  • Effects of chitosan on growth of tomato plant, and suppression of Fusaruim wilt caused by Fusarium oxysporum f. sp. lycopersici and late blight casued by Phytophthora infestans, were examined. Both late blight and fusarium wilt were suppressed by spray and irrigation of chitosan, respectively. Inhibition of mycelial growth was not greatly affected by molecular size of chitosan but, concentration dependent effects was observed. Ninty percent of P. infestans and 80% of F. oxysporum f. sp. lycopersici of mycelial growth was inhibited by 1,000 ppm of chitosan (MW 30,000~50,000) when amended in plate media. Induction of defense-related gene expression in plant by chitosan treatments were observed when chitosan treated tobacco and tomato RNA samples were hybridized with several defense-related genes as probes. The results revealed that $\beta$-1,3-glucanase and chitinase genes were strongly induced, while pathogenesis-related protein-1, 3-hydroxy-3-methylglutaryl coenzyme A reductase, anionic peroxidase, phenylalanine ammonia lyase genes were weakly induced by chitosan treatment. These results suggest that chitosan have dual effects on these host-pathogen interactions. Possible roles of chitosan in suppression of tomato diseases by inhibition of mycelial growth and activation of plant defense responses are discussed.

Nutrient dynamics study of overlying water affected by peroxide-treated sediment

  • Background: Loading of excess nutrient via bioremediation of polluted sediment to overlying water could trigger anoxia and eutrophication in coastal area. The aim of this research was to understand the changes of overlying water features such as dissolved oxygen (DO); pH; oxidation reduction potential (ORP); $chlorophyll-{\alpha}$ ($Chl-{\alpha}$); and nitrogen nutrients ammonia ($N-NH_4{^+}$), nitrate ($N-NO_3{^-}$), and nitrite ($N-NO_2^-$) when the sediment was not treated (control) and treated by calcium peroxide for 5 weeks. Methods: The water samples were analyzed for measuring physical and chemical properties along with the sediment analyzed by polymerase chain reaction (PCR) including denaturing gradient gel electrophoresis (DGGE) for identifying the phylogenetic affiliation of microbial communities. Results: Results showed that due to the addition of calcium peroxide in sediment, the overlying water exposed the rise of dissolve oxygen, pH, and ORP than control. Among the nitrogen nutrients, ammonia inhibition was higher in calcium peroxide treatment than control but in case of nitrate inhibition, it was reversed than control. $Chlorophyll-{\alpha}$ was declined in treatment column water by 30% where it was 20% in control column water. Actibacter and Salegentibacter group were detectable in the calcium-peroxide-treated sediment; in contrary, no detectable community ware found in control sediment. Both phylogenetic groups are closely related to marine microflora. Conclusions: This study emphasizes the importance of calcium peroxide as an oxygen release material. Interaction with peroxide proved to be enhancing the formation of microbial community that are beneficial for biodegradation and spontaneity of nutrient attenuation into overlying water.

Comparative study on response of thiocyanate shock load on continuous and fed batch anaerobic-anoxic-aerobic sequential moving bed reactors

  • A comparative study on response of a toxic compound thiocyanate ($SCN^-$) was carried out in continuous and fed batch moving bed reactor systems. Both systems had three sequential anaerobic, anoxic and aerobic reactors and operated at same hydraulic retention time. Feed $SCN^-$ was first increased from 600 mg/L to 1,000 mg/L for 3 days (shock 1) and then from 600 to 1,200 mg/L for 3 days (shock 2). In anaerobic continuous reactor, increase of effluent COD (chemical oxygen demand) due to shock load was only 2%, whereas in fed batch reactor it was 14%. In anoxic fed batch reactor recovery was partial in terms of $SCN^-$, phenol, COD and $NO{_3}{^-}$-N and $NO{_2}{^-}$-N removals and in continuous reactor complete recovery was possible. In both systems, inhibition was more significant on aerobic reactors than anaerobic and anoxic reactors. In aerobic reactors ammonia removal efficiency deteriorated and damage was irreversible. Present study showed that fed batch reactors showed higher substrate removal efficiency than continuous reactors during regular operation, but are more susceptible to toxic feed shock load and in nitrifying reactor damage was irreversible.

Effect of 3,3',4',5-Tetrachlorosalicylanilide on Reduction of Excess Sludge and Nitrogen Removal in Biological Wastewater Treatment Process

  • A metabolic uncoupler, 3,3',4',5-tetrachlorosalicylanilide (TCS), was used to reduce excess sludge production in biological wastewater treatment processes. Batch experiments confirmed that 0.4 mg/l of TCS reduced the aerobic growth yield of activated sludge by over 60%. However, the growth yield remained virtually constant even at the increased concentrations of TCS when cultivations were carried out under the anoxic condition. Reduction of sludge production yield was confirmed in a laboratory-scale anoxic-oxic process operated for 6 months. However, it was found that ammonia oxidation efficiency was reduced by as much as 77% in the presence of 0.8 mg/l of TCS in the batch culture. Similar results were also obtained through batch inhibition tests with activated sludges and by bioluminescence assays using a recombinant Nitrosomonas europaea (pMJ217). Because of this inhibitory effect of TCS on nitrification, the TCS-fed continuous system failed to remove ammonia in the influent. When TCS feeding was stopped, the nitrification yield of the process was resumed. Therefore, it seems to be necessary to assess the nitrogen content of wastewater if TCS is used for reducing sludge generation.

  • Helicobacter pylori infection is associated with type B gastritis, peptic ulcer, and gastric cancer. The vacuolation of cells induced by H. pylori is thought to be essential for the initiation and maintenance of gastric infection. The roles of H. pylori cytotoxin, urease, and ammonia in the vacuolation of HeLa cells were determined. Ammonium chloride augmented the neutral red uptake induced by H. pylori toxin. Acetohydroxamic acid (AHA) failed to block the neutral red uptake induced by H. pylori toxin. Leweifang significantly prevented the vacuolation of HeLa cells induced by H. pylori toxin or H. pylori toxin and ammonium chloride. Further investigation is required to determine the mechanisms of Leweifang for the inhibition of vacuole formation of eukaryotic cells in response to the H. pylori toxin.

A comparison of individual and combined $_L$-phenylalanine ammonia lyase and cationic peroxidase transgenes for engineering resistance in tobacco to necrotrophic pathogens

  • This study tested the relative and combined efficacy of ShPx2 and ShPAL transgenes by comparing Nicotiana tabacum hybrids with enhanced levels of $_L$-phenylalanine ammonia lyase (PAL) activity and cationic peroxidase (Prx) activity with transgenic parental lines that overexpress either transgene. The PAL/Prx hybrids expressed both transgenes driven by the 35S CaMV promoter, and leaf PAL and Prx enzyme activities were similar to those of the relevant transgenic parent and seven- to tenfold higher than nontransgenic controls. Lignin levels in the PAL/Prx hybrids were higher than the PAL parent and nontransgenic controls, but not significantly higher than the Prx parent. All transgenic plants showed increased resistance to the necrotrophs Phytophthora parasitica pv. nicotianae and Cercospora nicotianae compared to nontransgenic controls, with a preponderance of smaller lesion categories produced in Prx-expressing lines. However, the PAL/Prx hybrids showed no significant increase in resistance to either pathogen relative to the Prx parental line. These data indicate that, in tobacco, the PAL and Prx transgenes do not act additively in disease resistance. Stacking with Prx did not prevent a visible growth inhibition from PAL overexpression. Practical use of ShPAL will likely require more sophisticated developmental control, and we conclude that ShPx2 is a preferred candidate for development as a resistance transgene.

Study on the optimization of partial nitritation using air-lift granulation reactor for two stage partial nitritation/Anammox process

  • This study aimed to develop a compact partial nitritation step by forming granules with high Ammonia-Oxidizing Bacteria (AOB) fraction using the Air-lift Granulation Reactor (AGR) and to evaluate the feasibility of treating reject water with high ammonium content by combination with the Anammox process. The partial nitritation using AGR was achieved at high nitrogen loading rate ($2.25{\pm}0.05kg\;N\;m-3\;d^{-1}$). The important factors for successful partial nitritation at high nitrogen loading rate were relatively high pH (7.5~8), resulting in high free ammonia concentration ($1{\sim}10mg\;FA\;L^{-1}$) and highly enriched AOB granules accounting for 25% of the total bacteria population in the reactor. After the establishment of stable partial nitritation, an effluent $NO_2{^-}-N/NH_4{^+}-N$ ratio of $1.2{\pm}0.05$ was achieved, which was then fed into the Anammox reactor. A high nitrogen removal rate of $2.0k\; N\;m^{-3}\;d^{-1}$ was successfully achieved in the Anammox reactor. By controlling the nitrogen loading rate at the partial nitritation using AGR, the influent concentration ratio ($NO_2{^-}-N/NH_4{^+}-N=1.2{\pm}0.05$) required for the Anammox was controlled, thereby minimizing the inhibition effect of residual nitrite.

Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism

  • The growth responses of a variety of human Intestinal bacteria to extracts of Pun(1.vKy'n.ieny and five other oriental medicinal Araliaceae were evaluated in vitro and in vivo. The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.

Growth and nutrient removal of Chlorella vulgaris in ammonia-reduced raw and anaerobically-digested piggery wastewaters

  • This study was aimed to investigate the possibility of using raw and anaerobically-digested piggery wastewater as culture media for a green microalga Chlorella vulgaris (C. vulgaris). Due to high concentration of ammonia and dark color, the microalga did not grow well in this wastewater. In order to solve this problem, air stripping and NaOCl-treatment were applied to reduce the concentration of NH3-N and the color intensity from the wastewater. Algal growth was monitored in terms of specific growth rate, biomass productivity, and nutrient removal efficiency. As a result, C. vulgaris grew without any sign of inhibition in air-stripped and 10-folds diluted anaerobically-digested piggery wastewater with enhanced biomass productivity of 0.57 g/L·d and nutrient removal of 98.7-99.8% for NH3-N and 41.0-62.5% for total phosphorus. However, NaOCl-treatment showed no significant effect on growth of C. vulgaris, although dark color was removed greatly. Interestingly, despite that the soluble organic concentration after air stripping was still high, the biomass productivity was 4.4 times higher than BG-11. Moreover, air stripping was identically effective for raw piggery wastewater as for anaerobic digestate. Therefore, it was concluded that air stripping was a very effective method for culturing microalgae and removing nutrients from raw and anaerobically-digested piggery wastewaters.

유입부하가 DEPHANOX 및 Modified-DEPHANOX 공정에 미치는 영향 (Effect of Loading Rate in the Operation of DEPHANOX and Modified-DEPHANOX Processes)

  • This study was initiated to evaluate the efficiencies of DEPHANOX and Modified-DEPHANOX, which were devoloped to enhance nitrogen removal efficiency in municipal wastewater treatment. In the results, removal efficiency of organic matters was not affected much by increased loading rate of organic matters which is contained in influent. The nitrogen removal efficiencies according to the loading rate of influent TN was decreased drastically in conditions of over $0.2kg/m^3{\cdot}day$, which is T-N loading rate, and the DEPHANOX process was affected more sensitively than the M-DEPHANOX was. When the temperature was altered from $25^{\circ}C$ to $16^{\circ}C$ at HRT 6hrs, the removal efficiency of ammonia nitrogen was still over 90% and it was concluded that both DEPHANOX and M-DEPHANOX were strong enough to endure temperature variation. Moreover, both processes showed over 90% in ammonia removal efficiencies in over HRT 5hrs, so it was concluded that they were strong in HRT variation. M-DEPHANOX process showed a higher value than DEPHANOX did in T-N removal efficiency to the extent of 4~21 %, which resulted from differency of denitrification rates and the biosorption efficiency of organic matter in both processes. In the condition of HRT less than 4hrs, concentrations of ammonia nitrogen contained in effluents and nitrification reactors, might be sensitively affected by biosorption efficiency of organic matters in first separation tank. In the effect of effluent nitrate concentration in phosphorus removal, the more effluent nitrate concentration was decreased, the more phosphorus removal efficiency was increased. This result is related to the decrease of concentration of effluent nitrate which resulted from nitrification inhibition by decreased HRT.
