Effects of Ascaris suum Extract and Sulfamethoxazole on Allergic Airway Inflammation

  • Allergic asthma is complex inflammatory airway disorder caused by genetic and environmental factors. Sulfamethoxazole, a sulfonamide, is the cause of drug rash with eosinophilia and systemic symptoms syndrome. Parasites infection also related with eosinophilia and allergic diseases. In the present study, we investigated the modulating effects of parasitic derivative and sulfamethoxazole (SMX) on allergic airway inflammation in the ovalbumin (OVA)-induced murine asthma model. Histopathological changes, cytokine secretion, and total and allergen-specific IgE were investigated. BALB/c mice were treated with Ascaris suum extract or SMX for 4 weeks before sensitized and challenged to ovalbumin. Pre-treatment of Ascaris suum extract decreased allergic inflammation in lung tissue and IL-4, total IgE, and OVA-specific IgE levels in bronchoalveolar lavage fluid. However, pre-treatment of SMX did not show any effects on allergic airway inflammation. These results indicate that parasitic infection has protective effects on allergic asthma, but the sulfamamides may not relate with allergic asthma.

The Effects of Bojungikgi-tang on the Nasal Tissue in Allergic Rhinitis of Ovalbumin0inhalation Rats. (보중익기탕이 알레르기 비염 유발 白鼠의 과립구 및 조직학적 변화에 미치는 효과)

  • Allergic rhinitis is an allergic reaction characterized by sneezing, coughing, itchy nose, mouth and throat, congestion and/or nasal discharge. The offending allergens are usually pollens, molds, dust mites and animal allergens. We aimed to observe inhibitory effects of Bojungikgi-tang on incerase of the number of eosinophil and neutrophil, and protective effects of nasal mucosal tissue in the allergic rhinitis. 1. In Inhibitory Effect of Bojungikgi-tang on Increase of the number of Eosinophil in Serum, Bojungikgi-tang showed significant effects.(p〈0.05) 2. In Inhibitory Effect of Bojungikgi-tang on Increase of the number of Neutrophil in Serum, Bojungikgi-tang showed significant effects.(p〈0.05) 3. In histopathologic change of nasal mucosal tissue, Bojungikgi-tang showed significant protective effects. Depending upon above results, it is considered that Bojungikgi-tang has the inhibitory effects on the process of allergic rhinitis and that it could be used in reliving patients of the symptoms caused by allergic rhinitis.

Anti-allergic Effects of Schizonepeta tenuifolia on Mast Cell-Mediated Allergy Model

  • Immediate-type hypersensitivity is involved in many allergic diseases such as asthma, allergic rhinitis and anaphylaxis. The discovery of drugs for the treatment of allergic disease is an important subject in human health. Stimulation of mast cells releases inflammatory mediators, such as histamine and pro-inflammatory cytokines with immune regulatory properties. We investigated the effect of the aqueous extract of Schizonepeta tenuifolia (AEST) (Labiatae) on the immediate-type allergic reaction. AEST inhibited compound 48/80-induced systemic allergic reaction. AEST attenuated immunoglobulin E (IgE)-mediated skin allergic reaction and histamine release from human mast cell line (HMC-1) cells. In addition, AEST decreased the gene expression and secretion of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated HMC-1 cells. Our results indicate that AEST inhibits the mast cell-derived allergic reactions and involvement of histamine and pro-inflammatory cytokines in these effects.

The Effects of Sinichengpae-um in Allergic Rhinitis Model Rats (신이청폐음(辛荑淸肺飮)이 알레르기 비염 동물 모델에 미치는 영향)

  • Objective : Allergic rhinitis is an allergic reaction characterized by sneezing, coughing, itchy nose, mouth and throat, congestion and/or nasal discharge. We aim to observe effect of Sinichengpae-um on protective effects of nasal mucosal tissue in th allergic rhinitis. Method : For this purpose, we oberserved number of leukocyte and erythrocyte in blood, ratio of neutrophil and lymphocyte in leukocyte, activity of GOT and GPT, and histopathologic change of nasal mucose. Result and Conclusion : Sinicheogpae-um showed effects on immune reaction with no harms liver. And in histopathologic change of nasal mucosal tissue, Sinichengpae-um showed significant protective effet against allergic rhinitis.

Anti-allergic Effects of Artemisia iwayomogi on Animal Models of Allergic Reactions

  • The effects of aqueous extract of Artemisia iwayomogi (Compositae) (AIAE) on the mast cell-dependent allergic and inflammatory reactions were investigated. AIAE (0.05 to 1 g/kg) dose-dependently inhibited systemic allergic reaction induced by compound 48/80 in mice. AIAE (0.1 and 1 g/kg) also significantly inhibited local allergic reaction activated by anti-DNP IgE. AIAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. Moreover, AIAE inhibited the secretion of interleukin (IL)-6 in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187-stimulated human mast cell line (HMC-1) cells. These results provide evidence that AIAE may be beneficial in the treatment of allergic diseases.

The Effects of Sochungyong-tang plus Okbyeong-san Extract on Allergic Rhinitis of Rats (소청룡탕합옥병풍산(小靑龍湯合玉屛風散)이 백서(白鼠)의 알레르기 비염(鼻炎)에 미치는 효과)

  • Background and Objectives : Although the incidence of allergic rhinitis has increased specific therapeutics with a certain medicine is not well developed yet. In therapeutics of allergic rhinitis, limited medicines just have been used in the clinin. Sochungyong-tang has been a widespread medicine for allergic rhinitis. However, okbyeongpung-san has been taken for raising immunity. In medical treatment of allergic rhinitis, raising immunity is a necessary part of the process. In this reason, this study aimed to find out therapeutic effects of combination of sochungyoung-tang and okbyeongpung-san extract on allergic rhinintis of nasal tissue in rats. Materials and Methods : The Sprague-Dawley male rats, about 200g of body weight, were divided into three groups: the normal group, the control groups, and the sample groups. With the Levine and Vaz method, the allergic reaction was induced with ovalbumin as antigen in the intraperitoneal route. Rats of the sample groups were treated with 2cc of sochungyoung-tang plus okbyeongpung-san extract once a day for 28days. I observed historical changes in nasal tissue. I also found changes in the segment of lympocyte and neutrophil of Leukocyte and erythrocyte. I used the statistical methods of post Hoc test(p<0.05) Results : The increase of the goblet cell and mucous secretion in the sample groups was rare when compared to the control groups. Effects of sochungyoung-tang plus okbyeongpung-san extract on the liver function were also studied in rats. Treatment of sochungyoung-tang plus okbyeongpung-san extract did not affect GOT and GPT. The number of lymphocyte was decreased, while the number of erythrocyte was increased in sample groups. Conclusion : This study shows that sochungyoung-tang plus okbyeongpung-san plus okbyeongpung-san extract has considerable effects on allergic rhinitis.

An Experimental Study on the Anti-inflammatory Effects of Cheonggisan Extract in Allergic Late Inflammation (알레르기 후기 반응 염증 억제효과에 관한 청기산(淸肌散)의 실험적 연구)

  • Objectives : Allergic diseases have a various symptoms of hyperresponsiveness and recently hyperresponsive reaction in the chronic phase is reported as the important mechanisms. Cheonggisan(CGS) is used in oriental clinics for curing various skin diseases due to effect of controlling of pruritus. There have been studies on the anti-allergic effect and anti-inflammatory effect of CGS, but there had no study of anti-allergic effects in allergic late inflammation of CGS, so we aimed to find out the effects of CGS in allergic late inflammation in our study.Methods : To investigate the anti-allergy effect and anti-inflammatory effect of CGS, RAW 264.7 macrophage cells and CSG water-extracts were used. Cytotoxic effect of CSG was examined by MTT assay, an oxidative product of NO was measured in the culture medium by the Griess reagent assay. The level of prostaglandin E2(PGE2) was measured by competitive enzyme-linked immunoassay. Cytokine(PGE2, IL-1β, IL-6, TNF-α) was measured by Bio-Plex suspension assay system and quantitative multiplexed cytokine/chemokine assay.Results : We investigated that there was no cytotoxic effect of CGS water-extract at any levels of concentration on RAW 264.7 macrophage cells by MTT assay. CGS water-extracts significantly suppressed the levels of the inflammatory mediators such as NO and PGE2, cytokine of IL-1β, TNF-α at the level of 400 ㎍/㎖ CGS concentration. But there was no significant effect on IL-6 production suppression.Conclusions : These results suggest that CSG water-extract has and anti-inflammatory effects in allergic reaction. These properties may contribute to the allergic diseases and inflammatory related disease care.

The Effects of Tongkwansan on the Changes of Total IgE and Specific IgE in Allergic Rhinitis Mouse Model (알레르기성 비염 모델에서 통관산(通關散)이 Total IgE, specific IgE 생산에 미치는 영향)

  • Background &Objectives : Rhinitis is an inflammation of nasal mucosa and the major symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Rhinitis is classified into allergic rhinitis and nonallergic rhinitis. Allergic rhinitis is an immune reaction by allergen, and vasomotor rhinitis which is nonallergic and noninfectious is hypersensitive reaction. The incidence of allergic rhinitis has increased and the rate of vasomotor rhinitis is high. However there have been no studies about vasomotor rhinitis compared with allergic rhinitis. And there have been no studies so far performed on the effect of Tongkwansan. Therefore this study is aimed to find out the effects of Tongkwansan on allergic rhinitis and vasomotor rhinitis. Materials and Methods : Fifteen BALC/c mouses were divided into three groups : normal group, control group and sample group. To induce the allergic rhinitis in control group and sample group, mouses were sensitized intrapertioneally 0.1% ovalbumin solution three times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1 % ovalbumin solution 3 times at intervals of 2 days. After that time, mouses in the sample group were oral administration treated by Tongkwansan for 28 days. We observed changes in the segment of IL-4, IL-5, $IFN-{\gamma}$, Total IgE, and ovalvumin specific IgE in blood. We used the statistical methods of ANOVA test(p<0.05). Results : There were no significant changes statistically in $IFN-{\gamma}$, IL-4, and IL-5 in blood(p<0.05). There were also no significant changes statistically in Total IgE, OVA-specific IgE in blood(p<0.05). Conclusion : According to above results, it is supposed that Tongkwansan has no significant effects on allergic rhinitis. But it is supposed that Tongkwansan has significant effects on vasomotor rhinitis which is nonallergic and noninfectious

  • Objectives : This study was carried out to analyze the trends of clinical studies of allergic contact dermatitis published in the Journal of Korean Medical Ophthalmology & Otolaryngology & Dermatology. Methods : We studied 10 research papers on allergic contact dermatitis published in JKOOD. We analyzed about experiment methods, pharmacodynamic effects, therapeutic effects in published papers. Results : 1. The number of searched journal is 10 papers. 2. All experiments was performed using DNCB primed mouse. To demonstrate therapeutic effect of drugs, previous researches observed dermatological and histopathogical conditions, erythema, melasma, white blood cell count, Lymphocyte ratio to white blood cell. 3. Most of drugs have heat-clearing and detoxifying effects. 4. Every drugs have positive effect in treating allergic contact dermatitis. Conclusions : Figuring out how experiment proceeded and tendency of drugs in researching allergic contact dermatitis is significance of this paper. Further research about drugs and their treating methods should be carried out to improve curative effectiveness for allergic contact dermatitis.

Ambient air pollution and allergic diseases in children

  • The prevalence of allergic diseases has increased worldwide, a phenomenon that can be largely attributed to environmental effects. Among environmental factors, air pollution due to traffic is thought to be a major threat to childhood health. Residing near busy roadways is associated with increased asthma hospitalization, decreased lung function, and increased prevalence and severity of wheezing and allergic rhinitis. Recently, prospective cohort studies using more accurate measurements of individual exposure to air pollution have been conducted and have provided definitive evidence of the impact of air pollution on allergic diseases. Particulate matter and ground-level ozone are the most frequent air pollutants that cause harmful effects, and the mechanisms underlying these effects may be related to oxidative stress. The reactive oxidative species produced in response to air pollutants can overwhelm the redox system and damage the cell wall, lipids, proteins, and DNA, leading to airway inflammation and hyper-reactivity. Pollutants may also cause harmful effects via epigenetic mechanisms, which control the expression of genes without changing the DNA sequence itself. These mechanisms are likely to be a target for the prevention of allergies. Further studies are necessary to identify children at risk and understand how these mechanisms regulate gene-environment interactions. This review provides an update of the current understanding on the impact of air pollution on allergic diseases in children and facilitates the integration of issues regarding air pollution and allergies into pediatric practices, with the goal of improving pediatric health.
