  • We implemented DPU(Double-Pass Gain-Clamped) L-band EDFA for highly efficient amplification. A lasing signal generated within the linear cavity, can minimize the fluctuation of surviving channels when several WDM(Wavelength Division Multiplexing) channels are added or dropped. The new method measuring the characteristics of transient response of surviving channels quantitatively is suggested. It is to measure the ratio of lasing output before add or drop to that after add or drop. We investigated dynamic characteristics by using this method according to lasing wavelengths and propagation directions within the cavity. Experimental measurements show that the short lasing wavelength and backward propagation direction is the best condition for small fluctuation of surviving channels.

  • A simple and convenient device for deflection-type direct reading the variations in electrolytic conductance is described and applied to the analysis of sulfur by combustion-$H_2O_2$ oxidation method. The apparatus consisted of a high resistance-ratio bridge in which the other adjacent arms are the differential cells. By adopting unusually high a-c voltage source for the bridge excitation, the a-c method for unbalanced bridge is established, decreased sensitivity owing to reduced bridge factor, 0.01, is overcome and also the absolute sensitivity and linearity are greatly improved. Over 50% variations in impedance of the balanced cell, within 1% deviation from the linearity can be attained with a volt (rms)order of output which was detected directly with VTVM without further amplification. Analysis of the bridge shows that these useful features are natural result of the constant current character of the high source impedance generator and the performance of the device agreed with the theoretical predictions. A standard procedure for the rapid analysis of sulfur using the bridge is also given, the analytical accuracy was approximately 1%. A determination takes not more than 5 minutes.

Resonant Oscillations in Mukho Harbor (묵호항의 항내 진동)

  • Three Pressure type wave gauges were installed for about 10 days for the analysis of long wave agitations in Mukho Harbor. Helmholtz and second resonant periods of seiche in Mukho Harbor are shown to be approximately 10.0-14.3 and 3.3 minutes from the spectral analysis of measured wave data. Amplification ratio at Helmholtz period reaches about 6.8 and the wave amplitudes in the harbor were in the range of 5-10 cm during the measurement period. Helmholtz and second resonant periods of seiche in Mukho Harbor agree very well with those computed using Jeong dt al. (1993b)'s model. The model gives rise to the first and third resonant peaks at 7.5 and 1.9 minutes, respectively.

  • Site coefficients in IBC and KBC codes have some limits to predict the rational seismic responses of a structure, because they consider only the effect of the soil amplification without the effects of the structure-soil interaction. In this study, upper and lower limits of site coefficients are estimated through the pseudo 3-D elastic seismic response analyses of structures built on linear or nonlinear soil layers considering the structure-soil interaction effects. Soil characteristics of site classes of A, B, and C were assumed to be linear, and those of site classes of D and E were done to be nonlinear and the Ramberg-Osgood model was used to evaluate shear modulus and damping ratio of a soil layer depending on the shear wave velocity of a soil layer. Seismic analyses were performed with 12 weak or moderate earthquake records, scaled the peak acceleration to 0.1g or 0.2g and deconvoluted as earthquake records at the bedrock 30m beneath the outcrop. With the study results of the elastic seismic response analyses of structures, new standard response spectrum and upper and lower limits of the site coefficients of Fa and Fv at the short period range and the period of 1 second are suggested Including the structure-soil interaction effects.

Application of Engineered Zinc Finger Proteins Immobilized on Paramagnetic Beads for Multiplexed Detection of Pathogenic DNA

  • Micro-scale magnetic beads are widely used for isolation of proteins, DNA, and cells, leading to the development of in vitro diagnostics. Efficient isolation of target biomolecules is one of the keys to developing a simple and rapid point-of-care diagnostic. A zinc finger protein (ZFP) is a double-stranded (ds) DNA-binding domain, providing a useful scaffold for direct reading of the sequence information. Here, we utilized two engineered ZFPs (Stx2-268 and SEB-435) to detect the Shiga toxin (stx2) gene and the staphylococcal enterotoxin B (seb) gene present in foodborne pathogens, Escherichia coli O157 and Staphylococcus aureus, respectively. Engineered ZFPs are immobilized on a paramagnetic bead as a detection platform to efficiently isolate the target dsDNA-ZFP bound complex. The small paramagnetic beads provide a high surface area to volume ratio, allowing more ZFPs to be immobilized on the beads, which leads to increased target DNA detection. The fluorescence signal was measured upon ZFP binding to fluorophore-labeled target dsDNA. In this study, our system provided a detection limit of ≤ 60 fmol and demonstrated high specificity with multiplexing capability, suggesting a potential for development into a simple and reliable diagnostic for detecting multiple pathogens without target amplification.

Experimental Study and Confinement Analysis on RC Stub Columns Strengthened with Circular CFST Under Axial Load

  • As the excellent mechanical performance and easy construction of concrete filled steel tubes (CFST) composite structure, it has the potential to be used to strengthen RC pier columns. Therefore, tests were conducted on 2 reinforcement concrete (RC) stub columns and 9 RC columns strengthened with circular CFST under axial loading. The test results show that the circular CFST strengthening method is effective since the mean bearing capacity of the RC columns is increased at least 3.69 times and the ductility index is significantly improved more than 30%. One of the reasons for enhancement is obvious confinement provided by steel tube besides the additional bearing capacity supplied by the strengthening materials. From the analysis of the enhancement ratio, the strengthening structure has at least an extra 20% amplification except for taking full advantage of the strength of the strengthening material. Through the analysis of confining stress provided by steel tube and the stress-strain relationship of confined concrete, it is found that the strength of the core concrete can be increased by 21-33% and the ultimate strain can be enhanced to beyond $15,000{\mu}{\varepsilon}$.

Implementation of Memory Efficient Flash Translation Layer for Open-channel SSDs

  • Open-channel SSD is a new type of Solid-State Disk (SSD) that improves the garbage collection overhead and write amplification due to physical constraints of NAND flash memory by exposing the internal structure of the SSD to the host. However, the host-level Flash Translation Layer (FTL) provided for open-channel SSDs in the current Linux kernel consumes host memory excessively because it use page-level mapping table to translate logical address to physical address. Therefore, in this paper, we implemente a selective mapping table loading scheme that loads only a currently required part of the mapping table to the mapping table cache from SSD instead of entire mapping table. In addition, to increase the hit ratio of the mapping table cache, filesystem information and mapping table access history are utilized for cache replacement policy. The proposed scheme is implemented in the host-level FTL of the Linux kernel and evaluated using open-channel SSD emulator. According to the evaluation results, we can achieve 80% of I/O performance using the only 32% of memory usage compared to the previous host-level FTL.

  • The design response spectrum presented in the seismic design standard reflects the characteristics of the tectonic environment at a site. However, since the design response spectrum does not represent the ground motion with a specific earthquake magnitude or distance, input ground motions for response history analysis need to be selected reasonably. It is appropriate to use observed ground motions recorded in Korea for the seismic design. However, recently recorded ground motions in the Gyeongju (2016) or Pohang (2017) earthquakes are not compatible with the design response spectrum. Therefore, it is necessary to convert the recorded ground motion in Korea to a model similar to the design response spectrum. In this study, several approaches to adjust the spectral acceleration level at each period range were tested. These are the intrinsic and scattering attenuation considering the earthquake environment, magnitude, distance change by the green function method, and a rupture propagation direction's directivity effect. Using these variables, the amplification ratio for the representative natural period was regressed. Finally, the optimum condition compatible with the design response spectrum was suggested, and the validation was performed by converting the recorded ground motion.

Cyclic testing of a new visco-plastic damper subjected to harmonic and quasi-static loading

  • Visco-Plastic Damper (VPD) as a passive energy dissipation device with dual behavior has been recently numerically studied. It consists of two bent steel plates and segments with a viscoelastic solid material in between, combining and improving characteristics of both displacement-dependent and velocity-dependent devices. In order to trust the performance of VPD, for the 1st time this paper experimentally investigates prototype damper behavior under a wide range of frequency and amplitude of dynamic loading. A high-axial damping rubber is innovatively proposed as the viscoelastic layer designed to withstand large axial strains and dissipate energy accordingly. Test results confirmed all assumptions about VPD. The behavior of VPD subjected to low levels of excitation is elastic while with increasing levels of excitation, a significant source of energy dissipation is provided through the yielding of the steel elements in addition to the viscoelastic energy dissipation. The results showed energy dissipation of 99.35 kN.m under a dynamic displacement with 14.095 mm amplitude and 0.333 Hz frequency. Lateral displacement at the middle of the device was created with an amplification factor obtained ranging from 2.108 to 3.242 in the rubber block. Therefore, the energy dissipation of viscoelastic material of VPD was calculated 18.6 times that of the ordinary viscoelastic damper.
