Difference in Physiological Responses to Environmental Stress in Protox Inhibitor Herbicide-Resistant Transgenic Rice and Non-transgenic Rice (Protox 저해형 제초제 저항성 형질전환벼와 비형질전환벼의 환경스트레스에 대한 생리적 반응 차이)

  • The objective of this research was to confirm the difference in physiological responses to environmental stresses such as chilling, high temperature, NaCl, and chemical stress (paraquat) in Protox inhibitor resistant-transgenic rice (MX, PX, and AP37) and its non-transgenic counterpart (WT). Transgenic and non-transgenic rice plants were exposed to a chilling temperature of $5^{\circ}C$ for 1 day or a high temperature of $45^{\circ}C$ for 4 days and allowed to recover at $25^{\circ}C$ for 6 days after the chilling treatment or 8 days after the high temperature treatment. Leaf injury, shoot fresh weight, porphyrin biosynthesis substances, and chlorophyll content were investigated in transgenic and non-transgenic rice at 6 days after 0.5% and 1% NaCl treatments or at 5 days after 0~300 ${\mu}M$ paraquat treatments. No significant difference in leaf injury and shoot fresh weight were observed between transgenic and non-transgenic rice during chilling and recovery. Plant height and shoot fresh weight were also similar between transgenic and non-transgenic rice during the high temperature and recovery period (0~5 days). However, plant height and shoot fresh weight in transgenic rice line MX and PX were lower than in non-transgenic rice at 6 days for recovery. Leaf injury, chlorophyll, and Mg-Proto IX ME contents had no significant difference between transgenic rice and non-transgenic rice after NaCl treatment, but Proto IX content for AP37 and shoot fresh weight for PX and AP37 in 0.5% NaCl treatment were significantly reduced compared with non-transgenic rice. There was no difference in leaf injury and shoot fresh weight when comparing transgenic rice and non-transgenic rice after paraquat treatment. Although transgenic rice and non-transgenic rice showed a little difference at a particular measurement period in certain environmental stresses, there was generally no difference in physiological responses between transgenic rice and non-transgenic rice.

Differences in Rice Quality and Physiochemical Component between Protox Inhibitor-Herbicide Resistant Transgenic Rice and Its Non-transgenic Counterpart (Protox 저해형 제초제 저항성 형질환벼와 비형질전환벼의 미질 및 이화학적 성분 차이)

  • Characteristics related to grain quality and physiochemical components such as mineral, total amino acid, free amino acid, and free sugar composition were investigated in Protox inhibitor resistanttransgenic rice (MX, PX, and AP37) and its nontransgenic counterpart (WT). Head rice, palatability, protein, and whiteness (except for MX and AP37) of milled transgenic rice were high or similar to those of the non-transgenic counterpart. Immature rice, unfilled grain, and cracked kernels (PX and AP37) of milled transgenic rice were lower than those of its non-transgenic counterpart. However, there were no significant differences in damaged grain between the transgenic rice lines and its counterpart. Potassium content in PX and copper contents in PX and AP37 were only low compared with their non-transgenic counterparts, but other mineral contents in transgenic rice lines were high or showed no significant differences compared with non-transgenic counterparts. Contents of most total amino acid composition in transgenic rice lines were high or similar to those in non-transgenic counterparts, but the content of isoleucine in AP37 was only low compared with its non-transgenic counterpart. On the other hand, free amino acid, leucine and tyrosine in PX and AP37, and total free amino acid in PX were low compared with their non-transgenic counterparts. However, the content of free amino acid in other kinds in transgenic rice lines were similar to those in their non-transgenic counterparts. Contents of sucrose in MX and PX were low compared with non-transgenic counterpars, but contents of fructose, glucose, and maltose in transgenic rice lines were high or similar compared with their non-transgenic counterparts. This results indicated that Protox genes had no negative affect on the nutritional composition of rice.

An Analysis of Factors Associated with Private Owner's Attitude toward Forest Management (사유림소유자(私有林所有者)의 시업의식(施業意識)에 영향(影響)을 미치는 제요인(諸要因)의 분석(分析))

  • The study has been carried out to investigate the relationship of certain factors to the private owners' attitudes toward forest management. The dependent variable is the attitude toward forest management, and the six independent variables are owner's age, educational level, forestland area, socioeconomic status, communicational and sociopsychological factors. The dependent variables were measured by Likert attitude scale. The data used in the study were obtained from 262 common-and model-forest owners, and analyzed primarily by multiple correlation statistical technique. Major findings of the study are summarized as follows; 1. The average score of common-owners' attitudes toward forest management is significantly lower than that of model-owners'. The scores of such independent variables as age, forestland area, socioeconomic status and communication of common-owners are also significantly lower than those of model-owners', while the significant difference of sociopsychological scores is not recognized between two owner groups. 2. The common-owners' attitudes toward forest management are negatively correlated with age variable, arid positively with other independent variables. On the other hand, the model-owners' are negatively correlated with education variable, arid positively with age, communicational and sociopsychological factors. 3. The multiple correlation coefficient between common-owner' management-attitude scores and six independent variables is 0.6971. About 41 percent of the variance of this attitude cart be explained by sociopsychological factor, and other 7 percent by communicational variable. In case of model-owners, the multiple correlation coefficient is 0.5207, and about 16.2 percent of the variance of the attitude can be explained by sociopsychological factor and 6 percent by age variable. 4. The multiple correlation coefficient between the attitude scores of all-owners arid such seven different personality factors as fatalistic, trustful, cooperative, ambitious, conservative, scientific and traditional familism personalities is 0.6566. In this case, 15 percent of the variance of the management-attitude can be explained by ambitious personality, and 11.2 percent by traditional familism. 5. The private forest owners' attitudes toward forest management are greatly affected by sociopsychological and communicational factors. The emphasis should be therefore put on the forestry extension works to induce private owners to management practices.

Effect of Rice Bran and Barley Bran Application on Growth and Yield of Chinese Chive (Allium tuberosum Rottler) and Taro (Colocasia esculenta) and Weed Control (쌀겨, 보릿겨 처리가 부추와 토란의 생육과 수량 및 잡초방제에 미치는 영향)

  • This study was carried out to examine the effect of rice bran, barley bran, burned rice bran, and burned barley bran on the growth and yield of Chinese chive (Allium tuberosum Rottler), taro (Colocasia esculenta), and weed control. When the above 4 brans were examined 13, 27, 41 and 57 days respectively after application, the plant height of Chinese chive applied with burned barley bran was significantly higher than non-treated control, whereas the other brans did not have any distinct effect on the plant height or population number of Chinese chive. However, when examined 57 days after the application of the above 4 brans, all the plants applied with brans showed more than twice the improvement in shoot fresh weight compared with non-treated control. A chemical analysis of soil 57 days after the application of the above 4 brans showed that the soils were richer in available phosphate and organic matter. Shoot fresh weight of Chinese chive at 2 weeks after cutting was significantly higher in barely bran treated plot than in non-treated plot. In the case of taro, only taro plots transplanted when 10 cm tall and applied with barley bran showed an improvement in growth increment of both the underground and above parts. However, when sowed seeds after the application of the 4 brans, the yield of taro was reduced by the brans. Thus this research indicates that the effect of brans is differ based on the amount of bran application as well as crops. The effect of weed control on Echinochloa crus-galli, Digitaria clliaris, Chenopodium album, and Solanum nigrum as affected by brans was very low in pot conditions. Weed efficacy of the brans was also very low in field conditions. Growth of Chinese cabbage and garland chrysanthemum was inhibited 63% and 37% by rice bran at $4,000kg\;ha^{-1}$, respectively, but other crops such as maize, squash, cucumber, and Chinese chive were inhibited by 0-20%. These results were similar to that of barley bran except for Chinese cabbage.

Quality Changes as Affected by Storage Temperature and Polyamide Film Packaging in Paprika (Capsicum annuum L.) (파프리카 저장 온도 변화와 폴리아미드 필름 포장 적용에 따른 품질 변화)

  • The purpose of this study was to examine the effect of packaging on quality maintenance of paprika (Capsicum annuum L. cv. Nagano RZ) stored at three different temperatures. In Korea, paprika is stored and distributed under ambient conditions. To ensure the freshness maintenance, determining optimal storage temperature is necessary. Paprika were unpacked (control) or packed with polyamide film and stored at 5℃, 10℃ and 20℃ for 35 days. Quality characteristics such as weight loss and appearance were examined. Paprika packed with polyamide film showed less quality changes compared to unpacked paprika under all the storage temperatures. The commercial properties tended to decrease rapidly during storage at 20℃ regardless of packing. The degree of weight loss was significantly lower in packed paprika compared to unpacked paprika. It was found that soluble solids, pigments, hardness, etc. were complexly affected by storage temperature and film packaging. For paprika, the storage temperature of 5℃ or 10℃ was effective in maintaining freshness; paprika packed in polyamide film packing maintained greater freshness than unpacked paprika. Our results showed that, packaging is required to preserve the freshness and to improve the marketability of paprika in the domestic market. It seems that it is necessary to continuously search for an effective packaging method.

Comparison of the Bacterial and Fungal Colonies from Rana dybowskii which Collected from Inside and Outside Frog Farms and Identification of the Bacteria from the Tadpoles (개구리 증양식장 내·외부에서 채집된 북방산개구리(Rana dybowskii)로부터 검출된 세균과 곰팡이 콜로니 수의 비교 및 유생으로부터 확인된 세균 규명)

  • There are many ongoing studies of infectious diseases as the major factor responsible for global declining of the amphibian population. Although some point out the amphibian rearing facilities like frog farms as one of the important sources of harboring and spreading amphibian infectious pathogens in the wild, there have been few related studies in South Korea. In this study, we investigated the bacterial and fungal colonies on the skin and in the internal organs of frogs and tadpoles collected inside and outside of Dybowski's brown frog farms in Inje, Goesan, and Gongju to compare the difference according to the region and between inside and outside the farm. We also intended to classify the bacteria collected from the tadpoles into species by analyzing 16s rDNA gene sequences. The result showed that the number of bacterial colonies found in the skin and gut of frogs and the number of fungal colonies found in the skin and liver of frogs collected in Goesan was significantly greater than those in the frogs in Inje. However, there was no difference between the frogs collected inside and outside of farms in both regions. In the case of tadpoles, the number of fungal colonies in the tadpoles collected from Gongju was greater than that in the tadpoles collected from Inje. The comparison of inside and outside frog farms showed that there were more bacterial colonies on the skin of the tadpoles collected from inside than outside the frog farm in Inje and more bacterial colonies in the organs of the tadpoles collected from outside than inside the farm in Gongju. The frogs with higher condition factor (body weight/snout-vent length*100) showed fewer bacterial colonies on the skin and fewer fungal colonies in the heart, but there were no significant relationships in tadpoles. We identified the total of 15 genera and four phyla of bacteria, but the difference according to regions and between inside and outside farm was not evident. The result of this study indicates that the different conditions according to the locality of farm and between inside and outside farm cause the difference in the population sizes of bacterial and fungal colonies and that it can affect the overall health condition of Dybowski's brown frogs in the farm. Moreover, the result suggests that effective disease control in the facility is greatly necessary to ensure successful operation of amphibian rearing facility and to prevent the possible spread of diseases from the facility to the wild.
