We isolated and cultured bacteria that inhabited marine biofilms, and identified them by phylogenetic analysis using 16S rDNA sequences. In the marine environment, biofilms cover most subtidal and intertidal solid surfaces such as rocks, ships, loops, marine animals, and algae. The bacteria in most biofilms are embedded in extracellular polymeric substances that comprise mainly of exopolysaccharides. The exopolysaccharides are excreted from multiple bacterial species; therefore, biofilms are a good source for screening exopolysaccharide-producing bacteria. Thirty-one strains were cultured, and a total of 17 unique strains were identified. Phylogenetic analysis using 16S rDNA sequences indicated that the 17 strains belonged to ${\alpha}$-Proteobacteria (Ochrobactrum anthropi, Paracoccus carotinifaciens); ${\gamma}$-Proteobacteria (Pseudoalteromonas agarovorans, P. piscicida, Pseudomonas aeruginosa, Shewanella baltica, Vibrio parahaemolyticus, V. pomeroyi); CFB group bacteria (Cytophaga latercula, Tenacibaculum mesophilum); high GC, Gram-positive bacteria (Arthrobacter nicotianae, Brevibacterium casei, B. epidermidis, Tsukamurella inchonensis); and low GC, Gram-positive bacteria (Bacillus macroides, Staphylococcus haemolyticus, S. warneri).
Spatial and temporal changes in algal population in Yongdam reservoir and ecological factors that induced the changes in the size and composition of algal population were investigated by monthly sampling at ten locations in the reservoir. Nutritional state of the reservoir was identified to be phosphorus-limited with nitrogen to phosphorus (N : P) ratio much greater than 17 in most samples. Algal population was dominated by three taxonomic groups, diatoms, chlorophytes and cyanobacteria. Although explosive algal growth was not observed in the summer, algal population showed transition with time of the dominant algal type from diatoms in the winter to cyanobacteria in the summer. Chlorophyta was not the dominant group in the reservoir although they maintained relatively stable number of cells in the reservoir and showed increase in population from March to May. The application of statistical methods revealed that the factors inducing changes in cell number of each group were water temperature for diatoms and cyanobacteria and phosphorus concentration for chlorophyte. Fluctuation of cyanobacterial population was mainly observed near the inlet of tributaries while diatoms showed higher variation inside the reservoir.
DNA barcoding is becoming a widely applied tool to accurately discriminate red algae. We tested the effectiveness of DNA barcoding for identification and discovery of Champia species in Korea and clarified the phylogenetic relationships using the plastid rbcL gene. As results, we described four species of Champia such as C. inkyua sp. nov., C. recta Noda, C. bifida Okamura, and C. expansa Yendo. A new species, C. inkyua, is characterized by entangled thallus, terete and irregular branches, hooked apices, and longitudinal filaments running throughout the frond periphery only. Longitudinal filaments were composed of a complete cell with two half cells between diaphragms in the cavity. C. recta and C. bifida were reinstated with previously used names of C. parvula and C. compressa, respectively. C. recta is the first recorded species from Korea and is characterized by an erect thallus, terete and irregular branches, and straight apices. C. bifida is characterized by compressed thallus, pinnate or alternate branches, and bifid apices. C. expansa is characterized by flabellate thallus and dichotomous branches. Molecular analyses of COI and rbcL genes revealed sufficient sequence divergence to warrant species recognition in the genus Champia.
Plants dissipate excess excitation energy from their photosynthetic apparatus by a process called non-photochemical quenching (NPQ). The major part of NPQ is energy dependent quenching (qE) which is dependent on the thylakoid pH and regulated by xanthophyll cycle carotenoids associated with photosystem (PS) II of higher plants. The acidification of the lumen leads to protonation and thus conformational change of light harvesting complex (LHC) proteins as well as PsbS protein of PSII, which results in the induction of qE. Although physiological importance of qE has been well established, the mechanistic understanding is rather insufficient. However, recent finding of crystal structure of LHCII trimer and identification of qE mutants in higher plants and algae enrich and sharpen our understanding of this process. This review summarizes our current knowledge on the qE mechanism. The nature of quenching sites and components involved in this process, and their contribution and interaction for the generation of qE appeared in the proposed models for the qE mechanism are discussed.
진해만에서 분리한 무독성 Alexandrium tamarense와 이미 보고된 독성을 가진 A. tamarense종 간의 접속공, 제1상판, 후속공의 형태를 상호 비교하였다. 형태적으로 독성종과 거의 일치되지만, HPLC나 생체실험에서 진해만에서 새롭게 분리한 종은 독성이 전혀 나타나지 않았다. lectin반응결과 무독종 A. tamarense은 PNA와 강한 반응을 보여, 세포표면에 lactose나 galactose와 같은 물질이 많이 분포하고 있음을 보였다. 또한, 단백질 전기영동시 무독종은 유독종과 달리 21 kDa 정도 부위에서 종 특이적 밴드를 보였다. 따라서 PNA lectin이나 면역학적 방법을 이용하면 무독종 A. famarense을 신속하게 동정하는데 이용될 수 있다.

Lichen studies, including biodiversity, phylogenetic relationships, and conservation concerns require definitive species identification, however many lichens can be challenging to identify at the species level. Molecular techniques have shown efficacy in discriminating among lichen taxa, however, obtaining genomic DNA from herbarium and fresh lichen thalli by conventional methods has been difficult, because lichens contain high proteins, polysaccharides, and other complex compounds in their cell walls. Here we report a rapid, easy, and inexpensive protocol for extracting PCR-quality DNA from various lichen species. This method involves the following two steps: first, cell breakage using a beadbeater; and second, extraction, isolation, and precipitation of genomic DNA. The procedure requires approximately 10 mg of lichen thalli and can be completed within 20 min. The obtained DNAs were of sufficient quality and quantity to amplify the internal transcribed spacer region from the fungal and algal lichen components, as well as to sequence the amplified products. In addition, 26 different lichen taxa were tested, resulting in successful PCR products. The results of this study validated the experimental protocols, and clearly demonstrated the efficacy and value of our KCl extraction method applied in the fungal and algal samples.
Microcystis aeruginosa is common form of cyanobacteria (blue-green algae) capable of producing toxic heptapeptide (microcystin) that cause illness or death. The comparison of molecular genetic method with the morphological characteristics of cyanobacteria was conducted. We have designed PCR primers (JJM98F, JJM1141R) for cyanobacterial 16S rRNA and phycocyanin intergenic spacer (PC-IGS) gene domain. To confirm the production of microcystins, PCR primers for the N-methyltransferase (NMT) domain of microcystin synthetase gene mcyA were designed using 21 cyanobacteria strains Most of isolated strains from the Nakdong River was classified as Microcystis aeruginosa and the similarities were $99\%$ with M. aeruginosa AF 139292. $38.1\%$ of isolated strains contained microcystin synthesis gene. NMT (N-methyltransferase) were not detected in isolated strain in several strains, which means non-toxic. However, the NMTs of the strains were detected during the cultivation.
해조류에서 생합성되는 3급 함황화합물인 DMPT를 창자파래로부터 추출하기 위하여 시료의 상태, 용매 의 극성, 초음파 처리, boiling 및 autoclaving 처리에 의한 추출조건을 검토하였다. 용매추출, 초음파 처리후 용매추출, 열수추출 및 가압 열수추출법을 이용하여 창자파래로부터 DMPT를 추출한 결과, 초음파 처리후 가압 열수처리한 경우가 가장 효과적이었고, 다음으로는 초음파 처리후 열수 처리하는 방법이었다. 창자파래로부터 DMPT를 추출 해내는 최적조건으로 분말시료에 물과 acetone (2:1, v/v)의 혼합용매를 15배량 가하여 초음파 처리 후 121$^{\circ}C$에서 60분간 autoclaving하는 방법으로서, 이 때 추출된 DMPT 함량은 시료 g당 311.200ng이었다. 창자파래에서 추출한 DMPT 수용액을 조제하여 온도별로 저장시킬 때 DMPT 분해에 의한 DMS의 발생량은 저장온도에 비례하여 증가되었지만 저온에서는 매우 낮은 분해율을 나타내었다. DMPT 1mg을 저장하였을 때의 잔존율은 6$0^{\circ}C$ 저장의 경우78.5%로 고온에서는 다소 불안정하였으나 $0^{\circ}C$에서는 DMPT의 잔존율이 99.8%로 매우 높게 나타났다. 창자파래에서 추출한 DMPT 수용액을 3$0^{\circ}C$에서 방치하였을 때 pH가 5 이하의 산성영역에서는 DM-PT의 분해가 거의 일어나지 않았으나 pH가 올라감에 따라서 분해가 서서히 일어나기 시작하여 pH 9.5에 이르러서는 DMS의 양이 급속하게 증가하였다.
Carotenoid는 천연 지용성 색소이며, 세균, 조류, 식물 등이 생산한다. 세계 시장의 대부분을 차지하는 합성 염료의 대안으로서 현재는 조류나 세균, 갑각류 등의 원료로부터 아스타잔틴의 생산, 정제, 이용이 주목 받고 있다. 이 연구는 UV와 EMS를 이용하여 P. haeundaensis의 돌연변이를 유도하고, 결과적으로 astaxanthin을 과잉 생산하는 돌연변이주를 선별하고 특성을 확인하기 위해 다양한 배양 및 영양 조건을 이용하여 astaxanthin 생산량을 확인하였다. 실험 결과 UV 조사 시간이 증가하거나, EMS 농도가 증가할수록 균주의 생존율이 감소하였다. Astaxanthin 과잉 생산 돌연변이 균주의 경우 400 mM EMS와 UV 20분을 순차적으로 처리한 방법에서 선별된 변이주가 가장 높은 astaxanthin 생산량을 보이는 것을 확인하였으며, 이 균주의 이름을 PUE로 명명하였다. PUE의 최적 배양 조건은 $25^{\circ}C$, pH 7-8, 3% NaCl이며, 1% raffinose, 3% potassium nitrate 첨가 시 astaxanthin 생산량이 증가하는 것으로 밝혀졌다. PUE에서는 wild type 균주에 비해 astaxanthin 생산량이 1.58배 증가함을 확인할 수 있었다. 본 연구의 실험 결과, 돌연변이 유도에 의해 선별된 변이주는 astaxanthin의 산업적 생산에 활용 가능한 후보가 될 수 있을 것으로 사료된다.
남조류의 대번성은 특정 생물종의 감소와 물고기의 서식처를 감소하게 하는 결과를 가져와서 생태계에 상당한 교란을 가져오고 4대강의 수질을 위협하고 있다. 조류의 대번성에 영향을 미치는 인자를 해석하기 위해서 전통적으로 클로르필 a의 농도와 환경인자간의 교차상관함수를 계산하는 방식이 수행되어왔다. 교차상관함수에 사용되는 원 시계열 자료는 추계구조에 의해 영향을 받기 때문에 시계열 데이터의 추계학적인 구조를 파악하고 외부의 영향을 제거하는 선백색화 기법을 도입하였다. 이와 같은 모의과정은 모형구조의 파악, 매개변수추정, 선택된 모형의 자가진단수행등의 일련의 과정으로 진행된다. 선백색화 처리된 데이터를 이용하여 배타적 상관분석을 실시하였고 원데이터의 결과와 비교하였다. 이와 같은 과정은 조류농도 발생의 영향 인자를 구분하기 위해서도 유용한 과정이고 다른 환경인자들 사이에서의 인과성을 규명하는데도 유용하다.
