  • By in vivo labelling of AMHP using[35S]-methionine, fat body culture and immunological analysis, it is proved that Bombyx adult fat body synthesizes 18K and 20K subunits of AMHP and releases them into haemolymph. Also these peptides are assembled to form native AMHP in the adult haemolymph.

  • A persistent major haemolyruph protein (MHP) was confirmed, and its ontogeny and physicochemical charadedstics were investigated in Helicoverpa assulta. The MHP existed continually during larval-pupal-adult development, and its ontogeny was similar to that of total haemolymph protein concentration during development. Its content increased with larval growth, and kept to high level during pupal-adult development except for temporary decrease at the early pupal and adult stages. The MHP was purified by ammonium sulfate precipitation, gel filtration and ion exchange chromatography. The purified MHP was determined to be hexamer glycolipoprotein (pI 5.9, M.W. 414kDa) consisted of single type subunit (69kDa). Amino acid analysis suggested that the MHP contained a relatively high content of aromatic amino acids (18.27 mole % of tryptophan, 7.47 mole % of tyrosine and 6.51 mole % of phenylalanine) compared to storage proteins from other insects. Immunodiffusion test and electrophoretic analysis of the organ proteins (gut, fat body, and Malphigian tubule) suggested that the major haemolymph protein was present in the fat body.

  • Isolation, purification and molecular properties of adult specific protein (ASP) were investigated in the silkworm, Bombyx mori. ASP was purified from adult haemolymph by gel filtration on Sephadex G-100 and ion exchange chromatography on CM52. The preparation was shown to be homogeneous by native-PAGE and immunoelectrophoresis. The purified protein was consisted of two different subunits with molecular weights of 19.5kDa and 17.5kDa, and contained no sugars and no lipids.

  • Tolerance limits of the adult crab were determined by exposing them to different temperatures (5~2$0^{\circ}C$) and salinities(12~32$\textperthousand$). The tolerance limit for high temperature was determined at 14$^{\circ}C$ and 16$^{\circ}C$ for male and female snow crab, respectively and that for salinity was 21 $\textperthousand$ at 5 $^{\circ}C$. Osmolality concentration, Na$^{+}$ and $K^{+}$ ions in the haemolymph of especially, the females was positively related to the salinity changes. Increased water temperature resulted in a marked decreased in ionic and osmolality concentrations near the limit of tolerance in male, however, such a clear pattern was not apparent for the females. The concentration of glucose in the haemolymph was positively related to water temperature but negatively to salinity.ity.

  • Apolipophorin-III (ApoLp-III) was purified from adult haemolynph of Hyphantriu cuneo and their molecular weight and synthetic place were investigated. ApoLp-III purification was performed by KBr-density gradient ultracentrifugation followed by gel permeation chromatographv (Sephadex G-1001 and ion-exchange chromatography (CM-52) and their purity was confirmed on 10% SDS-PAGE. ApoLp-III has the molecular weight of 18 ItDa and is synthesized by fat body.

  • A storage protein(SP-2) was confirmed in haemolymph during larval-pupal-adult development of tobacco burdworm(Helicoverpa assulta Guenee), and its biochemical characteristics were investigated. The titer of SP-2 showed a peak at mature larva, decreased gradually through the late pupal stage, and became undetectable at adult period. As the results from electrophoretic mnysis, SP-2 was confirmed to be glycolipoprotein(M.W. 332kDa) relatively stable to heat( $\leq$ 68$^{\circ}C$ ). This storage protein was determined to be a tetramer composed of a single subunit with MW of 83kDa, and the isoelectric point was 5.7. The amino acid composition of the SP-2 was characterized. It has relatively high content of methionine and histidine, whereas the contents of tyrosine and phenylalanine were relatively low.

  • After investigating the developmental process of galls forming on leaf of Zelkova serrata and analyzing the composition and contents of the free amino acids in leaves, galls and haemolymph of aphids, we have come to the following results. Galls induced were mostly distributed on the middle of lower part of leaf to the petiole along vein and most of investigated leaves had one gall only. The growth of galls continued in length and width until Paracolopha morrisoni habitants which transformed into alate adult left the gall. These results also indicate that galls are closely connected with a portion of plant tissues as well as foods and habitats of aphids. 26${\sim}$29 kinds of free amino acids such as proline, alanine and valine and so on, detected in leaves, gall and haemolymph of aphids. The composition of free amino acids were similar in three materials examined, but proline and asparatic acid showed the highest quantity in the leaves and falls, respectively. In case of aphids, alanine was extremely higher as 18.4% of total contents of free amino acids, in 1st instar of fundatrix stage. Total contents of free amino acids in the apteral adult stage of aphid were also increased than in the 1st instar stage. This investigation implies that nitrogen source such as free amino acids is apparently a major growth-material like carbohydrates during development of galls in Zelkova serrata.

담배나방의 발생과 휴면에 따른 혈림프 생체물질의 변화

  • Changes of biological components in haemolymph of Helicoverpa assulta Guenee were investigated during the development and pupae of cold-stress periods. Protein content was the highest in the larval stage, whereas it showed somewhat lower level at the cold-stressed pupae. The level of free amino acids was high in the larval and adult stages. Althougth decresed in pupal stage, the level of free amino acid remarkably increased at the cold-stressed pupae. Total lipids were a little increased at the pupal stage, after small quantity decreased in I-day old adults, and then remarkably decreased at the cold-stressed pupae. Glycogen was increased at the pupal stage, and somewhat decreased in the stage of adult, but remarkably decreased at the cold-stressed pupae. From thin layer chromatographic analysis, glucose was detected in small quantity at 1-day old pupae, one-day old adults, and cold-stressed pupae, respectively. Each .activity of acid and alkaline phosphatases was the highest in the 1 and 7-day old pupae, respectively, and both enzyme activities were increased about two-fold compared to the activity in the cold-stressed pupae. A total of 2 phosphatase isozymes were observed in throughout the developmental stage, and a new isozyme specific to cold-stressed pupae was identified.

Synthesis of New Uracil-5-Sulfonamide Derivatives and Immuno-Stimulatory Effect of a Chemically Modified Hemolymph of Biomphalaria alexandrina on Schistosoma mansoni Infected Mice

  • Some N-p-substituted phenyl uracil-5-sulphonamide derivatives have been synthesized to be evaluated as molluscicides against Biomphalaria alexandrina snails, the intermediate host of Schistosoma mansoni. Schistosoma mansoni infected mice were treated with hemolymph obtained from pre-treated Biomphalaria alexandrina snails with the products 4a, 10a, 10b and 4b or obtained from non-treated snails. The selection of the concentration based on the predetermined dose which caused mortality of less than 50% of snails/24 h. $LC_{50}$ of compounds 4a, 10a, 10b and 4b was 50, 100, 200 and 50 ppm respectively. The result showed that immuno-stimulatory effect of treated hemolymph with compounds 4a, 10a and 4b was related to significant protective effects (44.4, 34.6 and 50.4% reduction in worm burden respectively). In addition, mean total worm burdens were significantly reduced in non treated hemolymph by 33.8%. The effect of hemolymph obtained from treated or non treated snails on S. mansoni adult worms antigens was studied by indirect immunofluorescence technique using chronic mouse sera (CMS). The results indicated that there was a strong reaction with epitopes in gut epithelium, tubercles, teigument and subtegumental musculature of untreated and treated S. mansoni adult worms antigens. Therefore, treatment of hemolymph obtained from pre-treated snails with compounds 4a, 10a, and 4b can stimulate specific immune response and induce protective effects against S. mansoni infection.

Effects of 20-hydroxyecdysoneon Adult Development of Diapausing Fall webworm (Hyphantria cunea Drury) Pupae (휴면중인 미국흰불나방 번데기의 성충발육에 미치는 20-hyderoxyecdysone의 영향)

  • Studies were carried out to elucidate the efficacy of exogeneous 20-hydroxyecdysone treatment on terminating pupal diapause in the fall webworm. Hyphantria cunea Drury. And the difference was also investigated between normal adult development and post-diapause development after 20-hydroxyecdysone treatment. In the diapause termination rate of pupae treated with 20-hydroxyecdysone after storage for various periods at 16L:8D and $25\pm$$1^{\circ}C$, at the highest rate was observed from the group stored for the longest period and the lowest rate from those stored for 1.5 months. The time needed for adult emergence was inversely proportional to the chilling (at $0\pm$$1^{\circ}C$) period, and the longer its exposure period at low temperature, the higher its sensitivity to 20-hydroxyecdysone treatment. Pupal diapause duration was almost the same, regardless of storage period in the total darkness or at the photoperiod of 16L:8D, and also they successfully emerged to adult even without any experience at low temperature. The oxygen consumption rate in normally developing pupae showed nearly a typical U-form. But, that of diapausing pupae treated with 20-hydroxyecdysone slowly increased and jumped 14 days after the treatment. Pupal diapause began before formation of adult tissues, an a timing of adult tissue formation coincided with ascending timing of the metabolic rate in both normally developing pupae and diapausing pupae treated with 20-hydroxyecdysone. The diapausing pupae treated with 20-hydroxyecdysone was similar to normally developing pupae in band patterns of proteins from haemolymph or fat body.
