Determination of ginsenosides in Asian and American ginsengs by liquid chromatography-quadrupole/time-of-flight MS: assessing variations based on morphological characteristics

  • Background: Asian ginseng and American ginseng are functional foods that share a close genetic relationship and are well-known worldwide. This article aims to investigate the correlation between morphological characteristics and the inherent quality of Asian and American ginsengs. Methods: In this study, an ultra-HPLC-quadrupole/time-of-flight MS (UHPLC-Q/TOF-MS) method was established for the quantitative analysis of 45 ginseng samples. The method developed for determination was precise and accurate. Results: The results showed that Asian ginseng samples with the same growing time (with the same or similar number of stem scars) that had a thinner main root, a longer rhizome and more branch roots contained greater amounts of ginsenosides. For American ginseng, two tendencies were observed in the relationship between the diameter of the main root and contents of ginsenosides. One tendency was that samples with thinner main roots tended to contain higher levels of ginsenosides, which was observed in the samples sold under the commercial name pao-shen. Another tendency was that samples with thicker main roots contained higher contents of ginsenosides, which was observed in the samples sold under the commercial name pao-mian, as well as in samples of American ginseng cultivated in Jilin, China. Conclusion: An approach using ultra-HPLC-quadrupole/time-of-flight MS was successfully established to link morphology and active components for evaluating the quality of Asian and American ginsengs. Clear correlation between visible morphological features and quality of Asian and American ginsengs was found. People can see the difference; this means consumers and vendors can evaluate ginseng by themselves.

한국인과 중국인에서 인삼과 서양삼 복용후 전신 증상 -이중맹검 무작위배정 임상시험- (The Effects of Ginseng and American Ginseng on General Symptom in Koreans and Chineses - Double-blind Randomized Controlled Trials -)

  • 본 연구는 인삼이나 서양삼이 상열감이나 코피 같은 전신증상에 미치는 영향을 임상적으로 비교 연구한 것이다. 복용군을 다섯 군(위약군, 한국 양평산 홍삼 6년근 복용군, 한국 금산산 백삼 6년근 복용군, 중국 요녕산 서양삼 4년근, 미국 뉴욕산 서양삼 6년근 복용군)으로 나누어 이중 맹검 임상시험을 하였다. 한국인과 중국인을 대상으로 인삼과 서양삼 복용 후 전신증상에 미치는 영향을 비교한 결과 부작용은 한국인에서 유의하게 나타난 것은 없었으나 중국인에서 구순건조가 위약군에서, 가슴 답답함(흉민)이 위약군과 4년근 서양삼군에서 유의하게 많았다. 개선효과는 한국인에서 유의하게 나타난 것은 없었으나 중국인에서 가슴 답답함(흉민)이 4년근 서양삼군과 6년근 서양삼군에서 유의하게 개선되었다 본 연구 결과는 현재 인삼과 서양삼에 대한 인식이 아마도 한의학에서 본 초학의 기미의 개념을 상업적인 의도를 가지고 왜곡시킨 것임을 시사한다고 하겠다.

  • This study was devised to observe the inhibitory effects of 3 kinds of petroleum ether extracts (percolation by petroleum ether) from Korean red ginseng, Chinese red ginseng and American white ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been Inoculated with sarcoma-180. The yields of petroleum ether extract from Korean red ginseng, Chinese red ginseng and American white ginseng were 0.64, 0.47 and 0.58 and respectively, indicating that the yield of Korean red ginseng was the highest. In vitro, at the concentration of 2 mg /ml, the inhibition rate of lipolysis by the petroleum ether extract of Korean red ginseng, Chinese red ginseng and American white ginseng were 55.1, 50.0 and 44.9% respectively, and the total inhibitory activity per gram of ginseng material were 18, 12 and 13 unit respectively, indicating that the Korean red ginseng was the most effective in the inhibition of the lipolysis.

인삼제품의 수출시장 (Export Markets for Ginseng Products)

  • The current size, characteristics. and channels of distribution of the offshore export (non North American) market for ginseng products are described. The apparent size of the worlds export market for ginseng products in 1987 is 3,363 tonnes. The major export markets for ginseng are Hong Kong (1,972.4 tonnes) and Japan (540.9 tonnes). The volume of ginseng imported into Hong Kong has doubled over the past five years from 976.3 tonnes in 1983 to 1,972.4 tonnes in 1987. This increase is reflective of an expanding Chinese market. In terms of products forms, the toast majority of ginseng imported into China is in root form, while Japan imports both red and white ginseng as well as ginseng beverages and medical products. Hong Kong is the destination of over 86f; of the volume of North American ginseng exported between 1983 and 1987. North American ginseng products are distributed by ginseng brokers or importers/exporters.

Effect of Two Glomus Species Inoculations on Survival, Photosynthetic Capacity, Growth, Morphology ana Root Ginsenoside Content of Panax quinquefolius L.

  • Vesicular-arbuscular mycorrhizae (VAM) fungi naturally colonise American ginseng roots and this relationship is highly beneficial to enhance plant productivity. Our goal was to determine the effect of adding two Glomus species (Glomus etuticatum, G. intraradices) on survival, photosynthetic capacity, growth, morphology and root ginsenoside content of one-year-old American ginseng plants grown in a broadleaf forest. While our study revealed that VAM inoculations significantly affected root morphology and Re ginsenoside content, the survival, photosynthetic capacity and root growth of American ginseng plants were not significantly influenced by VAM inoculations. Surface area and volume of rootlets were 16-25% higher for ginseng grown in VAM-inoculated soil compared to those grown in the control plots. Also, Re ginsenoside content was 18 % higher in YAM-inoculated roots compared to controls.

Stem-leaves of Panax as a rich and sustainable source of less-polar ginsenosides: comparison of ginsenosides from Panax ginseng, American ginseng and Panax notoginseng prepared by heating and acid treatment

  • Background: Ginsenosides, which have strong biological activities, can be divided into polar or less-polar ginsenosides. Methods: This study evaluated the phytochemical diversity of the saponins in Panax ginseng (PG) root, American ginseng (AG) root, and Panax notoginseng (NG) root; the stem-leaves from Panax ginseng (SPG) root, American ginseng (SAG) root, and Panax notoginseng (SNG) root as well as the saponins obtained following heating and acidification [transformed Panax ginseng (TPG), transformed American ginseng (TAG), transformed Panax notoginseng (TNG), transformed stem-leaves from Panax ginseng (TSPG), transformed stem-leaves from American ginseng (TSAG), and transformed stem-leaves from Panax notoginseng (TSNG)]. The diversity was determined through the simultaneous quantification of the 16 major ginsenosides. Results: The content of ginsenosides in NG was found to be higher than those in AG and PG, and the content in SPG was greater than those in SNG and SAG. After transformation, the contents of polar ginsenosides in the raw saponins decreased, and contents of less-polar compounds increased. TNG had the highest levels of ginsenosides, which is consistent with the transformation of ginseng root. The contents of saponins in the stem-leaves were higher than those in the roots. The transformation rate of SNG was higher than those of the other samples, and the loss ratios of total ginsenosides from NG (6%) and SNG (4%) were the lowest among the tested materials. In addition to the conversion temperature, time, and pH, the crude protein content also affects the conversion to rare saponins. The proteins in Panax notoginseng allowed the highest conversion rate. Conclusion: Thus, the industrial preparation of less-polar ginsenosides from SNG is more efficient and cheaper.

Rapid Somatic Embryogenesis and Plant Regeneration in American Ginseng: Effete of Auxins and Explants

  • The efficacy of three auxins, viz. 2,4-0, NAA and dicamba, were compared for the induction of somatic embryogenesis in American ginseng (Panax quinquefolium L.). Somatic embryos (SEs) formed on ginseng cotyledonary, zygotic embryo and shoot explants after 8 weeks of induction by the auxin stimuli. Significantly more somatic embryos were induced by culture of any of the ginseng explants on media supplemented with $5{\mu}M$ 2,4-0 than any other auxin treatment. Shoots derived from somatic embryos had the greatest regenerative potential and zygotic embryos the least. Explants generated from green (unstratified) seeds gave similar or higher frequency of embryogenesis as the explants derived from stratified seeds. Histological and SEM studies confirmed that the regenerimts were somatic embryos. Somatic embryos germinated and developed into normal plants in $3\~6$ months. About $10\%$ of plantlets from second generation SEs formed flowers within 10 weeks, particularly on media supplemented with $GA_3$ The development of a regeneration system for ginseng through somatic embryogenesis is a necessary first step for mass propagation and genetic improvement of American ginseng.

Effects of American Ginseng Cultivation on Bacterial Community Structure and Responses of Soil Nutrients in Different Ecological Niches

  • American ginseng (Panax quinquefolium L.) is a perennial herbaceous plant widely cultivated in China, Korea, the United States, and Japan due to its multifunctional properties. In northwest China, transplanting after 2-3 years has become the main mode of artificial cultivation of American ginseng. However, the effects of the cultivation process on the chemical properties of the soil and bacterial community remain poorly understood. Hence, in the present study, high-throughput sequencing and soil chemical analyses were applied to investigate the differences between bacterial communities and nutrition driver factors in the soil during the cultivation of American ginseng. The responses of soil nutrition in different ecological niches were also determined with the results indicating that the cultivation of American ginseng significantly increased the soluble nutrients in the soil. Moreover, the bacterial diversity fluctuated with cultivation years, and 4-year-old ginseng roots had low bacterial diversity and evenness. In the first two years of cultivation, the bacterial community was more sensitive to soil nutrition compared to the last two years. Proteobacteria, Actinobacteria, Gemmatimonadetes, Acidobacteria, Firmicutes, and Bacteroidetes dominated the bacterial community regardless of the cultivation year and ecological niche. With the increase of cultivation years, the assembly of bacterial communities changed from stochastic to deterministic processes. The high abundance of Sphingobium, Novosphingobium, and Rhizorhabdus enriched in 4-years-old ginseng roots was mainly associated with variations in the available potassium (AK), total phosphorus (TP), total potassium (TK), and organic matter (OM).

한국인삼과 북미산 서양상의 Sesquiterpene 성분 비교 (Comparison of Sesquiterpenes in Korean and American Ginsengs)

  • Sesquiterpenes of Korean ginseng (Panax ginseng C.A. Meyer, KG) and American ginseng (Panax quinquefolium, AG) were Isolated by simultaneous distillation and extraction (SDE) with 30% methanol and n-hexane or adsorption using C18 Sep-Pak . The sesquiterpenes in KG and AG were compared each other by using TLC and GC/MS. Thin layer chromatogram of KG gave 4~5 spots of terpenes colored by vanillin-sulfuric acid, while AG did one major spot. Total ion chromatogram of KG showed about 30 peaks of sesquiterpene having molecular weight 204. Among these, 9 components such as alloaromadendrene, germacrene B, isocaryophyllene, $\alpha$-neoclovene, ${\gamma}$-muurolene and $\beta$-panasinsene together occupied 81.5% of total sesquiterpenes identified. In AG, however, only 3 components, i.e., isocaryophyllene, $\beta$-bisabolene, $\beta$-sesquiphellandrene occupied 79.3% of total sesquiterpenes identified. Especially isocaryophyllene was a dominant component of AG occupying 56.6%. Eight sesquiterpenes, including alloaromadendrene, germacrene B and $\alpha$-humulene, were not detected in AG. These results indicate that sesquiterpenes could be used as indices for the chemical difference between KG and AG.
