  • 유전자총(Particle bombardment) 방법에 의해 snowdrop lectin (Galanthus nivalis agglutinin; GNA) 유전자가 도입된 해충저항성 형질전환 벼를 개발하였다. GNA 유전자가 도입된 많은 형질전환 식물체가 재분화되었으며, 형질전환 벼의 GNA 유전자의 integration, expression 및 inheritance는 Southern 및 western analysis분석 방법에 의해 증명되었다. 벼 genome 내 도입 유전자 수는 one 혹은 five copies이었다. 형질전환 $R_1$$R_2$ 식물체의 GNA 단백질 발현 분석 결과, 함량은 총 단백질 중 0.01%부터 2.0%까지 포함하고 있었다. 형질전환 식물체 중 GNA 과발현 형질전환 벼를 이용한 생물 검정 결과, 대조구와 비교하여 벼의 주요 해충인 벼멸구(Nilaparvata lugens St${\aa}$l)에 저항성을 나타내었다. 위의 실험 결과를 통해, 해충저항성 유전자인 GNA가 도입된 형질전환 벼는 해충의 증식을 억제하는데 이용될 수 있을 것이다.

Field Control of Phytophthora Blight of Pepper Plants with Antagonistic Rhizobacteria and DL-$\beta$-Amino-n-Butyric Acid

  • Treatment with antagonistic rhizobactera Burkholderia cepacia strain N9523 or an inducer of resistance DL-$\beta$-amino-n-butyric acid (BABA) effectively inhibited Phytophthora capsici infection on pepper plants in artificially infested pots. Treatment with BABA alone at $1,000\mu\textrm{g}$/ml or together with B. cepacia in combination induced a strong protection from the Phytophthora disease in the greenhouse. In artificially infested field, protection of pepper plants against the Phytophthora epidemic by BABA treatment was maintained at a considerable level. In contrast, soil drench with the antagonist B. cepacia alone, or in combination with BABA did not suppress the Phytophthora epidemic in the field. Mortality of pepper plants caused by P. capsici infection was significantly reduced by treatment with the antagonist Pseudomonas aeruginosa strain 950923-29 and BABA (12-29% plants diseased) relative to the untreated control (41-91% plants diseased) in the naturally infested field. Treatment with the antagonist Ps. aeruginosa strain 950923-29 and BABA also resulted in high levels of protection against Phytophthora blight in pepper plants. In the plastic filmhouse test, the average percentage of plants diseased was significantly low relative to the naturally infested field. Treatment with the antagonist Ps. aeruginosa strain 950923-29 and BABA in combination was most effective in suppressing the Phytophthora disease in field and plastic filmhouse.

토마토에 염류 내성을 유도하는 바실러스 균주 처리 후 근권 미생물 군집 구조 연구 (Assessment of Rhizosphere Microbial Community Structure in Tomato Plants after Inoculation of Bacillus Species for Inducing Tolerance to Salinity)

  • BACKGROUND: Soil salinity causes reduction of crop productivity. Rhizosphere microbes have metabolic capabilities and ability to adaptation of plants to biotic and abiotic stresses. Plant growth-promoting bacteria (PGPB) could play a role as elicitors for inducing tolerance to stresses in plants by affecting resident microorganism in soil. This study was conducted to demonstrate the effect of selected strains on rhizosphere microbial community under salinity stress. METHODS AND RESULTS: The experiments were conducted in tomato plants in pots containing field soil. Bacterial suspension was inoculated into three-week-old tomato plants, one week after inoculation, and -1,000 kPa-balanced salinity stress was imposed. The physiological and biochemical attributes of plant under salt stress were monitored by evaluating pigment, malondialdehyde (MDA), proline, soil pH, electrical conductivity (EC) and ion concentrations. To demonstrate the effect of selected Bacillus strains on rhizosphere microbial community, soil microbial diversity and abundance were evaluated with Illumina MiSeq sequencing, and primer sets of 341F/805R and ITS3/ITS4 were used for bacterial and fungal communities, respectively. As a result, when the bacterial strains were inoculated and then salinity stress was imposed, the inoculation decreases the stress susceptibility including reduction in lipid peroxidation, enhanced pigmentation and proline accumulation which subsequently resulted in better plant growth. However, bacterial inoculations did not affect diversity (observed OTUs, ACE, Chao1 and Shannon) and structure (principle coordinate analysis) of microbial communities under salinity stress. Furthermore, relative abundance in microbial communities had no significant difference between bacterial treated- and untreated-soils under salinity stress. CONCLUSION: Inoculation of Bacillus strains could affect plant responses and soil pH of tomato plants under salinity stress, whereas microbial diversity and abundance had no significant difference by the bacterial treatments. These findings demonstrated that Bacillus strains could alleviate plant's salinity damages by regulating pigments, proline, and MDA contents without significant changes of microbial community in tomato plants, and can be used as effective biostimulators against salinity stress for sustainable agriculture.

Effect of the Microalga Chlorella fusca CHK0059 on Strawberry PGPR and Biological Control of Fusarium Wilt Disease in Non-Pesticide Hydroponic Strawberry Cultivation

  • The purpose of this study was to identify strawberry wilt pathogens and evaluate the efficacy of Chlorella fusca CHK0059 for improving plant growth and suppressing Fusarium wilt. We identified 10 isolates of wilt pathogens of non-pesticide Seolhyang strawberry plant, including Fusarium oxysporum f. sp. fragariae, using morphological and molecular analysis. On the 15th day after 0.4% CHK0059 treatment, the plant height of the untreated control strawberry plants was significantly greater than that of the CHK0059-treated strawberry plants. After 85 days, both treatments showed a similar tendency regarding the height of the strawberry plants. However, the thickness of strawberry leaves treated with the CHK0059 was found to be 1 mm thicker than that of the untreated control. The flowering percentage of the CHK0059 plants was also 40.2% higher on average than that of the untreated control. The chlorophyll content of strawberry leaves treated with the CHK0059 was also, on average, 6.63% higher than that of the untreated control. After 90 days of the CHK0059 treatment, the incidence of Fusarium wilt in the CHK0059-treated plants had reduced by 9.8% on average compared to the untreated control. The population density of F. oxysporum f. sp. fragariae was also reduced by approximately 86.8% in the CHK0059-treated plants by comparison to the untreated control at 70 days after treatment. The results indicate that the microalga C. fusca CHK0059 is an efficient biological agent for improving strawberry plant growth and suppressing Fusarium wilt disease in organic strawberries.

  • Recently, attempts to use indigenous plants have been made for the recovery of nature of slopes and making permanent vegetation group, harmonizing with surroundings from the ecological perspective. For surface failure prevention of slopes, indigenous plants are known as superior to those of exotic grasses, but studies how much indigenous plants influence shear strength of soil is lacking. Therefore, in this study, strength increasement of the root-soil system were investigated for four indigenous plants.

Over-expression of OsHsfA7 enhanced salt and drought tolerance in transgenic rice

  • Heat shock proteins play an important role in plant stress tolerance and are mainly regulated by heat shock transcription factors (Hsfs). In this study, we generated transgenic rice over-expressing OsHsfA7 and carried out morphological observation and stress tolerance assays. Transgenic plants exhibited less, shorter lateral roots and root hair. Under salt treatment, over-expressing OsHsfA7 rice showed alleviative appearance of damage symptoms and higher survival rate, leaf electrical conductivity and malondialdehyde content of transgenic plants were lower than those of wild type plants. Meanwhile, transgenic rice seedlings restored normal growth but wild type plants could not be rescued after drought and re-watering treatment. These findings indicate that over-expression of OsHsfA7 gene can increase tolerance to salt and drought stresses in rice seedlings.

  • Farmers need alternatives for weed control due to the desire to reduce chemicals used in farming. However, conventional mechanical cultivation cannot selectively remove weeds located in the seedline between crop plants and there are no selective heribicides for some crop/weed situations. Since hand labor is costly , an automated weed control system could be feasible. A robotic weed control system can also reduce or eliminate the need for chemicals. Currently no such system exists for removing weeds located in the seedline between crop plants. The goal of this project is to build a real-time , machine vision weed control system that can detect crop and weed locations. remove weeds and thin crop plants. In order to accomplish this objective , a real-time robotic system was developed to identify and locate outdoor plants using machine vision technology, pattern recognition techniques, knowledge-based decision theory, and robotics. The prototype weed control system is composed f a real-time computer vision system, a uniform illumination device, and a precision chemical application system. The prototype system is mounted on the UC Davis Robotic Cultivator , which finds the center of the seedline of crop plants. Field tests showed that the robotic spraying system correctly targeted simulated weeds (metal coins of 2.54 cm diameter) with an average error of 0.78 cm and the standard deviation of 0.62cm.

  • Selectable marker gene systems are vital for the development of transgenic plants, but the presence of selectable marker genes encoding antibiotic or herbicide resistance in genetically modified plants poses a number of problems. A lot of research results and various techniques have been developed to produce marker-free GM plants. The aim of this review is to describe the principal methods used for eliminating selectable marker genes to generate marker-free GM plants, concentrating on the three significant methods(co-transformation, site-specific recombinase-mediated excision, non-selected transformation) in several marker-free techniques.

Leaf Spot of Yam Caused by Pseudophloeosporella dioscoreae in Korea

  • Leaf spot symptoms were frequently observed on yam plants grown in the Yeoju area in Korea during a disease survey in 2008. A total of five isolates of Pseudophloeosporella sp. were obtained from the infected leaves of yam plants. All of the isolates were identified as Pseudophloeosporella dioscoreae based on their morphological and cultural characteristics. A phylogenetic tree derived from the internal transcribed spacer sequences of the fungal isolates showed that the fungus is distinctly separated from species in other related genera. P. dioscoreae isolates caused very tiny spots on leaves of yam plants two weeks after artificial inoculation which were similar to those observed in the field. This is the first report that Pseudophloeosporella dioscoreae causes leaf spot in yams in Korea.

  • Water Quality of effluent from wastewater treatment plants was reviewed to reuse effluent for agricultural water as alternative water resources. Among 2004, wastewater treatment plants, 19 plants are found to be used as wastewater resources applicable to irrigation. The total effluent capacities are 9,293 thousand $m^3$per day, which may be used to irrigate paddy fields. In order to know how much the effluent can be use for agricultural water, we classified the effluent according to the river basin area and evaluated the water quality of the effluent.
