Effect of Culture Conditions on Microbial Cellulose Production by Acetobacter sp. A9 in Shaking Cultures

  • Several culture conditions affecting cellulose production by a newly isolated Acetobacter sp. A9 were examined by cultivating cells under shaking cultures. The inoculum size in the range of 1-10% (v/v) did not influence cellulose production. Maximum cellulose production was obtained with 200 rpm of agitation speed. The cells grown in the 75 ml of medium in a 250-ml conical flask produced the highest level of cellulose. The strain was able to produce cellulose at 25-3$0^{\circ}C$ with a maximum at 3$0^{\circ}C$. Cellulose production occurred at pH 4.5-7.5 with a maximum at pH6.5.

  • Brewer's worts were fermented under five different conditions in each of which one of the five elemental factors involved in the conventional batch fermentation, i. e., fermentation period, heterothermal condition, spontaneous agitation, stratification, and foam covering, was forced to alter remaining other factors unchanged. The resulting beers were analyzed for their flavor components gas-chromatographycally and all of the five factors were found to be necessary for the development of the characteristic flavor of traditional beer.

Properties of Bismuthoxynitrate as a Synthetic Pearl Pigment (인공 진주 안료로서 염기성 질산비스무트에 관한 연구)

  • The purposes of this investigation were to study the characteristic properties of bismuthoxynitrate formed by hydrolysis of bismuthnitrate according to various reaction conditions and to propose the property difference between synthesized pearl pigment and bismuthsubnitrate as pharmaceuticals by means of X-ray diffraction, IR, DTA and TGA. The pearl pigment could be obtained by reaction of bismuth nitrate-diluted nitric acid solution with cold water (5.deg.) agitation at lower pH. The pearl pigment was BiO. NO$_{3}$. 2H$_{2}$O. in composition and crystalline form was thin plate of monoclinic system and its combination with water was assumed to be hydrated hydrous form between Bi-H$_{2}$O bond. On the other hand, bismuthsubnitrate was 5BiO.4NO$_{3}$.6H$_{2}$O in composition and crystalline form was pillar of rhombic system and the bond between Bi and H$_{2}$O molecules was assumed to be hydrous form. The different properties between two compounds in structure are presumed to be caused by the hydrolysing conditions of bismuthnitrate respectively.

  • In synthesizing hydrated zirconia powder by hydrolysis of Zr-alkoxides using ethanol as mutual solvent, three experimental parameters, namely, concentration of alkoxides and hydrolysis water and addition rate of hydrolysis water were varied systematically. Spherical, monodispersed, nonagglomerated and submicrometer sized powders were prepared at 0.3 M of Zr(n-OPr)4 and 0.05M of Zr(n-OBu)4 with wide ranges of hydrolysis water conditions i.e. 0.5-2.0M concentration and 1-20ml/min addition rate. During the hydrolsis, careful attention have to be paid to maintain homogeneous reaction by controlling the agitation of the reactant and the addition of the hydrolysis water. For more improved condition of monodispersity it was found that the key point is to shorten the self-nucleation time within several seconds as rapid as possible. In both alkoxides system, with higher concentration of alkoxide and hydrolysis water and with slow addition rate of hydrolysis water, hydrated zirconia powders synthesized showed tendency to fall in worse powder conditions.

  • The present paper investigated the effect of ultrasonic vibrations on the melting process of a phase-change material (PCM). Furthermore, the present study considered constant heat-flux boundary conditions unlike many of the previous researches, which had adopted constant wall-temperature conditions. Therefore in the study, modified dimensionless numbers such as Stefan and Rayleigh were adopted to represent heat transfer results. The experimental results revealed that ultrasonic vibrations accompanied the effects like agitation, acoustic streaming, cavitation, and oscillating fluid motion, accelerating the melting process as much as 2.5 times, compared with the result of natural melting (i. e., the case without ultrasonic vibration). Such effects are believed to be a prime mechanism in the overall melting process when ultrasonic vibrations were applied. Subsequently, energy could be saved by applying the ultrasonic vibrations to the natural melting In addition, various time-wise dimensionless numbers provided a conclusive evidence of the important role of the ultrasonic vibrations on the melting phenomena of the PCM.

  • The effect of AcNPV infection conditions such as serum concentration, pH, CaCl2, lysosomotropic agent, cell density at infection, agitation, aeration and nutritional supplementattion on recombinant protein production in Spodoptera frugiperda 21 cells was investigated using tissue culture flask, bottle and spinner flask. It was shown that serum, CaCl2, pH and cell density at infection affected recombinant production. The lysosomotropic agent did not significantly influence recombinant protein production.

  • This study was carried out to produce the maltooligosaccharides directly from the culture medium containing high concentration of soluble starch as carbohydrate source by Bacillus cereus IAM 1072. Optimum conditions for the production of maltopentaose and maltooligosaccharides were predicted as 10.62 and 10.92 in C/N ration, 115.74 and 116.51 rpm in agitation speed, 30.19 and 30.9$0^{\circ}C$, respectively. And at these conditions, products of maltopentaose and maltooligosaccharides were 23.23 and 50.33g/L, respectively. From the results of continuous culture for maltopentaose, the productivity increased up to 6.9 times, showing 6.6g/L/hr compared with 0.96g/L/hr batch culture. Maltopentaose showed lower sweetness at 3% concentration representing 1/5 of that sugar. Also, swelling power of maltooligosaccharides was reached to the same point with sugar after fermentation.

  • The effect of ultrasound on the acid hydrolysis of inulin was studied under significantly mild reaction conditions, at which sugar degradation products were not detected. Reaction conditions were i the range of 50~$60^{\circ}C$ and 0.1~0.3%(w/w) of HCl concentrations. The effects of reactor position inside water bath and mechanical agitation under ultrasound were investigated. The production rates of fructose with/without ultrasound irradiation were compared. The activation energies for both control and ultrasound reaction were the same, i. e., 25kca1/mo1, and ultrasound enhancement was average 22%.

  • The objective of this study was to investigate the effects of nutrients and culture conditions on morphology during the seed culture of C. acremonium ATCC 20339 Morphological factors such as hyphal length number of tips number of arthrospores were observed to investigate the relationship between seed morphology and CPC production. During the time course of seed culture, hyphal length was shortened and the number of arthrospores increased rapidly On the other hand the number of tips deceased rapidly and this was closely related to the hyphal length Mixed nitrogen sources of 3% solybean meal and 1% cotton seed flour were determined as the proper organic nitrogen sources, in terms of the morphological factors in the seed culture. This fact was proven in batch culture for the production of Cephalosporin C. It was also found that a proper agitation speed enhanced the morphological differentiation of C. acremonium ATCC 20339, thus improving the production of Cephalosporin C.

  • The graft copolymerization of methyl methacrylate(MMA) onto natural rubber latex(NRL) initiated by t-butyl hydroperoxide(t-BHPO) was investigated in aquous medium. The grafting percentage, grafting efficiency and total conversion were observed in various reaction conditions such as monomer, initiator and emulsifier concentration, reaction temperature, reaction time and agitation speed. The optimum conditions for the graft copolymerization onto natural rubber latex were as follows ; At given monomer concentration of $3{\times}10^{-2}mole/l$, the maximum grafting percentage was appeared in the case of grading in initiator concentration of $4{\times}10^{-2}mo1e/l$ and emulsifier concentration of 0.2wt.% at $40^{\circ}C$ for 5hrs.
