Antimicrobial Susceptibility of Vibrio parahaemolyticus and Vibrio alginolyticus from Fish Farms on the Southern Coast of Korea (남해안 어류양식장에서 분리된 Vibrio parahaemolyticus와 Vibrio aiginolyticus의 항균제 감수성)

  • The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit,imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).

  • Vibrio spp. are aquatic bacteria that are ubiquitous in warm estuarine and marine environments. Especially, V. vulnificus and V. cholerae are currently known to cause potentially fatal infections in humans. This study investigated the distribution and antibiotic resistance of V. vulnificus and V. cholerae isolated from coastal areas of Gyeongsangnam-do in 2022. A total of 252 samples of water, shellfish and coastal sediment were collected from 7 locations along the coast, and 124 samples of fishery products were collected from markets. Among the 252 samples, forty-four V. vulnificus (11.7%) and fourteen V. cholerae non-O1/non-O139 (3.7%), none of which carried the ctx gene, were isolated. Out of the 124 samples, 6 (4.8%) tested positive for V. vulnificus and V. cholerae was not detected. The isolation rates of V. vulnificus and V. cholerae showed a significant correlation with environmental factors such as seawater temperature and salinity. In an antibiotic resistance test, V. vulnificus was susceptible to amikacin, gentamicin, imipenem trimethoprim/sulfamethoxazole, and ciprofloxacin, but resistant to cefoxitin (100.0%), followed by tetracycline (9.1%). Multidrug resistance was also observed. Continuous monitoring of Vibrio pathogens with water temperature and salinity is expected to help reduce the outbreaks, and rational use of antibiotic agents is needed to prevent the accession of antibiotic-resistant microorganisms in aquatic ecosystems.

Prevalence and Characterization of Pathogenic Escherichia coli Isolated from the Livestock Environment in Chungcheongnam-do Province of South Korea (충남지역 농장환경에서 분리된 병원성대장균 분포 및 특성 분석)

  • Background: One of the major causes of pathogenic E. coli is the feces of infected livestock, and the management of the livestock environment is necessary to prevent pathogenic E. coli. Objectives: The prevalence of pathogenic E. coli was identified from livestock environments, and the molecular characteristics and antibiotic resistance profiles of the isolated pathogenic E. coli strains were analyzed. Methods: In 2022 and 2023, nine points of livestock houses at sites in Chungcheongnam-do Province were selected, and 100 cow feces or soil samples around the livestock houses were collected once per month. Pathogenic E. coli was isolated by selective culture and identified using multiplex PCR. Antibiotic resistance was tested on the isolated strains by using VITEK-2, and candidate strains were selected to perform 16s rRNA sequencing and phylogenetic analysis. Results: A total of 100 samples were tested, and 60 pathogenic E. coli strains were isolated. Of these, 45 and 15 isolates were determined to be single and hybrid pathogenic E. coli , respectively. Among the 15 hybrid pathogenic E. coli strains, eight, five, and two strains were respectively identified as EHEC/ETEC, EHEC/EPEC, and EHEC/ETEC/EPEC hybrids. All 45 isolates showed resistance to at least one antibiotic, and they were susceptible to cefotaxime, amikacin, nalidixic acid, and ciprofloxacin. The highest resistance was against cefalotin, tetracyclin, and ampicillin (20.0%~58.3%). The 16s rRNA sequences of candidate isolates revealed nucleotide sequence identities of 99.1% to 100%. Conclusions: In order to manage pathogenic E. coli from the One Health animal environment perspective, the characteristics of the occurrence of pathogenic E. coli from the livestock environment and molecular biology and antibiotic resistance to isolated strains were analyzed. In order to prevent and manage the occurrence of pathogenic E. coli, these monitoring studies must be continuously conducted.

  • Background: Nosocomial pneumonia in an intensive care unit (ICU) is associated with a high mortality rate. Diagnosing a respiratory tract infection in critically ill patients is still difficult but detailed information for the pathogens is needed to establish an adequate antimicrobial treatment. This study examined the causative organisms and their antimicrobial resistance using bronchoalveolar lavage (BAL) from patients suspected of having pneumonia in the ICU. Methods: From January 2004 to June 2006, ICU patients with diffuse lung infiltration were prospectively enrolled. The BAL was used to diagnose the respiratory infection, with 104 > or = organisms considered a positive result. The most common organisms and their antimicrobial resistances were analyzed from the quantitative BAL cultures in the burn ICU and non-burn ICU. Results: A total 72 patients were included, 35 (M 29, F 6) in the burn ICU and 37 (M 26, F 11) in the non-burn ICU. 27 patients (77.1%) in the burn ICU and 22 patients (59.5%) in the non-burn ICU met the criteria for a positive BAL culture. The major pathogens were Staphylococcus aureus, Acinetobacter species and Pseudomonas aeruginosa. All strains (100%) of Staphylococcus aureus isolated from BAL (9 cases) were methicillin-resistant (MRSA) in the burn ICU, but 5 strains (71.4%, 7 cases) were MRSA in the non-burn ICU. Regarding Pseudomonas aeruginosa, the rate of resistance to amikacin, ciprofloxacin, cefepime, imipenem, ceftazidime, piperacillin/tazobactam in the burn and non-burn ICU ranged from 45.5% to 90% and 25% to 50%, respectively. In addition, the rate of resistance of Acinetobacter species to the above drugs in the burn and non-burn ICU ranged from 81.8% to 100% and 62.5% to 100%, respectively. Conclusions: These results are expected to provide useful guidelines for choosing the effective empirical antimicrobial therapy in patients with lung infiltrations in the burn and non-burn ICU.

  • To investigate the epidemiological trait of intestinal diseases of animals caused by thermophilic Campylobacter spp., isolation of etiological agent was carried out. Isolated Campylobacter spp. were biotyped, serotyped and the susceptibility of the isolates to antimicrobial agents were examined. Th results were as follows. 1. Isolation rates of Campylobacter spp. from 649 fecal materials of 208 cattle, 300 pigs and 141 chickens were 25.5%, 23.7% and 38.3%, respectively. 2. The majority of the 130 isolates of C jejuni was classified as biotype I(50.6%) and biotype II (34.6%). Most of the 46 isolates of C coli were biotype I (71.7%). 3. Isolated C jejuni strains showed 14 different serotype, and serotype 4, 26, 36 were most frequent. Isolated C coli strains showed 5 different serotype and serotype 31 and 21 were relatively common. 4. Isolated Campylobacter spp. were highly susceptible to nalidixic acid, amikacin, gentamicin, colistin and chlorampehnocol.

Molecular typing of epidemiologically unrelated Staphylococcus epidermidis recovered from dogs by pulsed-field gel electrophoresis

  • A total of 16 Staphylococcus epidermidis isolates collected from 14 dogs admitted to the Veterinary Medicial Teaching Hospital in Seoul National University over eleven months were examined for in vitro antibiotic susceptibility pattern with minimum inhibitory concentration (MIC) and slime production, a virulence-associated phenotype, and were genetically characterized by pulsed-field gel electrophoresis (PFGE). The frequency of resistance to antimicrobial agents tested was not high, with a susceptibility ranging from 56.3% to 100%. Three strains exhibited multiple drug resistance against amikacin (MIC, $32-64{\mu}g/ml$), ampicillin ($32{\mu}g/ml$), fosfomycin ($32-128{\mu}g/ml$) and gentamicin ($16{\mu}g/ml$). Vancomycin, ciprofloxacin and rifampin were effective antibiotics against the isolates. All isolates were slime producers ; strains isolated from dogs which died of bacteremia were more likely to produce slime than those isolated from dogs which survived. Chromosomal DNA fingerprinting of the isolates yielded 16 different genomic types with few common bands, indicating a variety of clones of S epidermidis were prevalent in the hospital. This study revealed that PFGE is an useful method for the genotype characterization of S epidermidis strains and this organism could probably be pathogenic in some dogs with severe disorders. Further works on a larger number of epidemiologically defined strains are required to assess these results.

  • Biochemical and antimicrobial susceptibility tests were conducted on 40 strains of Pseudomonas aeruginosa originated from diseased chicken submitted for diagnosis to this Institute during 1978-80. An extensive study of the biochemical properties revealed that the tested strains can be identified with Pseudomonas aeruginosa. Antibiogram showed that all the strains were susceptible to gentamicin, colistin and amikacin but resist to nitrofurantoin, trimethoprim+sulfamethoxazole, ampicillin, methicillin and kanamycin, and had varing degreed of resistance to other antimicrobials including carbenicillin, sulfomamides, neomycin, streptomycin, tetracycline and chloramphenicol. Three of the most frequent resitance patterns observed were FM SXT AM ME KM CM TC SM NE SSS Pattern, FM SXT AM ME KM CM TC SM NE SSS CB Pattern and FM SXT AM EKM CM TC SM Pattern, and these resistance patterns contained 72.5% of the tested strains.

Genetic Organization of a 50-kb Gene Cluster Isolated from Streptomyces kanamyceticus for Kanamycin Biosynthesis and Characterization of Kanamycin Acetyltransferase

  • A 50-kb chromosome DNA region was isolated from Streptomyces kanamyceticus by screening the fosmid genomic library, using the 16S rRNA methylase gene (kmr) as a probe. Sequence analysis of this region revealed 42 putative open reading frames (ORFs), which included biosynthetic genes such as genes responsible for 2-deoxystreptamine (2­DOS) biosynthesis as well as genes for resistance and regulatory function. Also, the kanamycin acetyltransferase gene (kac) was characterized by in vitro enzyme assay, which conferred E. coli BL21 (DE3) with 10, 50, and 80-times higher resistance to kanamycin A, tobramycin, and amikacin, respectively, than the control strain had, thus strongly indicating that the isolated gene cluster is very likely involved in kanamycin biosynthesis. This work provides a solid basis for further elucidation of the kanamycin biosynthesis pathway as well as the productivity improvement and construction of new hybrid antibiotics.

Serological Distribution and Antibiotic Susceptibility of Salmonella sp. Isolated from Pusan Area in 2000 (2000년도 부산지역에서 분리된 살모넬라속균의 혈청학적 분포 및 항균제 감수성)

  • A total of 79 Salmonella spp. were isolated from Pusan area in 2000. The serotypes of 79 Salmonella isolates were classified as 42 strains of S. typhi(53.1%), 24 strains of S. enteritidis(30.4%), 9 strains of S. montevideo(11.4%), 2 strains of S. typhimurium(2.5%), 1 strain of S. infantis(1.3%) and 1 strain of S. indiana(1.3%) strains(16.5%) of Salmonella sp. were isolated at May July, respectively. The isolates of S. typhi were sensitive to most sntibiotics except streptomycin. All isolates of S. typhi were especially sensitive to tobramycin, gentamicin, colistin, kanamycin, samikacin, sulfamethozazole/ trimethoprim, cefriaxone, ceftazdime, cifrofloxacin, cefoxitin and cefotaxime. Isolates of S. enteritidis wer presented higher resistance than isolates of S. typhi. Twenty-four strains of S. enteritidis were sensitive to kanamycin, amikacin cifrofloxacin, cefoxitin and cefotaxime, however 13 strains(54.2%) of S. enteritidis were resistant to carbenicillin, ampicillin and ticarcillin. Nine strains of S. montevideo were sensitive to most antibiotics except carbenicillin and streptomycin. Each 1 stain of S. indiana and S. infantis was sensitive to most antibiotics used in this study except streptomycin. Three kinds of resistant pattern (CB, SM, TE, AM, TC). In the case of S. enteritidis isolates, 9 kinds resistant pattern were detected. Most frequent resistant pattern of S. enteritidis isolates was CB, AM, TC type(16.7%)

Characterization and Antimicrobial Resistance of Vibrio parahaemolyticus Strains Isolated from Seawater of Geum River Estuary Area, West Coast of Korea (금강 하구 해역의 해수에서 분리한 장염비브리오(Vibrio parahaemolyticus) 균의 특성 및 항균제 내성)

  • Seventy-five Vibrio parahaemolyticus isolates from the surface seawater of the Geum River Estuary area, on the west coast of Korea, were analyzed for the presence of virulence genes and susceptibility to 17 different antimicrobials. All 75 isolates were examined for the presence of two virulence genes (tdh or trh) using polymerase chain reaction; Only one of the isolates possessed the tdh or trh gene. According to the results of disk diffusion susceptibility tests, all of the strains were resistant to penicillin G, 92.0% were resistant to ampicillin, 82.7% were resistant to amoxicillin, 2.7% were resistant to ciprofloxacin, 2.7% were resistant to trimethoprim, 1.3% were resistant to cephalothin, and 1.3% were resistant to erythromycin. However, all of the strains were susceptible to amikacin, cefoxitin, chloramphenicol, gentamycin, kanamycin, nalidixic acid, nitrofurantoin, rifampin, streptomycin, and tetracycline. The average minimum inhibitory concentrations for ampicillin for V. parahaemolyticus was 557.6 ㎍/mL. These results not only provide novel insight into the necessity for seawater sanitation in Geum river estuary area, but they help reduce the risk of contamination of antimicrobial-resistant bacteria.
