Modulation of DNA methylation by one-carbon metabolism: a milestone for healthy aging

  • Healthy aging can be defined as an extended lifespan and health span. Nutrition has been regarded as an important factor in healthy aging, because nutrients, bioactive food components, and diets have demonstrated beneficial effects on aging hallmarks such as oxidative stress, mitochondrial function, apoptosis and autophagy, genomic stability, and immune function. Nutrition also plays a role in epigenetic regulation of gene expression, and DNA methylation is the most extensively investigated epigenetic phenomenon in aging. Interestingly, age-associated DNA methylation can be modulated by one-carbon metabolism or inhibition of DNA methyltransferases. One-carbon metabolism ultimately controls the balance between the universal methyl donor S-adenosylmethionine and the methyltransferase inhibitor S-adenosylhomocysteine. Water-soluble B-vitamins such as folate, vitamin B6, and vitamin B12 serve as coenzymes for multiple steps in one-carbon metabolism, whereas methionine, choline, betaine, and serine act as methyl donors. Thus, these one-carbon nutrients can modify age-associated DNA methylation and subsequently alter the age-associated physiologic and pathologic processes. We cannot elude aging per se but we may at least change age-associated DNA methylation, which could mitigate age-associated diseases and disorders.

Korean Red Ginseng Tonic Extends Lifespan in D. melanogaster

  • Aging is the single most important risk factor that increases susceptibility to many forms of diseases. As such, much effort has been put forward to elucidate the mechanisms behind the processes of aging and to discover novel compounds that retain anti-aging activities. Korean red ginseng has been used for a variety of medical purposes in eastern countries for several thousands of years. It has been shown that Korean red ginseng affects a number of biological activities including, but not limited to, anti-inflammatory, anti-oxidative and anti-diabetic pathways. However, few studies have been performed to evaluate its anti-aging effects with an in vivo system. Here Drosophila melanogaster as an in vivo model organism demonstrates that Korean red ginseng tonic extends lifespan, increases resistance to starvation stress and prevents weight gain. This data suggest that Korean red ginseng may regulate organisms' metabolism in favor of extending lifespan.

  • Magnesia partially stabilized zirconia(Mg-PSZ) solid electrolytes for an improvement of thermal shock resistance, which is suitable for the measurement of oxygen activity in a molten steel, were prepared by post-thermal aging treatment. The steelmaking oxygen sensor elements were formed by an injection molding method, sintered at $1650^{\circ}C$, and then thermal aged ranged from 1250 to $1400^{\circ}C$. Sintered density and porosity were decreased as increasing the magnesia content in a zirconia-magnesia solid solution. Fractions of cubic phase to the synthesized Mg-PSZ solid electrolytes were ranged from 13.13 to 79.54.% after post-thermal aging treatment. Very dense microstructure without voids in the grains was obtained by the post-thermal aging process. Fine tetragonal phase crystallites precipitated on the cubic surface during post-thermal aging up to $1300^{\circ}C$ improve a thermal shock resistance and reappearance of electro motive force(EMF) curve.

Effects of Aging Methods and Periods on Quality Characteristics of Beef

  • The objective of this study was to determine effects of aging methods (wet-aged, dry-aged, and packaged dry-aged) during 60 d on quality traits and microbial characteristics of beef. Wet-aged beef was packed by vacuum packaging and stored in a 4℃ refrigerator. Dry-aged beef was used without packaging. Packaged dry-aged beef was packaged in commercial bags. Dry-aged and packaged dry-aged samples were stored in a meat ager at 2℃-4℃ with 85%-90% relative humidity. Meat color, crust thickness, aging loss, cooking loss, Warner-Bratzler shear force (WBSF), texture profile analysis, Torrymeter, meat pH, water activity, volatile basic nitrogen (VBN), thiobarbituric acid reactant substances (TBARS), and microbial analysis were measured or performed every 15 d until 60 d of aging time. Meat color changed significantly with increasing aging time. Differences in meat color among aging methods were observed. Aging losses of dry-aged and packaged dry-aged samples were higher than those of wet-aged samples. Wet-aged beef showed higher cooking loss, but lower WBSF than dry-aged and packaged dry-aged beef. VBN and TBARS showed an increasing tendency with increasing aging time. Differences of VBN and TBARS among aging methods were found. Regarding microbial analysis, counts of yeasts and molds were different among aging methods at the initial aging time. Packaged dry-aged and dry-aged beef showed similar values or tendency. Significant changes occurred during aging in all aging methods. Packaged dry aging and dry aging could result in similar quality traits and microbial characteristics of beef.

Changes of Carbide Characteristics and Magnetic Properties in Artificially Aging Heat Treated 2.25CrMo Steel (경년열화 열처리된 2.25CrMo 강에서의 탄화물 특성 및 자기적 성질의 변화)

  • Artificial aging was performed to simulate the microstructural degradation in 2.25CrMo steel arising from long time exposure at $540^{\circ}C$. The carbide morphologies were classified as acicular, pipe and globular type, and the number of carbides per unit area was measured for each type of carbides. The fine acicular carbides were found to diminish drastically in the initial stage of aging. An attempt was made to evaluate the microstructural degradation in artificially aging heat treated 2.25CrMo steel by the magnetic property measurements such as saturation magnetization, coercivity and remanence. The saturation magnetization showed no distinct trend with aging time. However, the coercivity and remanence were observed to decrease rapidly in initial 920 hours of aging time and then decrease slowly afterwards.

  • This study investigates the effects of long-term artificial-aging on hardness variation in the dissimilar metal weldments for nuclear power plant facilities. These dissimilar welds are inevitably required to join the components in nozzle parts of pressurized vessels, such as austenitic stainless steels and ferritic steels. A artificial thermal aging was conducted in an electrical furnace to simulate material degradation at high temperatures. The test materials were held at the temperature of $600^{\circ}C$ for 10000 hours and interrupted at various levels of degraded specimens. The degradation of hardness is a well-known phenomenon resulting from long-term aging or high-temperature degradation of structural materials. In this study, the variation of hardness at each position was different, and complicated in relation to microstructures such as twins, grains, precipitates, phase transformations, and residual stresses in dissimilar weldments. We discussed the variation of hardness in terms of microstructural changes during long-term aging.

Defensive Behavior against Noxious Heat Stimuli Is Declined with Aging Due to Decreased Pain-Associated Gene Expression in Drosophila

  • Aging is defined as a collective process that alters organism's functional capacity and appearance over the course of life. Apart from an increase in susceptibility to many diseases, aging affects the cellular system that is responsible for decoding painful stimuli. Yet, aging-associated molecular mechanisms of pain perception remains elusive. Using Drosophila, we showed a decrease in temperature tolerance and a reduction in high temperature thermal avoidance with aging. Locomotor activity assay demonstrated that the age-dependent changes in heat nociception did not stem from the general decline in muscular activity. However, we identified pain-related gene expression alteration with aging. We anticipate that our findings would help opening a new window onto developing the optimal pain treatment for the elderly.

Acid sphingomyelinase-mediated blood-brain barrier disruption in aging

  • Although many studies have reported that the breakdown of the blood-brain barrier (BBB) represents one of the major pathological changes in aging, the mechanism underlying this process remains relatively unexplored. In this study, we described that acid sphingomyelinase (ASM) derived from endothelial cells plays a critical role in BBB disruption in aging. ASM levels were elevated in the brain endothelium and plasma of aged humans and mice, resulting in BBB leakage through an increase in caveolae-mediated transcytosis. Moreover, ASM caused damage to the caveolae-cytoskeleton via protein phosphatase 1-mediated ezrin/radixin/moesin dephosphorylation in primary mouse brain endothelial cells. Mice overexpressing brain endothelial cell-specific ASM exhibited acceleration of BBB impairment and neuronal dysfunction. However, genetic inhibition and endothelial specific knock-down of ASM in mice improved BBB disruption and neurocognitive impairment during aging. Results of this study revealed a novel role of ASM in the regulation of BBB integrity and neuronal function in aging, thus highlighting the potential of ASM as a new therapeutic target for anti-aging.

  • An aging society in which the proportion of the aged of a society is growing faster than the of the young is becoming an worldwide phenomenon owing to low birth rate, the upgraded medical art and so on. Korea is one of the most rapidly aging societies in the world today. Since aging society is totally different from the traditional society in the many field, potential problems must be expected and countermeasure must be considered. One of the potential problems it may cause is the issue of supporting subject of the aged. Parent-supporting and welfare for the aged become important social issues. Especially the supporting of the aged woman is more important issue because of their longer average life than man and their exposure to sexual discrimination and their welfare is very important issue of social welfare system. Accordingly the purpose of this study is to present the practical counter measure to the supporting of the aged woman. These studies can be divided into traditional approach which insists on the family supporting and modern approach which asserts government(or social) supporting. Comprehensive literature study reveals that both of government and family are responsible for the supporting of the aged woman.

Aging Anxiety and Related Factors of Middle-Aged Adults (중년기 성인의 노화불안과 영향요인)

  • Purpose: The purpose of this study was to identify aging anxiety and influencing factors of middle-aged adults. Methods: Descriptive correlation design was used. Three hundred and three middle-aged adults participated in the study and completed the questionnaires in the period from September 11 to October 15 of 2011. The data was analyzed using independent t-test, one-way ANOVA, Scheff$\acute{e}$ test, Pearson correlation coefficients, and hierarchial multiple regression analysis. Results: Aging anxiety of middle-aged adults was higher than a moderate level. Socio-demographic variables such as age, religion, education, economic level, perceived health status and experience of living with older adults influenced significantly on aging anxiety. In contrast, general self-efficacy, aging knowledge and attitude toward older adults were negatively correlated with aging anxiety. Hierarchial multiple regression analysis incorporating these variables showed an explanation rate of aging anxiety at 21%. And religion, perceived health status, attitude toward older adults, and general self-efficacy were statistically significant among variables that influence on aging anxiety. Conclusion: The findings of this study suggest that senior preparation programs that can enhance self-efficacy, induce positive attitude on older adults and promote health should be implemented to reduce aging anxiety of middle-aged adults.
