Quality Characteristics of Tofu with Added Alfalfa (Medicago sativa L.) Extracts (알팔파 추출물을 첨가한 두부의 품질 특성)

  • This study was conducted to investigate the quality characteristics of tofu (soybean curd) prepared with added alfalfa extracts (Medicago sativa L.). The moisture content, yield, and pH of tofu decreased according to the increase of an added quantity of alfalfa extract, whereas the crude protein and turbidity increased. The tofu made with additional alfalfa extracts gave a similar amount of crude ash with that of the control. The Hunter's L (lightness) color value decreased as the content of alfalfa extract increased, but the a (redness) and b (yellowness) values increased with increasing alfalfa extract concentration. Textural properties for hardness were higher but cohesiveness, springiness, gumminess, and brittleness were lower in alfalfa extracts with added tofu compared to those of control. Isoflavones such as daidzein and genistein content were significantly increased with increasing concentration of alfalfa extracts. A sensory evaluation indicated that the group with a 0.5% alfalfa extract showed the best color, flavor, taste, and overall acceptability results. In conclusion, adding alfalfa extracts could improve the physicochemical and sensory characteristics of tofu.

Effect of Lactic Acid Bacteria and Chlorella on Nutritive Values and Quality of Italian Ryegrass-Alfalfa Silage (젖산균과 클로렐라 첨가가 이탈리안라이그라스-알팔파 혼파 사일리지의 사료가치 및 품질에 미치는 영향)

  • To provide the basic data on the utilization of chlorella as a method to improve the quality of forage, we examined the effect of the addition of lactic acid bacteria (LAB) and chlorella on the change of nutritive value, quality and microbes on Italian ryegrass and alfalfa mixture silage (IRG-Alfalfa silage). The nutritive values of IRG-Alfalfa silage were not significantly different in all treatments. The pH of IRG-Alfalfa silage decreased in both LAB and chlorella treatments. Further the content of lactic acid in the LAB treatment increased as compared to the control, and also significantly increased in the treatment of LAB with chlorella (p<0.05). In addition, the number of LAB increased in lactic acid bacteria treatment as compared to the control, and also increased in the treatment of LAB with chlorella (p<0.05). Therefore, we suggest that LAB and chlorella would improve the quality of IRG-Alfalfa silage.

Transformation of Alfalfa by BcHSP17.6 Gene using Agrobacterium tumefaciens (BcHSP17.6 유전자 도입에 의한 알팔파의 형질전환)

  • This study was conducted to obtain the transformed alfalfa (Medicago sativa L.) plants with thermotolerance gene (BcHSP17.6) using Agrobacterium tumefaciens LBA4404 and we confirmed the transformed gene from the regenerated alfalfa plants. The expression vector, pBKH4, harboring BcHSP17.6 gene was used for production of transgenic alfalfa plants. In a process for transformation, the callus of alfalfa was cocultivated with Agrobacterium tumefaciens and transformed calli were selected on kanamycin-containing SH-3-kc medium to regenerate into into the plant. The complete transgenic alfalfa plants were produced by cultivation for about 4 months on several regeneration media, SH-nk-c, SH-l lb-c, SH-sp-c, and SH-IBA. The transgenic alfalfa plants were analyzed by isolation of genomic DNA and PCR/Southem blot.

Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli (알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy)

  • The aim of this study was to separate or purify some bioactive compounds from flowers of alfalfa(Medicago sativa L.) and to test of the isolated compounds on alfalfa for their autotoxicity and on Echinochloa crus-galli for their allelopathy for seed germination and seedling weight. Using thin layer chromatography(TLC) of $CHCl_3$ extracts, the most inhibitory band to alfalfa seed germination was determined. Germination inhibition of this extract suggested a complex chemical interaction. Separation and purification of compounds with CHCl$_3$ extract of fresh alfalfa flowers were conducted by a silica gel TLC, and microcrystalline cellulose TLC(MCTLC), followed by droplet countercurrent chromatography(DCCC) bioassay. Preliminary identification was done by high perfomance liquid chromatography(HPLC) on the most inhibitory fractions in DCCC. Ferulic acid, caffeic acid, vanillic acid, rutin, narringin were identified in fraction 5 and ferulic acid, caffeic acid, vanillic acid, rutin, coumarin in fraction 6. The phytotoxicity of their individual compound was tested on alfalfa and Echinochloa crus-galli seed germination and seedling weight. Coumarin and ferulic acid showed the most inhibitory effect on alfalfa seed germination and Echinochloa crus-galli seedling fresh and dry weight. These compounds may be, at least in part, involved in autotoxicity and allelopathy.

Molecular Characterization of Silicon (Si) Transporter Genes, Insights into Si-acquisition Status, Plant Growth, Development, and Yield in Alfalfa

  • Silicon (Si) has the potential to improve plant growth and stress tolerance. The study aimed to explore Si-involving plant responses and molecular characterization of different Si-responsive genes in alfalfa. In this study, the exogenous supplementation of Si enhanced plant growth, and biomass yield. Si-acquisition in alfalfa root and shoot was higher in Si-supplemented compared to silicon deficient (-Si) plants, implying Si-acquisition has beneficial on alfalfa plants. As a consequence, the quantum efficiency of photosystem II (Fv/Fm) was significantly increased in silicon-sufficient (+Si) plants. The quantitative gene expression analysis exhibited a significant upregulation of the Lsi1, Lsi2, Lsi3, NIP5;1, and NIP6;1 genes in alfalfa roots, while BOR1, BOR4, NIP2, and NIP3 showed no significant variation in their expression. The MEME results further noticed the association of four motifs related to the major intrinsic protein (MIP). The interaction analysis revealed that NIP5;1 and Lsi1 showed a shared gene network with NIP2, BOR1, and BOR4, and Lsi2, Lsi3 and NIP3-1, respectively. These results suggest that members of the major intrinsic proteins (MIPs) family especially Lsi1, Lsi2, Lsi3, NIP5;1, and NIP6;1 genes helped to pass water and other neutral solutes through the cell membrane and those played significant roles in Si uptake and transport in plants. Together, these insights might be useful for alfalfa breeding and genome editing approaches for alfalfa improvement.

SEPARATION, IDENTIFICATION OF BIOACTIVE COMPOUNDS FROM ALFALFA PLANT (알팔파의 생리활성물질 분리 및 동정)

  • To isolate, purity and identify of bioactive compounds involved in alfalfa allelopathy and/or autotoxicity, experiment was conducted. Isolation and separation procedures used from an 80% methanol extract of fresh alfalfa leaves(1kg), silica gel thin layer chromatography(TLC), followed by Droplet Counter Current Chromatography(DCCC). Preliminary identification was examined by high preformance lipid chromatography(HPLC). Four phenolic compound, salicylic acid, scopoletin, rutin, and quercetin, were identified and identified all compounds were phytotoxic to alfalfa seed germination and seedling growth. Among these compounds, quercetin treatment($10^{-3}M$) was most inhibitory to alfalfa seed germination and seedling growth. These compounds may be, at least in part, involved autotoxicity and allelopathy.

  • The productivity of natural flavors in alfalfa suspension cultures is low partially due to their low solubility. Thus, by formation inclusion complex of cyclodextrin(CD) with flavor compounds enhancement of the productivity of flavor was attempted in this research. we observed that CD formed inclusion complex with flavor compounds by GC/MS analysis. Also, there was indication that CD played a role as an elicitor in alfalfa suspension cultures.

  • This study was conducted to investigate the effects of pathogenic hngi(Fusarium culmorurn, Fusarium oxyspomm fsp. lycopersici, Fusaiurn solani, Rhizoctonia solani), pregermination time(0, 6, 9, 12, 18, 24, 48 hrs.) and temperature($12^{\circ}C, 18^C{\circ}, $28^C{\circ}$) on growth of alfalfa seedling. The survival rate of alfalfa was decreased by pathogenic hngi and treatment of pregermination and survival rate of alfalfa resulted in higher than that of nontreatment of pregermination. Pathogenicity of fungi was increased as increased the temperature(P<0.05). The emergence rate of alfalfa was decreased when pathogenic fungi were inoculated. However, the emergence rate was increased when seeds were pregerminated(P<0.05)

Extreme pH Reduced Vegetative Growth and Biomass Accumulation in Alfalfa

  • Soil acidity or alkalinity are serious limitations for crop production. The purpose of this study was to clarify the negative effects of extreme pH stress (low and high) on alfalfa vegetative growth (VG) and biomass accumulation (BA). Two-week-old alfalfa seedlings were exposed to different pH (4.0, 4.5, 7.0, 8.0 and 8.5, respectively) levels for 72 hours. Alfalfa grown at pH 4.0 and 8.5 significantly reduced VG and BA, wherein as neutral pH (7.0) comparably exhibited better growth and biomass yield. These results indicate that extreme acidic or alkaline level are critical limiting factors for growth and biomass yield in alfalfa.

  • Effects of nitrogen applied to grown alone and in mixture with grass and legumes of forage crops on the yield of dry matter, absorptivity, and the amount of nitrogen fixation were studied using $^{15}N$ ammonium sulfate. 1. The amount of nitrogen fixation in legume was decreased in order of alfalfa > red clover > birdsfoot trefoil, and red clover gave highest soil-nitrogen dependency. 2. Fertilizer nitrogen for the yield of dry matter was highly effective only in orchardgrass, and effective at the early stages of alfalfa and birdsfoot trefoil in case of a mixed sowing. 3. Alone and mixed sewings showed the yield of dry matter in decreasing order of alfalfa > red clover > orchardgrass > birdsfoot trefoil and orchardgrass + alfalfa > orchardgrass + red clover > orchardgrass + birdsfoot trefoil, respectively. 4. The plot of with nitrogen showed greater nitrogen uptake than the plot of without-nitrogen throughout the plots. In case of grown alone, however, difference in the absorptivity of various forage crops was decreased in order of alfalfa > red clover > orchardgrass > birdsfoot trefoil. 5. Regardless of nitrogen application, nitrogen uptake amount of orchardgrass was higher in the plot of mixture then in the plot grown alone, except the with nitrogen plot of red clover grown in mixture at the ratio of 3 : 7.
