analysis of the sediments showed that COD content was higher in the sediments collected from Ansan-cheon and Shiwha-ho than those collected from sea area nearby. Particularly noticeable was the fact that heavy metal concentration was much higher in the sediments of Shiwha-ho area contaminated by heavy-metal, although they were rich in electron donor and electron acceptor for Fe(III)-reducing bacteria using lactate as an electron donor was in the range of 1.1$\times$106-4.6$\times$107MPNs/ml in the sediments collected from the sea-side of the lake, which were lower in heavy-methal concentration and higher in Fe-Mn content than those from other region. The number of Fe(III)-reducing bacteria using acetate as an electron donor was in the rang eof 4.3$\times$102-8.1$\times$105MPNs/ml in the same sediments. Chromate-reducing bacteria were more populated(4.6$\times$104-8.1$\times$105MPNs/ml) in the sediments contaminated by heavy metals. The number of sulfate-reducing bacteria wee counted in the sediments collected from the more contaminate inner-side than those from the sea-side of the lake.

Objectives of this experiment were to select of the optimum water plant and to investigate the removal efficiency of the major nutrients in the constructed wetland by natural purification method for municipal sewage treatment in the farming and fishing village. For the experiment we used "constructed wetland" which was consisted of aerobic (vertical filter system) and anaerobic systems (horizontal filter system). Both systems were filled with gravel and filter media, and grew water plants on top of them. And then, we investigated several items such as sewage treatment efficiency, growth status of water plants and the absorbed amount of inorganic element in water plants with periodical periods. In aerobic area, removal efficiencies of BOD, COD, T-N and T-P were over 92%, 74%, 25% and 57%, respectively, and then when the water is passed through anaerobic area, the efficiency was over 96%, 84%, 44% and 71%, respectively, which was increased more treatment efficiency than that of aerobic area. Absorption amount of nitrogen and phosphorous in Miscanthus sacchariflorus BENTH were the highest in the water plants as 17.7 and 2.41 g/plant in the aerobic area, respectively. Absorption amounts of nitrogen and phosphorous in Scirpus tabernaemontani GMEL were the highest in the water plants as 8.7 and 1.13 g/plant in the anaerobic area, respectively. For the selection of optimum water plants in the constructed wetland by natural purification method for municipal sewage treatment, it were observed that there were Miscanthus sacchariflorus BENTH, Phragmites japonica STEUD and Phragmites communis TRINIUS in the aerobic area and were Zizania latifolia TURCZ, Scirpus tabernaemontani GMEL, Typha orientalis PRESL, Iris pseudoacorus L and Cares dispalata BOOTT in the anaerobic area.
Polynuclear aromatic hydrocarbon (PAH) compounds are highly carcinogenic chemicals and common groundwater contaminants that are observed to persist in soils. The adherence and slow release of PAHs in soil is an obstacle to remediation and complicates the assessment of cleanup standards and risks. Biological degradation of PAHs in soil has been an area of active research because biological treatment may be less costly than conventional pumping technologies or excavation and thermal treatment. Biological degradation also offers the advantage to transform PAHs into non-toxic products such as biomass and carbon dioxide. Ample evidence exists for aerobic biodegradation of PAHs and many bacteria capable of degrading PAHs have been isolated and characterized. However, the microbial degradation of PAHs in sediments is impaired due to the anaerobic conditions that result from the typically high oxygen demand of the organic material present in the soil, the low solubility of oxygen in water, and the slow mass transfer of oxygen from overlying water to the soil environment. For these reasons, anaerobic microbial degradation technologies could help alleviate sediment PAH contamination and offer significant advantages for cost-efficient in-situ treatment. But very little is known about the potential for anaerobic degradation of PAHs in field soils. The objectives of this research were to assess: (1) the potential for biodegradation of PAH in field aged soils under denitrification conditions, (2) to assess the potential for biodegradation of naphthalene in soil microcosms under denitrifying conditions, and (3) to assess for the existence of microorganisms in field sediments capable of degrading naphthalene via denitrification. Two kinds of soils were used in this research: Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS). Results presented in this seminar indicate possible degradation of PAHs in soil under denitrifying conditions. During the two months of anaerobic degradation, total PAH removal was modest probably due to both the low availability of the PAHs and competition with other more easily degradable sources of carbon in the sediments. For both Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS), PAH reduction was confined to 3- and 4-ring PAHs. Comparing PAH reductions during two months of aerobic and anaerobic biotreatment of MHS, it was found that extent of PAHreduction for anaerobic treatment was compatible with that for aerobic treatment. Interestingly, removal of PAHs from sediment particle classes (by size and density) followed similar trends for aerobic and anaerobic treatment of MHS. The majority of the PAHs removed during biotreatment came from the clay/silt fraction. In an earlier study it was shown that PAHs associated with the clay/silt fraction in MHS were more available than PAHs associated with coal-derived fraction. Therefore, although total PAH reductions were small, the removal of PAHs from the more easily available sediment fraction (clay/silt) may result in a significant environmental benefit owing to a reduction in total PAH bioavailability. By using naphthalene as a model PAH compound, biodegradation of naphthalene under denitrifying condition was assessed in microcosms containing MHS. Naphthalene spiked into MHS was degraded below detection limit within 20 days with the accompanying reduction of nitrate. With repeated addition of naphthalene and nitrate, naphthalene degradation under nitrate reducing conditions was stable over one month. Nitrite, one of the intermediates of denitrification was detected during the incubation. Also the denitrification activity of the enrichment culture from MHS slurries was verified by monitoring the production of nitrogen gas in solid fluorescence denitrification medium. Microorganisms capable of degrading naphthalene via denitrification were isolated from this enrichment culture.

This study was carried out to quantify the rates of anaerobic mineralization and sulfate reduction, and to discuss the potential effects of benthic fauna on sulfate reduction in total anaerobic carbon respiration in Ganghwa intertidal flat in Korea. Anaerobic carbon mineralization rates ranged from 26 to 85 mmol $C\;m^{-2}\;d^{-1}$, which accounted for approximately 46 tons of daily organic matter mineralization in the intertidal flat of southern part of the Ganghwa Island (approximately $90\;km^2$). Sulfate reduction ranged from 22.6 to 533.4 nmol $cm^{-3}\;d^{-1}$, and were responsible for $31{\sim}129%$ of total anaerobic carbon oxidation, which indicated that sulfate reduction was a dominant pathway for anaerobic carbon oxidation in the study area. On the other hand, the partitioning of sulfate reduction in anaerobic carbon mineralization in October decreased, whereas concentrations of Fe(II) in the pore water increased. The results implied that the re-oxidation of Fe(II) in the sediments is stimulated by macrobenthic activity, leading to an increased supply of reactive Fe(II), and thereby increasing Fe(III) reduction to depress sulfate reduction during carbon oxidation.
Effect of organic loading rate on UASB performance was evaluated under the renditions of some surface area/reactor volume ratio and different reactor diameter. At the low leading rate of 0.4 kg $COD/m^3{\cdot}d$, reactor performance was not affected by reactor diameter. At the organic loading rate of 6 kg $COD/m^3{\cdot}d$, however, volatile acid accumulation and low COD removal efficiency is observed in reactor having 6.4 cm diameter, while volatile acid is not accumulated at all and high COD removal efficiency is observed in reactor having 3 cm diameter. Such a difference of reactor performance depending on reactor diameter can be explained that sludge bed can be fluidized by evolved gas bubble in narrow reactor, while sludge bed ran not be fluidized by evolved gas bubble only in wide reactor. At a high organic loading rate of 20 kg $COD/m^3{\cdot}d$, it can be judged that there is no relation between reactor configuration and reactor performance because all reactors showed very low COD removal efficiencies regardless of reactor diameter. Narrow and tall type reactor is favorable condition for making sludge bed fluidization at a constant surface area/reactor volume ratio. Thus, it can be judged that reactor configuration and sludge bed fluidization have great influence to reactor performance.

To develop small-scale sewage treatment apparatus for detached house of agricultural village, a small-scale sewage treatment apparatus by natural purification method that consisted of aerobic and anaerobic bed was constructed. To reduce the area of a sewage treatment apparatus, four different fitter media were used and each filter medium was coarse sand, broken stone, steel slag, and mixed fitter media (coarse sand : broken stone : steel slag = 1:1:1). The efficiency of sewage treatment according to the depth of aerobic and anaerobic bed and the volume ratio of aerobic to anaerobic bed were investigated in small-scale sewage treatment apparatus. The removal rate of pollutants according to the depth of aerobic and anaerobic bed in small-scale sewage treatment apparatus was high in the order of 50 cm < 70 cm < 90 cm. The removal rate of pollutants according to the ratio of aerobic to anaerobic bed in small-scale sewage treatment apparatus was high in the order of 1:1 < 1:2 $\fallingdotseq$ 1:3. Under the optimum conditions, removal rate of BOD, COD, SS, T-N and T-P were $98{\sim}99,\;95{\sim}97,\;99,\;65{\sim}66\;and\;96{\sim}99%$ respectively, in small-scale sewage treatment apparatus.

The aim of this study was to confirm the feasibility of the electro-coagulation process as a pre-treatment for the membrane separation of anaerobic digestion effluents to minimize membrane fouling. The reduction of membrane fouling was evaluated according to the number of electrodes (immersed surface area of electrodes), current density and contact time. In the case of the small surface area of electrodes, the increased electric field strength resulted in a soluble COD increase due to the destruction of the microbial flocs and/or cells, whereas large changes in the soluble COD were not observed in the case of the high surface area of electrodes. On the other hand, the T-P concentration decreased as a result of the precipitation of aluminum ions and phosphates. The membrane permeation flux increased and the fouling resistance (Rc+Rf) decreased with increasing electric current density. Although the particle size of the anaerobic digestion effluent increased slightly, it was not related directly to the reduced fouling phenomena. The main mechanism for the enhanced flux was attributed to the inorganic particulate produced during electrocoagulation, such as $AlPO_4$, which acted as a dynamic membrane deposited on the membrane surface.

Soils in agricultural lands contain large amount of organic matter and nutrients due to the injected fertilizers and manure. During heavy rain, surface water and base runoff pollutants flows into a nearby stream or lake with eroded soil from agricultural lands. On the other hands, agricultural lands near the lake are inundated due to the increase of the water level in the lake, leading to organic matter and nutrient release from the inundated soil. In this study, releasing rates of nutrient salts and organic substances were analyzed for the soil in the agricultural land, where cultivation activities has been carried out and periodically flooded, to account for the possibility of contamination from the inundated agricultural land in reservoir areas The experiment results have shown that COD was released from the soil in anaerobic conditions, and T-P was released in both anaerobic and aerobic conditions. However, in the case of T-N, it was found that the runoff by soil was not made before the rainfall occurred, and when the soil was impound due to rainfall, the elution occurred under the aerobic conditions. Through the results of this study, it was possible to account for the effect of flooded agricultural lands on the water quality in the lake, and this could be reflected in an efficient agricultural non-point pollution management policy. In order to determine the precise releasing rate for each agricultural land, it is believed that the leaching experiment for paddy fields and grasslands are needed.
