Dual positional substrate specificity of rice allene oxide synthase-1: insight into mechanism of inhibition by type II ligand imidazole

  • Phylogenetic and amino acid sequence analysis indicated that rice allene oxide synthase-1 (OsAOS1) is CYP74, and is clearly distinct from CYP74B, C and D subfamilies. Regio- and stereo-chemical analysis revealed the dual substrate specificity of OsAOS1 for (cis,trans)-configurational isomers of 13(S)- and 9(S)-hydroperoxyoctadecadienoic acid. GC-MS analysis showed that OsAOS1 converts 13(S)- and 9(S)-hydroperoxyoctadecadi(tri)enoic acid into their corresponding allene oxide. UV-Visible spectral analysis of native OsAOS1 revealed a Soret maximum at 393 nm, which shifted to 424 nm with several clean isobestic points upon binding of OsAOS1 to imidazole. The spectral shift induced by imidazole correlated with inhibition of OsAOS1 activity, implying that imidazole may coordinate to ferric heme iron, triggering a heme-iron transition from high spin state to low spin state. The implications and significance of a putative type II ligand-induced spin state transition in OsAOS1 are discussed.

토하(土蝦) 및 젓새우의 함질소(含窒素) 엑스성분(成分)에 관(關)한 연구(硏究) (Extractive Nitrogenous Constituents of Toha (Caridina denticulata denticulata, Freshwater Shrimp) and Jeotsaeu (Acetes japonicus , Seawater Shrimp))

  • 본 연구는 토하(Caridina denticulata denticulata)의 식품학적인 우수성을 밝히기 위하여 식품성분 중 맛과 밀접한 관계를 가지고 있는 함질소 엑스성분을 상세히 분석한 후 그 성분조정을 파악하는 한편, 이와 비교하기 위하여 바다에서 어획되는 젓새우(Acetes japonicus)에 대해서도 동일한 이화학 성분들을 분석하여 그 성분 차이를 검토함으로써 토하에 대한 맛의 특성을 구명하고자 하였다. 토하는 1994년 12월부터 1995년 11월 사이에 전남 나주군에서 양식된 것을, 그리고 젓새우는 1994년 12월과 1995년 11월 전남 신안군에서 어획된 것을 시료로하여 엑스분질소, 유리아미노산, oligopeptide류, 핵산관련물질, betaine류, 4급 암모늄염기, 구아니시노화합물 등을 분석하였다. 토하는 젓새우보다 엑스분질소, 유리아미노산, oligopeptide류, 핵산관련물질의 함량이 높은 편이었고, 젓새우는 토하보다 homarine, trimethylamine oxide, trimethylamine이 높은 편이었으며, creatine과 creatinine은 토하와 젓새우에서 미량으로서 큰 차이가 없었다. 토하의 중요한 유리아미노산으로는 arginine, lysine, glutamine, glutamic acid, pheylalanine, leucine, alanine, asparagine, glycine 등이었고, 젓새우의 중요한 유리아미노산은 arginine, glutamic acid, proline, glutamine, lysine, leucine, glycine, alanine, taurine 등이었다. 그리고 methionine, glutamine, valine 등은 토하와 젓새우에서 그 함량이 같은 수준이었다. 토하에서 가장 함량이 많은 핵산관련물질은 AMP였으며, 젓새우에서는 inosine이었다. 이상의 결과로부터 토하는 맛을 나타내는데 중요한 역할을 하는 대부분의 함질소 엑스성분들의 함량이 젓새우보다 높았기 때문에 식품학적으로 더 우수한 가공원료라고 판단되었다.

Amino-style 유도체를 이용한 분자 전자 소자의 전류-전압 특성에 관한 연구 (Current-Voltage Characteristics of Molecular Electronic Devices Using a Amino-Style Derivatives)

  • Organic molecules have many properties that make them attractive for electronic applications. We have been examining the progress of memory cell by using molecular-scale switch to give an example of the application using both nanoscale components and Si-technology. In this study, molecular electronic devices were fabricated with amion style derivatives as redox-active component to compare to the devices using Zn-Porphyrin derivatives. This molecule is amphiphilic to allow monolayer formation by the Langmuir-Blodgett (LB) method, and then this LB monolayer is inserted between two metal electrodes. According to current-voltage (I-V) characteristics, it was found that the devices show remarkable hysteresis behavior and can be used as memory devices at ambient conditions, when aluminum oxide layer was existed on bottom electrode. Diode-like characteristics were measured only, when Pt layer was existed as bottom electrode. It was also found that this metal layer interacts with the organic molecules and acts as a protecting layer, when thin Ti layer was inserted between the organic molecular layer and the top Al electrode. These electrical properties of the devices may be applicable to active components for the memory and/or logic gates in the future.

Apparent Amino Acid and Energy Digestibilities of Common Feed Ingredients for Flounder Paralichthys olivaceus

  • Flounder were fed a reference diet and test diets containing various feed ingredients: mackerel fish meal, meat meal, soybean meal, wheat flour, wheat gluten, corn gluten meal and brewer's yeast. Apparent digestibility was determined using a reference diet with 0.5% chromic oxide indicator and test diets contained 70% reference diet and 30% of the feed ingredient being evaluated. Apparent digestibility coefficients for amino acid and energy in the reference and test diets were determined, and digestibility coefficients for the test ingredients were calculated based on differences in the digestibility of test diets relative to the reference diet. The fish averaging 300 g were held in 2000 L tanks at a density of 20 fish per tank. Feces were collected from three replicate groups of fish using a fecal collection column attached to fish rearing tank. Apparent total amino acids digestibilities(90-95%) of mackerel fish meal, soybean meal, wheat gluten, corn gluten meal and brewer's yeast were higher than those of meat meal and wheat flour(P<0.05). Apparent energy digestibilities(86-98%) of mackerel fish meal, meat meal, soybean meal, wheat gluten and corn gluten meal were significantly higher(P<0.05) than those of wheat flour and brewer's yeast. These results provide useful information about nutrient and energy utilization for flounder.

Standardized Ileal Digestibility of Amino Acids in Feed Ingredients for Broiler Chickens

  • The objective of this experiment was to determine the apparent (AID) and standardized ileal digestibility (SID) of crude protein (CP) and amino acids (AA) in five feed ingredients for male broiler chickens (Ross 308). The test feed ingredients consisted of corn, wheat, soybean meal (SBM), canola meal (CM), and corn distillers dried grains with solubles (DDGS). The birds were fed a standard starter diet for the first 19 d. On day 19, nine hundred sixty birds were assigned to six dietary treatments with eight replicate cages (20 birds/cage) in a randomized complete block design. All experimental diets contained chromic oxide (0.5% in the diet) as an indigestible index. The diets were offered ad libitum for 4 d. At the end of the experiment, all birds were euthanized and ileal digesta samples were collected immediately. The indispensable AA with the least AID and SID were Thr for all the ingredients used in the present study, except for DDGS, in which Lys had the lowest digestibility. For indispensable AA, the AID and SID of Leu, Phe, Arg, and Met were the greatest in corn, DDGS, wheat, SBM, and CM, respectively. The results from the present study showed that there are variations in both the AID and SID of CP and AA among feed ingredients.

도판트를 이용한 적색 유기 발광 다이오드의 제작 및 특성 연구 (Fabrication and Characterization of Red Organic Light-Emitting Diodes Using Red Fluorescent Material)

  • 본 연구에서 새로 합성한 적색 도판트 Red-1을 Physical vapor Deposition (PVD) 법을 이용하여 다층구조의 유기 발광 다이오드를 제작하였다. 적층된 유기물 층으로 정공주입층은 4,4',4"-tri [2-naphthyl(phenyl)amino]triphenylamine(2-TNATA) 정공 수송층으로4-4bis [N-(1-napthyl-N-phenyl-amino)biphenyl] (NPB)를 사용하였으며 전자 수송층은 tris (8-quinolinolato)-aluminum ($Alq_3$), 발광층에서의 host 재료로 사용한 물질은 $Alq_3$. 4,4'- N-N'-dicarba zole-biphenyl (CBP), 게스트재료는 Red-1, 정공저지층으로 2,9-dimethyl-4, 7-diphenyl -1 10-phen antroline (BCP), 전자 주입층으로는 lithiumquinolate (Liq)를 사용하여 보다 향상된 전기적, 발광특성을 보이는 소자를 제작하였다. 전하를 주입하는 전극으로 일함수가 큰 투명전극인 ITO (indium-tin-oxide)를 양전극으로, Al을 음전극으로 사용하였다. 그리하여, 발광층 내에서의 host재료 $Alq_3$와 CBP와의 energy transfer의 관점에서 그 특성을 연구하였다.

아미노실란화 철산화물 나노입자를 이용한 Human DNA의 초고속 자성분리 (High Throughput Magnetic Separation for Human DNA by Aminosilanized Iron Oxide Nanoparticles)

  • This work describes the preparation of functionalized magnetic nanoparticles(MNPs) and their bioapplication to human DNA separation. Silica coated MNPs were prepared by changing the volume ratio of tetraethyl orthosilicate(TEOS) for controlled coating thickness on the original nanoparticle of MNPs. The sol-gel process in silica coating on MNPs surface was adapted for relatively mild reaction condition, low-cost, and surfactant-free. And then amino functionalized magnetic nanoparticles were synthesized using amine groups as surface modifiers. The result of adsorption efficiency for human DNA with amino-functionalized silica coated MNPs was calculated as a function of the number of amine groups.

Ammonium Chloride Solution Leaching of Crude Zinc Oxide Recovered from Reduction of EAF′s Dust

  • EAF's dust has been treated mainly by pyrometallurgical reduction process in rotary kiln furnace to recover valuable metal elements such as Zn and to avoid the disposal of hazardous materials to waste. Recently, hydrometallurgical eletrowinning of zinc from a zinc-amino chloride solution obtained by the leaching of EAF's dust was developed to recover high grade zinc metal from EAF’s dust. But there are some disadvantages in each process such as difficulty of operation condition control and sticking problem in kiln process and low extractability and recovery of zinc owing to insoluble zinc-ferrite in electrowinning process. We propose a new combined process of pyrometallurgical one and hydrometallurgical one to treat EAF's dust efficiently and economically. In this study, ammonium chloride solution leaching of crude zinc oxide recovered from reduction of EAF's dust was carried out to find out the efficiency of zinc extraction from it and the possibility for performance of eletrowinning in the proposed process. Effects of various leaching variables ruck as leaching temperature, concentration of leaching solution and leaching time were investigated. And the leaching results of the crude zinc oxide were compared with those of EAF's dust. The extraction percents of zinc in ammonium chloride solution leaching of the crude zinc oxide recovered from reduction of EAF's dust were above 80% after 60 minutes of leaching under the leaching condition of 4M NH$_4$CI concentration and above leaching temperature of 7$0^{\circ}C$. And the concentrations of zinc in the leached solution were obtained above 50g/$\ell$. The activation energy calculated for zinc extraction in NH$_4$CI leaching was 58.1 KJ/㏖ for EAF's dust and 15.8 KJ/㏖ for the crude zinc oxide recovered from reduction of EAF's dust.

열 이미드화 온도에 따른 작용기화 그래핀/폴리이미드 나노복합재료 (Functionalized Graphene/Polyimide Nanocomposites under Different Thermal Imidization Temperatures)

  • 폴리이미드(PI) 나노복합체 필름 제조에 사용된 작용기화 4-amino-N-hexadecylbenzamide graphene sheets (AHB-GSs)는 graphene oxide 분산액에 4-amino-N-hexadecylbenzamide(AHB)를 반응시켜 합성하였다. AHB-GS의 주사탐침 현미경(atomic force microscope, AFM) 이미지와 모식도를 통해서 AHB-GS의 평균 두께가 약 3.21 nm임을 확인하였다. PI는 4,4'-biphthalic anhydride와 bis(4-aminophenyl)sulfide를 사용하여 합성하였다. PI 나노복합체는 0-10 wt%의 다양한 함량의 AHB-GS를 용액 삽입(solution intercalation) 방법을 사용하여 합성하였고, 이미드화는 각각 $250^{\circ}C$$350^{\circ}C$까지 열 처리하였다. AHB-GS는 대부분 고분자 매트릭스에 잘 분산되었고 약간 뭉친 것도 있었지만 마이크로미터 수준의 입자는 관찰되지 않았다. TEM으로 관찰하였을 때, 평균적으로 입자의 두께는 10 nm 미만이었다. PI 복합체 필름 중 소량의 AHB-GS만으로도 가스 투과도와 전기 전도도는 향상되었지만, 반대로 유리전이 온도와 초기 분해 온도는 AHB-GS의 함량이 10 wt%까지 증가함에 따라 지속적으로 감소되는 경향을 보였다. 전체적으로는, $250^{\circ}C$까지 이미드화한 PI에 비해 $350^{\circ}C$까지 열처리한 PI 필름이 보다 향상된 특성을 보였다.

Prediction of Digestible and Metabolizable Energy Content and Standardized Ileal Amino Acid Digestibility in Wheat Shorts and Red Dog for Growing Pigs

  • Two experiments were conducted to evaluate the effects of chemical composition of wheat shorts and red dog on energy and amino acid digestibility in growing pigs and to establish prediction models to estimate their digestible (DE) and metabolizable (ME) energy content and as well as their standardized ileal digestible (SID) amino acid content. For Exp. 1, sixteen diets were fed to thirty-two growing pigs according to a completely randomized design during three successive periods. The basal diet was based on corn and soybean meal while the other fifteen diets contained 28.8% wheat shorts (N = 7) or red dog (N = 8), added at the expense of corn and soybean meal. Over the three periods, each diet was fed to six pigs with each diet being fed to two pigs during each period. The apparent total tract digestibility (ATTD) of energy in wheat shorts and red dog averaged 75.1 and 87.9%. The DE values of wheat shorts and red dog averaged 13.8 MJ/kg (range 13.1 to 15.0 MJ/kg) and 15.1 MJ/kg (range 13.3 to 16.6 MJ/kg) of dry matter, respectively. For Exp. 2, twelve growing pigs were allotted to two $6{\times}6$ Latin Square Designs with six periods. Ten of the diets were formulated based on 60% wheat shorts or red dog and the remaining two diets were nitrogen-free diets based on cornstarch and sucrose. Chromic oxide (0.3%) was used as an indigestible marker in all diets. There were no differences (p>0.05) in SID values for the amino acids in wheat shorts and red dog except for lysine and methionine. Apparent ileal digestibility (AID) and SID values for lysine in different sources of wheat shorts or red dog, which averaged 78.1 and 87.8%, showed more variation than either methionine or tryptophan. A stepwise regression was performed to establish DE, ME and amino acid digestibility prediction models. Data indicated that fiber content and amino acid concentrations were good indicators to predict energy values and amino acid digestibility, respectively. The present study confirms the large variation in the energy content and amino acid digestibility in wheat shorts and red dog, and describes the factors that influence this variation and presents equations based on chemical composition that could probably be used to predict the DE and ME values as well as the amino acid digestibility of wheat shorts and red dog.
