Development and validation of an analytical method for the quantification of 2,6-diisopropylnaphthalene in agricultural products using GC-MS/MS

  • An analytical method was developed and optimized for the quantification of a plant growth regulator, 2,6-diisopropylnaphthalene (2,6-DIPN), in agricultural products using gas chromatography-tandem mass spectrometry. The samples were extracted, partitioned, and were purified using a Florisil® cartridge. To validate the analytical method, its specificity, linearity, limit of detection (LOD) and limit of quantification (LOQ) of the instrument, LOQ of the analytical method (MLOQ), accuracy, and repeatability were considered. The method displayed excellent results during validation, and is suitable for the determination and quantification of the low residual levels of the analyte in the agricultural samples. All of the results with the optimized method were satisfactory and within the criteria ranges requested in the Codex Alimentarius Commission guidelines and the Ministry of Food and Drug Safety guidelines for pesticide residue analysis. The developed method is simple and accurate and can be used as a basis for safety management of 2,6-DIPN.

Quantitation of CP4 5-Enolpyruvylshikimate-3-Phosphate Synthase in Soybean by Two-Dimensional Gel Electrophoresis

  • Changes of CP4 5-enolpyruvylshikimate-3-phosphate synthase (CP4 EPSPS) in the glyphosate-tolerant Roundup Ready soybean were examined using purified CP4 EPSPS produced in cloned Escherichia coli as a control. CP4 EPSPS in genetically modified soybean was detected by twodimensional gel electrophoresis (2-DE) and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and electrospray ionization tandem mass spectrometry (ESI-MS/MS) with databases. CP4 EPSPS in soybean products was resolved on 2-DE by first isoelectric focusing (IEF) based on its characteristic pI of 5.1, followed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) based on its molecular mass of 47.5 kDa. We quantified various percentages of soybean CP4 EPSPS. The quantitative analysis was performed using a 2D software program on artificial gels with spots varying in Gaussian volumes. These results suggested that 2-DE image analysis could be used for quantitative detection of GM soybean, unlike Western blotting.

  • The phosphoenolpyruvate-dependent carbohydrate phosphotransferase system (PTS) is responsible for the simultaneous transfer and phosphorylation of various carbon sources in Escherichia coli. The ptsG gene encoding the enzyme $IICB^{Glc}$, the membrane component of the glucose-specific PTS, is repressed by Mlc and activated by the CRP cAMP complex; various other factors, such as Fis, FruR, and ArcA, are also known to be involved in ptsG regulation. Thus, in an attempt to discover a novel gene affecting the regulation of ptsG, a mutant with a decreased ptsG transcription in the presence of glucose compared with the wild-type strain was screened using transposon random mutagenesis. The mutant was found to have a transposon insertion in yhjV, a putative gene encoding a transporter protein whose function is yet unknown.

Analysis on the Value of Attributes of Agricultural Products for Chungnam School Food Service: Focused on Potato, Bean Sprouts and Tomato (충남 친환경학교급식의 농식품 속성 가치 분석: 감자, 콩나물, 토마토를 중심으로)

  • The objective of this study is to analyze the value of attributes of agricultural products in school food service using hedonic price model and choice experiment. In the case of potatoes, bean sprouts, and tomatoes, environment-friendly agricultural products rather than conventional ones, and domestic price premiums exist rather than Chungnam. The higher the price, the higher the margin rate. Conventional agricultural products were more profitable than environment-friendly ones, and domestic products were more profitable than Chungnam. In consumer preferences, environment-friendly agricultural products are preferred over conventional agricultural products and local products (Chungnam, Cheonan, Asan) rather than domestic ones. This is the opposite result of the Hedonic price model estimation that Chungnam has a lower price premium than domestic in the case of origin. This study is meaningful by analyzing the attribute value of agricultural products used for school meals in Chungnam and comparing them with the willingness to pay consumers. With the results of this study, it is necessary that discussions on supplementing the value of local products used in Chungnam school food service.

Immune Enhancing Effect by Ethanol Extract of Ailantias altissima (가죽나무 에탄올 추출물에 의한 면역증강 효과)

  • The aim of this study was to investigate the immune activity of Ailantias altissima as an active ingredient on the immune enhancement by mixing ethanol extract with lactic acid bacteria (LAB). The activity of $NF-{\kappa}B/AP1$ transcription factor increased by $NF-{\kappa}B$ activity when mixed with LAB samples rather than with the extract alone. Nitric oxide (NO) production was similar in ethanol extract alone group and LPS treatment group. Mixing Ailantias altissima extract and lactic acid bacteria led to low NO production. The cytokine productivity of $TNF-{\alpha}$ significantly increased in Ailantias altissima extract when treated with LPS, and increased even more when mixed with lactic acid bacteria. The $IL-1{\beta}$ cytokine production was high when the Ailantias altissima extract were treated alone, but no $IL-1{\beta}$ cytokine was produced in the mixtures with isolates. The combination of the ethanol extract of the Ailantias altissima and the lactic acid bacteria was found to be effective in the immune function. Consequently, the ingredient to combine Ailantias altissima extract and lactic acid bacteria can be effectively used for development of the health functional food on the prevention and treatment of hypoimmunities.

Alpha-Lipoic Acid Inhibits Glycogen Synthesis and Modifies Glucose Metabolism and Signaling Pathways in Soleus Muscles from Healthy Rats

  • Alpha-lipoic acid is a known hypoglycemic agent that may be useful in the treatment of diabetes. The objective of this study was to investigate the fate of glucose in isolated muscles incubated with lipoic acid by determining its direct effects on specific metabolic and signaling pathways. Soleus muscles from healthy rats were incubated with lipoic acid in the absence or presence of insulin. Glucose transport, glycogen synthesis, glucose oxidation and lipid synthesis were determined and affects on major pathways associated with insulin signaling were evaluated. Glucose transport was not significantly altered by the addition of lipoic acid to the incubation medium. However, lipoic acid decreased glycogen synthesis in comparison to controls. Glucose oxidation was moderately increased while de-novo lipid synthesis from glucose was inhibited. Wortmannin repressed insulin stimulation of glucose incorporation into glycogen, an effect that was augmented by the combined treatment of wortmannin and lipoic acid. Basal and insulin-stimulated serine phosphorylation of Akt was not changed by the addition of lipoic acid to the incubation medium. These data show that in this in vitro model, lipoic acid did not significantly affect glucose uptake but dramatically modified pathways of glucose metabolism within muscle tissue.

Formulation and Quality Evaluation of Chicken Nuggets Supplemented with Beef and Chicken Livers

  • This study explores the potential of utilizing meat byproducts, specifically chicken and beef liver, to enhance the nutritional value of processed foods like chicken nuggets. Proximate analysis was conducted on the livers, including moisture, ash, fat, and protein content, and degradation potential was observed. Antioxidant potential was analyzed through 2,2-diphenyl-1-picrylhydrazyl (DPPH). The total phenolic content (TPC), oxidative stability through peroxide value (POV), and free fatty acid (FFA) were performed to evaluate quality changes during seven-day storage. The radical scavenging activity showed that beef liver has excellent antioxidant capacity (61.55%- and 195.89- mM gallic acid equivalent for DPPH and TPC, respectively) compared to chicken liver and significantly increased the antioxidant potential of nuggets by 5%-10%. POV and FFA values increased with increased storage days for the liver and its incorporation in nuggets. However, the values remained under the 10 meq/kg threshold. Incorporating the livers into chicken nuggets led to a significant (p=0.000) improvement in nutritional content, particularly a 1.5%-2% increase in protein, with a similar increase in mineral content. Texture and sensory evaluations indicated favorable consumer acceptability for liver-enriched nuggets. Overall, this research shows the value of adding liver as a functional ingredient to enhance the nutritional profile of processed foods.

A Study on the Improvement of Effluent Treatment from Small Scale Agro-food Processes (소규모 농산가공시설 배출수 처리시설 개선방안)

  • This study examined the regulation status on wastewater treatment in agro-food processing and a case study on effluent treatment m ethods was carried out to evaluate any change after the mitigation of regulation. First, in order to clarify the area of investigation, the definition of small-scale agro-processing facilities was reviewed through literature survey and local government ordinance. The current law were separately analyzed into four areas; effluent treatment facilities, development of agro-processing industries, land use and food processes equipment. The exclusion clauses on wastewater discharging facility in the enforcement regulation were defined in detail, which can be served in practice. Site survey, after the questionnaire survey of the person in charge of the local unit, was carried out. As the result, this survey confirmed the positive effects of the deregulation on promoting sewer system service in rural areas, introducing the new processing construction and so on. In addition, it was found that some matters to be considered to determine whether to introduce wastewater treatment plan for public food processing facilities.

Quality characteristics and bread-making properties of Korean whole wheat flour produced at different addition levels of wheat bran

  • Whole wheat flour is produced by grinding wheat grains and blending each milling fraction such as wheat bran and white flour with no specific standard for the proportions. An exploration on classifying and utilizing Korean whole wheat flour is required to increase the practical use of many different types of Korean wheat flour products. This study aimed to examine factors for categorization and processability of Korean whole wheat flour by investigating quality characteristics and bread-making properties of Korean hard wheat flour samples produced with different levels of wheat bran (0% as control, 5%, 10%, 15%, 20%, and whole grain wheat flour). The ash content of flour samples was divided into six gradational categories, which could be suggested as classification criteria for Korean whole wheat flour. The main parameters of dough and gelatinization properties of the flour samples exhibited downtrends, but nutritional constituents demonstrated uptrends with increasing bran addition level. The bread specific volume, hardness, chewiness, and brittleness of bread had strong correlations with dough stability and pasting parameters, which could be presented as significant factors for predicting bread-making properties of Korean whole wheat flour.

  • To internalize climate-related external costs from agricultural production and food consumption Pigou taxes and carbon credits increase private costs for food. Voluntary GHG reduction program for carbon-neutral food can be advantageous over such policy measures since they avoid higher food prices for the poor. The pilot project of low-carbon agriculture certification system is to reduce GHG emissions from agricultural production and enhance the competitiveness of domestic agricultural products. This study examines producers', distributors', and consumers' perceptions of the low-carbon agriculture certification system and analyzes alternatives to promote the low-carbon certificated agricultural products.
