알칼리 펙티나제를 이용한 면직물의 효소정련 (Bioscouring of 100% Cotton Fabric with Alkaline Pectinase)

  • Study on the use of enzymes for textile wet processing has been very active. The exploratory research conducted herein is related to the bioscouring process for cotton fabric. The optimum concentration of alkaline pectinase(BioPrep) was in the range of 0.05~0.2 g/l, the proper treatment time was 30~60 minutes, the appropriate treatment temperature was $60^{\circ}C$ for both the batch method and the padding method. The simultaneous desizing/bioscouring by padding method did not give water absorbency as good as the bioscouring after desizing. Color of fabrics which were bioscoured and dyed with direct dyes and a reactive dye was just a little darker than that of NaOH scoured one. K/S and Lab values of the bioscoured fabrics, regardless of the degree of water absorbency, were quite similar to each other.

  • Treatment with the enzyme pectinase has been reported to lower the cationic demand of thermomechanical pulp(TMP) bleached with alkaline peroxide in the laboratory. We have extended this discovery to bleached TMP produced industrially, and shown that commercial enzyme preparations can treat pulp within 15 minutes at the at the temperature and pH values prevalent in paper mills. About half of the cationic demand in the bleached pulp can be destroyed by pectinase. Dynamic drainage jar experiments show that the enzyme treatment improves the effectiveness of several cationic polymers to increase retention in the absence of retention aids or with non-ionic polymers, and does not damage the strength properties of the pulp. Pectinase could be easily incorporated into paper machine stock preparation systems to lower the charges of cationic retention aids needed in furnishes containing peroxide-bleached mechanical pulp.

  • 효소는 주로 면직물의 전처리 및 염색가공에 사용되고 있는데, 공업적으로 적용되고 있는 예로는 아밀라제에 의한 호발, 셀룰라제에 의한 감량 및 유연가공을 들 수 있고, 최근 미국과 일본을 중심으로 연구되고 있는 펙티나제에 의한 정련은 공업화 가능성이 매우 높은 것으로 보고되고 있으며, 글루코스 옥시다제에 의한 표백은 아직연구 초기 단계에 있다. 효소정련이란 기존의 수산화나트륨 대신에 효소를 사용하여 행하는 정련을 말하며, 장점으로는 50-6$0^{\circ}C$ 정도의 온도에서 행하기 때문에 기존 정련법과 비교시 에너지와 용수를 절약할 수 있는 것, 폐수 중에 방출되는 오염물질의 양을 현격하게 줄일수 있을 것, 우수한 생분해성으로 인한 수질오염을 감소시킬 수 있을 것, 부드럽고 스무스하여 자연스런 태를 부여하는 것 및 재현성이 높은 것 등이다. Aspergillus niger CF-34로부터 분리한 대두세포벽분해효소 복합체 중의 Protease의 선택적인 제거 (Selective Removal of Protease from Soybean Cell Wall Degrading Enzyme Complex Isolated from Aspergillus niger CF-34)

  • Aspergillus niger CF-34가 생산하는 대두세포벽분해 효소 복합액을 알칼리로 처리하여 조효소액 중에 함유된 protease만을 선택적으로 제거할 수 있었다. 조효소액을 알칼리로 처리하였을 때 세포벽분해활성에는 영향을 적게 주고, 대두 단백질을 분해시켜 쓴 맛의 peptide를 생성하는 protease만을 선택적으로 불활성화시킬 수 있었다. 조효소액의 pH를 $9.0{\pm}0.1$로 조절하여 $20^{\circ}C$에서 약 30분 동안 서서히 교반한 후 다시 pH를 5.0로 조절하는 것이 최적 조건이었다. 이때 조효소액 중 protease 활성은 초기의 약 10% 미만으로 감소하였으며, 각 효소의 잔존 활성을 살펴보면 pectinase는 약 80%, polygalacturonase는 약 85%, xylanase는 약 95%, carboxymethyl cellulase는 약 100% 정도이었고, 대두세포벽분해활성은 초기의 약 90% 정도 유지할 수 있어 protease만이 선택적으로 제거되었다. 이렇게 처리된 효소액을 사용하여 비지 중의 대두 단백질을 추출할 경우 생산효율은 비록 감소하였지만, 대두단백질의 분해를 막고, 쓴맛 생성을 억제하여 품질 및 관능적으로 우수한 제품을 생산할 수 있었다.

  • To develop a scouring method for the high-grade quality and the high value added of flax for spinning, it was measured and analyzed weight loss and degree of injury with change of scouring concentration, time, temperature and pH. We got the f ollowing results; Firstly, in case that flax was scoured at a fixed condition, the weight loss and degree of injury of scoured flax under the no-cutting condition were lower than that of scoured flax under the cutting condition. Secondly, when flax was scoured with $Na_{2}CO_{3}$, it was obtained that the higher weight loss and degree of injury and weight loss with the increase of scouring time and condition. The degree of injury was lower than any other under the treatment condition of 4% $Na_{2}CO_{3}$concentraion for 2 hours. Finally, when alkaline scoured flax was rescoured by pectinase, enzyme activation effect was the most active under the treatment condition of pH 4 for 2 hours. The scouring effect was better than that of alkaline scouring only.
