계피(Cinnamomum cassia Presl) 추출물과 cinnamaldehyde의 alcohol dehydrogenase 저해 효과 (Inhibitory effect of cinnamon (Cinnamomum cassia Presl) extract and cinnamaldehyde on alcohol dehydrogenase)

  • 계피(Cinnamomum cassia Presl)의 열수 추출물은 alcohol dehydrogenase (ADH)의 활성을 저해하였고 IC50값은 45.6 ㎍/mL이었다. 계피 추출물의 ADH 저해 성분은 산과 열에 비교적 안정하였으나 휘발성을 가진 물질로 나타났다. 계피에서 ADH 저해 물질의 추출을 위한 최적 온도와 시간은 각각 80 ℃와 2시간이었다. 계피의 정유 성분 중 cinnamaldehyde가 ADH 저해의 주된 물질이었다. Cinnamaldehyde는 기질인 에탄올에 대해 ADH 활성의 경쟁적 저해제로 여겨진다. 따라서 계피 열수 추출물과 cinnamaldehyde는 ADH의 활성을 효과적으로 저해하는 숙취해소 천연소재로서의 활용가능성을 보여주었다.

Variation of Nutritional and Antioxidant Characteristics of Extract of Lycium barbarum produced by using Different Extraction Processes

  • Lycium barbarum extract has a high potential to be developed as a health functional food due to the various health-promoting effects of Lycium barbarum. This study analyzed changes in nutritional and functional components depending on the extraction solvent (purified water and a mixture of purified water and alcohol) and the condition of the sample. The nutritional components (carbohydrates, protein, fat, ash), organic acids, amino acids, total phenolic compounds, and total flavonoids of the extract produced during the extraction process were analyzed. The nutritional composition and functional substances of the extracts showed some differences depending on the type of solvent and the condition of the sample. The amounts of crude protein (7.61%), crude fat (1.63%), carbohydrate (90.22%), and ash (0.54%) of dried Lycium barbarum extract using purified water as a solvent were similar to those of the powder sample extract. The highest content of citric acid was 4.31 mg/mL, similar to the case of acetic acid, when the powder sample used a mixture of purified water and alcohol as a solvent. The highest amino acid content was 357.39 mg/mL when the powder sample was mixed with purified water and alcohol as a solvent. The total amount of phenolic compounds was 686.16 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent. The highest total flavonoid content was 111.32 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent.

Oenanthe javanica extract accelerates ethanol metabolism in ethanol-treated animals

  • The effect of water dropwort (Oenanthe javanica DC) extract in eliminating ethanol was evaluated in New Zealand white rabbit and ICR mice. When a hot-water extract of water dropwort extract and ethanol was injected into New Zealand white rabbit, the plasma ethanol level was rapidly reduced, similar to metadoxine treatment. Specifically, the n-butanol fraction of hot-water extract was the strongest in eliminating plasma alcohol in ICR mice. When ethanol was orally ingested, administration of the hot-water extract eliminated up to 44% of the plasma ethanol in mice while the n-butanol fraction eliminated around 70%. Alcohol removal behaved in a dose-dependent manner in response to 50-200 mg/kg of n-butanol fraction. These data show O. javanica extract is effective in overcoming alcohol intoxication by the accelerating ethanol metabolism.

흑삼의 인삼 사포닌 분석 (Analysis of Ginsenosides of Black Ginseng)

  • The objective of this study is to provide the basic information for developing a high-value ginseng product using ginseng saponin and prosapogenin. In order to achieve such aim, Ginsenoside compositions of black ginseng (BG) extracts with various solvent conditions were examined by HPLC. The total saponin and the prosapogenin content of 95$\%$ ethyl alcohol extract were higher than that of the either 50$\%$ ethyl alcohol extract or distilled water extract. As a result, the order of the total saponin and the prosapogenin content was 1) 95$\%$ ethyl alcohol,2) 50$\%$ ethyl alcohol,3) the first and second mixture of 95$\%$ ethyl alcohol, distilled water, and 4) distilled water extract. In the case of fine black ginseng (FBG), the first and second mixture extracts of 95$\%$ ethyl alcohol and distilled water were the highest. In addition, the ratio of the protopanaxadiol group and the protopanaxatriol group (PD/PT) showed that the ratio of BG ranged from 0.304 to 0.601, while the ratio of FBG ranged from 1.166 to 1.657.

마두령(馬兜鈴)의 흰쥐 간염(肝炎) 억제(抑制) 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究) (Inhibitory effects of Maduryung(Aristolochiae Fructus) on alcohol, acetaminophen and galactosamine induced hepatitis in rats)

  • Maduryung Extract, one of herbal medicine was tested for inhibitory effect to alcohol, acetaminophen and d-galactosamine induced hepatitis in rats. The results were as follows. 1. Increaced serum GOT, GPT levels by alcohol were significantly decreased by Maduryung extract.(p<0.01) 2. Maduryung extract decreaced GOT value in acetaminophen induced hepatitis and this effect may be due to increace of GSH level in liver tissue.(p<0.05) 3. Repeat administration of Maduryung extract showed inhibitory effect on s-GOT, s-GPT levels in d-galactosamine induced hepatitis. (p<0.05) According to the above results, it seems that Maduryung could be use as drug for alcohol or drug induced hepatitis treatment.

  • Petroleum ether, benzene and alcohol extracts of the seeds of C. juncea were tested for antiimplantation and pregnancy interruption activities in female albino rats. Of these three extracts, the alcohol extract was found to be the most effective in causing antiimplantation and pregnancy interruption activities. These adverse effects on fertility are reversible upon withdrawal of the extract treatments. The alcohol extract was found to possess estrogenic activity. After subjecting to preliminary phytochemical screening, the alcohol extract showed positive tests for alkaloids, steroids, glycosides, saponins, flavonoides, fixed oils, phenols and tannins.

Mechanism of Alcohol Decrease by Acid Hydrolsis of Hovenia dulcis Extract

  • This work was a method that used an acid hydrolysis for increasing the efficacy of decreasing alcohol concentration from Hovenia dulcis extract. The best pH was 2.0 to obtain a maximum alcohol dehydrogenase activity at fixed reaction temperature and time. At pH 2.0, reaction temperature $80^{\circ}C$ and reaction time 4hr gave the highest activity which was 124% of control. The bioactive compound, (+)-dihydromyricetin, content increased to 30% after acid hydrolysis. This is very simple and efficient method to increase the efficacy of decreasing alcohol concentration from Hovenia dulcis extract. The mechanism that increase the efficiency of alcohol decrease be examined through hydrolysis.

Effects of Extract and Isorhamnetin Glycoside from Brassica juncea on Hepatic Alcohol-Metabolizing Enzyme System in Rats

  • The effects of methanol extract of the leaves of Brassica juncea and its major component, isorhamnetin 3-O-${\beta}$-D-glucopyranoside on hepatic alcohol metabolizing enzymes were investigated. The methanol extract and isorhamnetin 3-O-${\beta}$-D-glucopyranoside supplementations increased the activities of microsomal ethanol oxidizing system and aldehyde dehydrogenase in a dose-dependent manner, and had mild effects on the activities of alcohol dehydrogenase and catalase. Isorhamnetin 3-O-${\beta}$-D-glucopyranoside alleviated the adverse effect of ethanol ingestion by enhancing the activities of alcohol oxidizing emzymes, microsomal ethanol oxidizing system and aldehyde dehydrogenase.

Effects of Plant Vinegar Extract on the Reduction of Blood Concentration of Alcohol and Acetaldehyde in Alcohol Administrated Rats

  • Excessive drinking causes 'alcohol hangover' within 8-16 hours. The cause of 'hangover' has not been elucidated exactly until now, but it is reported that it is caused by the creation of blood ethanol and acetaldehyde as ethanol metabolites. In this study vinegar extract of wood (VE) or OC-1, to which the powder extract of green tea leaves extract is added, was administered to the rats 30 minutes before the oral administration of ethanol (3 g/kg) and the blood ethanol and acetaldehyde concentration was measured in order to evaluate the efficacy of the beverage material for detoxification. As a result, the blood ethanol concentration in the group of the VE-1(vinegar crude extract) and VE-2 (double diluted solution) is statistically lower (P,0.05) than the exclusive alcohol administered control group. The blood acetaldehyde concentration of all groups of VE and OC-2, which is the double dilution of OC-1, is statistically low after 7 hours following ethanol administration. Especially, the AUC value of OC-2 group is statistically low compared to the control group. Accordingly, it indicates the conclusion that VE and OC-1, reducing the blood ethanol and acetaldehyde concentration which are two leading factors of 'hangover' after drinking, and worthwhile to be developed as beverage materials to eliminate 'hangover'.

Single Oral Dose Toxicity Study of an Alcohol Extract of Bumblebee, Bombus ignitus Larvae in Rats

  • The alcohol extract of the larvae of Bombus ignitus, otherwise known as the Bumblebee, was orally administered to rats at doses of 0, 0.04, 0.2, 1 or 2 g/kg as a single oral dose. There were no observed clinical signs or deaths related to treatment in all the groups tested. Therefore, the approximate lethal dose of the alcohol extract of B. ignitus was considered to be higher than 2 g/kg in rats. Mild decreases in body weight gain in male rats were observed dose-dependently within the B. ignitus treated groups over 2 weeks. Throughout the administration periods, no significant changes in diet consumption, ophthalmologic findings, clinical pathology (hematology, clinical chemistry and coagulation) or gross pathology were detected. Minor changes in male rats were found with in the hematological parameters in groups treated with the 0.04 g/kg, 1 g/kg or 2 g/kg of B. ignitus larvae extract, however, all the changes observed were within the physiological range. From these results, it was concluded that there was no evidence of specific toxicity related to the ingestion of alcohol extract of B. ignitus larvae.
