Improved Poly-${\varepsilon}$-Lysine Biosynthesis Using Streptomyces noursei NRRL 5126 by Controlling Dissolved Oxygen During Fermentation

  • The growth kinetics of Streptomyces noursei NRRL 5126 was investigated under different aeration and agitation combinations in a 5.0 l stirred tank fermenter. Poly-${\varepsilon}$-lysine biosynthesis, cell mass formation, and glycerol utilization rates were affected markedly by both aeration and agitation. An agitation speed of 300 rpm and aeration rate at 2.0 vvm supported better yields of 1,622.81 mg/l with highest specific productivity of 15 mg/l.h. Fermentation kinetics performed under different aeration and agitation conditions showed poly- ${\varepsilon}$-lysine fermentation to be a growth-associated production. A constant DO at 40% in the growth phase and 20% in the production phase increased the poly-${\varepsilon}$-lysine yield as well as cell mass to their maximum values of 1,992.35 mg/l and 20.73 g/l, respectively. The oxygen transfer rate (OTR), oxygen utilization rate (OUR), and specific oxygen uptake rates ($qO_2$) in the fermentation broth increased in the growth phase and remained unchanged in the stationary phase.

  • One of the most important factors which affect the impact strength of high impact polystyrene (HIPS) is the rubber-phase particle size and size distribution. In this study, HIPS was prepared from a batch reactor to observe the influence of reaction conditions such as rubber content, agitation speed and prepolymerization time on the particle size and size distribution. Measurements concerning the particle size distribution were conducted using a particle size analyzer. Due to swelling, the particle suspended in toluene increases in size with lower heat-treatment temperature and shorter heat-treatment time, while the particle in methyl ethyl ketone shows quite reasonable size without any effort of heat-treatment. As rubber content increases, the average particle size increases substantially, but the increase in agitation speed at lower rubber contents does not have much influence on the size. However, the polystyrene-phase particles occluded in rubber-phase become more uniform as agitation speed increases. Longer prepolymerization time produces rubber-phase particles with narrower particle size distribution.

Scale- Up of Water-Oil Hydrolysis System

  • Scale-up experiments for hydrolysis of beef tallow, fat, and palm kernel with lipase derived from Candida cylindracea were carried out in 1-1, 100-1, and 10,000-1 reactors. The optimum agitation speed for the hydrolysis of the 1-1 reactor was investigated and found to be 350rpm, and this was a basis for the scale-up of agitation speed. The hydrolysis system in this work was the oil-water system in which the hydrolysis seems to process a heterogeneous reaction. An emulsion condition was the most important factor for determining the reaction rate of hydrolysis. Therefore, the scale-up of agitation speed was performed by using the power n = 1/3 in an equation of the rules of thumb method. The geometrical similarity for scaling-up turned out to be unsatisfactory in this study. Thus, the working volume per one agitator was used for the scale-up. In the case of scale-up from a 1-1 reactor to a 100-1 reactor, the hydrolysis of palm kernel was very much scaled-up by initiating the rules of thumb method. However, the hydrolysis of fat and beef tallow in a 100-1 reactor was a little higher than that of the 1-1 reactor because of the difference of geometrical similarity. The scale-up of hydrolysis from the 100-1 reactor to the 10,000-1 reactor was improved compared to that of the 1-1 to 100-1 reactor. The present results indicated that the scale-up of hydrolysis in the oil-water system by the rules of thumb method was more satisfactory under the condition of geometrical similarity. Even in the case where geometrical similarity was not satisfactory, the working volume per one agitator could be used for the scale-up of a heterogeneous enzyme reaction.

Optimization of Bioreactor Operation by Mass Transfer Coefficient (물질전달계수를 이용한 생물 반응기 운전 최적화)

  • The effects of various operating parameters(agitation speed, impeller type, antiform agents, impeller spacing etc.) on air-liquid mass transfer was characterized by volumetric mass transfer coefficient($k_La$). Also, the dual-impeller agitated systems are compared with single-impeller agitated systems with a special focus on its applications for bioreactors, $k_La$ was take over a range of 200~450 rpm of agitation speed, and 0.5~2.5 vvm of air flow rates, for four single impeller and impeller combinations consisting of four impeller types, namely rushton, pitched blade, scaba, intermig were tested. The rushton impeller showed the best $k_La$ as compared with other single impellers. The dual impeller system are found to be superior as compared to single impeller in all aspects, The best combination of the dual impeller was a intermig of axial flow type as an upper impeller and a rushton of radial flow type as a lower part. Also, the control of the DO level with the variation of agitation speed was more efficient than that with an increase in air flow rate. The addition of antiform dropped the $k_La$ very large up to 1g/L regardless the type. PPG was less effect on $k_La$ than other antiforms. The impeller spacing and presence of solute are found very effective on $k_La$. When the $NaNO_3$is presented as solute, the $k_La$ increased approximately 50% then control.

  • In this study, the synthesis of succinic acid from hydrogenation of maleic anhydride over Pd/C were performed in aqueous solution at various reaction conditions. We confirmed that the distribution of product was different according to process parameters. When increasing the reaction pressure and agitation speed, the by-product decreased and the purity of succinic acid increased. From the result, we may conclude that the reaction pressure and agitation speed are important factors for promoting the mass transfer rate of gas-liquid interface by increasing gas-liquid solubility in liquid hydrogenation. When the reaction pressure increased from 5 bar to 10 bar, the reaction rate increased 2.14 times. When the agitation speed increased from 300 rpm to 700 rpm, the reaction rate increased 2.75 times.

The Recovery of Silver from Thiourea Leaching Solution by Cementation Technique (침전법을 이용한 Thiourea 용출용액으로부터 Silver 회수)

  • In order to apply the silver cementation method using Fe powder from pregnant thiourea leaching solution. Parameters such as the amount of Fe powder addition, agitation speed, and temperature were investigated. The silver cementation rate was increased by the increasing of Fe powder addition, agitation speed, and temperature. The highest silver cementation rate was found when the addition of Fe powder was 50 g/L at the agitation speed of 500 rpm. The silver cementation rate increase with increasing temperature according to the Arrhenius equation and obeys $1^{st}$ order kinetics. The activation energy from the kinetics data was found to be between 13.73 KJ/mol and 17.02 KJ/mol. In the XRD analysis, goethite was detected in the precipitate of the thiourea leach solution. This indicates that an oxidation-reduction reaction had occurred in the thiourea solution due to the addition of the Fe powder.

Enhanced Production of Gellan by Sphingomonas paucibilis NK-2000 with Shifts in Agitation Speed and Aeration Rate after Glucose Feeding into the Medium (Sphingomonas paucibilis NK-2000 균주가 생산하는 젤란의 생산 농도 향상을 위한 포도당 첨가 및 교반속도와 통기량 변화 방법의 최적화)

  • Optimal agitation speed and aeration rate for the production of gellan by Sphingomnas paucibilis NK2000 in a 7 l bioreactor were found to be 400 rpm and 1.0 vvm. The best time for glucose feeding into the medium for enhanced production of gellan by S. paucibilis NK2000 was 36 hr after cultivation. The concentrations of gellan produced by S. paucibilis NK2000 from 1) 20.0 g/l glucose without additional feeding, 2) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr, in which the final concentration in the medium was 10.0 g/l, 3) 20 g/l glucose with feeding of 200.0 g/l glucose and a shift in an agitation speed from 400 to 600 rpm, 4) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 1.5 vvm, 5) and 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 2.0 vvm, were 5.19, 5.74, 6.73, 7.93, and 9.40 g/l, respectively, and their conversion rates from glucose were 26.0, 19.1, 22.4, 26.4, and 31.3%, respectively. Compared to those developed using a normal process, production of gellan by S. paucibilis NK2000 from 20.0 g/l glucose was 1.81 times higher, and and its conversion rate was 1.20 times higher when the optimized process developed in this study was used.

Agitation Effects of an Ultrasonic Standing Wave on the Dynamic Behavior of Methane/Air Premixed Flame (메탄/공기 예혼합화염의 동역학적 거동과 정상초음파의 교반)

  • This study has been conducted to scrutinize agitation effects of an ultrasonic standing wave on the dynamic behavior of methane/air premixed flame. The propagating flame was caught by high-speed Schlieren images, through which local flame velocities of the moving front were analyzed in unprecedent detail. It is revealed that the propagation velocity agitated by the ultrasonic standing wave is greater than that without agitation at the stoichiometric ratio: the velocity enhancement diminishes as the equivalence ratio approaches upper flammability limit or lower flammability limit. Also, vertical locations of the wave-affected frontal distortions do not vary appreciably, unless the propagating-mode characteristics (pressure amplitude and driving frequency) of ultrasonic standing wave were not changed.

The Determination of TRC using an Electrochemical Method (II: Pt electrode) (전기화학적 방법의 TRC(Total residual chlorine) 측정 연구(II: Pt전극 이용))

  • The conventional methods for total residual chlorine such as iodometry and DPD colorimetric can cause secondary pollution due to additional agents, also have a wide error range. As for alternative, electrochemical method can measure TRC(Total residual chlorine), and is not required as additional agents, also very suitable for using the fields of ballast water because test time is relatively fast. Therefore, this study was investigated for changing charge by agitation, salt concentration, and temperature change. Charge showed differences based on changes of reduction peak with or without agitation. In contrast, TRC and charge were well correlated in constant agitation speed. As TRC and charge were analyzed with high correlations in constant salinity and temperature of ocean, thereby conductivity was firstly measured, and charge had high correlation for TRC in spite of changing salinity and temperature Pt electrode revealed high reliability ($r^2=0.960$) because it was rarely effected by TRC, On the other hand, Au electrode appeared inadequate ($r^2=0.767$) to use sensor in less than 1.0 ppm of TRC. For high accuracy and detection of TRC, Pt and Au electrodes for test time were, respectively, 14 and 22 seconds. As a result, Pt electrode was more valuable than Au electrode in terms of response time.

The Fermentation Characteristics of Newly Selected Thermotolerant Yeasts at High Temperature

  • In order to develop a method of economical production and to reduce energy-consumption in fuel alcohol production, we investigated the fermentation characters of two newly selected thermotolerant yeasts. The RA-74-2 showed stable and superior fermentability between 30 and $40^{\circ}C$ in 20% glucose media in comparison to the industrial strains. The optimum concentration of glucose for economical fermentation at $40^{\circ}C$ was 15-18%, and organic nitrogen was necessary for a satisfactory fermentation. The optimum pH was 4.0 and aeration was adversed for high temperature fermentation. Agitation was an important factor at $40^{\circ}C$ and the addition of magnesium ion 0.2% was required in this experiment. When the inoculum was increased, ethanol productivity as well as the speed of fermentation increased. On the other hand RA-912, which can grow at $48^{\circ}C$, showed similar fermentability between 30-$45^{\circ}C$ in 20% glucose media As the concentration of substrate decreased, fermentation ratio increased at $45^{\circ}C$ (45%, 65%, 95% fermentation ratio in 20%, 15%, 10% glucose media, respectively). Also, requirement of organic nitrogen and magnesium ion in RA-912 was similar in RA-74-2. The optimum pH for fermentation was 5.0, and the effects of agitation were enhanced at $37^{\circ}C$ than at $45^{\circ}C$. As the inoculum was increased, fermentation speed became more enhanced but the ethanol productivity was less affected. RA-912 showed fermentability with various substrates. Among the substrates used, inulin was the most promising substrate for the high-temperature fermentation. When 14.5% inulin was used as the substrate, 93% and 55% fermentation ratios were shown at $37^{\circ}C$ and $45^{\circ}C$, respectively.
