Antimicrobial Activities of Scutellariae Radix Extract against Vibrio parahaemolyticus (Vibrio parahaemolyticus에 대한 황금추출물의 항균활성)

  • To investigate the antimicrobial effects of Scutellariae Radix extract against Vibrio parahaemolyticus from food samples, Vibrio parahaemolyticus strains isolated from Tapes philippinarum were examined for their sensitivity to Scutellariae Radix extract. Total 66 Vibrio parahaemolyticus strains were isolated from Tapes philippinarm 72 samples (91.7%). The serotypes of isolated Vibrio parahemolytics were K-I group 7 strains (10.6%), K-IV group 5 strains (7.6%), K-II group 2 strains (3.0%), K-V group 2 strains (3.0%), K-VII group 2 strains (3.0%), K-VI group 1 strains (1.5%), K-VIII group 1 strains (1.5%) and antisera UT K-group 46 strains (69.7%) on antisera agglutination test, but K-III group and K-IX group strains were not found. The growth curves of isolates showed lag phase, logarithmic phase, stationary phase and death phase as typical sigmoid curve on the shellfish samples. After 6 hours, the group containing Scutellariae Radix extract differs from the control on shellfish samples in the growth inhibition curves, and Vibrio parahaemolyticus were inhibited in more than 1000 ppm Scutellariae Radix extract. The morphological changes were observed by transmission electron microscope and the microbial cells membrane was destroyed by Scutellariae Radix extract.

  • Purpose: The antimicrobial resistance of S. pneumoniae has encountered with increasing frequency from around the world. In our country, penicillin resistant strains of S. penumococci are rapidly increasing. It has been known that colonized pneumococci in upper respiratory tract cause sinisitis, otitis media, meningitis and pneumonia. We tried to reveal the colonization rate of pneumonocci in upper respiratory tract, their antimicrobial resistance and DNA fingerprinting pattern in normal children. Methods: We got specimens from 117 children of day-care center in Seoul through oropharyngeal swab. After incubation on BAP, optochin test and slide latex agglutination test were used for identification. Antimicobial susceptibility test to penicillin, vancomycin, erythromycin and TMP-SMZ was done with disk diffusion method. Penicillin MIC was gotten through the broth microdilution method. Genotyping of 45 pneumococci was done by rep-PCR using REP1R-Dt and REP2-Dt primer. Results: The carriage rate of pneumococci in the day-care center children was 38%(45/117). The resistance of penicillin, erhthromycin, TMP/SMZ, vancomycin by the disk diffusion method are 89%, 91%, 64% and 0%, respectively. 64% of the isolates showed multiple resistance. 7 types of DNA fingerprinting were gotten and 78% of isolates belonged to three types. Conclusion: We found that the antimicrobial resistance of children attending the day-care center in Seoul was much higher than expected. We assumed that this might be due to their easy and frequent exposure to antimicrobial agents and crowded day-care center environment.

  • To investigate the antimicrobial effects of Scutellariae Radix extract against L.monocytogenes from foods, L. monocytogenes strains isolated from livestock, processed food from meat and milk, and frozen foods, were examined for their sensitivity to Scutellariae Radix extract. 30 L. monocytogenes strains were isolated from total 178 samples(16.9%); 13(14.0%) strains from beef 6(20.7%) strains from pork, 9(39.2%) strains from chicken and 2 (16.7%) strains from frozen foods but was not found from processed products, The serotypes of isolated L.monocytogenes were serotype O-1 strains (23, 76.7%) and serotype O-4 strains(7, 23.3%) on antisera agglutination test. The growth curves of isolates were shown lag phase, logarithmic phase, stationary phase and death phase as typical sigmoid curve on the preservative-free hams. After 6 hours. Scutellariae Radix extract contain group differ from control group on preservative-free ham samples, and the isolates were inhibited in more than 1000 ppm Scutellariae Radix extract on the inhibitory growth curve of L.monocytogenes. The mor-phological changes were observed by transmission electron microscope and the microbial cells membrane was destroyed by Scutellariae Radix extract.

  • This study was conducted to select the media for the formalin killed vaccine (FKC) production of Vibrio harveyi and its application for olive flounder, Paralichthys olivaceus. For this, we have investigated the immune effects of Vibrio harveyi FKC vaccines grown in 3 different media i.e. Tryptic Soy Broth (TSB), TSB containing 2-2'-dipyridyl (TSB-D), Brain Heart Infusion Broth (BHIB) on the production of agglutinating antibody and protection against Vibrio harveyi in olive flounder. Additionally, a dual vaccine was prepared by combining Streptococcus parauberis vaccine to V. harveyi vaccine and its efficacy was also analyzed with the determination of optimal administration dosage. Consequently, olive flounder immunized with FKC grown in TSB-D showed the same protection with the vaccine grown in BHIB and the optimal dose of the vaccine was 10mg/kg of body weight. Indeed the dual vaccine showed higher agglutination titer and protection than control fish. The optimal dose for dual vaccine was 10mg/kg body weight for each of two vaccines.

Case of Acute Hemolytic Transfusion Reaction due to Anti-Fya Alloantibody in a Patient with Autoimmune Hemolytic Anemia (자가면역용혈환자에서 항-Fya 동종항체에 의한 급성용혈성수혈반응 1예)

  • A 72-year-old man with general weakness visited the outpatient clinic of the hematology department. The patient had been treated under the diagnosis of autoimmune hemolytic anemia for 2 years. His hemoglobin level at the time of the visit was 6.3 g/dL, and a blood transfusion was requested to treat his anemia. The patient's blood type was A, RhD positive. Antibody screening and identification test showed agglutination in all reagent cells with a positive reaction to autologous red blood cells (RBCs). He had a prior transfusion history with three least incompatible RBCs. The patient returned home after receiving one unit of leukoreduced filtered RBC, which was the least incompatible blood in the crossmatching test. After approximately five hours, however, fever, chills, dyspnea, abdominal pain, and hematuria appeared and the patient returned to the emergency room next day after the transfusion. The $anti-Fy^a$ antibody, which was masked by the autoantibody, was identified after autoadsorption using polyethylene glycol. He was diagnosed with an acute hemolytic transfusion reaction due to $anti-Fy^a$ that had not been detected before the transfusion. In this setting, it is necessary to consider the identification of coexisting alloantibodies in patients with autoantibodies and to become more familiar with the method of autoantibody adsorption.

Evaluation for the Usefulness of Automated Blood Typing Analyzer (혈액은행 자동화 장비 도입의 유용성 평가)

  • In this study, we evaluated the usefulness of an automatic blood typing analyzer using QWALYS-2 up (Diagast, Loos Cedex, France). During a month( 01OCT2013 - 31OCT2013) we performed 1,636 tests for ABO & RhD blood typing, 1,374 tests for antibody screen & identification tests and compared the results by automatic blood type analyzer with previous manual methods and column agglutination tests. And we analyzed the economic performance by comparison the test unit price between automatic blood type analyzer and manual methods. In ABO & RhD blood typing tests, there were complete concordances between manual and automated blood typing analyzer for 200 clinical samples. In Antibody screen tests, the concordance rate between manual and automated blood typing analyzer was 98.5% and more strong reaction in automated blood typing analyzer than manual methods. Therefore, the introduction of an automated blood typing analyzer, reagents costs were increased but labor costs were decreased. Considering the importance of transfusion safety and economic advantages, the introduction of an automated blood typing analyzer was very useful.

Risk Factors of Nosocomial Rotavirus Infections in Children (소아 로타바이러스 원내 감염의 위험 인자)

  • Purpose :Rotavirus is one of the most important causes of nosocomial infections among children. The aim of this study is to determine the risk of nosocomial rotavirus infections and to evaluate the effectiveness of breast-feeding and probiotics in the prevention of nosocomial rotavirus infections. Methods : This study was carried out on admitted children without diarrhea between March 1, 2003 and February 29, 2004. Three hundred ninety patients aged 4 days to 13 years during this study were available. We examined the feces of all children for rotavirus by latex agglutination on admission, during hospital, and after discharge, to see whether they developed diarrhea or not. Results : Nosocomial rotavirus infections was significantly increased with children under 12 months of age(P=0.008). The monthly attack rate was great between December and March(P=0.046). Prolonged hospital stay was associated with an increased attack rate of nosocomial rotavirus infections (P=0.003). The risk of nosocomial rotavirus infections was not associated with the number of roommates and whether or not they were breast-fed or fed on probiotics. Conclusion : Nosocomial rotavirus infections are significantly more likely to occur in children under 12 months of age, admitted between December and March, and with prolonged hospital stays. Prompt identification and isolation of children with nosocomial rotavirus infections, even without diarrhea, may decrease rates of nosocomial rotavirus infections.

Comparison of Resistance to Fowl Typhoid among Crossbreed (산란계 합성종의 가금티푸스 저항성 비교 연구)

  • The present study was conducted to compare the disease resistance to fowl typhoid among White Leghorn (WL) known as possessing greater genetic resistance to Salmonella, Rhode Island Red (RIR) susceptible to the disease and their mating crossbreed, WL ${\times}$RIR. Also, it was carried out to compare fowl typhoid-resistance among Hyline white${\times}$Hyline brown(HwHb), Hw${\times}$lsa brown(Hwlb), Isa white${\times}$lb(Iwlb), Iw${\times}$ Hb(IwHb) and Ib ${\times}$ Iw(IbIw) in order to select crossbreed chickens which have disease resistance to fowl typhoid and simultaneously produce the brown eggs which our domestic consummers prefer There was no death of WL chickens challenged with Sal. gallinarum at each 2, 5 and 8 wk of age, while average 86.2 to 64.1% of RIR chickens tested died at the same challenge age during the test period of 2 weeks. Their maiting crossbreed, WL ${\times}$ RIR were intermediate between the two breeds. weight gains of tested chickens were significantly higher in WL and WL ${\times}$RIR without difference between them than RIR. In recovery rate of tested strain of Sal. gallinarum from braun, heart, fiver, spleen and cecum of chickens challenged, WL ${\times}$RIR fell between the other two breeds, with somewhat higher rate then WL but much lower then RIR. In the disease resistance determination test using HwHb, Hwlb, Iwlb, IwHb and IbIw$.$ crossbreed chickens experimentally infected with Sal. gallinarum, it was recognized that all of crossbreeds tested developed marked resistance to fowl typhoid, based on the results judged by the measurement parameters of mortality, weight gain, recovery rate of challenged strain of bacteria, and positive rate of agglutination antibody, and HwHb had a tittle higher disease resistance then ethers.

Sexual Reproduction in Unicellular Green Alga Chlamydomonas (수염녹두말속(Chlamydomonas) 단세포 녹조의 유성생식)

  • The sexual reproduction of the unicellular green alga Chlamydomonas is reviewed for a comprehensive understanding of the complex processes. The sexual life cycle of C. reinhardtii is distinguished into five main stages: gametogenesis, gamete activation, cell fusion, zygote maturation, and meiosis and germination. Gametogenesis is induced by nitrogen starvation in the environment. C. reinhardtii has two mating types: mating type plus ($mt^+$) and mating type minus ($mt^-$), controlled by a single complex mating type locus ($MT^+$ or $MT^-$) on linkage group VI. In the early gametogenesis agglutinins are synthesized. The $mt^+$ and $mt^-$ agglutinins are encoded by the autosomal genes SAG1 (Sexual AGglutination1) and SAD1 (Sexual ADhesion1), respectively. The agglutinins are responsible for the flagellar adhesion of the two mating type of gametes. The flagellar adhesion initiates a cAMP mediated signal transduction pathways and activates the flagellar tips. In response to the cAMP signal, mating structures between two flagella are activated. The $mt^+$ and $mt^-$ gamete-specific fusion proteins, Fus1 and Hap2/Gcs1, are present on the plasma membrane of the two mating structures. Contact of the two mating structures leads to develop a fertilization tubule forming a cytoplasmic bridge between the two gametes. Upon fusion of nuclei and chloroplasts of $mt^+$ and $mt^-$ cells, the zygotes become zygospores. It is notable that the young zygote shows uniparental inheritance of chloroplast DNA from the $mt^+$ parent and mitochondrial DNA from the $mt^-$ parent. Under the favorable conditions, the zygospores divide meiotically and germinate and then new haploid progenies, vegetative cells, are released.

Epidemiologic study of rotaviral gastroenteritis in Daejeon, Korea, 2001-2005 (대전 지역에서 로타바이러스 위장관염의 역학적 연구(2001-2005년))

  • Purpose : We evaluated the epidemiologic characteristics of rotavirus gastroenteritis in Daejeon, Korea from January 2001 through December 2005. Methods : We retrospectively analyzed 540 medical records of children with gastroenteritis, who had been hospitalized at The Catholic University of Korea, Daejeon St. Mary's Hospital and were tested positive by rotavirus stool examinations (latex agglutination or immunechromatography). Results : The mean age was $21.8{\pm}15.3$ months (ranged from 2 months to 9 years of age) and the distribution of children were 144 cases (26.7%) in <1 year of age, 228 (42.2%) in 1 year of age, and 93 (17.2%) in 2 years of age, and the 505 children (93.5%) were less than 4 years of age. The male-to-female ratio was 1.4:1. The mean annual number of cases was 108, with a relative even occurrence in each year. Although the patients with rotaviral gastroenteritis occurred through the year, a higher rate of occurrence was seen in spring (March to May, 40.8%) and winter season (December to February, 37.9%), without significant differences in each year. Conclusion : In Daejeon, Korea, rotaviral gastroenteritis have occurred in children less than 4 years of age, with slight annual variations and similar seasonal occurrence in the recent years. The data indicate that the recent trend in peak rotavirus activity is different from that observed decades ago in Korea.
