Effects of Temperature and Urea on in vitro Aggregation of Tryptophan Synthase $\alpha$-Subunits

  • Protein aggregation could be problematic as causes of diseases and hindrance in the production of useful recombinant proteins. Aggregation of mutant tryptophan synthase $\alpha$-subunits was examined by treatment with urea and at high temperature. Large amorphous aggregate seemed to appear by heat treatment, while more various aggregates in size were formed by treatment with urea at low concentration. The result indicates that different aggregate in size could be formed depending on the treatment condition, suggesting different mechanisms underlying aggregation processes.

  • This study shows basic data for using as the structural lightweight aggregate. This will be the procedural method of recycling environmental close waste PET bottle lightweight aggregate(PBLA) that is rapidly increased the amount of production of waste PET bottle recently, the quality of developed PBLA and the fundamental properties by analyzing of mortar containing with PBLA. After experiment, the result shows the PBLA quality that have oven dry specific gravity of 1.39, unit volume weight of 844 kg/m$^3$ and absorption rate of 0% is satisfied with qualify regulation of lightweight aggregate. The flowability of mortar containing PBLA is increased maximum 16% with increasing mixing ratio of PBLA, however the compressive strength of mortar is decreased maximum 35% with increasing mixing ratio of PBLA.

Bloating Mechanism of Lightweight Aggregate with the Size

  • The purpose of this study was to investigate the bloating mechanism of artificial lightweight aggregates with different sizes (ESA, effective surface area). Aggregates were produced using hard clay, stone sludge, and a bloating agent in order to observe the effect of the gradation of the artificial lightweight aggregates. Kerosene and amorphous carbon were used as bloating agent. The particle size of the produced aggregate ranged from 3 mm to 9 mm. With regard to the amount of bloating agent to be used, 2 ~ 6 parts by weight were used. The specific gravity, absorption rate, and the type of aggregates produced by rapid sintering at $1075{\sim}1200^{\circ}C$ were determined. Microstructures were observed. When ESA had a value of 1 or below, kerosene, which has a high burning rate, was found to be advantageous for use as a bloating agent. When ESA had a value of 1 or above, carbon, which has a relatively low burning rate was found to be an advantageous bloating agent. It is thought that kerosene is more advantageous, as ESA decreases, for the production of aggregates having low water absorption rate.

  • Technological advancements in the field of building materials are achieved day by day. In this study, a new lightweight concrete aggregate is produced by mixing certain rates of colemanite (0%, 7.5%, 12.5%, 17.5%), cement and coating the surface of pumice aggregate with this mixture. Thin aggregate sections are analyzed with specific gravity, unit weight, water absorption, impact, and crushing experiments. In this way, the production of cement and cement+colemanite coated lightweight concrete aggregates is investigated and an opinion on the likely behavior of these concrete types is provided.

  • This study was performed in order to obtain the effect of the compressive strength of the cured product with manufacturing conditions (amounts of fine aggregate and different types of alkali activator). Material which is the basis of the cured product was used for the blast furnace slag, which has a latent hydraulic activity. Consequently, when using sodium hydroxide as the alkali activator, it is possible to obtain a higher compressive strength than using the calcium hydroxide. And also, it can be added a 10% of fine aggregate with blast furnace slag to improve the compressive strength.

Effects of Nutrients and Cell Aggregate Size on the Biosynthesis of Carotenoid in Daucus carota Suspension Culture (Daucus carota 현탁 배양에서 배지 조성 및 세포 응집 크기가 Carotenoid 생합성에 미치는 영향)

  • The effects of nutrients and cell aggregate size on the cell growth of D. carota and the biosynthesi of carotenoid were investigated. Highest carotenoid content was obtained with sucrose as a carbon source and the equal ratio of ammonium to nitrate. High phosphate concentration stimulated the carotenoid biosynthesis in D. corota. 2,4-D inhibited the cell growth but stimulated the specific carotenoid content at high concentration. By modifying the medium composition based on these findings, three times higher specific carotenoid content and 2.5 times higher total carotenoid content were obtained as compared with the results obtained with basic MS media. Biosynthesis of carotenoid was found to be affected by cell aggregate size; high carotenoid production was obtained from the large aggregated cells resulted from high sucrose concentration.

Bloating Mechanism for Artificial Light Weight Aggregate of Surface Modification with Coal ash

  • We manufacture artificial lightweight aggregates (ALWAs) using bottom-ash as the primary raw material. We coat the ALWA surfaces with low-melting point materials in order to enable them to bloat, which is essential to reduce the bulk density of the aggregate. Then, we sinter the prepared aggregates at 1000, 1100, and $1200^{\circ}C$ using either the direct or two-step firing schedules. Finally, we evaluate the properties of the fired samples through analyzing their bulk density, water absorption, and microstructure. The surface-modified samples result in a reduction of their bulk density by $0.3{\sim}0.4g/cm^3$ regardless of the firing method used. Based on these results, we conclude that this approach could provide a viable method for the mass-production of ALWAs from industrial waste such as bottom-ash.

  • In this study, earlier analyzed tetrapod, one of an original group of offshore constructions studied for manufacturability of the concrete using the properties of steel making slag aggregate. steel making slag aggregate assessment, RCS and Blast Furnace Slag : the 20 mm air-cooled slag and combinations by 50 %, aggregate properties on the most appropriate for the properties of recycled aggregate concrete optimal mix, and assessing it. Properties of concrete used to be derived are judged as to bury the studies show that the hollowing-out of the RCS, plastic sole use is in the workability of the aggregate, plastic in the 20 mm slag also assessed to be a slight disadvantage, but RCS by mixing air-cooled coarse and 50 percent to 20 percent 50 mm. Thus, steel making slag marine structures using recycled aggregate, in rapid chilled slag or air-cooled slag. The sole use of the aggregate them than to combine the aggregate of concrete. After they satisfy the quality standard quality shall be used will aggregate steel making slag who meet the criteria concrete manufacturing in general or par with the aggregate of concrete. Performance was assessed as to develop a more than that.

Effects of Aggregate Grading on the Performance of High-Flowing Concrete with General Strength (일반 강도용 고유동 콘크리트에서의 골재 입도 영향)

  • The high-flowing concrete requires additionally or excessively more expensive admixture than conventional concrete. So, the concrete has not to be widely used in practical field due to the increase of production price, need of additional facilities, and excessive development of concrete strength in associate with addition of too much cementitious material even though it has more significant advantages than conventional concrete. Thus, this study aims at developing high-flowing concrete with general strength unlike high strength which has been carried out in conventional study. To observe the role of aggregate in the concrete quantitatively and to increase the performance of high-flowing concrete effectively, parametric studies were carried out such as W/C, s/a, fineness modulus of aggregate, contribution degree of particle sizes, and the effect of 13mm aggregate and fine stone powder as a partial replacement of aggregates. And the effect of these factors on performance of the concrete was evaluated by measuring slump-flow and gap of penetration height in U-typed instrument. As a result, it was found that flowability of high-flowing concrete depends upon grading of fine aggregate more significantly than that of coarse aggregate and is enhanced greatly as fineness modulus of fine aggregate decreases and the value of s/a increases. In addition, the application of 13mm aggregate and fine stone powder are expected as a partial replacement of aggregate in order to increase the performance of high-flowing concrete more effectively.

  • Recently, the study is progressing actively about manufacture skill of concrete for promoted recycled aggregate and concrete made into recycled aggregate in the construction production field. But, application and study about recycled fine aggregate insufficient compared to recycled coarse aggregate. So, in this study, it presents basic data for development of environmental load reduction fiber reinforcement recycled fine aggregate concrete by comparison and investigation about engineering properties and shrinkage cracking of fiber reinforcement recycled find aggregate concrete for increasing shrinkage cracking reduction and long term stability of environmental load reduction concrete used recycled fine aggregate. In the result of the study, compared to natural fine aggregate, a crack-extent increased by applying recycled fine aggregate, moreover, as a water cement ratio increased, the crack size increased, as well. In addition, it's shown that the specimen mixed with PVA and Nylon, among all kinds of fibers, showed the smallest crack size, so it's verified that the mix of fiber had an effect on decreasing crack-extent.
