Techno-functional and rheological properties of Tenebrio molitor larvae protein by different extraction methods

  • Alkaline- or salt-assisted extractions have been widely used to extract edible insect proteins, however, there is a need for extraction techniques that balance cost-efficient production as well as preserving the protein properties. Mealworm proteins (Tenebrio molitor larvae) were extracted using three different extraction methods-alkali (AMP), salt (SMP), and water (WMP)-and then physicochemical and techno-functional properties were examined. AMP had high yield, protein, and amino acid contents, whereas WMP had high moisture, ash, and fat contents. SDS-PAGE showed a wide range of molecular weights in WMP whereas mostly low molecular weights were observed in AMP and SMP. AMP had poor protein solubilities compared to SMP and WMP across all pHs. AMP had enhanced water-holding capacity and emulsion stability, whereas WMP had improved oil-holding capacity and foaming properties. WMP formed a gel with and without the transglutaminase. The physicochemical and techno-functional properties demonstrated that water-soluble mealworm protein was superior to alkali-and salt-soluble mealworm proteins. Considering the cost efficiency and minimal impact on the environment as well, a cold press juicer could be utilized for mass production of mealworm protein compared to the conventional methods of protein extraction using alkali and salt.

Strength Properties of Alkali-Activated Lightweight Composites with Alkali Activators of Different Types and Amounts (알칼리 자극제의 종류 및 첨가율에 따른 무시멘트 경량 경화체의 강도특성)

  • In order to reduce the emission of carbon dioxide($CO_2$), this research use blast furnace slag in concrete manufacture, as 100% replacement of cement. The aim of this study is to investigate the density and strength properties of alkali-activated lightweight composites with alkali activators of different types and different amounts. The bubble for achieving the lightweight of alkali-activated lightweight composites was generated in the reaction between the paper ash and the alkali activators instead of using a foaming agent. Lightweight formed concrete was conducted basic experimental for determining replacement ratio of paper ash. Then, the density and strength were measured according to the types and the contents of the alkali accelerator that can react with the paper ash. As results, the optimum replacement ratio of the paper ash was 5%. The alkali activator containing NaOH 12.5% obtained the lowest weight of $1.13g/cm^3$. Also, compressive strength were relatively high. Therefore, this study demonstrated that alkali accelerator with a certain amount of NaOH can achieve relatively high strength and lightweight alkali-activated lightweight composites.

Petrology of Alkali Volcanic Rocks in Northern part of Ulrung Island (울릉도(鬱陵島) 북부(北部) 알칼리 화산암류(火山岩類)에 대(對)한 암석학적(岩石學的) 연구(硏究))

  • The study revealed that the sequence of volcanism in Ulrung island can be classified into 5 stages, and the volcanic history is summerized as follow: 1st stage: Eruption of basaltic agglomerates, tuffs and lavas, 2nd stage: Eruption of trachytic and trachyandesitic agglomerates and tuffs, 3rd stage: Eruption of trachyte lavas and their lapilli tuffs, 4th stage: Eruption of trachyte lavas and nepheline phonolites, 5th stage: Eruption of pumice, trachytic ash and lapilli, and plutonic ejecta (fragments of alkali gabbro, monzonite and alkali feldspar syenite) and a subsequent caldera formation. Finally, a small scale eruption of leucite bearing trachyandesite lava in the caldera. Several evidences show that there have been long erosional intervals between the 1st and 2nd stages and between the 4th and 5th stages. A K-Ar age for trachybasalt lava of the 1st stage was determined to be 1.8 Ma, and a $C^{14}$ age, 9300Y. (Machida, 1981) is available for these volcanic events. Therefore, it is considered that volcanic activity of the island above sea level began at least in early Pleistocene, and continued to until 9300 years ago exploding large amount of pumice, prior to pouring out of leucite bearing trachyandesite from the inner caldera. Using solidification index (SI) of Kuno, microscopic texture and mineral composition as criteria of the classification, the volcanic rocks are classified into alkali basalt, trachybasalt, trachyandesite, trachyte and phonolite. These are mostly prophyritic in texture. Main constituent minerals of alkali basalt and trachybasalt are plagioclase, olivine, Ti-augite and magnetite. Principal minerals of trachyandesite are plagioclase, anorthoclase, clinopyroxenes, kaersutite, biotite and magnetite. Trachyte and phonolite consist mainly of anorthoclase, clinopyroxene and magnetite, showing typical trachytic texture in groundmass. In solidification index, alkali basalt ranges from 39 to 27, trachybasalt 17 to 14, trachyandesite 12 to 9 and trachyte 8.15 to 0.72. A trend of compositional variation showing a typical alkali volcanic rock series is revealed on $SiO_2$-oxides and SI-oxides diagrams. In $SiO_2$-total alkali diagram, alkali lime index and An-Ab'-Or diagram, the samples fall into the fields of potassic series of the alkali volcanic rock series, whereas in A-F-M diagram show a trend toward the alkali enrichment with a curve approaching toward the iron apex. In particular, trachybasalt lavas in this island have higher total iron contents which is comparable to alkali rocks in other areas, e. g. as Gough and Tristan volcanic islands located near the Mid-Oceanic ridge in South Atlantic Ocean.

Setting Time and Strength Characteristics of Cement Mixtures with Set Accelerating Agent for Shotcrete (숏크리트용 급결제를 첨가한 시멘트 모르타르의 응결 및 강도특성)

  • Although set accelerating agents are used generally in New Austrian Tunneling Method, the standards for test methods and quality of set accelerating agents are not prescribed domestically. In this study, the proprieties of the various standards and the characteristics of set accelerating agents for shotcrete were evaluated. The alkali contents of set accelerating agents based on silicate, aluminate and cement were higher than those of alkali-free ones. From the result, it is thought that the quality control of aggregate should be enhanced and that the number of test cycle of alkali-aggregate reaction should be increased. The setting times of cement paste with set accelerating agents based on silicate and alkali-free ones were different largely with mixing methods. Compressive strength of mortar with set accelerating agents based on silicate, aluminate and cement at one day satisfied the specifications of Korea Concrete Institute. However, the strength ratio compared to control mix at 28 days showed as $50{\~}65\%$ except for the alkali-free set accelerating agents. As a results of setting time and strength test, the establishment of domestic standards that can reflect the characteristics of materials and construction methods of tunnels and that can increase quality of set accelerating agents is required immediately.

Effect of indole acetic acid on the contents of various phosphate compounds in the growth phase of saccharomyces uvarum (Saccharomyces uvarum의 배양시기에 따른 여러가지 인산화합물의 함량에 미치는 IAA의 효과)

  • In order to interpret the effect of IAA on the phosphate metabolism and biosynthesis of organic compounds, Saccharomyces uvarum were cultured in the media treated with various concnetration of IAA $(10^{-3}M,\;10^{-5}M,\;10^{-7}M)$. Sampling at the beginning and intervals of culture, yeast cells fractionated were traced the contents of inorganic phosphate and organic compounds of various fractions. 1. Growth of Saccharomyces uvarum were enhanced by IAA $(10^{-3}M,\;10^{-5}M)$ and phosphate contents in DNA and RNA fractions treated with IAA were accelerated 2.3 times and 2 times in comparison with those of control. 2. Amounts of poly-P"A" and poly-p"B" were increased but poly-P"C" decreased during the culture. Therefore, it is considered that poly-P"C" play on most important role as a phosphate pool. 3. It is suggested that because phosphate contents in DNA, protein and lipid fractions increased, inorganic phosphates required phosphates required RNA were transferred from phosphates in cytoplasm, because these increased slowly during the culture. 4. Alkali-labile protein were accelerated by IAA and alkali stable protein only were inhibiction were enhanced by IAA while, ethanol : ether soluble fraction was induced by $10^{-7}M$ IAA in comparison with those control.X> IAA in comparison with those control.

Physicochemical Properties of Depolymerized Barley β-Glucan by Alkali Hydrolysis (알칼리 가수분해에 따른 보리 β-Glucan의 이화학적 특성)

  • This study was performed to investigate the changes of total and soluble ${\beta}$-glucan contents, purity and physicochemical characteristics of alkali hydrolyzed barley varieties: Saessalbori (SSB), Saechalssalbori (SCSB) and Hinchalssalbori (HCSB). The barleys were hydrolyzed at different concentrations of sodium hydroxide (0.2~1.0 N) for 12 hours. Total ${\beta}$-glucan contents of raw SSB, SCSB and HCSB were 8.40, 7.77 and 8.28%, and soluble ${\beta}$-glucan contents were 4.80, 4.16 and 4.61%, respectively. The total ${\beta}$-glucan contents after alkali hydrolyzed at 1.0 N NaOH were 7.54, 6.89 and 7.54%, also soluble ${\beta}$-glucan contents were 4.82, 4.30 and 4.55%, respectively. The degree of purity of soluble ${\beta}$-glucan in SSB, SCSB, and HCSB were 35.79, 30.91 and 33.90%, respectively. They were increased to 74.02, 75.28 and 81.41% after hydrolyzed at 1.0 N NaOH, respectively. The molecular weight and viscosity of soluble ${\beta}$-glucan solutions were decreased as sodium hydroxide concentration was increased. The re-solubility of raw barley ${\beta}$-glucan was about 50%; however, it was increased to approximately 87% as sodium hydroxide concentration was increased.

  • This study was carried out to investigate the papermaking characteristics of paper mulberry(Broussonetia kazinoki) for new Korean traditional paper(Hanji). Hanji has been traditionally made only with the bast fiber of paper mulberry. Besides mulberry bast part, wood core was utilized separately or in mixed forms as raw materials for the new Hanji. Hanjis made from the solfomethylated pulp were shown higher brightness and sheet strengths than those from alkali and alkali-hydrogen peroxide pulps. The brightness of solfomethylated pulp was found to be high enough not to need additional bleaching. In the physical properties of the Hanjis mixed with bast part and wood core pulps, the handsheet strengths were decreased as the increase of the wood core pulp contents. But the sheet strengths of Hanjis containing 10∼30% of wood core pulps were higher than those of the Hanji containing bast part pulp only. The sheet formations were improved as the increase of wood core pulp contents, while the sheet strengths were decreased.

  • Traditionally, Hanji had been made only with the bast fiber of paper mulberry (Broussonetia kazinoki). Nowadays, Hanji has been made mainly in the mixed forms of paper mulberry bast fiber and waste paper, and consequently it has raised many problems using them. This study was carried out to investigate the characteristics of Hanji mixed with paper mulberry bast part and whole stalk pulps. Hanjis made from the solfomethylated pulp were shown higher brightness and sheet strengths than those from alkali and alkali-hydrogen peroxide pulps. The brightness of solfomethylated pulp was found to be high enough not to need additional bleaching. The sheet formations were improved as the increase of whole stalk pulp contents, while the sheet strengths were decreased. In the physical properties of the Hanji mixed with bast part and whole stalk pulps, the handsheet strengths were decreased as the increase of the whole stalk pulp contents. The tensile strength and folding endurance of the Hanji containing 40∼60% of whole stalk pulps were higher as compared to the others.

  • Anatomical and chemical properties of three mulberry species (Morus spp) were analyzed as an alternative row material for the paper mulberry (Broussonetia kazinoki) . The pulping and papermaing characteristics of bast fiber and whole stalks by three different pulping processes, conventional alkali, alkali-hydrogen peroxide, and sulfomethylated, for the Hanji were investigated. The fiber dimension of M.bombycis was the biggest of the species. The fiber length of upper part and the fiber width and cell wall thickness of lower part were bigger than the others. The extractives and ash contents of bast fiber were higher than those of whole stalk , and holocellulose and lignin contents of whole stalk were higher than those of bast fiber. The pulp yields of M.alba bast fiber, M.bombycis whole stalk, and sufomethylated pulping were higher than the others.

Antitumor and Antioxidant Activities of the Extracts from Fruiting Body of Phellinus linteus

  • Fruiting bodies of Phellinus linteus were extracted by hot water and alkali methods. Sugar contents of PL-H (hot water extract) and PL-A (alkali water extract) were 81.1%, 37.4% and protein contents were 6.2%, 21.8%, respectively. Amino acid pattern showed that two extracts contained large amount of aspartic acid and alanine. Two extracts showed characteristic IR absorption pattern for glycosidic bond at $890\;cm^{-1}$. PL-H was divided two fractions by gel filtration chromatography and the molecular weights of each fraction were estimated to be about 10 kD and 225 kD, respectively and also PL-A was estimated 10 kD. Two extracts showed strong antitumor, immunomodulating and antioxidant activities, and were compared with commercialized glycopeptide anticancer drugs.
