This study was performed to investigate the medicinal effects of the herbal combination extracts-2 (HCE-2), consisting of Artemisia capillaris Thunb., Lonicera japonica Thunb., Prunella vulgaris var. lilacina, and Hovenia dulcis Thunb. on the alcohol-induced liver injury in rats. The rats were randomly divided into four groups: normal group (n =6), non-treated control group (n =6), saline-treated group (n =6) and the herbal combination extract (HCE-2)-treated group (n =6). The rats in the alcohol-loaded groups were orally administered with ethanol at a daily dose of 4 g/kg-body weight for 5 weeks. Thirty minutes before the ethanol injection, saline or herbal combination extracts was administered by using a gastrogavage. Blood and liver tissue samples were taken out from the hearts and livers of the rats, respectively, on 15th and 38th days. The activities of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were measured using an enzyme-linked immunosorbent assay (ELISA). We also investigated the protective effect of the herbal combination extracts by Hematoxylin-Eosin staining on histological sections of rat liver. In this study, the oral administration of the herbal combination extracts significantly reduced the serum levels of AST and ALT, which had been raised by alcohol-induced liver injury. Histological analysis and apparent observation of liver also showed the preventive effect of the herbal combination extracts in a chronic alcohol-induced rat model. Theses results revealed that the herbal combination extracts effectively prevented hepatic damage consequent to the chronic exposure to repetitive administration of ethanol and could be used as a primary resource of a health beverage or herbal medicine, alleviating the alcohol-induced hepatic injury and hangover symptoms.

We have previously reported that Acer tegmentosum extract, which is traditionally used in Korea to reduce alcohol-related liver injury, suppresses liver inflammation caused by excessive alcohol consumption and might improve metabolism. The active ingredient, 6-O-galloylsalidroside (GAL), was isolated from A. tegmentosum, and we hypothesized that GAL could provide desirable pharmacological benefits by ameliorating physiological conditions caused by alcohol abuse. Therefore, this study focused on whether GAL could ameliorate alcoholic fat accumulation and repair liver injury in mice. During chronic alcohol consumption plus binge feeding in mice, GAL was administered orally once per day for 11 days. Intrahepatic lipid accumulation was measured in vivo using a noninvasive method, 1H magnetic resonance imaging, and confirmed by staining with hematoxylin and eosin and Oil Red O. The serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured using a Konelab system, and the triglyceride content was measured in liver homogenates using an enzymatic peroxide assay. The results suggested that GAL alleviated alcohol-induced steatosis,e as indicated by decreased hepatic and serum triglyceride levels in ethanol-fed mice. GAL treatment also correlated with a decrease in the Cd36 mRNA expression, thus potentially inhibiting the development of alcoholic steatosis via the hepatic de novo lipogenesis pathway. Furthermore, treatment with GAL inhibited the expression of cytochrome P450 2E1 and attenuated hepatocellular damage, as reflected by a reduction in ALT and AST levels. These findings suggest that GAL extracted from A. tegmentosum has the potential to serve as a bioactive agent for the treatment of alcoholic fatty liver and liver damage.
In order to investigate whether or not the alcohol-treated rat liver cells can be protected by Korean Citrus junos and medicinal herbs, We compared the serum biochemistry of rats administered both alcohol and the complex of Korean Citrus junos and medicinal herbs to control rats treated with alcohol alone. The activities of Aspartate aminotransferase (AST) and Alanine aminotransferase (ALT) in the citron 3 (Citron 3, less mellowed citron which was ripened for three months)+Phellinus linteus, Alnus japonica, Dendropanax morbifera and citron 4 (Citron 4, completely mellowed citron which was ripened fer four months)+Phaseolus radiatus, Cordyceps militaris group were significantly low when compared with the negative control group (p<0.05). The levels of triglyceride (TG) in all experimental groups were significantly lower than the negative control group (p<0.05). The concentrations of total cholesterol in the citron 3+Phellinus linteus, Atnus japonica, Dendropanax morbifera and citron 4+Phaseolus radiatus, Cordyceps militaris, Phellinus linteus were lower than the negative control group (p<0.05). The activities of alcohol dehydyogenase (ADH) in all experimental groups were significantly high when compared with the normal control group (p<0.05). These results suggest that the complex of Korean Citrus junos and medecinal herbs could be an excellent candidate for protecting vat liver cell damage induced by alcohol.
Excessive alcohol use causes oxidative stress in the liver, and antioxidant therapy has been an attractive approach for the treatment of ethanol-induced liver damage. The present study examined the hepatoprotective effect of Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) in C57BL/6 mice intoxicated with ethanol. Mice were intraperitoneally administered with ethanol alone, or together with test materials three times at 12-h intervals. At 3 h after the last dosing, hepatotoxicity was assessed based on serum activities of aspartate aminotransferase and alanine aminotransferase, and hepatic contents of thiobarbituric acid-reactive substances and glutathione. Sasa quelpaertensis extract mitigated the acute ethanol hepatotoxicity as effectively as silymarin. Its n-butanol fraction was more active than methylene chloride or aqueous fraction. p-Coumaric acid, a major constituent of S. quelpaertensis, was found to effectively prevent the ethanol-induced hepatotoxicity. These data suggest that S. quelpaertensis and p-coumaric acid could be useful for the prevention of liver disease caused by alcohol abuse.
For the evaluation of protective effect on hepatic damage by Korean red ginseng mixed formula, we used GR(Korean red ginseng), GRF-A(Korean red ginseng-mixed formula) as a materials. The study was performed on protective effect against hepatic damage induced by $CCl_4$. In vitro assay with 1.1 mM galactosamine, protection(%) was 44%(GR), 58%(GRF-A) at 50ug/ml, while maxium protection(%) was 5%(GR) and 24% (GRF-A) against acute hepatotoxicity by $CCl_4$. GRF-A significantly protected ethanol induced-liver damage by lowering ALT and ALP and fatty degenertion in liver tissue.
This study investigated the effects of an ethanol extract of Allium monanthum MAX. (AME) on ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 100~150 g, were divided into 5 groups; normal group (NOR), AME 200 mg/kg treated group (S1), ethanol (35%, 10 mL/kg) treated group (S2), AME 200 mg/kg and ethanol (35%, 10 mL/kg) treated group (S3) and AME 400 mg/kg and alcohol (35%, 10 mL/kg) treated group (S4). AME was fractionated by the following solvents: n-hexane, chloroform, EtOAC and n-BuOH. Antioxidant index of the n-BuOH fraction was 600 ppm, highest among fractions. The growth rate and feed efficiency ratio were decreased by ethanol, but gradually increased to the corresponding level of the normal group by administering AME. The serum ALT activities that were elevated by ethanol were significantly decreased by AME administration. It was also observed that the hepatic activities of SOD, catalase, xanthine oxidase and GSH-Px that were increased by ethanol were also markedly decreased in the AME treated group with compared to ETB. These results suggest that ethanol extracts of Allium monanthum MAX. may have a protective effect on ethanol-induced hepatotoxicity in rat liver.

Objectives : This study investigated the liver-protective effects of MJY2018, a Herbal formula, against alcoholic fatty liver disease and anti-oxidative effects. Methods : Its effects were investigated in an alcoholic fatty liver disease model in male C57BL/6 mice, which were fed Lieber-DeCarli liquid diet containing ethanol. MJY2018 (100 and 200 mg/kg bw/day) or silymarin (50 mg/kg bw/day) were orally administered daily in the alcoholic fatty liver disease mice for 16 days. Results : The results indicate that MJY2018 promotes hepatoprotection by significantly reducing aspartate transaminase (AST) and alanine transaminase (ALT) levels as indicators of liver damage in the serum. Furthermore, MJY2018 reduced accumulation of triglyceride and total cholesterol, increased levels of superoxide dismutase (SOD) and glutathione (GSH) in the livers of the alcoholic fatty liver disease mice model. Additionally, it improved the serum alcohol dehydrogenase (ADH) activity. Conclusions : These results indicate that MJY2018 were effective in improving and protecting oxidative stress and alcoholic liver disease.
Hovenia dulcis which is distributed in Korea, China and Japan is known to show hepatoprotctive effect and reduce the acute alcohol toxicity. In this study, the hepatoprotective effect against the chemically induced experimental liver injury models and lowering effect of blood alcohol level in animal models acutely administered alcohol by the peduncle extracts of Hovenia dulcis var. koreana were investigated. HdfHW-1, and HdfM-1 which are the extracts of fruit peduncles and young branches with hot water or 70% methanol and followed with 100% methanol, were significantly reduced the $CCl_4$ or D-galactosamine/LPS induced damage in sliced liver. The hot water or methanol extracts of fruit peduncle protected dose-dependently against $CCl_4$ induced toxicity in primary hepatocyte culture and particularly, the amount of LDH release was reduced to the control level at 500 $\mu\textrm{g}$/ml of hot water extracts. HdfHW-1 also decreased the hepatotoxicity induced by $CCl_4$ in rats. The active components of HdfHW-1 seemed to be high molecular weights because 0.2 M NaCl HdfHW-1 fraction was the most effective among NaCl fractions of HdfHW-1 eluted with various concentrations of NaCl on DEAE 650C column chromatography. HdfM and HdfHW were significantly reduced the levels of blood alcohol in rats and mice administered 40% alcohol. These results indicated that the hot water or methanol extracts of fruit peduncle of Hovenia dulcis var. koreana have hepatoprotective effect and may be reduce alcohol toxicity.
This study was performed to investigate the effect of dietary intakes and nutritional status for fatty liver male patients with fatty livers. General characteristics, nutritional knowledge, and dietary intakes were obtained from fatty liver male patients aged 20-60yr living in Seoul and Kyunggi province by individual interview and blood samples. The Results were as follows : the mean age of the subjects was 39yrs. Old and over half the subjects were overweight. Although nutritional knowledge, recognition, and accuracy scores were good, these did not affect the subject's health. The average alcohol intake of subjects was 79.8g/day and the drinking duration was 5-40yrs(average : 17yrs). The subjects drank alcohol over 4tiles/week and preferred Soju. Most side dishes taken with alcohol drinking by the subjects were foods from animal sources. Half of the subjects also drank alcohol the next day for breakfast as a hangover-chaser. The reasons for drinking were due to social relationships and by habit. Vitamin D and folate were lower than the RDA for Koreans, and other nutrient amounts were higher than the RDA. The amounts of aspartate amino-transferase(AST), Alanine amino-transferase(ALT), ${\gamma}$-glutamyl transpeptidase(${\gamma}$-GTP), Triglyceride(TG) for the subjects were higher. In conclusion, fatty liver patients drank excessively and frequently. It seems that heavy alcohol drinking is more prone to overweight or obese people and contributes to fatty livers. Therefore, the subjects need to be taken effectively educated about alcohol-induced liver damage and the importance of a balanced diet for on healthy liver.
Objectives : Alcohol-induced oxidative stress has been known to injure various tissues or organs. This stress is related with free radicals which are produced as the result of long-term alcohol consumption. Malonyldialdehyde(MDA) is produced by the interaction of free radicals and cell membrane lipids, and indicates the degree of lipid peroxidation indirectly. The purpose of this study was to investigate the relationship between red blood cell(RBC) membrane lipid peroxidation by free radicals, and associated hepatic injuries and hematologic changes. Methods : Thirty-three subjects diagnosed as alcohol dependence according to DSM-IV diagnostic criteria were evaluated within 72 hours after discontinuing alcohol drinking. Clinical characteristics were evaluated by CAGE questionnaire and Korean Michigan Alcoholism Screening Test(MAST). RBC membrane MDA level was measured as the marker of RBC membrane lipid peroxidation. Aspartate aminotransferase(AST), alanine aminotransferase(ALT) and gamma-glutamyltransferase(GGT) were used as the biochemical markers of liver damage due to alcohol ingestion. The alcohol-induced hematologic change was assessed by mean corpuscular volume(MCV). Results : The results were as follows. Clinical characteristics were not different between two groups having normal and abnormal levels of AST, ALT, GGT or MCV. The levels of MDA were not correlated with the clinical characteristics and serum levels of AST, ALT and GGT. However, there was a significant correlation between the levels of MDA and the value of MCV(p=0.017). Conclusions : These findings suggest that oxidative stress in alcohol dependence may not be reflected in liver enzyme markers such as AST, ALT and GGT, but may be reflected in MCV.
