BACKGROUD/OBEJECTIVES: The mechanism of how black garlic effects lipid metabolism remains unsolved. Therefore, the objectives of this study were to determine the effects of black garlic on lipid profiles and the expression of related genes in rats fed a high fat diet. MATERIALS/METHODS: Thirty-two male Sqrague-Dawley rats aged 4 weeks were randomly divided into four groups (n=8) and fed the following diets for 5 weeks: normal food diet, (NF); a high-fat diet (HF); and a high-fat diet + 0.5% or 1.5% black garlic extract (HFBG0.5 or HFBG1.5). Body weights and blood biochemical parameters, including lipid profiles, and expressions of genes related to lipid metabolism were determined. RESULTS: Significant differences were observed in the final weights between the HFBG1.5 and HF groups. All blood biochemical parameters measured in the HFBG1.5 group showed significantly lower values than those in the HF group. Significant improvements of the plasama lipid profiles as well as fecal excretions of total lipids and triglyceride (TG) were also observed in the HFBG1.5 group, when compared to the HF diet group. There were significant differences in the levels of mRNA of sterol regulatory element binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and glucose-6-phosphate dehydrogenase (G6PDH) in the HFBG1.5 group compared to the HF group. In addition, the hepatic expression of (HMG-CoA) reductase and Acyl-CoA cholesterol acyltransferase (ACAT) mRNA was also significantly lower than the HF group. CONCLUSIONS: Consumption of black garlic extract lowers SREBP-1C mRNA expression, which causes downregulation of lipid and cholestrol metahbolism. As a result, the blood levels of total lipids, TG, and cholesterol were decreased.
Recently, we have reported that the environmental pollutant 2-bromopropane (2-BP) induces a significant embryo-fetal developmental toxicity in rats. However, the cause of developmental toxicity and the relationship between maternal and developmental toxicities could not be elucidated because the developmental toxicity of 2-BP was observed only in the presence of maternal toxicity The in vitro teratogenicity study using whole embryo culture was carried out to understand the teratogenic properties and the possible mechanism of teratogenicity induced by 2-BP in rats. Rat embryos aged 9.5 days were cultured in vitro for 48 hrs at medium concentrations of 0, 1, 3, or 10 mg/ml of 2-BP. Embryos were evaluated for growth, differentiation, and morphological alterations at the end of the culture period. At 10 mg/ml, 2-BP caused a delay in the growth and differentiation of embryos and an increase in the incidence of morphological alterations, including altered yolk sac circulation, abnormal axial rotation, craniofacial hypoplasia, open neuropore, absent optic vesicle and kinked somites. At 3 mg/ml, only a delay in the growth and differentiation of embryos was observed. There were no adverse effects on embryonic growth and development at the concentration of 1 mg/ml. The results showed that the exposure of 2-BP to rat embryos results in a developmental delay and morphological alterations at dose levels of 3 mg/ml culture media or higher and that 2-BP can induce a direct developmental toxicity in rat embryos.
This study was done to investigate the effect of age and dietary linolenic acid content and the linolenic acid/linoleic acid(LNA/LA) ratio on the lipid metabolism and formation of PGI2 and TXA2. The male Sprague-Dawley rats were fed 6 different with 0.2, 0.4, 0.6 of LNA/LA ratio within either 8% LNA(high LNA) or 4% LNA(low LNA) of fatty acid content for different feeding period(1, 4, 12 month). The dietary fat used were sesame oil, perilla oil, soybean oil and beef tallow. The concentration of serum total lipid, total cholesterol and HDL-C were increased with aging. Triglyceride concentration was decreased in 0.2 ratio of LNA/LA. The lipid content of liver showed similar tendency to that of serum. The ratio of PGI2/TXA2 was increased in 1 month rats and antithrombotic effect was reduced significantly with increasing age. The PGI2/TXA2 ratio was tended to be higher in diet of 0.2 and 0.4 LNA/LA ratio at high LNA level and in diet of 0.6 LNA/LA ratio at low LNA level. Especially PGI2/TXA2 ratio was increased linearly with rising LNA/LA ratio at low LNA level. It seemed that the LNA content and LNA/LA ratio had interaction to increase the antithrombotic effect bychanging TXA2 synthesis. And the dietary fatty acid related effect lowering the serum and liver lipid content, excepting triglyceride, was increased when dietary n3/n6 ratio was high(0.6) at both high and low n3 level. Therefore, it could not be recommended to consume large amount of n3 fatty acid or high ratio of n3/n6 to prevent cardiovascular diseases. These results suggested that the dietary fatty acid ratio of n3/n6 could be determined based on the n-3 content of dietary fat to reduce the risk of cardiovascular disease.
The majority of laboratory animals were transported from commercial breeders to a research facility by ground transportation. During the transportation, many biological functions and systems can be affected by stress. In this experiment, the change of body weight during the transportation was measured and the recovery periods from the transportation stress established based on the body weight changes. Total 676 laboratory animals which were aged between 3 to 9 wk old were studied. The transportation time taken from container packing to unpacking the container was approximately 24 h. The temperature of animal container was constantly maintained by air-conditioning and heating equipment. Rats were found to be more sensitive than mice. The body weight of rats was significantly decreased 3.71% (p<0.05) compared to the body weight of mice which decreased 0.9% There was no significant difference between the strains in the same species. When the changes of body weights were compared between delivery days, C57BL/6 mice showed the most variable changes compared to other species and strains. Consequently, C57BL/6 was more sensitive to stress than the other strains and the transportation process needs to be standardized to reduce between day variability. To establish the recovery periods from transportation stress, the body weight changes were measured during the acclimation period. Although the body weight of animals decreased during transportation, animals recovered their weight loss after the next day.
This study was designed to compare the effect of dietary intakes on different quality protein & levels of caloric consumption supplemented by sugar at the level of 26% of total caloric intakes. 30 males and same number of females of Albino rats, aged 30-40 days were devided into following six groups, 5 rats each. ACO Group: Ad libitum, Casein, no sugar group ACS Group: Ad libitum, Casein, 26% sugar supplemented group RBO Group: 50% restriction,Bean, no sugar group RBS Group: 50% restriction, Bean, 26% sugar supplemented group RAO Group: 50% restriction, Anchovy, no sugar group RAS Group: 50% restriction, Anchovy, 26% sugar supplemented group The rats were kept in individual cage and given 6 different diet for 12 weeks. The result of this study were elucidated as follow. Body weight gained and organ weight showed no significant differences between sugar supplemented group and the others. It was noteworthy that the shrinkage of female sex organ supplemented by sugar in the diet showed lower degree than that of compared groups in this study. In other word, degree of shrinkage due to protein-caloric restriction was decreased by sugar supplementation. Nitrogen Metabolism and total nitrogen retention were not observed any significant differences between sugar supplemented group and the others. Dental caries showed higher incidence for sugar supplemented groups. Hematology and bone growth showed no differences in this study. The similar results on the metabolic effects concerned the above view Points were obtained in the different protein groups such as bean & anchovy as protein sources in the diet. Caloric restriction Produced a lower growth-rate, lower body weight and poorer long bone growth. But composition of bone ash, concentration of nitrogen, calcium and blood glucose, liver fats and liver water content maintenanced at the same levels of standard group.
The present study was undertaken to investigate the metabolic problems of high carbohydrate and cellulose diets of Korean. Forty males and same number of females of Albino rats, aged $45{\pm}5$days were divided into 95% high carbohydrate (H. CHO)group, 83.8% medium carbohydrate (M. CHO)group, 50% low carbohydrate (L. CHO) group and standard (Stand). group containing 72.2% sugar. Each group was divided into two again-1.55% cellulose group and non-cellulose group, 10 rats each of eight groups in both sexes. Cellulose was added to each of non-cellulose diets in the forms of spinach powder and rice bran. After 14 weeks the rats were sacrificed for chemical analysis and the results were elucidated as follows. (1) H. CHO+Cell. group showed the lowest value in body weight gained and shrinkage of almost all organs, in contrast with this group the L.CHO group showed higher body weight gained than Stand. group. M. CHO+Cell, group showed much the same body weight gained curve as Stand. group. (2) It was observed that cellulose group showed lower F.E.R and P.E.R value than non-cellulose group comparatively. (3) Total nitrogen retention and retention rate were decreaced in H. CHP groups compared with M. CHO or L. CHO groups. (4) The amount of feces was increased due to addition of cellulose to experimental diets and in accordance with the increasing total fecal excretion of nitrogen and glucose was also increased, especially noticeable in fecal glucose excretion. (5) It was noteworthy that serum cholesterol level was decreased due to addition of cellulose in H. CHO group and L. CHO group. (6) M. CHO+Cell. group was designed to reflect the average survey data of Korean diets and there was no significant differences on body weight gained, F.E.R, P.E.R, total nitrogen retention and hematology between M. CHO+Cell. and Stand. group. Total glucose excretion was increased due to dietary cellulose in M. CHO+Cell. group, but it seemed to be no metabolic problems in this group.
Benign prostatic hyperplasia (BPH) is one of the common diseases in elderly men. Recently, the old-aged population has increased, with the interest in the clinical importance of BPH ever growing. This study is designed to investigate the effects of KH-305 on BPH induced rat. The herb formulation KH-305 is consisted of Rubus coreanus, Cornus officinalis and Cuscuta chinensis. An experimental prostatic hyperplasia was induced in male rats by the administration of testosterone propionate, 3 mg/kg SC, for 2 months. The rats were divided into 3 experimental groups: the control, BPH-induced, oral KH-305 ingestion group. After 2 months, the prostates were removed, and analyzed for their prostatic weight and histological examination. The prostate weights were measured in each group, and found to be 820${\pm}$38mg, 3140${\pm}$26mg, 1880${\pm}$21mg in the control, BPH-induced, and oral KH-305 ingestion group, respectively. The BPH induced group showed statistically significant increases in their prostatic weights compared with control group(p<0.05) but oral KH-305 ingestion group showed more significant decreases than BPH-induced group statically(p<0.05). Histologically injected testosterone lead to prostatic hyperplasia in rats, but oral KH-305 ingestion decreased this change. These results suggest that KH-305 may be effective in treatment of BPH, and complementary medicine of BPH.
It was the aim of this investigation to evaluate some histologic aspect of rat pulp tissue after it had been compromised by an experimental orthodontic force. Experimental animals of thirty five Spraque-Dawley rats were employed. The first upper molars had been successively mesial moved (initial load 100 gr.) with a closed coil spring during 21 days. The experimental periods were set on immediate, 1 day, 1 week, 2 weeks, 3 weeks, 4 weeks following retention time. On each experimental period, the rats were killed and prepared for the light microscopy. After prepared with H/E stain and Gomori's one-step trichrome stain, the specimens were analyzed with evaluation criteria which were adopted in this study. The result may be summarized as follows; 1. The main pulp changes due to experimental orthodontic force included vacuolization of odontoblastic layer, circulation disturbance, root resorption, reduced pulp collagenous fiber density and mean cell count of pulp fibroblast in the immediate group. 2. The pulp tissue changes were revealed reversible because the relieved pulp tissues from experimental orthodontic force were recovered rapidly in each evaluation criteria during retention periods. 3. Compared with normal control group, pulp collagenous fiber density were decreased in immediated group (p < 0.01), but increased in each retention groups. These seem to suggest that the pulp tissues were aged after experimental orthodontic force conditions. 4. Compared with normal control group, mean cell counts of pulp fibroblasts were decreased in immediate group (p < 0.05), but increased continuous in each retention groups. These seem to indicate that the pulp tissues were highly regenerative after experimental orthodontic force conditions. 5. Compared with normal control group, root resorptions occurred in all immediate specimens (p < 0.01) and they were healed in each retention periods, but often observed in 4 weeks retention group. These seem to indicate that root resorptions were recovered slowly after experimental orthodontic force conditions.
The effect of radiation on the steroidogenesis of rat ovary was investigated. For this purpose, female rats aged 7-8 weeks were exposed to a single dose of ${\gamma}$ radiation(320 rad or 800 rad) by the cobalt-60. At fourth(day of the first estrus) and eighth(day of the second estrus) days after irradiation, the concentrations of serum steroid hormones were determined by radioimmunoassays (RIA). The correlation between survival rate(Y) and radiation dose(X) was Y=-0.06X+100(r=0.89, n=10). Lethal dose$(LD)_{50(30)}$ and $LD_{20(30)}$ were 833.33rad and 333.33rad respectively. The weights of body and ovary was decreased significantly by the $LD_{50(30)}$ irradiation during the 4 days, but both weights were recovered at day 8. The serum levels of 17a-hydroxyprogesterone(170HP) and estradiol($E_2$) in the irradiated rats were not generally different from those of control. However, the levels of testosterone(T) and 19-norterstosterone(19NT) in the irradiated rat at $LD_{50(30)}$ and $LD_{20(30)}$ radiation doses were decreased, while progesterone levels in serum of the irradiated rats were slightly increased. These results suggest that irradiation may inhibit the first step of steroidogenesis, especially the conversion of P to T, in the rat ovary.
The purpose of this study was to investigate effects of the natural coral(NC) and the hydroxyapatite/calcium sulfate hemihydrate(HA/CS) on an early stages of wound healing in the rat periodontal fenestration defects. In this experiment, twelve male rats(Mean : 520g in BW) aged 8 to 9 months were used. Experimental periodontal fenestration defects were surgically created with tapered fissure bur at the buccal surface of the left mandibular 1st, 2nd molars. The buccal aspects of molar roots were carefully denuded of their periodontal ligament through a bony window created in the left mandibles of rats under general anesthesia. Each experimental periodontal fenestration defect was grafted with natural coral and HA/CS, randomly. An area without bone graft was assigned for negative control group. At 10,35 days, rats were serially sacrificed via intracardiac perfusion with 2.5% glutaraldehyde and specimens were processed with Hematoxylin-Eosin stain for light microscopic evaluation. The results of this study were as follows : 1. The defect areas were filled with dense connective tissues at 10 days in control group. But in the test(NC, HA/CS)groups, the connective tissues around graft materials were formed more loosely and the response of inflammation by graft materials itself was not found. 2. The defect areas were filled with new osteoid tissues and new cementum was not formed on the cut root surface at 35 days in the control group. 3. New osteoid tissue formation was more prominent at 35 days in control than test groups. 4. The NC and HA/CS particles were encapsulated by loose connective tissues at 10 days and by dense connective tissues at 35 days, respectively. 5. In the test groups, resorption of graft particles was not found through the experimental time. From the above results, natural coral and hydroxyapatite/calcium sulfate hemihydrate may be biocompatible and osteoconductive and have a weak adverse reaction to the periodontal tissues.
