  • Oxindoles(6a-g) are obtained by rhodium(II)-catalyzed Wolff rearrangement of diazoquinolinediones in moderate yields. Treatment of 6a-g oxindole derivatives reacted with CAN in acetonitrile at room temperature to gave 7a-g isatins in good yields. The structures of these compound were identified by MP, IR and NMR-spectra.

The mobile phase used after degassing was composed of 1 mM ammonium acetate and acetonitrile (20:80, pH 6.0), with flow rate of 200 uL/min.   • This study was designed to identify and quantitate airborne isocyanate simultaneously by HPLC. These samples were collected using 1-(2-pyridyl)piperazine(1-2PP) coated glass fiber filter from polyurethane painting works at 8 wood furniture factories in Kimpo and Inchun. The results obtained were as follows : 1. The most suitable mobile phase condition of simultaneously analyzing isocyanates was 0.01 M ammonium acetate buffer ACN(70/30) adjusted to pH 6.2 from the beginning of the analysis to 20 min and 0.01 M ammonium acetate buffer/ACN(50/50) adjusted to pH 6.2 from 21 min to 40 min using the gradient mode. The peaks of isocyanates were able to obtain within 30 min. 2. The recovery efficiencies for 2,6-TDI, 2,4-TDI, HDI and MDI urea derivates spiked at the target concentration on coated glass fiber tillers were 91.00, 93.42, 91.31 and 94.21 %, respectively. 3. The qualitative analysis of the isocyanates samples from polyurethane spray painting works in wood furnture factories identified Ihree isocyanates, 2,6-TDI, 2,4-TDI and MOI. And their concentration ranges were 0-312.6, 0-56.3 and $0-62.1{\mu}g/m^3$, respectively. A disadvantage of using the colorimetric method for isocyanate analysis is its inability of separating isocyanates. This study identified such three isocyanates as 2,6-TDI, 2,4-TDI and MDI from polyurethane spray painting works in wood furniture factories. These isocyanates were successfully quantitated by HPLC by modifying the mobile phase condition and switching to gradient mode.

  • Bacillus licheniformis KMU-3 shown a strong antifungal activity was isolated from Swedish forest soils. B. licheniformis KMU-3 produced a maximum level of antifungal substance under incubation aerobically at $24^{\circ}C$ for 24 hours in LB broth containing $1.0\%$ sodium acetate, $1.0\%$ ammonium sulfate at 180 rpm and initial pH adjusted to 8.0. Chloroform extraction of culture broth was confirmed inhibitory zone by plate assay and Rf value 0.49 substance by thin layer chromatography (TLC) represented high antifungal activity against Rhizoctonia solani AG-1. This substance also exhibited against Rhizoctonia solani AG-4, Colletotrichum orbiculare, Colletotrichum gloeosporioides, Cladosporium cucumerinum, Fusarium oxysporum, and Fusarium graminearum.

  • The present work is devoted to study the interaction of $\beta$-aroylacrylic acid derivative (3) with malononitrile in (DMF) in the presence of piperidine and/or ammonium acetate, then using the formed compounds as starting materials for synthesizing fused and isolated heterocyclic systems. It has been established that the $\beta$-aroylacrylic acid (3) reacts with malononitrile in (DMF) in the presence of piperidine as a catalyst with the formation of 4H-pyran derivative (4). By changing the catalyst into ammonium acetate, pyridine derivative (5) has been obtained. Also the N-maleamic acid derivatives (19) and (27) have been synthesized via the interaction of (4) and (5) with maleic anhydride. The purpose of this step is to study the behavior of the formed maleamic acid derivatives – as analogies of $\beta$-aroylacrylic acids – towards different active methylene compounds under Michael addition reaction.

High Density Cell Culture of Bifidobacterium by Optimization of Medium Composition and Culture Conditions. (배지조성 및 배양환경 최적화에 의한 Bifidobacterium의 고농도 배양)

  • Bifidobacterium strain was isolated from the feces of brast fed infants. The isolated strain was identified as Bifidobacterium longum by 16S rRNA sequence analysis and named as Bifidobacterium SH2. The MRS medium was modified to obtain high density cells of Bifidobacterium SH2. The optimal medium was determined to be 50 g/L lactose, 10 g/L beef extract, 10 g/L peptone, 5 g/L yeast extract, 7 g/L sodium acetate, 2 g/L ammonium citrate, 2 g/L disodium phosphate,1 g/L tween 80, 0.2 g/L MnSO$_4$ and 0.5 g/L L-cysteine. The pH and temperature were optimized as 5.0 and $37^{\circ}C$, respectively. Through out the optimization of medium composition and culture conditions, the dry cell weight and viable cell count were 2.5 times and 1.8 times higer than those in MRS medium, respectively.

Development of Freeze-Dried DOTMP Kits for Labeling with 68Ga

  • Lyophilized DOTMP kits were prepared using DOTMP, ammonium acetate, and ascorbic acid. The $^{68}Ga$-DOTMP was prepared by incubating the kit dissolved in 0.5 ml of concentrated $^{68}Ga$ using NaCl method and 0.5 ml of DDW, at $100^{\circ}C$ for 7 min. The labeling yield was evaluated by two solvent systems of TLC. 1 MBq of concentrated $^{68}Ga$ was labeled with $0.8{\mu}g$ of DOTMP by high radiolabeling yield (>98%), which was determined by two TLC methods. The composition of the prepared freeze-dried vial is $400{\mu}g$ of DOTMP, 19.27 mg of ammonium acetate and 17.62 mg of ascorbic acid. ~555 MBq of $^{68}Ga$-DOTMP was prepared with excellent radiochemical purity (>98%) and it was stable for 4 hr at room temperature. In conclusion, Freeze-dried DOTMP kits for the convenient preparation of $^{68}Ga$-DOTMP have been developed. Availability of this kit is expected to stimulate the widespread use of $^{68}Ga$-DOTMP in the fields of nuclear medicine.

Improved HPLC-UV method for determination of five synthetic dyes in Typha orientalis

  • Synthetic azo dyes are used extensively in herbal medicines to render the medicines more visually attractive to consumers. This study developed and validated a rapid high-performance liquid chromatography (HPLC) method to determine whether synthetic colorants such as Tartrazine, Auramine O, Metanil yellow, Sunset yellow, and Orange II are used extensively in Typha orientalis. To increase the recovery of the synthetic dyes, this method employed containing 50 mM ammonium acetate in 70 % methanol at first extraction and 100 mM HCl in 70 % methanol at second extraction. Five synthetic pigments in Typha orientalis were separated by gradient elution with a mobile phase consisting of acetonitrile and 50 mM ammonium acetate in distilled water at ultra-violet (UV) detection 428 nm or 500 nm. Additionally, this study established the liquid chromatography tandem mass spectrometry (LC-MS/MS) method to confirm positive samples suspected by HPLC results. The HPLC-UV method had good linearity, indicating r2> 0.999. The recoveries of the samples spiked with three different concentration ranged from 73.8~91.5 %, and relative standard deviation values indicated 0.2~5.2 %. The established LC-MS/MS could successfully identify the synthetic pigments in herbal medicine samples. The study demonstrates that Typha orientalis adulterated by yellowish synthetic dyes can be successfully distinguished when using the HPLC-UV method.

Screening of Effective Medium Composition for the Cultivation of Lactobacillus plantarum and Lactobacillus reuteri Using Statistical Methods (통계적 방법을 이용한 Lactobacillus plantarum과 Lactobacillus reuteri 의 유효 배지 성분의 탐색)

  • This study was conducted to develop an economical optimum medium composition for the mass production of $Lactobacillus$ $plantarum$ and $Lactobacillus$ $reuteri$, livestock probiotics. Medium ingredient factors were selected on the basis of MRS broth composition, and the 15 ingredient variables were as follows: sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, dipotassium phosphate, manganese chloride, magnesium chloride, tween 80, sodium chloride, sodium acetate, ammonium citrate, sodium sulphate, and ferrous sulphate. The Plackett Burman design, consisting of 20 runs, was employed for the analysis of ingredient effects on cell growth of $L.$ $plantarum$ and $L.$ $reuteri$. As a result, sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, sodium acetate, and ammonium citrate positively influenced the growth of $L.$ $plantarum$. Additionally, yeast extract, soy peptone, $K_2PHO_4$, and tween 80 positively influenced the growth of $L.$ $reuteri$. Positive effects were found from sucrose, yeast extract, and soy peptone in the integrated analysis of the effects of both $L.$ $plantarum$ and $L.$ $reuteri$. Finally, effective medium components for both strains were found as follows: sucrose (20.0 g/l), glucose (5.0 g/l), soy peptone (11.0 g/l), yeast extract (5.0 g/l), $K_2PHO_4$ (0.2 g/l), $CH_3COONa$ (2 g/l), and $MgCl_2$ (0.02 g/l).
