Effects of Extraction and Processing Methods on Antioxidant Compound Contents and Radical Scavenging Activities of Laver (Porphyra tenera)

  • Laver is one of the most consumed edible red algae seaweeds in the genus Porphyra. Laver is primarily prepared in the form of dried, roasted, and seasoned products. We investigated the total polyphenol and flavonoid contents of laver products, and evaluated the in vitro antioxidant properties of solvent extracts from commercially processed laver products. Significant differences in the concentration of phenolic compounds were found among differently processed laver. The total phenolic content for laver extracts ranged from 10.81 mg gallic acid equivalent (GAE)/g extract to 32.14 mg GAE/g extract, depending on extraction solvent and temperature. Laver extracts contained very few flavonoids (0.55 mg catechin equivalent/g extracts to 1.75 mg catechin equivalent/g extracts). 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), hydroxyl radical, and superoxide anion scavenging assays were used to determine the radical scavenging capacities of laver extracts. These assays revealed that the processing method and extraction condition affected the antioxidant potentials of laver. Antioxidant activity of dried laver, roasted laver, and seasoned laver increased in a concentration-dependent manner ($100{\sim}1,000{\mu}g/mL$). The radical scavenging activities of $37^{\circ}C$ and $100^{\circ}C$ water extracts were lower than that of a $37^{\circ}C$ 70% ethanol extract. The highest radical scavenging capacity was observed in the $37^{\circ}C$ 70% ethanol extracts of dried laver, roasted laver, and seasoned laver. Overall, these results support that notion that laver contains bioactive compounds, such as polyphenols and flavonoids, which may have a positive effect on health.

Anticancer effects of Ulva compressa extracts on FaDu human hypopharangeal squamous carcinoma cells in vitro

  • Ulva compressa Linnaeus (UCL) is a green algae seaweed that performs photosynthesis and is used as a food material in some Asian regions including Korea. It is known to be the dominant species in copper ion-contaminated seas, and many studies on copper ion resistant mechanisms have been reported. UCL is known to have an excellent antioxidant effect, but limited information is available regarding its other physiological activities. In this study, we investigated the anticancer activity of 30% prethanol extracts of Ulva compressa Linnaeus (30% PeUCL) and the underlying mechanisms of its activity on human FaDu hypopharyngeal squamous carcinoma cells. The 30% PeUCL extracts suppressed FaDu cell viability without affecting normal cells (L929), as determined by MTT and viability assays. Furthermore, the 30% PeUCL extracts induced apoptosis, as determined by DAPI staining. The 30% PeUCL extracts inhibited colony formation effectively as well as wound-healing of FaDu cells, even at noncytotoxic concentrations. In addition, 30% PeUCL extracts induced apoptosis significantly through proteolytic cleavage of caspase-3, -7, and -9, and poly (ADP-ribose) polymerase, and by downregulation of Bcl-2 and upregulation of Bax in FaDu cells, as determined by Western blot analysis. Collectively, these results suggest that the inhibitory effect of 30% PeUCL extracts on the growth of oral cancer cells, colony formation and wound-healing may be mediated by caspase- and mitochondrial-dependent apoptotic pathways in human FaDu hypopharyngeal squamous carcinoma cells. Therefore, 30% PeUCL extracts can be administered as a natural chemotherapeutic drug for the treatment of human oral cancers.

한국산 해조류의 어류병원성세균에 대한 항균활성 (Antimicrobial Activities of Korean Marine Algae against Fish Pathogenic Bacteria)

  • 본 연구에서는 주요 어병세균인 Staphylococcus sp., Streptococcus sp., E. tarda 및 V. anguillarum에 대한 30 종의 해조류 추출물의 항균활성을 검색하였다. 그 결과, C. officinalis, D. simplex, G. furcata, G. lanceolata 및 G. turuturu의 경우는 그람 양성균인 Staphylococcus sp.에 대하여, 그리고 S. thunbergii 및 P. morrowii 는 그람 음성균인 E. tarda와 V. anguillarum에 대하여 항균활성을 나타냄을 밝혔다. 그 중, 가장 우수한 항균활성을 나타낸 P. morrowii의 항균활성분획물인 90% MeOH 분획물은 E. tarda의 증식을 매우 효과적으로 저해하며, MIC는 1 mg/ml으로 측정되었다. 뿐만 아니라, 다양한 pH 조건하에서도 항균활성이 유지되었으며 분획물의 주요활성 성분은 할로겐 함유 phenol계 화합물일 것으로 추정된다.

  • Marine Algae of order Chordariales are rich resources of bioactive metabolites. Methanolic extracts of the brown alga /shige Okamurae exhibited potent antioxidative and butyrylcholinesterase(BChE) inhibitory effects. Bio-guided purification [solvent partition, ODS flash, silica flash, gel-filtration on Sephadex LH 20, ODS HPLC] of them gave a compound 1. Its structure was elucidated by detailed analysis of spectroscopic data of 1 and comparison of literature data. A variety of bioassay for 1 is in progress.

거대녹조류 창자파래 추출물의 항균, 항산화 활성 (Antimicrobial and Antioxidant Activities of Extracts of Marine Green-algae Enteromorpha intestinalis)

  • In this study, we investigate the antimicrobial and antioxidant activities of extracts of Enteromorpha intestinalis. To evaluate the antimicrobial and antioxidant activities, three solvents (hexane, chloroform and methanol) were applied to obtain extracts. The extraction yields are hexane (A) 1.11%, chloroform (B) 0.94%, and methanol (C) 8.2%. Also, the contents of total phenolic compounds of extract A, B, C are 4.03%, 8.15%, and 2.33%, respectively. In the results of antimicrobial activity, extract A and B have higher activity than that of extract C. Especially, Vibrio vulificus, Pseudomonas aeruginosa, Bacillus subtilis, Escherichia coli are more sensitive than others. In antioxidant activity, extract A and B present the higher DPPH activity than that of control (BHA and ascorbic acid). The DPPH radical scavenging activity and SOD-like activity are ordered as B>C>A. However, extract C show high value in ferric reducing antioxidant power assay. In nitrite scavenging activity, extract A is relatively higher than others. However, the antioxidant activities are lower than that of controls (ascorbic acid and BHA). The antioxidant activities are presented the increasing pattern of increasing by the increasing of extract concentration.

  • We examined ethanol extracts prepared from the Korean marine algae belonging to the Sargassaceae family for their inhibitory effects on ${\alpha}$-glucosidase activity and free radicals in vitro. Among five marine algae, the extracts of Sargassum yezoense were found to possess strongly ${\alpha}$-glucosidase inhibition and free radicals scavenging activities. Two compounds were isolated via bioactivity guided isolation and tested for their effects on ${\alpha}$-glucosidase, DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ analysis. Their chemical structures were elucidated by spectral analysis and direct comparison with authentic compounds; their structures were identified as sargaquinoic acid (1) and sargahydroquinoic acid (2). The inhibitory effects of compound 1 and 2 ($IC_{50}$ value:14.2 and 12.8 ${\mu}M$, respectively) on ${\alpha}$-glucosidase were more potent that of deoxynojirimycin as a positive control ($IC_{50}$ value:18.0 ${\mu}M$). All compounds displayed antioxidative activity which was measured by DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ apparatus.

A comparative study of Sargassum horneri Korea and China strains collected along the coast of Jeju Island South Korea: its components and bioactive properties

  • Sargassum horneri is edible brown seaweed abundant along the coasts of Jeju Island, South Korea. In addition to the native S. horneri population, a large amount of S. horneri has been found to invade Jeju Island from the east coast of China. Thus, S. horneri of both Korea (SK) and China (SC) strains now inhabits along with the shore of Jeju Island and have become a threat to the coastal biodiversity. However, they could be used in obtaining functional ingredients for industrial level applications provided an optimized cost effective strategy. In the present study, we compared SK and SC strains for the extraction efficiency, components, antioxidant, and anti-inflammatory properties of 80% methanolic extracts and their partially purified fractions. According to the results, two strains indicated similar bioactive properties such as DPPH and alkyl radical scavenging activity as well as anti-inflammatory activities on lipopolysaccharide-stimulated RAW 264.7 cells. The yield of 80% methanol extract from SC was higher than SK. However, the yields of the ethyl acetate and chloroform fractions from SK were higher than those of SC strain. The major peaks in the high-performance liquid chromatography chromatograms, which was identified as Apo-9 fucoxanthinone, indicated that both methanolic extracts of SK and SC contains major target peaks but with different amounts. This study might be useful for developing functional materials from SC and SK in future.

유기농업자재와 유기합성 살균제(Iminoctadine tris + thiram) 교호살포에 따른 고추 탄저병 방제 효과 (Control of Pepper Anthracnose Caused by Colletotrichum acutatum using Alternate Application of Agricultural Organic Materials and Iminoctadine tris + thiram)

  • Collectotrichum acutaum 에 의해 발생하는 탄저병은 매년 고추 생산에서 수량과 품질을 감소시켜 고추 병해 중 가장 큰 피해를 주는 병해로 알려져 있다. 본 연구는 고추 탄저병 방제 시 유기농업자재와 화학농약의 교호살포로 화학농약의 사용량을 감소시키기 위한 방제 전략을 개발하기 위하여 수행되었다. 고추 탄저병에 대한 유기농업 자재와 화학농약의 교호살포 효과는 실내와 포장에서 확인하였다. 첫 번째로 13종의 미생물농약과 22종의 유기농업 자재들을 대상으로 탄저병 병원균의 생장억제시험과 생물검정을 실시하였다. 시험결과 1종의 미생물농약(Bacillus subtilis QST-713 액상수화제)과 3종의 유기농업자재(유황, 보르도액, 해조류 추출물)에서 탄저병 병원균에 대해 높은 억제효과를 나타내었다. 포장시험에서 유기합성 살균제(Iminoctadine tris+thiram)를 단용처리 하였을 때 89.5%의 방제효과가 있었으며, 반면에 유황, 보르도액, 구리, 해조류 추출물 그리고 Bacillus subtilis QST-713 액상수화제를 단용처리 하였을때에는 33.1-81.0%로 유기합성 살균제와 비교하여 방제효과가 낮은 것으로 조사되었다. 하지만 유기합성 살균제와 유기농업자재를 7일 간격으로 2회씩 교호살포하게되면 유기합성 살균제+보르도액 교호살포는 81.7%, 유기합성 살균제+해조류 추출물은 87.1%의 방제효과가 있는 것으로 확인되었다. 결론적으로 두 가지의 교호살포 방제체계는 화학농약의 살포횟수 및 사용량을 절감 할 수 있는 가능한 방안으로 추천할 수 있을 것이다.

해조단백질의 추출에 관한 연구 1. 수용성 단백질의 추출 (STUDIES ON THE EXTRACTION OF SEAWEED PROTEINS 1. Extraction of Water Soluble Proteins)

  • 분포량이 많고 식용되고 있는 중요 해조의 효율적인 이용과 영양학적인 의의를 구명하기 위한 기초자료를 얻기 위하여 동근돌김(Porphyra suborciculata). 잎파래(Enteromorpha linza). 톳(Hizikia jusiforme). 모자반(Sargassum fulvllum), 미역(Undaria pinnatifida), 청각(Codim fragile), 셀만모자반(Sargassum kjellmanianum), 구멍갈파래(Ulva pertusa)등 8종을 선정하여, 이들 해조의 불용성 단백질 추출에 영향을 주는 제요소 즉 조체처리별, 추출온도와 시간별, 시료일추출용매화, pH의 영향등을 검토하고 또한 추출액중의 단백질 심전조건에 대하여 검토하였다. 1. 식용 해조의 조단백질 함량은 둥근돌김(Porphyra suborbiculat) $24.14\%$, 미역(Undaria pinnatifida) $15.7\%$, 톳(Hizikia jusiforme) $7.66\%$, 모자반(Sargasum fulvellum) $12.61\%$, 셀만모자반(Sargassum kjellmanianum) $14.08\%$, 잎파래(Enteromorpha linza) $10.86\%$, 구멍갈파래(Ulva Pertusa) $16.60\%$, 청각(Codium fragile)$9.89\%$이었다. 2. 시료의 처리조건에 따른 추출효과는 dry icemethanol 한제 중에서 동결시켜, 해사를 첨가하여 마쇄한 시료가 수용성 단백질 추출성적이 월등하게 좋아, 막지사발에서 마쇄한 $1.5\~2.5$배의 추출성적을 보였다. 3. 시료일추출용매비는 점질물이 적은 구멍갈파래, 잎파래는 1:20(w/v)에서, 점질물량이 많은 청각,미역, 톳, 모자반, 둥근돌김 등은 1:30(w/v)에서, 셀만모자반은 1:40(w/v)일 때 추출성적이 좋았다. 4. 추출시간은 구멍갈파래, 잎파래는 1시간, 둥근돌김, 톳, 미역, 모자반, 셀만모자반은 2시간, 청각은 3시간 정도에서 가장 효과적이었다. 5. 추출온도는 톳이 $40^{\circ}C, 둥근돌김, 청각, 잎파래 미역, 모자반, 셀만모자반 $50\%$에서, 구멍갈파래는 $60^{\circ}C$에서 최고의 추출성적을 보였다. 6. pH의 영향은 각 시료마다 대동소이하여 $pH9\~12$에서 가장 좋은 결과를 얻었다. 7. 추출 최적조건에서의 수용성 단백질 추출율은 1차 추출에서 전조단백질에 대하여 평균 $63\%$, 2차 유출에서 평균 $8.6\%$ 합계 $74\%$ 정도의 수용성 단백질 추출이 가능했으며, 1차에 대한 2차의 추출비율이 7.5:1이었다. 8. 단백질 침전분리 조작으로는 Barnstein method, methanol 처리법, TCA 처리법, 가열처리법 순으로 침전효과가 있었다.
