Sugar Extraction by Pretreatment and Soda Pulping From Cattail (Typha latifolia L.) (1) Extraction of Sugar (부들의 전처리를 통한 당의 추출과 소다펄프화에 관한 연구 (1) 당 추출)

  • Cattail (Typha L.) was used as a raw material for producing both bio-ethanol and pulp for papermaking at the same time. Pretreatments of cattail stems and leaves with acid ($H_2SO_4$) and alkali (NaOH) in three different addition levels were studied before soda pulping. The acid pretreatment gave reducing sugar of 15.2% of initial weight, but alkali pretreatment close to 1%. Soda pulping of the pretreated cattail gave 3% reduction in pulp yield and less bonding properties in paper; however, refining of the pulp from the pretreated cattail with alkali restored their fiber bondings up to that of the pulp from no-pretreated cattail at equivalent freeness.

Optimum Extraction Conditions and Anticancer Effect of Functional Polysaccharide from Mycelia of Grifola frondosa (잎새버섯(Grifola frondosa) 균사체의 기능성 다당류 최적 추출방법 및 항암효과)

  • Grifola frondosa has been used as an herbal medicine for the treatment of cancer, diabetes mellitus and high blood pressure. In this study, functional polysaccharide was obtained from Grifola frondosa using four different extraction methods: hot water(HwFP), homogenize(HgFP), acid(AcFP), and alkali(AlFP) extraction methods. The effects of these extracts on KB and HepG2 cell lines were then examined for any anti-cancer activity. Alkaline extraction produced a yield of 0.175% and the total sugar content of the extract was 54.97%. We were able to confirm that the polysaccharide extracts from the mushroom produce an anti-cancer effect. The cytotoxicity of AlFP and AcFP against HepG2 cells were 22.86% and 28.88%, respectively, and the cytotoxicity of AlFP against the KB cell lines was 47.76% at a concentration of 1,000 ${\mu}g/m{\ell}$. Therefore, these results suggest that the optimum method for extracting functional polysaccharides from G. frondosa is the alkali extraction method.

Properties of Porphyran and Hemicellulose extracted with Different extract Solutions and Enzymatic Pretreatments from Porphyra (추출 용매 및 효소 전처리 방법에 따른 포피란과 헤미셀루로오즈의 특성)

  • Laver, Porphyra, is distinctive for its high content of proteins and polysaccharides such as porphyran and hemicellulose. The chemical properties of the polysaccharides extracted with different extraction methods such as hot water, dilute acid(pH 4.0) or alkali solution(2N NaOH) were examined to investigate the suitable extraction conditions for porphyran and hemicellulose from laver. For porphyran extraction, dilute acid solution was more preferable to hot water and alkali solution because of its higher 3,6-anhydrogalactose content and lower protein content. However, alkali solution was more suitable to extract the hemicellulose because of higher mannose content indicating the extraction of mannan. To decrease contamination of the polysaccharides with protein, the dried lavers were pretreated with enzymes (Protamex, Flavourzyme, Alcalase, Viscozyme) before hot water extraction. All enzyme pretreatments increased the yield of polysaccharides by compared with control (enzyme unpretreated) and Flavourzyme pretreatment was most effective to decrease protein contamination in the polysaccharide. All viscosities of porphyran solutions pretreated by enzymes were lower compared to the control porphyran solution and showed pseudoplastic behavior with yield stress. In case of alkali extraction of residues obtained after enzyme hydrolysis and hot water extraction, protease pretreatment increased the mannose contents in the polysaccharide while the xylose content was increased by Viscozyme pretreatment.

The Phase Separation of Low Alkali Borosilicate Glass by Substituting $Li_2O$ for $Na_2O$ (산화리튬의 치환에 따른 붕규산 유리의 분상에 관한 연구)

  • The phase separation of low-alkali borosilicate glass with the composition of $6.25Na_2O$.$18.75B_2O_3$.$75.00SiO_2$(mole%) substituting $Li_2O$ for $Na_2O$ was studied. The phase separation in the temperature range of transformation was examined with various heating temperatures and soaking times. Durability to water, thermal expansion and specific density of the specimen were investigated and the microstructure of the separated phase was also observed by transmission electron micrograph techniques. The maximum alkali extraction result with the best phase separation effect was obtained when $Na_2O$ of the base glass was replaced with $1.88Li_2O$ (mole %) and electron micrograph of carbon film replica of $1.88Li_2O$$4.37Na_2O$.$18.75B_2O_3$.$75.00SiO_2$ (mole %) glass showed that the glass consisted of homogeneous two phases. The minimum specific density was shown with the specimen treated at 57$0^{\circ}C$ and it was also shown that the longer the treating time the lower the specific density. The apparent activation energies of approximately 45 kcal/mole by the alkali extraction and 43kcal/mole by the thermal expansion method were derived from the Arrhenius plots, respectively.

  • Solvent extraction of alkali metal and alkaline earth metal cations by mono-crown ether(benzo-15-crown-5) and bis-(crown ether)s(ethylenediamine bis(4'-formyl benzo-15-crown-5)) containing benzo-15-crown-5 moieties were investigated with water-chloroform system at $25^{\circ}C$. The order of the extraction equilibrium constants($K_e$) and the complexation constants($K_c$) for the alkali metal and alkaline earth metal cations with mono-crown ether and bis-(crown ether)s were Ca(II)>Na(I)>Sr(II)>K(I)>Mg(II)>Rb(I) and Sr(II)>Ca(II)>K(I)>Rb(I)>Mg(II)>Na(I) respectively. These results were explained in terms of the size effect of metal cation and electron density effect. Also, the bis-(crown ether)s was found to extract metal cations more effectively than the corresponding mono-crown ether.

Removal of Arsenic From Closed Mine Tailings by Alkali-Leaching Method (알칼리 용출법에 의한 폐광산 광미중의 비소제거에 관한 연구)

  • Removal of the Arsenic components from the closed mine tailings has been attempted by the alkali-leaching method. Two tailings collected from the Daduck and Yuchon mine which were already closed many years ago were leached with caustic soda solutions. The Arsenic components in the leach liquor resulted from the alkali treatment of tailings could be removed fairly well in the form of insoluble calcium-Arsenic compound by the precipitation with calcium chloride. As a result, the extraction of about 60~90% Arsenic from the tailings could be obtained depending on the leaching conditions and the influence of temperature and the slurry density on the extraction of Arsenic was also found to be very small at the NaOH concentration more than 0.5N. In addition, it seemed that a caustic soda solution over 0.5N NaOH could be used repeatedly for the leaching of tailings since the consumption of NaOH was not so great in a leaching of them. As far as the precipitation of Arsenic components was concerned, more than 99% of Arsenic could be precipitated within 10 minutes by the addition of 2wt% CaC12 in to the leach liquor.

Optimization of Alkali Extraction for Preparing Oat Protein Concentrates from Oat Groat by Response Surface Methodology (반응표면분석법을 이용한 쌀귀리 단백질의 알칼리 추출 공정 최적화)

  • In this study, an attempt was made to produce oat protein concentrates from defatted oat groat by alkali extraction. Independent variables formulated by D-optimal design were NaOH concentration (X1, 0.005~0.06 N) for extraction and precipitation pH (X2, pH 4.0~6.0), and the dependent variable was extraction yield (Y1, %). Experimental results were analyzed by response surface methodology to determine optimized extraction conditions. Extraction yield increased both with an increase in NaOH concentration of the extraction solution and when approaching a precipitation pH of 4.9, and NaOH concentrations were a major influencing parameter. Solubility of oat protein concentrates showed a minimum value (i.e., 0.1%) at pH 5 and increased substantially at pH values in the range of ${\leq}$ pH 3 or ${\geq}$ pH 7, reaching a maximum value at pH 11 (i.e., 76%). Regression equation coincided well with the results of the experiment. Optimized extraction conditions to maximize extraction yield were 0.06 N NaOH (X1) for extraction and pH 4.7 (X2) for precipitation.

  • For elucidation of various parameters' effects on the reactive extraction of citric acids, citric-acetic-lactic acid system with various carriers, diluents, and modifiers were investigated. Carries used were secondary amines, tertiary amines, and solvation extract. Dileunts were n-butylacetate, methoisobutylketon(MIBK), kerosine, and xylene. Modifiers were TBP and isodecanol. The effects of temperature and pH in aquous phase were studied, and equilibrium constants for the reactive extraction were obtained. The re-extraction was performed by alkali solutions such as NaOH, $Na_2CO_3$, and $K_2HPO_4$. Based on the experiment Di-isotridecylamine(secondary amine ) gave higher degree of extraction and more selective than other extractants tested for the extraction of citric acid. N-butylacetate and TBP showed heigher performance for a diluent and modifier, repetitively. The degree of extraction was getting better with decreasing the pH in the aquous phase and the temperature of extraction system. The degree of re-extraction was getting higher with decreasing basicity in the stripping phase and the system temperature, and with increasing the concentration of alkali solution.

Quantification of Karanjin Using High Performance Liquid Chromatography in Raw and Detoxified Karanj (Pongamia glabra vent) Seed Cake

  • Various products of karanj (Pongamia glabra) are utilized for industrial, health and animal agriculture applications in the Indian subcontinent. Despite a rich source of protein (CP, 28-34%), karanj cake was found to be slightly bitter in taste and toxic owing to the presence of flavonoid (Karanjin), restricting its safe inclusion in the livestock diets. Feeding trials with raw cake revealed its poor palatability and adverse performance among different categories of livestock including poultry. The present study was, therefore, aimed to detoxify karanj cake by various physico-chemical methods like solvent extraction, water washing, pressure cooking and alkali and acid treatments. The level of residual karanjin in raw and variously processed cake was quantified using high performance liquid chromatography (HPLC). The raw expeller karanj cake was found to contain about 0.19% of karanjin. Though a non-polar solvent, soxhlet extraction of expeller pressed cake with petroleum ether drastically reduced karanjin content (0.01%). Soaking of cake for 24 h in 1% NaOH (w/w) solution was found to reduce karanjin to a major extent with little further benefit by increasing alkali level. Milder alkalies like lime and fertilizer grade urea reduced the karanjin levels marginally. Similar was the case with mineral acids such as HCl and glacial acetic acid. It was, therefore, concluded that solvent extraction of karanj seeds would be the best method of detoxification as well as for more recovery of oil and karanjin.

Effect of Crown Ring Size and Upper Moiety on the Extraction of s-Block Metals by Ionizable Calixcrown Nano-baskets

  • Eight ionizable nano-baskets of cone 25,26-di(carboxymethoxy)calix[4]arene-crown-3,4,5,6 were synthesized and were verified by $^1H$ and $^{13}C$ NMR spectroscopy, IR spectroscopy and elemental analysis. The competitive solvent extractions of alkali and alkaline earth metal cations were studied using such nano-baskets. The novelty of this study is including three binding units of calixarene's bowl, crown ether's ring and electron-donor ionizable moieties in a unique scaffold to assess the binding tendency towards the cations. The objective of this work is to study the extraction efficiency, selectivity and $pH_{1/2}$ of such complexes. The result of solvent extraction experiments indicated that these compounds were effective extractants of alkali and alkaline earth metal cations. Their selectivities were greatly influenced by the acidity of solution and the conformations of the calixcrown. One conformer was selective to $Na^+$ in pH ${\geq}$ 4, while the other was highly selective to $Ba^{2+}$ in pH 6 and upper.
