Biocatalytic production of chiral epoxide: Epoxide hydrolase-catalyzed enantioselective resolution

  • A newly isolated Aspergillus niger possessing the novel epoxide hydrolase(EHase) activity was investigated for the enantioselective hydrolysis of racemic aromatic epoxides. The gene encoding EHase was cloned by RT-PCR, and molecular characteristics of the EHase gene were compared with other microbial EHases. The cloned gene encodes 398 amino acids with a deduced molecular mass of 44.5 kDa and pI of 4.83, and sequence homology with other microbial EHase was low. Functional recombinant EHase could be obtained by heterologous expressions in E. coli. Enantioselectivity of recombinant EHase was tested for valuable aromatic epoxide intermediates. Reaction conditions of EHase-catalyzed asymmetric resolution were optimized for the production of chiral styrene oxide.

  • Nitrosative modifications regulate cellular signal transduction and pathogenesis of inflammatory responses and neuro-degenerative diseases. Protein tyrosine nitration is a biomarker of oxidative stress and also influences protein structure and function. Recent advances in mass spectrometry have made it possible to identify modified proteins and specific modified amino acid residues. For analysis of nitrated peptides with low yields or only a subset of peptides, affinity 'tags' can be bait for 'fishing out' target analytes from complex mixtures. These tagged peptides are then extracted to a solid phase, followed by mass analysis. In this review, we focus on protein tyrosine modifications caused by nitrosative stresses and proteomic methods for selective enrichment and identification of nitrosative protein modifications.

Optimization of Expression Conditions Enhances Production of Sepiapterin, a Precursor for Tetrahydrobiopterin Biosynthesis, in Recombinant Escherichia coli

  • Sepiapterin is a precursor for the synthesis of tetrahydrobiopterin (BH4), which is a wellknown cofactor for aromatic amino acid hydroxylation and nitric oxide synthesis in higher mammals. In this study, a recombinant Escherichia coli BL21(DE3) strain harboring cyanobacterial guanosine 5’-triphosphate cyclohydrolase 1 (GCH1) and human 6-pyruvoyltetrahydropterin synthase (PTPS) genes was constructed to produce sepiapterin. The optimum conditions for T7 promoter–driven expression of GCH1 and PTPS were 30℃ and 0.1 mM isopropyl-β-D-thioglucopyranoside (IPTG). The maximum sepiapterin concentration of 88.1 ± 2.4 mg/l was obtained in a batch cultivation of the recombinant E. coli, corresponding to an 18-fold increase in sepiapterin production compared with the control condition (37℃ and 1 mM IPTG).

  • Peroxynitrite(ONOO-), formed from the reaction of superoxide(O2-) and nitric oxide(NO), is a potent oxidant that contributes to oxidation of various cellular constituents including lipids. amino acids, sulphydryls and nucleotides. It can cause cellular injury such as DNA fragmentation and apoptotic cell death. Also. the toxicity of ONOO- has been reported to be involved in inflammatory and nurodegenerative diseases such as Alzheimer's disease, Parkinson's disease. and atherosclerosis. (omitted)

Simultaneous removal of nitrate and phosphate by micellar-enhanced ultrafiltration(MEUF) using PENTANOX 4X

  • The feasibility of PENTANOX 4X for the simultaneous removal of nitrate and phosphate was investigated using micellar-enhanced ultrafiltration. Because PENTANOX 4X has cationic property at low pH, anionic contaminants can be bound to PENTANOX 4X micelle by electrostatic interaction. At pH 3, 90% of nitrate and 72% of phosphate were removed by 27 mM of PENTANOX 4X, which were equivalent to 20 mM of CPC. PENTANOX 4X of > 80 % was rejected by ultrafiltration membrane and did not make any counter-ion such as chloride for CPC which might cause second-pollution.

  • Rosin maleic anhydride adduct (RMA)-bisester was prepared by the esterification of chlorinated RMA with hydroquinone. Phenylpyridinylcyclot.isiloxane ($D_3^{Ph,Py}$) was synthesized from phenylpyridinyldichlorosilane in the presence of zinc oxide catalyst, and amino group terminated polyphenylpyridinylsiloxane prepolymer was prepared by equilibrium polymerization of $D_3^{Ph,Py}$ with 1,3-bis(3-aminopropyl)-1,1,3,3-tetramethyldisiloxane. Rosinimide (PSMR) was prepared from the imidization of RMA-bisester with polyphenylpyridinylsiloxane prepolymer at 12$0^{\circ}C$ for 7 h using ${\gamma}$-butyrolactone/pyridine. It showed that PSMR had better thermal stalbility than rosinimide modified with polydimethylsiloxane.

  • ITO/4, 4', 4"-tris (N -3 - methylphenyl - N - phenyl - amino) - triphenylamine, [m-MTDATA] / Terbium Iris - (1 - phenyl - 3 - methyl - 4 - (tertiarybutyryl) - pyrazol - 5 - one) triphenylphosphine oxide [$(tb-PMP)_3Tb-(Ph_3PO)$] / Mg:Ag devices were made to investigate its electrical and light emission properties. The thickness of m-MTOATA layer was varied from 0 to 80 nm. There was a threshold thickness for the sufficient hole injection. The insertion of 20 nm thick m-MTDATA layer between ITO and Tb-complex resulted in the right shift of current-voltage curve because of the insufficient hole injection. The low operating voltage can be obtained above the 40 nm of m-MTDATA layer. The insertion of m-MTDATA induced the increase of the background in the electroluminescence spectrum which was dependent on the current density of the devices.

  • 우렁쉥이의 식품학적 가치와 성분적 특성을 밝히는 일환으로 정미성분을 포함한 엑스성분과 그 계절적 변화를 실험한 결과를 요약하면 다음과 같다. 1. 우렁쉥이의 계절적 Ex-질소량은 4월에 250mg/100g에서 9월에 330mg/100g으로 점점 증가하는 경향을 보였다. 2. 우렁쉥이의 유리아미노산중 taurine, proline, glutamic acid, alanine, glycine의 함량이 높았다. 3. 핵산관련물질중 AMP의 함량이 높았다. 4. Glycinebetain의 함량이 homarine보다 훨씬 높았으며 9월에 특히 많았고, 그 계절적 변화는 엑스분질소 및 유리아미노산의 변화와 비슷한 경향을 보였다. 5. 우렁쉥이의 엑스분질소중 함질소엑스성분이 차지하는 조성은 유리아미노산이 $60{\sim}62\%$로 가장 높았으며, 다음으로 betaine $12{\sim}16\%$, 핵산관련물질 $5{\sim}9\%$, TMAO 및 TNIA 총 creatinine의 순이었다. 6. 우렁쉥이의 유기산은 succinic, malic, lactic 및 pyroglutaric acid가 전체 유기산의 $84\%$ 이상을 차지하며 succinic acid의 함량이 가장 높았다. 7. Omission test 결과 우렁쉥이 맛은 유리아미노산, betaine류, 핵산관련물질, 불휘발성 유기산 순으로 영향을 미치는 것으로 나타났다.

담수어의 정미성분에 관한 연구 6. 메기의 정미성분 (Taste Compounds of Fresh-Water Fishes 6. Taste Compounds of Korean Catfish Meat)

  • 우리 나라 중요 담수어의 정미성분을 밝힐 목적으로 전보에 이어 메기배육중의 유리아미노산, 핵산관련물질, 유기염기, 당류, 유기산 및 무기질을 분석하였다. 또한 이 분석치를 기초로 하여 표준 시약을 가지고 합성엑스분을 조제하여 천연엑스분과 맛을 비교하였으며, omission test를 실시하여 각 성분들이 맛에 미치는 영향을 조사하였다. 유리아미노산의 함량은 glycine이 가장 맞아 전체 유리아미노산의 $25\%$를 차지하였고, 다음으로 alanine, lysine, taurine등이 많았다. 핵산관련물질은 IMP 함량이 98.9mg/100g으로서 가장 많았으며, 그 외 성분은 비교적 함량이 적었다. 유기염기는 creatinine이 351.7mg/100g으로서 가장 많았고 betaine은 18.0mg/100g이었으며 TMA 및 TMAO는 1.4mg/100g 이하로서 극미량이었다. 유기산의 경우는 숙신산, 브티르산, 프로피온산 등이 함량이 많았고 옥살산, 푸마르산, 말레산, 타르타르산 및 시트르산은 흔적량에 불과하였다. 당류의 함량은 포도당이 5.1mg/100g으로서 가장 많았으며, ribose, arabinose, fructose및 inositol은 약 1mg/100g으로서 미량이었다. 무기염류의 함량은 $K^+$$PO_{4}^{3-}$이 각각 210.0mg/100g, 305.8mg/100g으로서 월등히 많았고 다른 무기염류는 38.0mg/100g이하였다. 관능검사 결과로 본 매기의 주된 정미성분은 serine, alanine, IMP, 숙신산 및 $PO_{4}^{3-}$였다.

수확시기별 까마중 전초의 성분, 항산화활성 및 항염증 활성 (The chemical constituents, antioxidant activity and anti-inflammatory activities of Solanum nigrum Linne by different harvest time)

  • 본 연구는 까마중 수확시기에 따른 성분변화, 항산화효과 및 항염증 효과를 분석하여 최적 수확시기를 구명하고자 수행하였다. 까마중의 주요 아미노산은 수확시기에 관계없이 proline, histidine 과 serine으로 나타났으며, 구성아미노산 함량은 7월 10일과 8월 10일 수확한 까마중 지상부와 지하부에서 높게 나타났다. 4가지 유리당이 검출되었으며, 유리당 함량은 수확시기에 따라 편차를 나타내었다. 유리당 중 fructose는 수확시기가 늦어짐에 따라 감소하였으며, 총 폴리페놀 함량은 8월 30일 수확한 까마중 뿌리에서 가장 높게 나타났다. 수확기별 까마중 ethanol 추출물들의 항산화효과와 항염증효과 검정을 위하여 DPPH radical scavenging assay와 ABTS radical scavenging assay를 수행하였으며, 항염증 효과 측정을 위하여 lipopolysaccharide(LPS)로 염증을 유발시킨 C6 glioma cells을 NO 저해실험에 사용하였다. 항산화효과와 항염증 효과는 8월 30일 수확한 까마중 뿌리 ethanol 추출물에서 가장 높은 활성을 나타내었다. 8월 30일 수확한 까마중 뿌리는 총 폴리페놀 함량, 항산화 효과 및 항염증 효과간의 양적인 상관관계를 나타내었다.
