Early Germination Response of Soybean Seed to Accelerated Aging and Low Dose Gamma Irradiation

  • The responses of soybean seeds were evaluated to accelerated aging and gamma irradiation with regard to germination, seed leakage, seed leachate component and dry weight of hypocotyl and primary root of the germinating seed. Accelerated aging significantly reduced the final germination rate while gamma irradiation increased the final germination rate. Furthermore, the interactive effects occurred that the final germination rate of 5-day aged seeds increased considerably in response to 4 Gy of gamma irradiation. The extent to which the electrolyte was leaked from the seeds (conductivity) was significantly affected by accelerated aging and showed a close negative correlation with the germination rate. Gamma irradiation, however, did not significantly affect the electrical conductivity of seed leachate. The accelerated aging significantly increased the concentrations of the particular electrolytes leaked from the seeds while the gamma irradiation did not affect those concentrations. Of the electrolytes leaked from the seeds, Ca and Mg showed relatively lower concentrations while K showed greater concentrations than others. Moreover, N and P showed similar responses to aging treatment. Aging treatment significantly affected dry weight (DW) of hypocotyls and primary root. Also, gamma irradiation decreased DW of hypocotyls and primary root, particularly for 8 Gy associated with 5 days aging treatment. The data were discussed in terms of the relationships of seed vigor with aging treatment and gamma irradiation.

The Anticancer Properties of Lunasin Peptide from Aged Callus Induced by the Soybean Tissue Culture

  • Lunasin is small subunit peptide of coded from Gm2S-1 gene in soybean. It has been previously demonstrated that lunasin is a novel and promising cancer preventive peptide. Lunasin peptide is found only in the seed and not other tissues. And lunasin peptide starts to appear at 5 weeks after flowering and remains in the mature seed. We report here firstly lunasin peptide identified from soybean callus induced by the tissue culture and demonstrate its anticancer properties. The lunasin was identified and purified from soybean callus aged for 6 months. The callus lunasin($1{\mu}M$) inhibited the acetylation of histone H3 and H4 by 58.8% and 56.5%, respectively. And it fully inhibited foci formation compared to the values of the positive control(no lunasin) and negative control(no MCA). Purified lunasin was able to internalize into the cell and localized in the nucleus.

Development of Non-Destructive Sorting Technique for Viability of Watermelon Seed by Using Hyperspectral Image Processing (초분광 영상기술을 이용한 수박종자 발아여부 비파괴 선별기술 개발)

  • Seed viability is one of the most important parameters that is directly related with seed germination performance and seedling emergence. In this study, a hyperspectral imaging (HSI) system having a range of 1000-2500 nm was used to classify viable watermelon seeds from nonviable seeds. In order to obtain nonviable watermelon seeds, a total of 96 seeds were artificially aged by immersing the seeds in hot water ($25^{\circ}C$) for 15 days. Further, hyperspectral images for 192 seeds (96 normal and 96 aged) were acquired using the developed HSI system. A germination test was performed for all the 192 seeds in order to confirm their viability. Spectral data from the hyperspectral images of the seeds were extracted by selecting pixels from the region of interest. Each seed spectrum was averaged and preprocessed to develop a classification model of partial least square discriminant analysis (PLS-DA). The developed PLS-DA model showed a classification accuracy of 94.7% for the calibration set, and 84.2% for the validation set. The results demonstrate that the proposed technique can classify viable and nonviable watermelon seeds with a reasonable accuracy, and can be further converted into an online sorting system for rapid and nondestructive classification of watermelon seeds with regard to viability.

Nondestructive Vigor Test of Seed (비파괴적 방법에 의한 종자의 활력 검정)

  • Experiments were conducted to obtain information on separation of nonviable seeds from seed lots of rape, Chinese cabbage and radish, by nondestrutive ways. Seeds were artificially aged at 90% relative humidity(RH) and 45$^{\circ}C$ to get different seed qualities. Large amounts of amino acids, proteins and sinapine were leaked from the dead seeds into water in the course of 4~8 hours soaking, while high quality seeds did not leak in all crops. Percentage of normal seedlings from nonfluorescent cellulose coated seeds of rape, Chinese cabbage and radish under ultraviolet light was 96, 96 and 74%, while that of fluorescent seeds was 8, 9 and 1%, respectively. Cellulose coating and storage of Chinese cabbage and rape seeds at 60 and 75% RH for 3 months and 90% RH for 2 months did not deteriorate seed quality. But percentage of normal seedlings from nonfluorescent radish seeds stored at 60, 75 and 90% RHs for 3 months was 63, 64 and 2%, respectively.

Comparison of Indigenous Browses and Sunflower Seed Cake Supplementation on Intake and Growth Performance of Dual-purpose Goats Fed Buffel Grass (Cenchrus ciliaris) Hay

  • A study to compare the effects of supplementing Delonix elata, Grewia similis, Tamarindus indica and sunflower seed cake on intake and growth rate of dual-purpose goats fed low quality Buffel grass (Cenchrus ciliaris) hay was carried out. Twenty-eight male goats aged five to seven months (mean weight 12.93${\pm}$3.94 kg) were randomly allocated to four dietary groups in a completely randomised design. The diets were hay plus Grewia similis, hay plus Delonix elata, hay plus Tamarindus indica and hay plus sunflower seed cake. All diets were supplemented with maize bran. The experimental period was 90 days. Voluntary dry matter intake of the supplements was higher for Tamarindus indica (275.5 g/day) and Grewia similis (201.8 g/day) and lowest for sunflower seed cake (81g/day). Goats supplemented with Grewia similis had the highest hay intake (183.8 g/day) while those supplemented with sunflower seed cake had the lowest hay intake (98.9 g/day). Animals fed browse supplements gained significantly more weight (p<0.001) than those with sunflower seed cake. There were no significant differences in live weight change between goats fed the different browses. However, those fed Tamarindus indica gained an average of 20.79 g/d which was slightly higher than the gains for those on Grewia similis and Delonix elata while those fed sunflower seed cake lost weight. Correspondingly, goats supplemented with browse leaf meals had higher feed conversion ratios than those supplemented with sunflower seed cake and required 23.91 to 35.06 g DM of feed to produce one g of weight gain per day. In a separate study, the DM disappearance pattern indicated that Grewia similis and Delonix elata were highly degradable compared to Tamarindus indica. At 24 h of incubation, DM degradability was 627, 588 and 345 g/kg DM for Grewia similis, Delonix elata and Tamarindus indica, respectively. In another study in vivo DM digestibility ranged from 46.1% (for hay alone) to 56.2% (for hay plus Grewia similis). It was concluded that the addition of Tamarindus indica, Grewia similis and Delonix elata leaf meals to Cenchrus ciliaris hay resulted in increased total DM intake, in vivo digestibility and growth rate. Therefore, leaf meals of indigenous browses particularly Tamarindus indica and Grewia similis could be used as supplementary feeds for small ruminants grazing on poor quality roughages during the dry season rather than use of expensive, less effective and intermittently available sunflower seed cake.

Effect of Garlic on the Quality of Barley Kochuzang Brewed with Whole Red Pepper (통고추를 이용한 보리고추장 양조시 마늘이 품질에 미치는 영향)

  • This study was to improve the quality of Kochuzang and utilize red pepper seed. Kochuzang were prepared with the addition of red pepper seed and garlic (2%), compared the changes in the various chemical components and enzyme activity during the aging period of Kochuzang, and also organoleptic values of the products. Enzyme activities of liquefying and saccharogenic amylase, protease and lipase were increased by addition of garlic pulp and the suvival activities of enzyme except liquefying amylase were lasted high the late period of aging. Also the addition of red pepper seed was effective in maintaining the enzyme activities Change of titration acidity and pH of kochuzang were little when red pepper seed was added, but in case of a garlic additive it showed no difference at the late period of aging. Total nitrogen and amino nitrogen were increased by the addition of red pepper seed or garlic until the late period of aging, and ammonia nitrogen also increased during the middle period of againg, but showed no difference at the late period. Alcohol content was decreased by the addition of garlic or red pepper seed. but crude lipid was increased by the audition of red pepper seed. but crude lipid was increased by the addition of red pepper seed. Generally, taste, flavor and color of garlic added group were superior to the non-garlic added group for the products which aged for 10 weeks. Therefore, The quality of Barley Kochuazng may be improved by adding 2% garlic to the whole red pepper.

Viability Determination of Pinus rigida Seeds Using Artificially Accelerated Aging (노화처리를 이용한 리기다소나무 종자의 활력 평가)

  • We tested the seed viability of Pinus rigida using accelerated aging to discover optimum times and temperatures far artificially accelerated aging. Seeds were artificially aged at different temperatures and during different tines. The seed viability was affected by the accelerated aging and by temperature with a decline in germination and seed vigor. The aging index of P. rigida seed was 0.31 at $35^{\circ}C$ and seed viability was nearly lost after aging treatment at $40^{\circ}C$ for 15 days. The optimum temperature of P. rigida far the aging test was decided to be approximately $37^{\circ}C$ on the basis of the aging index. Inorganic materials and conductivity of leaching solution from aging seeds increased with the increase of aging period. The accelerated aging test was considered to be a suitable method to evaluate the seed viability of tree species. Because seed characters are much different among tree species, however, more studies need to be done to discover the optimum conditions for aging by tree species.

Growth and Spatial Distribution Characteristics of Rhus javanica Populations Sowed on Cut-Slopes - Focusing on the Dae-Ji Mountain Case Study - (비탈면에 파종된 붉나무(Rhus javanica) 개체군의 생육 및 공간분포 특성 - 용인 대지산 사례지역을 중심으로 -)

  • This study was to evaluate the growth and spatial distribution characteristics of Rhus javanica population. The study was conducted between 2015 January and July at the cut-slope revegetation site in Dae-Ji Mountain. Seeds of native species were sowed on the disturbed cut-slope in 2002. There were 119 vascular plant species 55 families and 94 genera on the cut-slope. Compositae, Rosaceae, and Leguminosae plants were dominant species, which are pioneer species in dry and barren site. Canopy of trees covered 44.1% total area, which was 8.3% increase from 2008. Rhus javanica, main species, dominated all DBH classes except DBH >24cm. Albizzia julibrissin showed high frequency in the 6~17cm DBH class and Salix koreensis was abundant at >12cm. Elaeagnus umbellata, Corylus heterophylla var. heterophylla, Alnus sibirica, and Acer pictum were not observed. Rhus javanica population was the most frequently observed on the south facing cut-slope. The average DBH of Rhus javanica was $7.3({\pm}3.7)cm$ and the average height of them was $5.2({\pm}1.7)m$. Annual average DBH growth was 7.9mm/yr and the maximum growth(12.0mm/yr) was the $3^{rd}$ year. It decreased after $6^{th}$ years. Although Rhus javanica population in the Dae-Ji Mountain was a seed originated plantation, the forest had reverse J shape diameter frequency distribution. It indicated that the forest is uneven-aged forest. The Rhus javanica population was expected to sustain.

Effects of Dietary Safflower Seed Powder and Chitosan on Hatching Egg Production and Eggshell Quality in Aged Egg-type Breeder Hens (홍화씨 분말 및 키토산의 사료 내 첨가가 종란 생산성 및 후기 난각질에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary safflower seed (SFS) powder and chitosan on hatching egg production and eggshell qualify in aged egg-type breeder hens. A total of four hundred 54-week-old Hy-Line Brown breeder hens were divided into five the groups and fed experimental diets either with addition of 0.25 and 0.50% SFS powder or 0.10 and 0.20% chitosan or devoid of all for 6 weeks. There were no significant differences in feed intake and laying performances among the groups. The rate of sellable egg in the groups fed diets containing SFS powder or chitosan was significantly increased compared to that of control (P<0.05), irrespective of dose-dependent manner. The addition of SFS powder or chitosan significantly improved the eggshell strength (P<0.01). Eggshell thickness was also significantly increased in the groups 134 diets containing chitosan as compared with that of control (P<0.01). Fertility and hatchability of egg set were significantly improved by dietary chitosan that those of control (P<0.05). ike contents of tibial Ca and P were significantly increased in the groups fed diets containing 0.5% SFS powder, 0.1 and 0.2% chitosan as compared with those of control. The levels of blood estrogen, calcitonin and parathyroid hormone were not affected by the dietary treatments. The overall results indicated that the proper use of some feed additives such as safflower seed powder and chitosan might provide means of improving eggshell quality and reproductive performances in aged egg-type breeder hens.

Leakage of Seed Reserve Nutrient in Artificially Aged Pepper Seeds and Enhancement of Seed Vigor by Priming (노화처리된 고추종자의 저장성분 누출과 priming 처리가 종자활력 증진에 미치는 영향)

  • Quantity of protein, amino acid, and sugar leaked from seeds was greater as the viability of seeds was dropped by the time elapsed of seed aging treatment. In the seeds with the artificial aging treatment for 20 days, 35.8 mg of protein was leaked on the 4th day after soaking, which was 6.9 times higher than that of control. Leakage of amino acid was also higher from low quality seeds treated with the aging treatment. In the seeds with the aging treatment for 20 days, $36.5{\mu}g$ fig of sugar was leaked on the 4th day after soaking, which was 2.8 times higher than that of control. The leakage of inorganic compound was higher from the low quality seeds, and leakage of total protein, amino acid, and sugar. According to .the quantity of leakage, water soluble compounds, which can be used for the assessment of seed quality without any destruction, were protein and potassium. Germination rate and percentage of seeds were dropped with the seed aging treatment, and the seed viability could be recovered by priming treatment. This phenomenon was very clear when the low quality seeds were germinated at low temperature.
