Hepatoprotective effect of Hippocampus abdominalis hydrolysate (Hippocampus abdominalis 유래 단백질 가수분해물의 간 보호 효과)

  • Recently, liver damage contributes to big percentage of the morbidity and mortality rates worldwide. Excessive intake of alcohol is one of the major causes of liver injury. When liver injury is repeated and becomes chronic, it leads to development of fibrosis and cirrhosis. In the liver, TGF-${\beta}$ is a profibrogenic cytokine, which participates in various critical events cause liver fibrosis. Seahorse (Hippocampus abdominalis) is a common traditional Chinese medicine and has been widely used for centuries. Seahorse has been known to have a variety of bioactivities, such as anti-oxidant, anti-fatigue, and anti-tumor. Peptide is one of the main compounds of seahorse. In this study, we isolated enzymatic hydrolysate from seahorse H. abdominalis by alcalase hydrolysis and investigated the effect of the hydrolysate on liver injury. In the present in vitro studies, the hydrolysate increases cell viability of Chang cells and protects Huh7 cells from ethanol toxicity. In addition, the hydrolysate inhibits TGF-${\beta}$-induced responses. In vivo studies show that the pretreatment of hydrolysate reduces alcohol-induced increases of serum Glutamic oxaloacetic acid transaminase and Glutamic pyruvate transaminase activities and increases liver weight and body weight. These results suggest that seahorse may have a hepatoprotective effect.

Biochemical Evaluation of Nutritional Status of Protein and Lipid in Patinets with Alcoholic Liver Disease (생화학적 지표로 본 알코올성 간질환 환자의 단백질 및 지질 영양상태)

  • The purpose of this study is to evaluate the nutritional status of protein and lipids in the patients with alcoholic liver disease, to find an effective way of dietary management for patients with alcoholic liver disease and to obtain the materials for nutritional education for alcoholics. The subjects consist of 80 patients with alcoholic liver cirrhosis(ALC) and 12 patients with alcoholic fatty liver(AFL). The control group included 57 alcoholics without liver disease(A), 32 patients with viral liver cir rhosis(VLC) and 194 normal(NL). Biochemical evaluation of nutritional status was performed by ana lyzing the profiles of protein and lipids in blood samples. The results are summarized as follows: 1) The frequency of subjects below the normal range of serum total protein was 26.3% in ALC, 16.7% in AFL and 3.5% in A. Serum albumin was lower in 41.3% of ALC. 2) The alcoholics without liver disease showed significantly higher level of serum triglycerides, total cholesterol and LDL cholesterol than the other groups. The alcoholic subjects had lower HDL cholesterol than normal subjects. Overall, the protein and lipids status of the alcoholic subjects in this study was evaluated to be very poor on the basis of biochemical assessments. The results suggest that alcohol abuse and poor dietary intake could cause malnutrition. An extensive nutritional education should be emphasized for the alcohol consuming population. High quality of protein and other dietary intakes from early stage of the disease may be effective in nutritional therapy for the patient with alcoholic liver disease.

Confusion Model Selection Criterion for On-Line Handwritten Numeral Recognition (온라인 필기 숫자 인식을 위한 혼동 모델 선택 기준)

  • HMM tends to output high probability for not only the proper class data but confusable class data, since the modeling power increases as the number of parameters increases. Thus it may not be helpful for discrimination to simply increase the number of parameters of HMM. We proposed two methods in this paper. One is a CMC(Confusion Likelihood Model Selection Criterion) using confusion class data probability, the other is a new recognition method, RCM(Recognition Using Confusion Models). In the proposed recognition method, confusion models are constructed using confusable class data, then confusion models are used to depress misrecognition by confusion likelihood is subtracted from the corresponding standard model probability. We found that CMC showed better results using fewer number of parameters compared with ML, ALC2, and BIC. RCM recorded 93.08% recognition rate, which is 1.5% higher result by reducing 17.4% of errors than using standard model only.

A Study on the Removal of Heavy Metals from Groundwater Using Permeable Reactive Barriers Based on Nano FeS (나노 FeS를 이용한 투과성반응벽체의 중금속 오염 지하수 처리에 관한 연구)

  • The acid mine drainage (AMD) and landfill leachates released into the subsurface environment can result in serious environmental problems like soil and groundwater contamination. The AMD and the leachates of landfill were known to contain many heavy metals. In this study, the author assessed the reactivity and ability of the FeS coated-ALC for the removal of contaminants (As, Cd, Cu, Pb, Ni, Zn, Al) in AMD and leachates in landfill. The synthetic nano-FeS and Autoclaved Lightweight Concrete (ALC) were used as reactive materials in the permeable reactive barriers(PRBs). The result of batch test indicated that synthetic nano-FeS can remove 99% of heavy metals for the 1hr of reaction time except for As and Ni(about 90%). However, the 80% of As and Ni was removed in column 1(FeS coated-ALC). The column 2(Ore FeS) removed more than 99% of heavy metals. The pH of the column 1 was increased from 3.51 to 6.39~6.50, and the pH with column 2 was increased from 3.51 to 9.20. As the result of this study, the author can surmise that the synthetic nano-FeS coated ALC will use as a very good reactive material of the PRBs to treat the contaminated groundwater with AMD and leachate of landfill.

Identification of Cuts-specific Myogenic Marker Genes in Hanwoo by DNA Microarray (DNA Microarray 분석을 통한 한우 부위별 특이 마커 유전자의 발굴)

  • Myogenic satellite cells (MSCs) are mononuclear, multipotent progenitors of adult skeletal muscle possessing a capacity of forming adipocyte-like cells (ALC). To identify the skeletal muscle type-specific myogenic and adipogenic genes during MSCs differentiation, total RNA was extracted from bovine MSCs, myotube-formed cell (MFC), and ALC from each of Beef shank, Longissimus dorsi, Deep pectoral, and Semitendinosus. DNA microarray analysis (24,000 oligo chip) comparing MSCs with MFC and ALC, respectively, revealed 135 differentially expressed genes (> 4 fold) among four cuts. Real-time PCR confirmed expression of 29 genes. Furthermore, the whole tissue sample RNAs analysis showed 6 differentially expressed genes in Beef shank. Among which, 1 gene in MSCs, 4 in MFC, and 1 in ALCs were highly expressed. This study will provide an insight for better understanding the molecular mechanism of differentiation of skeletal muscle type-specific MSCs. The identified genes may be used as marker to distinguish skeletal muscle types.

Blood Glucose Lowering Effects of Mulberry Leaves and Silkworm Extracts on Mice Fed with High-Carbohydrate Diet (고탄수화물 식이 섭취 마우스에서 상엽 및 누에 추출물의 혈당강하 효과)

  • Mulberry leaves(Mori folium) and silkworm(Bombyx mori) are potnet inhibiters of intestinal $\alpha$-glycosidase, and inhibit the digestion of starch and sucrose in the small intestine. They are able to prevent postprandial hyperglycemia and decrease blood insulin levels. In this study , a high-carbohydrate diet(CHO ; 67.5%, protein ; 20.8%, fat : 11.7%) was received by the control group. In contrast, the experimental groups received a high-carbohydrate diet with extracts of mulberry leaves and silkwork(50mg.100g diet), and acarbose(6.7mg/100g diet). after a 10 week study period , the experimental groups had lower blood glucose and triglyceride levels. The experimental groups tended to have lwer Hb Alc levels. Also, blood insulin levels were lower than the control groups in accordance with blood glucose levels. The activities of intestinal $\alpha$-glucosidase in the middle and distal parts of small intestine were induced by the extracts of mulberry leaves and silkworm in the experimental groups. However, the activities of liver lysosomal glucosidase and the contents of glycogen in the liver were not affected by the mulberry leave and silkworm extracts nor by acarbose. Mulberry leaves and silkworm were able to prevent sudden postprandial peaks in blood glucose as a result of $\alpha$-glycosidase, inhibition, there by decreasing unnecessary insulin secretion.

  • It was found that petroleum ether extract of White Ginseng had a very slightly stimulating effect on the RNA biosynthesis of Baker's yeast, while both ethanol extract of White Ginseng(WGpet-alc) and Red Ginseng Extract(RGE) showed a significant stimulating effect on the RNA biosynthesis and its activity to change mononucleotide composition of the yeast RNA was noticeable; RGE induced a remarkable decrease of AMP, CMP and especially of UMP, however a marked increase of GMP, and it was observed that the molar ratio of GMP was 78.5 % of all mononucleotides of the yeast RNA. It is of interest that these results are closely related with the stimulating effect of protein biosynthesis of Baker's yeast cells.

Effect of Telephone Follow-up on Blood Glucose Improvement of Diabetes Mellitus Patients with Depressive Symptoms (전화추후관리가 우울성향을 가진 당뇨병 환자의 혈당개선에 미치는 효과)

  • The aim of this study was to examine the effect of telephone follow-up on glycemic control, lipid profiles. and treatment method in diabetic patients with depressive symptoms over one year follow-up period. Twenty-two diabetic patients attended the structured diabetes education program between March 2004 and February 2005 and completed a questionnaire using the Beck Depression Inventory(BDI) to measure depressive symptoms. The patients showed depressive symptoms with the criterion being a BDI score at least 16. The intervention was applied to the telephone follow-up for one year. Data were analyzed using Wilcoxon signed rank test and McNemar test. The patients decreased their mean glycosylated hemoglobin(HbAlc) levels by 1.9%(p=0.001). There was a significant mean change in the fasting blood sugar(FBS) level, with a mean change of -61mg/$d{\ell}$(p=0.008). These findings indicated that the telephone follow-up may be effective in the HbAlc and FBS control.

  • The effects of alcohol. extract of acacia on total cholesterol, ${\beta}-lipoprotein$ content and S-GOT activity in serum of rabbit were examined. Rabbits were divided into four groups. A : Control group B : Cholesterol group C : Cholesterol+aca. alc. ex. (A) group D : Cholesterol+aca. alc. ex. (B) group Each group had four rabbits. A Group was fed with the stock diet only for the experimental period of 28 days. B Group was fed with the stock diet containing 500mg of cholesterol. C Group was fed with the stock diet containing 500mg of cholesterol and 300mg of acacia alcohol extract. D Group was fed with the stock diet containing 500mg of cholesterol and 500mg of acacia aocohol extract. Total cholesterol ${\beta}-lipoprotein$ content and S-GOT activity in serum was determined with spectrophotometer, spectronic 20. It was confirmed that the extract of acacia alcohol inhibits the increase of total cholesterol, ${\beta}-lipoprotein$ content and S-GOT activity in serum but the statistical probability was not significant.

Criteria of Impact Resistance of Lightweight Wall by the Large Soft Body (건식 경량벽체의 연질 충격체에 의한 내충격성 판정기준에 관한 연구)

  • Due to the nature of the existing load, the criteria of assessing the intensity of the lightweight wall's impact resistance has been though of as obscure. The current study, therefore, focuses on the standardized assessment of the impact resistance to the force of the large soft body applying to the lightweight wall. The gypsum board wall showed a low level of the maximum residual displacement. It is, however, required to be careful about the selection of the finishing process since the high level of the maximum displacement is likely to cause harm to finishing materials. Unlike the gypsum board, the ALC block wall displayed a considerable rigidity while showing almost no maximum residual displacement. Even with the low level of the maximum displacement due to the stiffness, the ALC block wall is still likely to be affected by the vibration derived from any impact on the surface, which demands a need for additional study. The future experimental study, accordingly, will focus on the impact of the vibration on finishing materials, consequently leading to the accurate prediction of the possibility of potential damage to the lightweight wall caused by the large soft body.
